백국균 Aspergillus kawachii Kitahara의 생태학적 연구 (The ecological studies on Aspergillus kawachii Kitahara.)

  • This study has been aimed to determine the ecological factors with relationship to the Jongkok production in view of fermentation technology by means of some strains, Asp. kawachii, which is now preserved by the author and the following factors are included during the study; inorganic salts, nitrogen, sugar, water contents and temperature. The results, are as follows: (a) Sugar among other above-mentioned factors is increasingly affecting the number of the short type of conidiophore on culture medium and the conidiophore is increased by direct ratio until glucose concentration of 50%, at which concentration is mostly effective for the short type of conidiophore, while other factors did not affect on it. (b) Until glucose concentration of 50% sugar component of culture medium is favorable for the spore formation of Asp. kawachii by direct ratio. And peptone or asparagine on nitrogen medium, calcium-phosphate among other inorganic salts, wheat bran and rice branare also favorable, but other factors rientioned earlier show no relationship with the spore formation. Sugar, however, also related with the spore color clearness of crimson and light brown, and spore color is mostly clear at the point of glucose concentration until 50%. And asparagine on nitrogen medium, calcium phosphate among other inorganic salts, rice bran did all affect on the color clearness, while other factors did not concern with color clearness. (c) Water, sugar and temperature have related with the acid formation which is promoted, by direct ratio at the point of water-saturated condition and glucose concentration of 50%, while temperature at $25^{\circ}C$favorably affected on the acid formation which is increased by inverse ratio at the temperature$25^{\circ}C$ to $45^{\circ}C$ And pH did not relate with the acid formation. (d) Cylindrical plate method devised by the author is mostly favorable for the preservation and isolation of culture, compared with the traditional slant medium method.

대형 인공호에서 무기 질소원에 대한 점오염원 및 유입수의 영향 (An Influence of Point-Source and Flow Events on Inorganic Nitrogen Fractions in a Large Artificial Reservoir)

  • 본 연구는 1993${\sim}$1994년 대청호 17개 조사지점에서 점오염원 및 계절적 유입수에 대한 무기질소원의 다변적 동태에 대하여 평가하였다. 연구기간 동안 총질소(TN)는 평균 1.53 mg/L으로, $0.70{\sim}2.56\;mg/L$ 범위에 있었다. 용존 무기질소(DIN)는 계절 및 조사지점에 관계없이 총질소의 90%이상을 차지하여 질소가 풍부한 부영양-과영양 상태의 호수임이 확인되었다. 용존 무기질소의 $67{\sim}94%$는 질산성-질소인 반면, 암모니아성-질소는 용존 무기질소의 5%이하로 구성되었다. 1993년 장마동안 질산성-질소는 빗물과 호수물의 혼합의 결과로서 상류역에서 희석된 반면, 암모니아성-질소는 장마 전에 비해 100%이상 증가를 보였다. 암모니아성-질소는 강우량과 정 상관관계(r=0.85; p<0.001)를 보였고, 수 체류시간(r = -0.90; p<0.001) 및 전기전도도 (r = -0.78, p<0.001)와는 역 상관관계를 보였다. 이런 결과에 따르면, 암모니아성-질소는 장마기에 호수 외부로부터 유입되었음을 제시한다. 연구기간 2년 모두 평균 총질소는 호수내 상류 및 하류에서보다 가두리 양식장 및 폐수 처리장이 위치한 중류역에서 높았다. 중류역에서 이런 특성은 유입량이 적은 1994년하절기 동안 점 오염원에서 축적된 오염부하 증가의 결과로서 가장 심화되었다. 본 인공호에서 총질소 분포는 호수내 상${\cdot}$하류역 사이에 큰 차이를 보이지 않았고, 유입량 보다는 점오염원에 의해 직접적으로 결정되는 것으로 사료된다.

점적관수시 관비와 표면시비된 중질소 표지요소의 행동비교 (Distribution of Inorganic N from Fertigated and Broadcast-applied 15N-Urea along Drip Irrigation Domain)

  • 본 연구는 점적관수시 토양수분 영역의 변화를 측정하고 이에 따른 관비와 표면시비로 투입된 중질소 표지요소에서 유래한 무기태 질소의 분포 양식을 비교하기 위하여 수행되었다. 이를 위해 비닐하우스 내에서 6주간 포트실험을 수행하였다. 관비구는 $117mg\;N\;L^{-1}$ 수준으로 4주간 1.41 g N 을 투입하였으며 표면시비구는 토양 5 kg에 1.81 g N 을 고루 섞어 표면 (3 cm) 에 충진하였다. 관개는 -50 kPa의 토양 수분 장력을 관수점으로 하여 유속 $0.5L\;hr^{-1}$로 공급하였다. 질소 동위원소비의 분석결과 관비구의 경우 투입된 1.41g N 중 89% 에 해당하는 1.25 g N 이 토양에서 무기태 질소로 회수되었으며 표면시비구의 경우 1.87 g N 중 단지 51% 만이 회수되었다. 이는 낮은 농도의 관비로 공급된 요소가 습윤구역에 분포하여 가수분해, 질산화가 일어나는데 반해 표면시비는 고농도로 집적된 요소가 지표에서 가수분해되어 암모니아 휘산이 많이 일어나기 때문이다. 관비로 공급된 요소에서 유래한 무기태 질소의 60%가 0-10 cm 깊이에 있었으며 깊어짐에 따라 농도가 점차 감소하였다. 반면 표면시비된 요소에서 유래한 무기태 질소는 91%가 지표에 집중되어 10 cm 밑으로는 거의 발견되지 않았다. 관비의 경우 요소에서 유래한 무기태 질소의 99%가 질산태 질소로 회수된 반면 표면시비된 요소에서 유래한 무기태 질소는 38%가 암모늄태로 남아있었으며 대부분 습윤구역 바깥쪽 표면에서 회수되었다. 이는 수분이동이 적고 건습이 반복되는 지표, 특히 습윤구역 외부에 투입된 요소의 경우 이동성이 적으며 질산화가 지연되기 때문이다. 본 실험의 결과로 보아 관비로 요소를 투입할 때 암모니아 휘산에 의한 질소의 손실을 최소화할 수 있었으며 발견된 요소유래 무기태 질소의 분포로 보아 식물이 흡수하기 용이한 무기태 특히 질산태 질소를 지속적으로 공급할 수 있었다.

  • The effect of nitrate, nitrite and ammonia as inorganic nitrogen sources on the modulation of nitrogen metabolism of Botryococcus braunii UTEX.-572 has been studied under aeration. The primary process in the regulation of nitrogen metabolism by this alga has the nitrate uptake system. This uptake of nitrate operation was immediately inhibited by the presence of 0.5 mM of ammonium and reversed by 0.2∼0.3 mM ammonium. When cell were exposed to 5 mM of ammonium for 24 hours the activity of nitrate reductase became inactive.

흰비단털버섯(Volvariella bombycina)의 배양적 특성 및 균사 배양 조건 설정 (Cultivation Characteristics and Optimal Conditions for Mycelial Growth of Volvariella bombycina)

  • 본 연구에서는 V. bombycina 자실체의 안정적인 대량생산에 기반되는 필수적인 균사체 생산을 위한 최적 배양조건을 구명하고자 하였다. V. bombycina 균사체의 효율적 배양을 위한 조건은 최적 배양온도는 30℃, pH 8.0, 탄소원 0.7% Soluble starch, 유기질소원 0.7% Soytone, 무기질소원 1.0% (NH4)2C4H4O6, 아미노산 0.1% Cysteine, 유기산은 0.07% Lactic acid, 그리고 무기염류는 7 mM CaCl2 조건이었다. 새로이 명명한 SSNA V. bombycina의 최적 배지는 추후 V. bombycina의 재배에 널리 활용될 수 있을 것으로 판단된다. 본 연구 결과에 따라 선발된 배양조건으로 흰비단털버섯의 반경을 측정한 결과 37 mm/7days로 24 mm/7days로 측정된 대조군인 PDA 배지보다 균사생육이 빨랐다(Fig. 16, 17). 흰비단털버섯의 기본적인 적정 생육환경을 조사하여 확인된 실험 데이터는 흰비단털버섯 자실체 발생을 위한 흰비단털버섯의 역할을 규명하는데 기여할 것으로 판단된다.

수생식물에 의한 축산폐수의 오염물질 감소 효과 (The Effect of Clarification by Aquatic Plant on Livestock Wastewater)

  • In general, livestock wastewater consists of many pollutants such as nitrogen, phosphorus, carbonic compounds and inorganic substances. Most carbonic and organic compounds are sufficiently removed by conventional secondary processes, but nitrogen, phosphorus and soluble inorganic compounds are little removed by traditional clarification process. These remained substances in wastewater, for instances, phosphorus and nitrogen are efficiently eliminated by advanced wastewater treatment or botanical removing process. Concentrations of $BOD_s$, SS, T-N and T-P in influent livestock wastewater used in this study were 126mg/l, 115mg/l, 45mg/l and 13mg/l, respectively. The hydraulic retention time(HRT) of wastewater was about 10 days in the pond packed with aquatic plants. A water-hyacinth and a water-dropwort were used as an experimental stuff plant. The removal ratios of nitrogen was 44.3% for the water-hyacinth and 40.2% for the water-dropwort. The removal efficiency of phosphorus in experimental ponds reached by 57.9% for the water-hyacinth and 58.5% for the water-dropwort for 10 days, respectively. Removal ratios of BODs and SS of livestock wastewater for 10 days were reached by 80.1%, 91.0% for he water-hyacinth, respectively. At the same condition, the removal ratios of BODs and SS were reached by 75.0%, 87.6% for the water-dropwort, respectively.

MBOD법에 의한 만경강 수계의 조류성장잠재력 평가 (Evaluation of Algal Growth Potential in the Mangyeong River by MBOD method)

  • The modified biochemical oxygen demand (MBOD) were conducted to evaluate the water quality and fertility in the Mangyeong river from november 2002 to april 2003. MBOD method was used to evaluate algal growth potentials and their limiting factors. MBOD depends on the amount of available inorganic nutrient and organic substrate during 5-day incubation in the dark condition at $20^{\circ}C.$ The MBOD assay depends on inorganic nutrients such as phosphorus and nitrogen as well as reduced carbon as called MBOD, MBOD-P, and MBOD-N, respectively. The concentration of pollutants were in the range of 3.08~48.36 mg/L for COD. The concentration of nutrients were in the range of 0.37~111.62 mg/L for dissolved inorganic nitrogen (DIN) and 0.00~1.03 mg/L for dissolved inorganic phosphorus (DIP). The results of MBOD bioassay showed that the MBOD, MBOD-P and MBOD-N values were 15~173 mg $O_2/L,$ 13~165 mg $O_2/L$ and 66~175 mg $O_2/L$ ranges, respectively. The MBOD values are found to be the highest in Iksan River and the lowest in Hari River throughout the Mangyeong River. The relationships of MBOD, MBOD-P and MBOD-N in MBOD method were generally found in MBOD$\risingdotseq$ MBOD-P$\risingdotseq$MBOD-N. But the result of Gosan was appeared to MBOD$\risingdotseq$MBOD-N > MBOD-P. The MBOD-N value was higher 3 to 5 times than the MBOD-P value in the Gosan station. The algal growth potentials expressed as the concentration of chlorophyll-a were maximum 20 times more than algal biomass in the water column.

지역별 전통된장과 개량된장의 품질특성 (Analysis of Quality Characteristics of Regional Traditional and Commercial Soybean Pastes (Doenjang))

  • Purpose: This study aimed to investigate the quality characteristics of traditional and commercial soybean pastes (Doenjang) for development of traditional soybean pastes. Methods: Proximate compositions, amino acid nitrogen contents, pH, acidity, salt contents, chromaticity, viable bacteria and inorganic substance contents of nineteen traditional and three commercial Doenjang were investigated. Results: Analysis of proximate compositions indicated a significant difference between samples. In moisture content, D10 did not meet the standard of food codex. The contents of moisture, fat, protein and ash were slightly higher in traditional than commercial Doenjang. However, the carbohydrate content was 2-fold higher in commercial than traditional, and thus, the calories were also higher. The amino nitrogen content was lower in commercial than traditional Doenjang. pH, acidity and salt contents of Doenjang were pH 4.67-6.15, 1.53-3.29%, and 9.01-18.78%, respectively. pH and acidity showed no significant differences between traditional and commercial Doenjang, however, salt contents were significantly higher in traditional than commercial Doenjang. In case of chromaticity, the averages of L, a, b values were higher in traditional Doenjang. In microbiological analysis, total bacteria counts were higher in traditional than commercial Doenjang. The number of Bacillus cereus in D12 and D19 exceeded the standard of food codex. Staphylococcus aureust was undetected in all samples. The number of fungi varied widely between samples, with no detection in five traditional and all commercial Doenjang. Results of inorganic substance analysis indicated that contents of inorganic substances in Doenjang were in order of Na, K, Mg and Ca. Overall, Na content was higher in traditional Doenjang. Contents of inorganic substances showed wide variations in traditional Doenjang. Conclusion: For the development of traditional Doenjang, quality standardization and ensuring safety are considered necessary.

GIS를 이용한 연안수질의 시공간적 분포 특성에 대한 연구 (A Study on Spatial and Temporal Distribution Characteristics of Coastal Water Quality Using GIS)

  • 연안수질의 시공간적 분포특성을 살펴보기 위하여 1997년부터 2004년 사이에 우리나라 연안해역에서 측정된 수질자료(chlorophyll-a, pH, DO, COD, SS, dissolved inorganic nitrogen, dissolved inorganic phosphorous, salinity, temperature)에 대해 GIS 공간분석 기법을 적용하였다. 본 연구에서 사용된 수질자료는 공간상 모든 위치에 그 값이 존재하나, 실제 간의 수집은 고정관측 위치에서만 수행된다. 따라서 공간적으로 연속적으로 존재하는 연안수질의 분포 특성을 파악하기 위해서는 미관측 지점의 값을 예측할 필요가 있다. 본 논문에서는 공간분석 기법인 IDW를 연안수질 측정자료에 적용하였으며, IDW 기법의 적용 가능성을 평가하였다. 또한 IDW 공간보간을 적용한 후, 전국 연안을 46개 구간으로 구분하여 각 구간의 수질특성을 GIS 시각화 기법을 이용하여 분석하였다. 적용결과 공간보간 및 GIS 기법이 다년간 광범위한 범위에서 수집된 수질 자료로부터 시공간적 분포 특성을 파악하는데 유용함을 확인할 수 있었다.

전통 고추장과 개량 고추장의 품질특성 (Analysis of Quality Characteristics of Traditional and Commercial Red Pepper Pastes (Gochujang))

  • Purpose: This study investigated the quality characteristics of traditional and commercial red pepper pastes (Gochujang) for development of traditional red pepper pastes. Methods: Proximate compositions, amino acid nitrogen contents, salt contents, color, viable bacteria, and inorganic substance contents of 19 traditional and three commercial Gochujang were investigated. Results: Analysis of proximate compositions showed huge differences between the samples. For moisture content, six kinds of traditional Gochujang did not meet the standard of the food codex. Contents of moisture, fat, and protein were significantly higher in traditional than commercial Gochujang. However, carbohydrate content was higher in commercial than traditional Gochujang, and thus calories were also higher. The amino nitrogen contents of the three kinds of traditional Gochujang did not meet the standard of the food codex. Salt contents and color were not significantly different between traditional and commercial Gochujang. In the microbiological analysis, total bacteria counts were higher in traditional than commercial Gochujang. The number of B. cereus did not exceed the standard of the food codex in all samples. S. aureus and fungi were not detected in any samples. In the inorganic substance analysis, inorganic substances contents in Gochujang were in the order of Na, K, Mg, and Ca. Overall, Na, K, Mg, and Ca were more abundant in traditional than commercial Gochujang. However, due to wide variations in inorganic substance contents of traditional Gochujang, there were no significant differences between traditional and commercial Gochujang. Conclusion: For the development of traditional Gochujang, quality standardization is considered necessary.
