The Recycling of Enokitake Cultural Waste and the Potentiality of 2nd Flush for Enokitake Production

  • The recycling method of enokitake cultural waste and the potentiality of second flush for enokitake were determined, because this fungus is not as prolific as the more commonly cultivated white rot fungi in the conversion of sawdust to mycelial mass. The mycelial growth of F. velutipes on several substrates, variously treated with rice bran was promoted at ratios of 10-20% (w/w) on all substrates, but suppressed at above ratios, although some difference was there. The mycelial densities generally increased correlated to the supplementation contents of rice bran. It could be concluded that F. velutipes preferred mild acidic to acidic conditions for mycelial growth, considering that the mycelial growth rate was highest on waste of pH 6.01, treated with 0.1 % Ca(OH)$_2$ and on populus mixed waste of pH 6.02, non treated. The ranges of substrate bulk densities, which was pertinent for mycelial linear growth were from B.D. (g/cc) 0.17 to 0.23 on waste and populus mixed waste all. The pertinent contents of rice bran supplementation in bottle cultivation was from 20 to 30% on waste and 20% on populus mixed waste, considering the requried duration for pinheading and fruiting yields. Standard bulk density for filling and utilizing the waste and populus mixed waste for commercial f. velutipes cultivation were B.D.(g/cc) 0.19 ~ 0.23, and 0.23~ 0.25, which could be conversed to 510~ 540g/900m1 and 520~ 570g/900m1, respectively, The second flush of F. velutipes was tried and the re-inoculation by sawdust and liquid spawn showed somewhat good results, indicating the potentiality of second crop and suggesting further research for it.

Effect of Northern Root-knot Nematode, Meloidogyne hapla, on Growth of Codonopsis Zanceolata (당근뿌리 혹선충(Meloidogyne hapla)이 더덕(Codonopsis lanceolata)의 생육에 미치는 영향)

  • This study has been conducted to assess and analyze the damage of Codonopsis lanceolata by differential inoculation levels of Meloidogyne hapla in pot and plot. As the density of M. hapla increased, early growth of C. lanceolata was inhibited 60% in vine and 54% in root. However, growth of C. lanceolata was not remarkably different by density of M. hapla in the plot test. This result may be attribute to low density of M. hapla in the plot.

  • We have investigated the characteristics of encephalomyocarditis(EMC) virus isolated in Korea. The CPE, buoyant density, polypeptide profile and the size of RNA of EMC virus were examined. The granulation, pyknosis and necrosis were observed from 30 to 48 hour's post inoculation of the virus into baby hamster kidney and lung cells. The buoyant density was 1.30 and $1.35g/m{\ell}$. Three different polypeptides, 26Kd, 32Kd, and 34Kd in size, were observed and the size of viral RNA was 7.7Kb.

Characteristics of Pinewood Nematode Trapping by Nematophagous Arthrobotrys spp. (선충포획성 Arthrobotrys속균에 의한 소나무재선충 포획 특성)

  • Pinewood nematode (PWN) trapping by nematophagous fungi, Arthrobotrys conoides, A. dactyloides and A. oligospora and the fungal growth were characterized. The three Arthrobotrys species each was inoculated into the PWN cultured on Botrytis cinera fungal colony on potato dextrose agar (PDA). The effects of temperature, pH, PWN inoculation density and nutrients on the growth of the three Arthrobotrys spp were measured. A. conoides grew fast, 13.9 mm/day while A. dactyloides grew slow, 3 mm/day. PDA medium was the best for the fungal growth at $25^{\circ}C$ and pH 4.5. The Arthrobotrys spp growth was stimulated by 500 nematodes inoculation but not by 1000 inoculation. A. dactyloides did not grow below pH 4.5 and at high PWN density. A. conoides and A oligospora formed trapping organs with thick constricting hyphal network only when PWN present, while A. dactyloides formed the organ with circular hyphae constitutively. A. conoides formed trapping organs faster than A. oligospora did. The nematode trapping hyphae of the fungi penetrated into PNW inside to form many tiny infection bulbs and to digest the nematode. However, A. dactyloides formed a few trapping organs but no trapping was observed. Infection rate of PWN was 95% by A. conoides, 80% by A. oligospora and 92% by the combination inoculation of A. conoides and A. oligospora. In contrast A. dactyloides increased PWN density without infecton. There was no interaction effect in any combination inoculation of the three Arthrobotrys spp. A. conoides enhanced PWN infection rate by rapid hyphal growth and early trapping, while A. oligospora did it by increasing hyphal density. In conclusion A. conoides is the most effective in both hyphal growth and infection, and thus these characteristics can be utilized as a biological control of PWN.

A New Method for Cultivation of Sclerotium of Grifola umbellata

  • Sclerotia of Grifola umbellata were cultivated by two methods such as burying and root inoculation methods. The sclerotia of G. umbellata produced by the burying method were $6.0{\sim}6.8{\times}3.4{\sim}4.6{\times}1.8{\sim}1.9cm$(Width$\times$Length$\times$Thickness) in size and $17.3{\sim}19.6g$ in weight, respectively. Their increase rate was $1.10{\times}1.12$ times. On the other hand, the sclerotia cultivated by the root inoculation method were $18.3{\sim}31.5{\times}12.5{\sim}26.4{\times}3.1{\sim}3.7cm(W{\times}L{\times}T)$ in size and $219.1{\sim}576.6g$ in weight, respectively. Their growth increment was $11.18{\sim}39.77$ times. The rhizomorphs of Armillaria mellea were developed with a high density under fallen leaves layer covering cultivation site, and distributed mainly between soil surface and soil depth of about 10 cm as well as colonized prominently on the inoculated wood logs. Fungal interaction between G. umbellata and A. mellea were observed mainly in the stage of white sclerotium of G. umbellata. The sclerotia of G. umbellata which were developed newly and harvested in the root inoculation method were twined with root hairs of host tree and rhizomorphs of A. mellea. The sclerotia of G. umbellata decomposing root hairs of host tree were confirmed through SEM examination. Physiochemical characteristics of soil in all cultivation sites had no significant differences. Soil pH were in the range of pH $3.98{\sim}4.40$. Organic matters were the range of $17.97{\sim}23.86%$ and moisture contents of soil were $12.00{\sim}18.20%$. Soil temperatures showed $12.9{\sim}13.8^{\circ}C$ in November and $22.0{\sim}23.9^{\circ}C$ in August, respectively. In conclusion, the root inoculation method seems to be a practical method for cultivating sclerotia of G. umbellata due to its many advantages such as simplicity of inoculation process, shortening of cultivation periods and facility of harvest.

  • For daily supply of rotifers to feed juvenile fish and/or larval crustaceans such as shrimp in their seedling culture procedures, the maximum daily increase of cultured rotifers in relations with the inoculation densities of rotifers and the concentrations of food organism, Chlorella sp. were initially investigated in an experimental culture unit. The best results were obtained from the 10 inds./ml of the rotifer inoculation density with $3\times10^6\;cells/ml$ of Chlorella showing the maximum daily increase of 73.0 inds./ml/day on the 7th day. Based on the best results of the initial experiments a trial was made for continuous daily harvests of rotifers. As in the initial experiments, the maximum daily increase of rotifer was observed on the 7th day, when the daily harvest was initiated and continued once a day for 6days. For the 6 days, the amount of the mean daily harvest was $1.42\times10^5$ individuals per culture flask ($3{\iota}\;filled\;with\;2{\iota}$culture water) shelving a possibility of continuous daily harvests.

  • Various susceptibility tests have been used to determine minimal inhibition concentration(MIC) of dermatophytes. They have limitations to apply practically because they need long time to determine MIC. Authors examined MIC of T. rubrum to ketoconazole and itraconazole using 96-well microplate and 24-well macroplate by method of Granade and Artis and tried to check the possibility of this method on clinical application. Nine strains of T. rubrum from patients with dermatophytosis were used. Evaluations of the factors affecting MIC were also tried. The results were as follows. 1. Effect of inoculation density on determination time and MIC : Determination of MIC were possible in 4th days after inoculation at higher inoculation density Caborbance 2.0, 1.0) compared to 6th days at lower inoculation density(absorbance 0.5, 0.25). 2. Effect of incubation temperature on MIC : When incubating at $37^{\circ}C$, MIC were below 0.006-$0.04{\mu}g/ml$ to ketokckonazole and below 0.006-$0.04{\mu}g/ml$ to itraconazole while at $25^{\circ}C$ 0.08-$5.68{\mu}g/ml$ to ketoconazole and 0.006-$0.71{\mu}g/ml$ to itraconazole. Significant reduction of MIC was observed at $37^{\circ}C$ compared to $25^{\circ}C$. 3. Effect of container size on determination time and MIC : When incubating in 96-well microplate and 24-well macroplate, determination of MIC was possible in 4th to 6th days after inoculation in broth-containig 96-well microplate compared to 8th to 12th days in broth-containing 24-well macroplate. But no difference in MIC was observed between different container size. 4. Effect of media on MIC : When using broth as media, MIC were below 0.006-$5.68{\mu}g/ml$ to ketoconazole, below 0.006-$0.36{\mu}g/ml$ to itraconazole in broth-containg 24-well macroplate. When using agar as media, MIC were below 0.006-$5.68{\mu}g/ml$ to ketoconzole, below 0.006-$5.68{\mu}g/ml$ to intraconzole in agar-containing 24-well macroplate. There was slight increase of MIC with agar media compared to broth media. 5. These findings confirm that determination of MIC of dermatophtes by method of Granade and Artis is fast and simple technique for antifungal susceptibility test.

  • Pine wood nematode (PWN), Bursaphelenchus xylophilus is one of the most serious pests of pine tree. Trunk injection of some nematicides in tree is well known as an effective control method. However there are some limitating factors which hindering the efficacy of trunk injection in field or potted tree. In this study we suggested easy and useful alternative screening methods of nematicides against PWN. Reproduction of PWN was influenced by tree twig moisture (high reproduction in high moisture trig) and paraffin coating was 78.6% reduced moisture loss in tested twig. There were no reproduction different in up and down site from infection site of twig at 1 month after inoculation of PWN and also distance (5 and 10 cm) from inoculation site of PWN did not influence the reproduction of PWN. Numbers of reproduced PWN were higher with decreasing diameter of twig. Numbers of reproduced PWN were similar to P. thunbergii and P. densiflora. However reproduction was increased depending on high inoculation density and longer propagation period. When inoculation of PWN on cut twig injected with emamectin benzoate 2.15% EC and morantel tartrate 8% SL in trunk of Pinus thunbergii in the field, PWN number were significantly reduced than untreated control. We suggest this screening method for PWN control agents.

Improvement of Strength and Chemical Resistance of Silicate Polymer Concrete

  • It has been known that acid-resistant concretes on the liquid glass basis have high porosity (up to 18${\sim}$20%), low strength and insufficient water resistance. Therefore they can not be used as materials for load-bearing structural elements. Significant increasing of silicate matrix strength and density was carried out by incorporation of special liquid organic alkali-soluble silicate additives, which block of superficial pores and reduces concrete shrinkage deformation. It was demonstrated that introduction of tetrafurfuryloxisilane additive sharply increases strength, durability and shock resistance of silicate polymer concrete in aggressive media. This effect is attributable to hardening of contacts between silicate binder gel globes and modification of alkaline component owing to "inoculation" of the furan radical. The optimal concrete composition with the increased strength, chemical resistance in the aggressive environments, density and crack resistance was obtained.

Characteristics of Recombinant CHO Cell Growth and Erythropoietin Production in Serum-Containing Media and Serum-Free Media (혈청배지와 무혈청배지에서의 재조합 CHO 세포 성장과 Erythropoietin 생산)

  • We have investigated the characteristics of recombinant CHO cell growth and erythropoletin(EPO) production at different concentrations of serum and inoculation density. Cell growth and EPO production were increased with the increase of serum concentration and inoculation density. Enhancement of CHO cell growth and EPO production by medium exchange using serum-free medium at the growth phase of cells was studied. It was found that the exchange of culture medium with serum-free medium was favorable for growth of cells and production of EPO. The maximum number of cell and concentration of EPO obtained by exchanging culture medium were $6.2{\times}105cells/$\textrm{cm}^2$ and 7,470units/m1, respectively, compared to $2.1{\times}105cells/\textrm{cm}^2$ and 2,380units/m1 in serum-containing medium without medium exchange. It was observed that CHO cell growth was correlated with EPO production in serum-free media.
