Analysis of in vitro 2D-COSY on Human Brain Metabolites for Molecular Stereochemistry

  • To investigate the 3-bond connectivity of human brain metabolites by scalar coupling interaction through 2D-correlation spectroscopy (COSY) techniques using high field NMR spectroscopy. All NMR experiments were performed at 298K on Unity Inova 500 or 600 (Varian Inc.) equipped with a triple resonance probe head with z-shield gradient. Human brain metabolites were prepared with 10% $D_2O$. Two dimensional 2D COSY spectra were acquired with 4096 complex data points in $t_2$ and 128 or 256 increments in $t_1$ dimension. The spectral width was 9615.4 Hz and solvent suppression was achieved using presaturation using low power irradiation of the water resonance during 2s of relaxation delay. NMR data were processed using VNMRJ (Varian Instrument) software and all the chemical shifts were referenced to the methyl resonance of N-acetyl aspartate (NAA) peak at 2.0 ppm. Total 10 metabolites such as N-acetyl aspartate (NAA), creatine (Cr), choline (Cho), glutamine (Gln), glutamate (Glu), myo-inositol (Ins), lactate (Lac), taurine (Tau), ${\gamma}$-aminobutyricacid (GABA), alanine (Ala) were included for major target metabolites. Symmetrical 2D-COSY spectra were successfully acquired. Total 14 COSY cross peaks were observed even though there were parallel/orthogonal noisy peaks induced by water suppression. Except for Cr, all of human brain metabolites produced COSY cross peaks. The spectra of NAA methyl proton at 2.02 ppm and Glu methylene proton ($CH_2(3)$) at 2.11 ppm and Gln methylene proton ($CH_2(3)$) at 2.14 ppm were overlapped in the similar resonance frequency between 2.00 ppm and 2.15 ppm. The present study demonstrated that in vitro 2D-COSY represented the 3-bond connectivity of human brain metabolites by scalar coupling interaction. This study could aid in better understanding the interactions between human brain metabolites in vivo 2D-COSY study. Also it would be helpful to determine the molecular stereochemistry in vivo by using two-dimensional MR spectroscopy.

Bioactive lipids in gintonin-enriched fraction from ginseng

  • Background: Ginseng is a traditional herbal medicine for human health. Ginseng contains a bioactive ligand named gintonin. The active ingredient of gintonin is lysophosphatidic acid C18:2 (LPA C18:2). We previously developed a method for gintonin-enriched fraction (GEF) preparation to mass-produce gintonin from ginseng. However, previous studies did not show the presence of other bioactive lipids besides LPAs. The aim of this study was to quantify the fatty acids, lysophospholipids (LPLs), and phospholipids (PLs) besides LPAs in GEF. Methods: We prepared GEF from white ginseng. We used gas chromatography-mass spectrometry for fatty acid analysis and liquid chromatography-tandem mass spectrometry for PL analysis, and quantified the fatty acids, LPLs, and PLs in GEF using respective standards. We examined the effect of GEF on insulin secretion in INS-1 cells. Results: GEF contains about 7.5% linoleic (C18:2), 2.8% palmitic (C16:0), and 1.5% oleic acids (C18:1). GEF contains about 0.2% LPA C18:2, 0.06% LPA C16:0, and 0.02% LPA C18:1. GEF contains 0.08% lysophosphatidylcholine, 0.03% lysophosphatidylethanolamine, and 0.13% lysophosphatidylinositols. GEF also contains about 1% phosphatidic acid (PA) 16:0-18:2, 0.5% PA 18:2-18:2, and 0.2% PA 16:0-18:1. GEFmediated insulin secretion was not blocked by LPA receptor antagonist. Conclusion: We determined four characteristics of GEF through lipid analysis and insulin secretion. First, GEF contains a large amount of linoleic acid (C18:2), PA 16:0-18:2, and LPA C18:2 compared with other lipids. Second, the main fatty acid component of LPLs and PLs is linoleic acid (C18:2). Third, GEF stimulates insulin secretion not through LPA receptors. Finally, GEF contains bioactive lipids besides LPAs.

완두 및 대두근류균(根瘤菌)의 분리(分離) 및 질소고정능력(窒素固定能力)의 비교(比較) (Isolation of Pea and Soybean Nodule Bacteria and Assessment of their Nitrogen-fixing Capacities)

  • 두과작물(荳科作物)의 증산(增産)과 질소비료(窒素肥料)의 소비절약(消費節約)을 위(爲)하여 근류균(根瘤菌)의 실용화(實用化)를 위(爲)한 일련(一連)의 연구(硏究)를 시도(試圖)하고 그 기초연구(基礎硏究)로서 완두와 대두군(群)을 대상(對象)으로 근류균(根瘤菌)을 분리(分離)하고 우수균주(優秀菌株)를 선발(選拔)하였다. 1. 대관령(大關嶺), 제주도(濟州道) 및 기타지역(其他地域)에서 수집(蒐集)한 근류(根瘤)로 부터 완두군(群) 22주(株), 대두군(群) 88주(株)의 근류균(根瘤菌)을 분리(分離)하였다. 2. 분리균류(分離菌瘤)에 대(對)하여 형태적(形態的) 및 배양적(培養的) 성질(性質)을 검토(檢討)하는 동시(同時)에 항원(抗原)의 특성(特性)을 비교(比較)하고 lysogeny 여부(與否)를 확인(確認)한 결과(結果) : (1) yeast mannitol broth 및 agar상(上)에서의 생육형태(生育形態) 및 증식속도(增殖速度)를 달리하였으며 congo red에 의한 착색정도(着色程度)에도 차이(差異)가 있었고 (2) 항원성(抗原性)의 상호(相互) 근록성(近緣性)이 인정(認定)되는 균주(菌株)는 대두군(群) G-8/G-9/D216 및 G-20/G-52이었으며; (3) latent phage의 감염균주(感染菌株)가 발견(發見)되지 않았다. 3. 항온조명실내(恒溫照明室內)에서 행(行)한 시험관(試驗管) 및 bottle법(法)에 의한 식물접종시험(植物接種試驗)을 통(通)하여 분리균주(分離菌株)의 근류형성(根瘤形成) 및 질소고정능력(窒素固定能力)을 비교한 결과, 각군별(各群別)로 공시숙주식물(供試宿主植物)에 대(對)하여 근류(根瘤)를 형성(形成)하지 못하는 즉친화력(卽親和力)이 없는 균주(菌株), 근류(根瘤)는 형성(形成)하나 숙주식물(宿主植物)의 생육(生育)에는 영향(影響)을 주지 못하는 무효균주(無效菌株) 그리고 숙주식물(宿主植物)의 생육(生育)을 크게 촉진(促進)하는 유효균주등(有效菌株等)으로 구분(區分)할 수 있었으며 특(特)히 각군별(各群別) 유효균주중(有效菌株中)에서 완두군(群)의 P-3, 4 및 8균주(菌株); 대구군(群)의 G-23 및 27 균주등(菌株等)은 질소시용무접종구(窒素施用無接種區) 수준(水準) 또는 그 이상(以上)으로 숙주식물(宿主植物)의 생육(生育)에 유효(有效)한 결과(結果)를 나타내었다.

포스트모던 연극의 새로운 극작술과 그 한계 : 미셀 비나베르(Michel Vinaver)의 작품을 중심으로 (Nouvelle Dramaturgie et limites du Théâtre postmoderne à travers les pièces de Michel Vinaver)

  • Depuis les $ann{\acute{e}}es$ 1990, sous l'influence de la $pens{\acute{e}}e$ postmoderne, des ${\oe}uvres$ $d{\acute{e}}constructivistes$ ont fait leur apparition dans le $th{\acute{e}}{\hat{a}}tre$ $cor{\acute{e}}en$. Cependant, cette approche est plus remarquable dans le domaine du $th{\acute{e}}{\hat{a}}tre$ pratique que dans celui de $l^{\prime}{\acute{e}}criture$ dramatique. Par exemple, ce mouvement a $commenc{\acute{e}}$ avec les metteurs en $sc{\grave{e}}ne$ OH Tae-suk et GUI Gook-seo au $d{\acute{e}}but$ des $ann{\acute{e}}es$ 1990. Leur ont $succ{\acute{e}}d{\acute{e}}$ ensuite les metteurs en $sc{\grave{e}}ne$ CHOI Yong-hoon, de la troupe de $th{\acute{e}}{\hat{a}}tre$ Chak Eun Shin Hwa, et PARK Kun-hyung. De plus, actuellement, de jeunes metteurs en $sc{\grave{e}}ne$ travaillent avec $perspicacit{\acute{e}}$, tels que KO Sung-woong, KANG Lyang-won et YOUN Han-sol. A $l^{\prime}oppos{\acute{e}}$ de ce $th{\acute{e}}{\hat{a}}tre$ pratique, $l^{\prime}{\acute{e}}criture$ dramatique des $pi{\grave{e}}ces$ de $th{\acute{e}}{\hat{a}}tre$ demeure au niveau de $l^{\prime}{\acute{e}}criture$ $repr{\acute{e}}sentative$ et est encore sans grande valeur. Pour $r{\acute{e}}fl{\acute{e}}chir$ ${\grave{a}}$ ce $probl{\grave{e}}me$, nous $consid{\acute{e}}rerons$ les $pi{\grave{e}}ces$ de Michel Vinaver dont la dramaturgie oppose $l^{\prime}{\acute{e}}criture$ $d{\acute{e}}constructiviste$ au principe $repr{\acute{e}}sentatif$ et ${\grave{a}}$ $l^{\prime}{\acute{e}}criture$ traditionnelle en ${\acute{e}}chappant$ au principe de $causalit{\acute{e}}$. Avec sa $strat{\acute{e}}gie$ de montage $sp{\acute{e}}cifique$, il $proc{\grave{e}}de$ avec une ${\acute{e}}criture$ $fragment{\acute{e}}e$ tout en $ins{\acute{e}}rant$ des dialogues $h{\acute{e}}t{\acute{e}}rog{\grave{e}}nes$. Il trouve ainsi le silence des mots, un $pr{\acute{e}}sent$ sensible, la $mat{\acute{e}}rialit{\acute{e}}$ des mots et des images ostensives ${\grave{a}}$ travers des dialogues $juxtapos{\acute{e}}s$, une $simultan{\acute{e}}it{\acute{e}}$ et une $discontinuit{\acute{e}}$ du dialogue. En examinant ses $pi{\grave{e}}ces$ de $th{\acute{e}}{\hat{a}}tre$, nous explorons cette nouvelle ${\acute{e}}criture$ qui $r{\acute{e}}siste$ au $r{\acute{e}}gime$ dramatique de la $repr{\acute{e}}sentation$. Toutefois, nous n'encenserons pas seulement cette ${\acute{e}}criture$ postmoderne. Plus $pr{\acute{e}}cis{\acute{e}}ment$, nous $rel{\grave{e}}verons$ ses limites, dans les circonstances $pr{\acute{e}}sentes$, en $r{\acute{e}}fl{\acute{e}}chissant$ aux limites du $th{\acute{e}}{\hat{a}}tre$ postmoderne depuis le $d{\acute{e}}but$ des $ann{\acute{e}}es$ 2000. Ce faisant, nous souhaitons $d{\acute{e}}finir$ de nouvelles $possibilit{\acute{e}}s$ $esth{\acute{e}}tiques$.
