In the present study, we investigated the anti-allergic effect of the water extract of Bopyeoyangyeong-jun(BYJ). To investigate the biological effect of BYJ, We examined cytotoxicity and inflammatory cytokine secretion with RBL-2H3. We examined Cell Viability, ${\beta}$-hexosaminidase, tumor necrosis factor-alpha(TNF-${\alpha}$), interleukin(IL)-4 secretion from RBL-2H3 cell after pre- treatment with Bopyeoyangyeong-jun of 1 mg/ml, 2 mg/ml. RBL-2H3 cell was stimulated with phorbol 12-myristate 13-acetate(PMA) and calcium ionophore A23187. BYJ by itself had no effect on viability of RBL-2H3 cell. We observed that Bopyeoyangyeong-jun reduced ${\beta}$-hexosaminidase, TNF-${\alpha}$, IL-4 secretion in RBL-2H3 cell. Taken together, these results indicate that Bopyeoyangyeong-jun has anti-histamic effect and controls TNF-${\alpha}$, IL-4 secretion on allergic reaction.
Type I allergic reaction and it's related clinical manifestations are known to occur by the effects of various chemical mediators. These chemical mediators are released from circulating basophils and tissue mast cells, which become 'sensitized' through the binding of antigens and antibodies of the IgE type to their cell surface receptors. Efforts to elucidate the mechanism of the release of these mediators, especially that of histamine, have been persued for years. The mechanism is not yet clarified at the present time. Recent reports of hyaluronidase, an enzyme known to be involved in the tissue inflammatory process, as possible participant in type I allergic reaction, initiated this study. Relationships between the hyaluronidase activity and histamine release from the sensitized rat peritoneal mast cells were investigated. Also anti-allergic agents, tranilast and disodium cromoglycate, along with known histamine releasers, morphine and compound 48/80, were used to observe the inhibitory and stimulatory effects of these substances on the hyaluronidase activity as well as histamine release from the rat mast cells. The results obtained are summarized as follows: 1) Hyaluronidase activity and histamine release from sensitiaed rat peritoneal mast cells started to increase on the 4th day of postsensitization. Hyaluronidase activity reached it's peak value on the 7th day of postsensitization and that of histamine release on the 14th day of postsensitization. 2) Hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells, pre-treated with tranilast revealed significant decrease in comparison with those of non-treated cells. 3) Hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells, pre-treated with tranilast, followed by morphine injection, revealed significant increase in comparison with those of tranilast treated cells. 4) In vitro study of hyaluronidase activity and histamine release from un-sensitized rat peritoneal mast cells, using morphine and compound 48/80 as activators, revealed significant increase compared to those of non-activator used cells. 5) In vitro study of hyaluronidase activity and histamine release from un-sensitized rat peritoneal mast cells, pre-treated with tranilast and disodium cromoglycate, using confound 48/80 and morphine as activators revealed significant decrease in comparison with those of tranilast and disodium cromoglycate treated cells. From above results, participation of enzyme hyaluronidase in the process of histamine release from sensitized rat pertioneal mast cells, could be suggested. It was also quite evident that the clinically used anti-allergic agents, tranilast and disodium cromoglycate, have significant inhibitory function on the hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells, while morphine significantly increased the hyaluronidase activity and histamine release from sensitized rat peritoneal mast cells.
A foreign body granuloma is a non-allergic chronic inflammatory reaction that is mainly composed of multinucleated giant cells. Foreign body granulomas may occur after the administration of any dermal filler. Factors such as the volume of the injection, impurities present in the fillers, and the physical properties of fillers affect granuloma formation. The formation of granulomas involves five phases: protein adsorption, macrophage adhesion, macrophage fusion, and crosstalk. The clinical and pathologic features of granulomas vary depending on the type of filler that causes them. Foreign body granulomas can be treated effectively with intralesional corticosteroid injections. Surgical excisions of granulomas tend to be incomplete because granulomas have ill-defined borders and moreover, surgical excisions may leave scars and deformities.

Atopic dermatitis is a chronic, inflammatory skin disease characterized by intense pruritus and the course marked by exacerbation and remission. It has the characteristic symptoms of dry, itchy, scaly skin with cracks by typical distribution of lesions. Even though the etiology of atopic dermatitis is not fully understood, it is considered an allergic reaction whose onset is frequently and strongly associated with both hereditary and environmental factors. In the present clinical study, we studied the efficacy of internal herb-medicine, external herb medicine - ex. dermal oints, aqueous solutions, etc -, and steam therapy. The patient's SCORAD index was lowered 55.8 than before of which the SCORAD index was 74.8. On the basis of this study, it should be considered that the treatment of atopic dermatitis should be focused on the external therapy as steam therapy, dermal oint, a aqueous solution etc.

Lonicerae Flos containing formulation, Geumeumyoungji-tang (GYT), is an herbal prescription prepared using 5 different herbal drugs, namely, Lonicerae Flos, Ganoderma, Lactucae Herba, Xanthii Fructus, and Smilacis Rhizoma. This study was focused on the investigation of the pharmacological effects of GYT on allergic reactions. As the first step of the study, GYT was administered BALB/c mice which were sensitized by 2,4-dinitrofluorobenzene (DNFB). As the result GYT ameliorated dermatitis provoked by DNFB. The serum IgE level of the DNFB sensitized-mouse was significantly decreased when GYT was administered. In order to confirm the moderating effect of this prescription on allergic reaction, GYT was pretreated to human mast cells (HMC-1) before they were stimulated by phorbol-12-myristate 13-acetate plus calcium ionophore A23187 (PMACI). GYT suppressed secretion of inflammatory cytokines, interleukin (IL)-6 and IL-8, from HMC-1. Additionally, pretreatment of GYT showed regulating effect on COX-2 expression. Collectively, these findings provide insights into the pharmacological actions of GYT as a potential agent for treatment of allergic dermatitis.

Gamipaidok-san(GPDS) is the Hyungbangpaidok-san prescription fortified with the additional ingredients known to be effective for dermatitis. So it has been used for atopic dermatitis in the clinic work actually. In this study, we investigated the effects of GPDS on in vitro and in vivo anti-allergic effect on RBL-2H3 rat basophilic leukemia Cells and on IgE-induced passive cutaneous anaphylaxis (PCA) in mice. The in vitro anti-inflammatory activity of GPDS in RAW 264.7 cells was investigated. GPDS potently inhibited $\beta$-hexosaminidase release from RBL-2H3 and the IgE-mediated PCA reaction in mice. GPDS inhibited LPS-induced NO and PGE2 production in a dose-dependent manner Furthermore, It also inhibited inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression in RAW 264.7 cells, and the activation of the transcription factor, NF-kB, in nuclear fraction. The antiallergic action of GPDS may originate from anti-inflammatory activities, and can improve the inflammation caused by allergies.
The number of patients with immune- mediated dermatitis such as contact dermatitis is increasing year by year. Allergic contact dermatitis is a complex phenomenon that involves resident epidermal cells, fibroblasts, and endothelial cells, as well as invading leukocytes that interact with each other under the control of a network of cytokines and lipid mediators. It is a cell-mediated immune reaction, which occurs after susceptible individuals are exposed to sensitizing chemicals, and characteristic eczematous reaction is seen at the point of contact with an allergen. In this study, we investigated the allergy prevention effects of quercetin on mast cell-mediated allergic inflammation in BALB/c mice. BALB/c mice were sensitized with 40 ${\mu}l$ of 1.5% picryl choloride (PCL) to the left and right ear each. Total serum IgE levels and histamine levels were measured by the sandwich ELISA method using mouse IgE, histamine measuring kit. For histopathological examination, paraffin sections were stained with hematoxylin and eosin(HE) or toluidine blue(TB). Ear swelling responses were much weaker in the high-dose group (100 mg/kg) than the control group (0 mg/kg). The number of mast cells showed a significant decrease in the high-dose group (100 mg/kg) compared to the control group (0 mg/kg). Degranulation of mast cells was also confirmed by Toluidine Blue (TB) staining method. Both total serum IgE and histamine levels were significantly decreased in the high-dose group (100 mg/kg) compared to other groups. These findings suggest a certain relationship between the elevation of IgE, histamine levels and the degranulation of mast cells. These results show that the pharmacological actions of quercetin indicate its potential activity for prevention of allergic inflammatory diseases through the down-regulation of mast cell activation.
Atopic dermatitis (AD) is an allergic, inflammatory skin disease characterized by chronic eczema and mechanical injury to the skin, caused by scratching. Korean red ginseng (RG) has diverse biological activities, but the molecular effects of RG on allergic diseases, like AD, are unclear. The present study was designed to investigate whether RG inhibits 1-chloro-2,4-dinitrobenzene (DNCB)-induced AD in a mouse model. DNCB was applied topically on the dorsal surface of Balb/c mice to induce AD-like skin lesions. We observed the scratching behavior and examined the serum IgE level and interleukin (IL)-4 and IL-10 in splenocytes compared with dexamethasone. We also evaluated the DNCB-induced mitogen-activated protein kinases (MAPKs), NF-${\kappa}B$, and Ikaros activities after RG treatment using reverse transcriptase-polymerase chain reaction, Western blotting, and ELISA. Our data showed that the topical application of RG significantly improved the AD-like skin lesions and scratching behavior. RG decreased not only the mRNA expression of IL-4 and IL-10, but also the secretion of IL-4 protein and serum IgE in mice. Additionally, RG treatment decreased the DNCB-induced MAPKs activity and subsequent Ikaros translocation irrespective of NF-${\kappa}B$. We suggest that RG may be useful as a therapeutic nutrition for the treatment of AD.

Mast cells play an important role in the pathophysiologlcal changes observed in acute cutaneous and inflammatory diseases In order to see whether Kum-Hwang-San has an influence on mast cell- mediated immediate cutaneous reactions, the author has undertaken an animal study. Ears of mice were treated with a compound 48/80 solution topically at 30 min after the cutaneous application of Kum-Hwang-San. At each point, an ear swelling response was measured with a digimatic thickness micrometer. Ear swelling response induced by compound 48/80 was significantly suppressed dose-dependently by Kum-Hwang-San 30 min before topical application as compared with that in nonapplicated control mice, and the value returned to normal levels by 120 min. Compound 48/80- induced mast cell degranulation by Kum-Hwang-San was also remarkably decreased in accordance with the suppression in ear swelling response. Kum-Hwang-San dose-dependently inhibited histamine release from the rat peritoneal mast cells activated by compound 48/80. Another way to test acute cutaneous reaction is to induce passive cutaneous anaphylactic reaction. Kum-Hwang-San significantly inhibited passive cutaneous anaphylactic reaction activated by anti-dinitrophenyl IgE on both topical application and intradermal injection. Kum-Hwang-San also inhibited histamine release from the rat peritoneal mast cells induced by anti-DNP IgE. This study provides evidence that Kum-Hwang-San will be beneficial in the treatment of acute cutaneous diseases.

Sopoongsan is an oriental medicinal composition including 12 medicinal herbs. Sopoongsan is known to have anti-inflammatory, antimicrobial, anti-allergic, and anticancer effects on human skin. The results of stability test showed that the creams containing Sopoongsan extracts were very stable at both accelerated temperature conditions and sun-light. And pH and viscosity of each cream did not change greatly for 56 days. From the particle size and rheological measurements, it can be known that when the amount of the Sopoongsan extracts increases, the mean value of particle size decreases, and the value of the complex modulus and loss angle increases, which means the Sopoongsan extracts help stabilize the emulsion. From the result of human patch test to assess the safety of cream containing Sopoongsan extracts, there was no stimulus of negative reaction on skin. In result of the user tests, it can be known that the higher concentration of the Sopoongsan was preferred by customers.
