Suppression of Adiposity and Improvement of Fat Metabolism in High-fat Diet-induced Obese Mice Treated with an Inonotus obliquus Extract (고지방식이 유도 비만 마우스에서 차가버섯(Inonotus obliquus) 추출물의 체중 증가 억제 및 지방대사 개선 작용)

  • Using high-fat diet-induced obese (DIO) mice, the mechanism of anti-adiposity and anti-obesity actions produced by Inonotus obliquus water extract (IOE) was investigated. Significant reduction in body weight in DIO mice orally administrated with IOE for 8 weeks compared to IOE-non-treated control mice was observed, which was attributed to the reduction of epididymal and mesenteric adipose tissue, but not the liver and skeletal muscle. Adiponectin synthesis in epididymal adipose tissue (EAT) and AMPK phosphorylation in the liver were significantly increased in IOE-treated DIO mice. Gene expression analysis showed that IOE-treated DIO mice had higher expression levels of lipogenic genes in EAT and fatty-acid oxidative genes in the liver, but lower expression levels of hepatic pro-inflammatory cytokines compared to IOE-non-treated controls. Our findings confirm a therapeutic potential of Inonotus obliquus for reducing adiposity and ameliorating hyperlipidemia to treat metabolic disorders.

Immunomodulatory Activity of the Water Extract from Medicinal Mushroom Inonotus obliquus

  • The immunomodulatory effect of aqueous extract of Inonotus obliquus, called as Chaga, was tested on bone marrow cells from chemically immunosuppressed mice. The Chaga water extract was daily administered for 24 days to mice that had been treated with cyclophosphamide (400 mg/kg body weight), immunosuppressive alkylating agent. The number of colony-forming unit (CFU)-granulocytes/macrophages (GM) and erythroid burst-forming unit (BFU-E), increased almost to the levels seen in non-treated control as early as 8 days after treatment. Oral administration of the extract highly increased serum levels of IL-6. Also, the level of $TNF-{\alpha}$ was elevated by the chemical treatment in control mice, whereas was maintained at the background level in the extract-treated mice, indicating that the extract might effectively suppress $TNF-{\alpha}$ related pathologic conditions. These results strongly suggest the great potential of the aqueous extract from Inonotus obliquus as immune enhancer during chemotherapy.

Anti-Tumor Metastasis Activity by Extracts of Inonotus obliquus (차가버섯 추출물의 종양 전이 억제 효과)

  • In experimental lung metastasis of colon26-M3.1 carcinoma cells, we found that the prophylactic or therapeutic admini-stration of Inonotus obliquus extracts significantly inhibited lung metastasis. In an in vitro cytotoxicity analysis, the extracts did not affect colon26-M3.1 cell growth at concentrations up to 1000 ${\mu}g/m{\ell}$. Peritoneal macrophages that were stimulated with the extracts produced $TNF-{\alpha}$. These data suggest that Inonotus obliquus extract has antitumor activity to inhibit tumor metastasis, and its antitumor effects are partially associated with macrophages activation.

  • Seven compounds were isolated from stationary cultured mycelia of Inonotus obliquus, of which several of them showed certain antitumor activities. Seven compounds were identified as lanosterol(1), inotodiol(2), trametenolic acid(3), 3β,22,25-trihydroxy-lanosta-8-ene(6), 3β,22-dihydroxy-lanosta-8,24-diene(A), 3β-hydroxy- lanosta-8,24-dien-21-al(B) and methyl trametenolate(C), respectively. The precursor compound of sclerotium obtained from shaking-cultured and stationary-cultured mycelia of Inonotus obliquus is lanosterol. Ergosterol and ergosterol peroxide were obtained by shaking-culture, and the substituted compounds of C-21 and C-22 of lanosterol were obtained by stationary culture.

Antioxidative Activity and Tyrosinase Inhibition Effect of Fractions from the Extract of Inonotus obliquus (차가버섯 용매별 분획물의 항산화 활성과 Tyrosinase 저해 활성)

  • This study was conducted to investigate the antioxidative activities and tyrosinase inhibition effects of fractions from the distilled water extract of Inonotus obliquus. Moreover, GC-MS based analysis with trimethylsilyl (TMS) derivatization was carried out for active compound in the extract of I. obliquus. In DPPH radical scavenging ability, $SC_{50}$ values of the ethyl acetate fraction was 0.393 mg/ml as a result of the most effective than other fractions. Meanwhile, aqueous fraction showed higher effect in tyrosinase inhibitoty activity. In GC-MS based analysis with TMS derivatization, 7 compounds including syringic acid, vanillic acid and protocatechuic acid were observed in ethyl acetate fraction, and oxalic acid is the main compound in aqueous fraction. As a result, it was confirmed that oxalic acid in aqueous fraction from the distilled water extract of I. obliquus was a compound showing tyrosinase inhibition effect.

Progress on Understanding the Anticancer Mechanisms of Medicinal Mushroom: Inonotus Obliquus

  • Cancer is a leading cause of death worldwide. Recently, the demand for more effective and safer therapeutic agents for the chemoprevention of human cancer has increased. As a white rot fungus, Inonotus obliquus is valued as an edible and medicinal resource. Chemical investigations have shown that I. obliquus produces a diverse range of secondary metabolites, including phenolic compounds, melanins, and lanostane-type triterpenoids. Among these are active components for antioxidant, antitumoral, and antiviral activities and for improving human immunity against infection of pathogenic microbes. Importantly, their anticancer activities have become a hot recently, but with relatively little knowledge of their modes of action. Some compounds extracted from I. obliquus arrest cancer cells in the G0/G1 phase and then induce cell apoptosis or differentiation, whereas some examples directly participate in the cell apoptosis pathway. In other cases, polysaccharides from I. obliquus can indirectly be involved in anticancer processes mainly via stimulating the immune system. Furthermore, the antioxidative ability of I. obliquus extracts can prevent generation of cancer cells. In this review, we highlight recent findings regarding mechanisms underlying the anticancer influence of I. obliquus, to provide a comprehensive landscape view of the actions of this mushroom in preventing cancer.

Effect of Beverage from Inonotus Obliquus on Serum Lipid Profile Improvement (체내 지질 농도 개선에 미치는 차가버섯 음료의 효과)

  • The purpose of this study was to investigate the possibility of Inonotus obiquus as an functional resources. We carried out to develop a beverage by using Inonotus obliquus, and we examined the effects of beverage on the reduction of serum lipid and improvement of blood parameters in rats fed high fat diet 5 weeks. Sprague-Dawley rat weigh $150\;g{\pm}15\;g$, were randomly assigned to 4 groups, basal diet only(BDG), high fat diet without beverage(FDCG), high fat diet and 10% beverage(FD10M), high fat diet and 20% beverage(FD20M). The result of this study were as follow. Hematological datas of 4 groups were same level, which were not significant. Serum GOT activity were reasonable levels in FD10M and FD20M groups compared to FDCG group. Total cholesterol, LDL-cholesterol, triglyceride in serum and atherogenic index were remarkably reduced in beverage supplemented groups as compared control group. These results imply that beverage from Inonotus obliquus could be used as possible for decrease of serum lipid concentration.

Antioxidative Activities of Phenolic Compounds Isolated from Inonotus obliquus (차가버섯으로부터 분리한 페놀성 화합물의 항산화효과)

  • ln our continued search for biologically active compounds from traditional medicine, we found that ethyl acetate fraction from Inonotus obliquus showed potent antioxidant activities on three different cell-free bioassay systems. Bioassay-directed chromatographic fractionation of the ethyl acetate fraction from Inonotus obliquus afforded four phenolic compounds, 4-(3,4-dihydroxyphenyl)-(E)-3-buten-2-one (1) , 3,4-dihydroxybenzaldehyde (2), protocatechuic acid (3) and caffeic acid (4) along with three sterols such as lanosterol (5), ergosterol peroxide (6) and 9,11-dehydroergosterol peroxide (7). Among isolates, phenolic compounds (1-4) were assessed for antioxidant activities. Almost phenolic compounds showed potent DPPH and superoxide anion radicals scavenging and lipid peroxidation inhibitory activities indicating that these phenolic compounds contribute to the antioxidative activities of I. obliquus. Compounds 2-3 and 7 were isolated for the first time from this plant.

Optimization of Microencapsulation of Inonotus obliquus Extract Powder by Response Surface Methodology and Its Application into Milk

  • This study was carried out to optimize microencapsulation conditions for Inonotus obliquus extract powder with mediumchain triacylglycerol (MCT) using response surface methodology (RSM), and to determine the possibility of adding encapsulated I. obliquus extract to milk toward improving the functionality of milk. and to investigate the effects of added encapsulated I. obliquus extract on the physicochemical and sensory properties of the product. The microencapsulation efficiency of I. obliquus extract powder was investigated with respect to three variables (ratios of core material to coating material, amounts of water added, and spray pressure) in RSM. The optimal conditions for microencapsulation of I. obliquus extract powder were obtained from the ratio of core material to coating material (2.92:10) and the amount of water added (0.5 mL); the spray pressure had an insignificant effect on the microencapsulation (p<0.05). Adding encapsulated I. obliquus extract into milk did not significantly affect either color (L, a and b values) or thiobarbituric acid (TBA) values. Sensory test revealed that there were significant improvements in brownish, herb flavor, and bitterness scores for encapsulated I. obliquus extract-added milk as compared with those for uncapsulated I. obliquus extract-added milk. Based on the data obtained from the present study, it is concluded that microcapsules of I. obliquus extract powder could be applicable to milk without remarkably adverse effects on physicochemical and sensory properties

Antioxidant and Genotoxic Inhibition Activity of Ethanol Extract from the Inonotus obliquus (차가버섯 분획물의 항산화활성 및 유전독성 억제효과)

  • This study was performed to observe the antioxidative and genotoxic effect of the fractions from Inonotus obliquus using DPPH test and micronucleus assay. Stepwise fractionation of the ethanol extract from Inonotus obliquus was done by using hexane, chloroform, ethyl acetate, butanol and water to obtain effective fraction. Each fraction was tested in 1.5 ${\times}$ 10$^{-4}$ DPPH. Among six fractions, the ethyl acetate fraction showed the highest electron donating activities (46.5 $\mu\textrm{g}$/mL). The results on genotoxic effects on insoluble fractions and most of fractions showed cytotoxic effects more than 90% activity. These results suggest that some components contained in the Inonotus obliquus showed such activities and much more studies have to perform.
