Biomass yield, nutritive value and nutrient utilization efficiency of different genera of duckweed (DW, Lemnaceae) under the same nutritional and management conditions were studied. Three genera of DW namely: Lemna perpusilla, Spirodela polyrhiza and Woljfia arrhiza, were cultivated in 18 plastic sinks each of $59{\times}54{\times}35cm^3$ size. Each sink contained approximately 80 L of water and was charged once with 6.648 kg of anaerobically fermented cow dung effluent. The seed rate for Spirodela, Lemna and Wolffia were 600, 400 and $600g/m^2$ respectively. Duckweed were harvested at 48 hours intervals. Media total N concentration for Lemna and Wolffia gradually increased with time. Growth of duckweed was measured by subtracting the inoculum from the total biomass production. Growth of Spirodela ceased within 4 days but Lemna and Wolffia continue to grow up to 34 days. Bio-mass yield was significantly (p<0.05) higher in Wolffia (906 kg/ha/d) than Lemna (631 kg/ha/d). Dry matter (DM) yield was very similar in Lemna (14.80 kg/ha/d) and Wolffia (14.57 kg/ha/d). The N content was non-significant higher in Lemna (5.45%) than Wolffia (5.00%) and Spirodela (4.6%). The crude protein (CP, $N{\times}6.25$) yield was non-significantly higher in Lemna (4.83 kg/ha/d) than Wolffia (4.32 kg/ha/d). The acid detergent fibre (ADF) content was the highest in Wolffia (28.59%), followed by Spirodela (19.47%) and Lemna (12.39%). Utilization efficiency of CP was 273 and 314% respectively for Wolffia and Lemna. However, similar efficiency values for Spirodela was only 1.5%. Considering the bio-mass yield, nutritive value and nutrient utilization efficiency, production performance of DW were in the order of Lemna>Wolffia>Spirodela under the present experimental conditions.
Under standard condition (Han, et al., 1990: glucose 1%-nitrate 0.1% minimal medium, 30 ml in 9 cm plate, $10^6$ cells of inoculum per plate), wild type of Aspergillus nidulans developed both sexual and asexual organs in ballance, while velA1 mutant developed asexual ones preferentially. Increase of glucose concentration did not significantly affect the asexual sporulation. However, development of sexual organs were largely affected. It was greatly enhanced when favorable nitrogen source, for example, casein hydrolysate was added, which is contrary to the case of Neurospora or Saccharomyces where limitation of N source induces sexual development. On most of moderate C sources asexual development in $velA^+$ strain was largely inhibited except acetate on which only asexual spores were produced, while that in velA1 mutant strain was not affected. Lactose promoted the sexual development even in velA1 mutant indicating that lactose itself or its metabolic intermediate may induce sexual development independent of allelic state of velA gene. On other moderate favorable C sources, glycerol, galactose and ethanol, asexual development was largely inhibited in $velA^+$ strain but not in velA1 mutant strain. Sexual organs were, however, never produced on acetate. These results suggested that asexual development of wild type is largely dependent on C sources and the velA gene is involved in the repression of asexual development in not-enough-grown (non-competent) thalli resulting in preferential progression of sexual development.
Gardenia callus was induced in MS medium containing $10{\;}{\mu}M$ of 2,4 diphenoxy acetic acid (2,4-D), $1{\;}{\mu}M$ kinetin, and 3% sucrose in the dark. $B_5$ medium was identified to be the most adequate medium for cell growth. Indole-3-acetic acid (IAA) was better growth regulator than 2,4-D not only for cell growth but slso for carotenoid production. Ligt also played a critical role on synthesis of carotenoid. Gardenia cells grown in $B_5$ medium could utilize a polysaccharide, soluble starch, as a carbon source. The cell growth was stimulated in $B_5$ medium fortified with 0.2% yeast extract. The optimum pH for cell growth was 5.7. High density cultures can be maintained by increasing inoculum size and medium concentration accordingly. Specific growth rate and mass doubling time were 0.095 $day^{-1}$ and 7.3 days, respectively. The cell immobilized in alginate tends to formulate more enlarged vacuoles containing yellow pigment compared with those of suspended cell. Carotenoid content of immobilized cell was about $264.4{\;}{\mu}g/g$ fresh weight (F.W.) corresponding twice of the content of suspended cell ($112.08{\;}{\mu}g/g$ F.W.). The color of gardenia cell was shifted from yellow to red when carbohydrase-secreting fungus, Trichoderma reesei, was co-cultivated with gardenia cells.
Escherichia coli O157:H7 and Salmonella ser. typhimurium are pathogens involved in food poisoning in numerous countries. This study aimed to obtain knowleges on the survival of E. coli O157:H7 KSC 109 and S. ser. typhimurium ATCC 14028 in fermentedmilk products which were on sale in Suwon Yakult supplier. To the final concentration of 103~104 cfu/$m\ell$ of E. coli O157:H7 KSC 109 or S. wer. typhimurium ATCC 14028 in the fermented milks, Metchnikoff, Ace, Yakult, Mastoni and Super 100 were inoculated with these pathogens and then were stored at 4$^{\circ}C$ and viable cells of these pathogens were periodically counted. The results showed that the survival of two pathogens differed in the different types of fermented milks tested. Number of suriviving E. coli O157:H7 KSC 109 and S. ser. typimurium ATCC 14028 cells (initial inoculum, 103~104 cfu/$m\ell$) were decreased to 101, 102 cfu/$m\ell$ in Ace after 100 hours, and were decreased gradually to 101 cfu/$m\ell$ in Yakult after 250 hours. In the other fermented milks, viable cells of E. coli O157:H7 KSC 109 was not drastically decreased but those of S. ser. typhimurium ATCC 14028 was decreased gradually to 102 (Mastoni), and to 101 cfu/$m\ell$ (Super 100) after 250 hours. It appeared that S. ser. typhimurium ATCC 14028 was more susceptible than E. coli O157:H7 KSC 109 at low pH. Vibale cells of E. coli O157:H7 KSC 109 was not drastically decreased in most of fermented milks tested except Ace and Yakult, but in general, S. ser. typhimurium ATCC 14028 was drastically decreased in most of the fermented milks. The major ingibition factor against these pathogens in the fermented milks during storage at 4$^{\circ}C$ appeared to be the acidity and the metabolites produced by the starters bacteria used in fermented milk products.
본 연구에서는 신 개념의 바이오필터 시스템인 RDBF를 이용하여 기상의 styrene을 제거하고자 하였다. 다양한 운전조건에서 시스템의 성능을 평가하고, 진공흡입을 통해 과다성장 미생물을 제어하고자 하였다. RDBF에서는 sheet 형태의 충진 담체를 사용하므로 영양원 및 공기의 균일한 공급이 가능하였고, 빠른 속도로 균일한 미생물 층을 담체 표면에 형성할 수 있었다. 또한 기-액 접촉면적을 증대 시켜 95% 이상의 안정적이고 높은 styrene 분해 효율과 $125g/m^3{\cdot}hr$의 높은 제거 용량을 가질 수 있게 해주었다. 하지만 우수한 성능은 미생물의 과다성장에 따른 기공의 폐쇄현상 때문에 오래 지속되지 않았고 반응기의 성능과 안정성은 급속히 저하되었다. 이를 해결하기 위해 진공흡입을 통한 미생물 제거를 시도하였으나 제한적인 효과만을 확인하였다. 향후 RDBF 반응기의 상업화를 위해서는 보다 효율적인 미생물 제어 방법이 개발될 필요가 있다.
하늘타리 (Trichosanthes kirilowii)로부터 조직배양 기법을 이용하여 약리작용을 나타내는 생리활성물질인 trichosantin을 생산하기 위한 연구로 Agrobacterium rhizogenes ATCC15834 의 감염을 통하여 기내 배양된 유묘의 잎절편으로부터 형질전환 모상근을 유도하였다. 유도된 모상근 clones으로부터 성장속도와 분지화 정도가 빠르며 단백질 합성능이 가장 우수한 TR-03 clones을 선발하였으며, 이를 사용하여 생체량과 배지내 가용성 단백질의 생산을 최대로 높일 수 있는 최적 배양조건을 조사하였다. TR-03 clone을 4% sucrose가 첨가된 MS 배지에 초기 접종농도 2 g (생중량)을 접종하여 100 rpm으로 배양시 생중량과 배지내 가용성 총단백질의 함량이 플라스크당 약 2.4 g (건중량)과 $28.3ug/\ell$으로 최대치를 나타내었다. 한편, 배지내 가용성 단백질에 대한 RIP활성 검정에서 배양 4주 후 21.3 unit로 최대치를 보였다.
두과목초의 근류균 접종방법 개선책의 일환으로써 3수준의 근류균농도, 즉 10, 20, 30%; 3종류의 접착제 즉 gum arabic, methyl cellulose 및 carboxy methyl cellulose; 그리고 4종류의 석회와 calcium carbonate를 포함한 5종류의 피복재료를 사용하여 근류균을 접종한 후에 plate count법으로 생존하고 있던 근류세포수를 측정하여 그 효과를 평가하였던 바 다음과 같은 결론을 얻었다. 1. New Zealand산 peat slurry의 근류균을 사용할 경우 근류균을 10%에서 3.5$\times$$10^9$ 개로써 표준에 가까운 근류균수를 확보할 수 있었다. 2. 접착제로서 gum arabic은 40% 용액에서 methyl cellulose는 5% 용액 그리고 carboxy methyl cellulose는 4% 용액으로 사용하였을 때 최대착생수를 나타내었으나 그 중에서 methyl cellulose 5% 용액이 적합한 것으로 보였다. 3. 피복재료로서 치과용석회(Lime A)는 타재료에 비하여 양호한 결과를 얻을 수 있었다.
This study was conducted to evaluate the effects of rumen-protected amino acid (RPAA) prototypes, which were chemically synthesized, on in vitro rumen fermentation and protection rate outcomes. Several RPAA prototypes were incubated with timothy hay and concentrate. Treatments consisted of 1) control (CON; no RPAA prototype supplement), and prototypes of 2) 0.5% RP-methionine (RPMet), 3) 0.5% RP-tryptophan (RPTrp), 4) 0.5% RP-valine (RPVal), 5) 0.5% RP-phenylalanine (RPPhe), 6) 0.5% RP-leucine (RPLeu), 7) 0.5% RP-histidine (RPHis), 8) 20% RPMet, and 9) 20% RPTrp (w·w-1 feed). The inoculum (50 mL) prepared with rumen fluid and McDougall's buffer (1 : 4) was dispensed in individual serum bottles and was anaerobically incubated for 0, 6, and 24 h at 39℃ in triplicate. The dry matter degradability did not differ among the groups, except for the 20% RPMet and the 20% RPTrp treatments at 6 and 24 h. The total volatile fatty acid concentration in the 20% RPMet was higher (p < 0.05) than the rest of the groups at 6 h, and 20% RPMet showed the highest molar proportion of acetate, whereas the lowest proportion of propionate was found at 6 h (p < 0.05). The protection rate of the RPAA prototypes ranged from 29.85 to 109.21%. at 24 h. In conclusion, the chemically synthesized RPAA prototypes studied here had no detrimental effects on rumen fermentation parameters. Further studies using animal models are needed for more accurate evaluations of the effectiveness of RPAA.
Corn cobs were fermented with Aspergillus niger to produce soluble dietary fiber (SDF) of high quality and excellent food safety. In this work, the fermentation process was optimized by single-factor test and response surface methodology (RSM). The optimal fermentation conditions were determined to be a material-liquid ratio of 1:30, an inoculum concentration of 11%, a temperature of 32℃, a time of 6 days, and a shaking speed of 200 r/min. Under these conditions, the SDF yield of corn cob increased from 2.34% to 11.92%, and the ratio of soluble dietary fiber to total dietary fiber (SDF/TDF) reached 19.08%, meeting the requirements for high-quality dietary fiber (SDF/TDF of more than 10%). Scanning electron microscopy (SEM) and Fourier-transformed infrared spectroscopy (FT-IR) analysis revealed that the fermentation effectively degraded part of cellulose and hemicellulose, resulting in the formation of a loose and porous structure. After fermentation the water swelling capacity, water-holding capacity, and oil-holding capacity of the corn cob SDF were significantly improved and the adsorption capacity of glucose, cholesterol, and nitrite ions all increased by more than 20%. Moreover, the total phenolic content increased by 20.96%, which correlated with the higher antioxidant activity of SDF. Overall, the fermentation of corn cobs by A. niger increased the yield and enhanced the functional properties of dietary fiber (DF) as well.
본 연구는 해산물 가공폐수를 대상으로 혐기성 미생물, S/I ratio (substrate/inoculum)와 염분농도에 따른 혐기성 최종 생분해도를 평가하였다. S/I ratio 0.9에서 혐기성 소화슬러지와 입상슬러지의 최종 생분해도는 각 72.0, 92.0%로 조사되었으며, 다중분해속도 상수 $k_1$은 소화슬러지가 $0.0478{\sim}0.1252\;day^{-1}$, 입상슬러지는 $0.0667{\sim}0.1709\;day^{-1}$로 조사되어 입상슬러지가 해산물 가공폐수의 혐기성 처리에 적합하였다. 혐기성 최종생분해도 실험을 통해 산정된 최적 S/I ratio는 0.9였으며, 염분농도에 따른 생분해도 실험 결과, $3,000\;mgCl^-/L$ 이하에서 85% 이상의 유기물 제거효율을 나타냈다. 다중분해속도 상수 $k_1$은, $3,000\;mgCl^-/L$ 이하에서는 $0.1603{\sim}0.1709\;day^{-1}$, $6,000\;mgCl^-/L$ 이상에서 $0.0492{\sim}0.0760\;day^{-1}$로 산정되었으며, $k_2$는 $6,000\;mgCl^-/L$ 이하에서는 $0.0183{\sim}0.0348\;day^{-1}$, $9,000\;mgCl^-/L$에서는 $0.0154\;day^{-1}$로 조사되어, 반응속도 상수($k_1$, $k_2$)는 $Cl^-$ 농도가 증가할수록 감소하였으며, 빠르게 분해되는 유기물 비율($S_1$)과 분해속도 또한 감소시키는 것으로 조사되었다.
