The Antiapoptic Effects of Hominis Placenta Extract

  • Purpose. Free radicals are implicated in the pathophysiology of aging, ischemic injury and neurodegenerative disorders. To deform]no whether Hominis Placenta extract prevents $H_2O_2$-induced apoptosis, we have performed morphological and biochemical analyses for the detection of apoptotic phenomena in the pineal tumor cell line $PGT-{\beta}$ We have also peformed cytochemical and immunocytochemical analyses for the detection of changes in nitric oxide synthase (NOS) activity and estimated the expression . of apoptotic genes using reverse transcription-polymerase chain reaction (RT-PCR) Methods. $PGT-{\beta}\;cells$ were pretreated with Hominis Placenta extracts $(0,\;10^{-2}\;{\mu}g/ml)$ for 2 hours and then exposed to $H_2O_2\;(0,\;50\;{\mu}M)$ for 3 hours. Appearance of apoptotic characteristics were monitored using 4, 6-diamidino-2-phenylindole dihydrochloride (DAPI) staining assay, terminal deoxynucleotidyl transferase-mediated dUTP-digoxigenin nick end labeling (TUNEL) assay and flow cytometric analysis. NOS activity was measured by NADPH-diaphorase cytochemistry. Expression of inducible NOS (iNOS) and nuclear factor kappa B (NF k B) was assessed via immunocytochemistry. The expression of apoptotic genes was examined by RT-PCR. Results. After 3 flours of exposure to $H_2O_2$, it was shown that $PGT-{\beta}\;cells$ treated with $H_2O_2(50\;{\mu}M)$ exhibit classical apoptotic features and increases in NOS activity and caspase-3 expression. Treatment with Hominis Placenta extract resulted in a reduced occurrence of apoptotic features. DAPI staining, TUNEL and flow cytometric assays revealed decreases in the occurrence of nuclear fragmentation and in the sub-Gl fraction in the $PGT-{\beta}\;cells$ treated with Hominis Placenta extract. Cells treated with Hominis Placenta extract also showed lower activity of NADPH-diaphorase and immunoreactivities of both iNOS and NF k B than those of $H_2O_2$-treated cells which were not treated with Hominis Placenta extract. By RT-PCR, it was shown that the level of caspase-3 mRNA was derreased In the cells treated with Hominis Placenta . extract. Conclusions. This study shows that Hominis Placenta extract prevents $H_2O_2$-induced apoptosis in $PGT-{\beta}\;cells$; inhibitions of iNOS and caspnse-3 are possible mechanisms of the protection against apoptosis.

Modulation of Nrf2/ARE and Inflammatory Signaling Pathways by Hericium erinaceus Mycelia Extract

  • Hericium erinaceus is an edible mushroom used as a medicinal food in Asian countries. In this study, the chemopreventive effects of H. erinaceus mycelia hot water extract (HEW) were evaluated. HEW remarkably induced the luciferase activity of the antioxidant response element (ARE), located in the promoter region of phase 2 and antioxidant genes and regulated by nuclear factor E2-related factor 2 (Nrf2). The up-regulation of ARE activity by HEW corresponded with the induction of Nrf2 and the antioxidant enzyme, hemeoxygenase-1. The inhibition of cyclooxygenase-2 (COX-2) activity is a promising effective approach in cancer chemoprevention, and HEW prominently suppressed COX-2 protein expression in HepG2 cells. Furthermore, HEW showed anti-inflammatory activity by modulating inflammatory mediators such as nitric oxide (NO), inducible NO synthase, tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, and the transcription factor, nuclear factor-${\kappa}B$, in lipopolysaccharide-stimulated RAW 264.7 cells. These results suggest that H. erinaceus possessed anti-tumor and anti-inflammatory effects via the modulation of Nrf2/ARE and inflammatory signaling pathways, and may therefore have potential use as a natural chemopreventive agent.

감귤류 과피 추출물의 항염증 효과 (Anti-Inflammatory Effect of Peel Extracts from Citrus Fruits)

  • 본 연구에서는 감귤류 중 온주밀감, 레몬, 한라봉의 과피추출물(PE)에 대한 항염증 효과를 조사하였다. 감귤류 세품종 과피추출물(PE)에 대한 murine macrophage cell line Raw 264.7세포를 배양하여 세포독성, NO생성량, 세포내 염증관련 단백질 발현 양상을 분석하였다. 그 결과 CUP, CHP, CLP 처리구의 경우 2 mg/mL 이하의 농도에서는 세포생육을 저해하지 않았다. LPS (100 ng/mL)를 처리한 세포의 NO생성량을 LPS를 처리하지 않은 대조구와 비교했을 때 상당량이 증가했음을 알 수 있었고, 추출물을 농도별로 처리했을 때 농도가 증가할수록 NO생성량은 감소하였다. 각 처리구에서 각 1 mg/mL 농도에서는 CUP > CHP > CLP 순으로 NO 저해활성이 강하게 나타났다. Western blot의 결과상으로는 CUP와 CHP의 iNOS 단백질 저해활성이 비슷한 강도로 나타남을 확인하였다. COX-2 효소의 저해활성은 CUP > CHP > CLP의 순으로 나타났다. 따라서 CUP, CHP, CLP 처리구는 모두 항염증 작용을 가지고 있으며, 또한 CUP 처리구가 세 개의 추출물 중에서 가장 저해활성이 강하였다.

인진추출물이 lipopolysaccharide주입에 의한 흰쥐 간의 염증반응에 미치는 혈청학적 및 면역조직화학적 영향 (The Serum and Immunohistochemical Analysis on the Anti-infammatory Effect of Aqueous Extract from Artemisia capillaris in the Liver of Lipopolysaccharide-injected Rat)

  • 인진추출물의 LPS에 의한 흰쥐 간의 염증반응에 미치는 영향을 혈청학적, 조직병리학적, 면역조직화학적으로 분석하였다. 혈청 GOT, GTP, LDH 는 LPS에 의해 증가하며 이는 인진추출물의 전처리에 의해 저해되며 특히 150 mg/kg 투여군의 GOT와 50 mg/kg 투여군의 LDH가 대조군에 비해 유의성을 보였다. 조직병리학적으로 보아 인진추출물의 전처리는 LPS에 의해 나타나는 혼탁종창, 수포성변성, 염증성 세포침윤, Kupper세포반응 등을 저해하였다. 염증관련 인자에 대한 면역조직화학적 분석으로 보아 인진추출물의 전처리는 LPS주사군의 NF-kBp65, TNF-$\alpha$, COX-2 면역반응보다 약한 반응을, I-kB$\alpha$는 LPS주사군보다 강한 세포질내 면역반응을 보였다. 이로 보아 인진추출물은 LPS에 의한 간손상에 대해 유효한 기능을 가지며 이에 NF-kB를 비롯한 염증관련 단백질의 발현이 작용함을 알 수 있다.

알코올성 간 손상에 대한 상황버섯 배양균사체 추출물의 효능 (Effects of Mycelial Extract of Phellinus linteus on Ethanol-Induced Liver Injury in Rats)

  • 상황버섯 배양균사체 추출물 (MCPL)이 알코올성 간 손상에 미치는 영향을 살펴보았다. Sprague-Dawley계 흰 쥐에 40% 알코올 3 ml을 MCPL (5 mg 및 15 mg/Kg)과 함께 1일 1회 10일간 투여하였다. 간 기능의 표지가 되는 혈청내 AST와 ALT값이 에탄올에 의해 현저히 증가하였으나 MCPL 투여에 의해 저하되며 특히 ALT 값이 유의성 있게 낮아졌다. 병리조직학적으로 살펴보면 알코올에 의해 염증세포의 침윤, 쿠퍼세포 반응 및 국소적 염증이 유발되나 MCPL투여에 의해 그 정도가 다소 완화되었다. 간소엽내 글리코겐 분포도 알코올에 의해 감소하나 MCPL투여에 의해 중심정맥주변 부위의 분포가 일부 회복되었다. 염증관련 단백질에 대한 Western blot 및 면역조직화학적 반응을 보면 에탄올 투여에 의해 COX-2, iNOS 및 $TNF-{\alpha}$ 면역반응이 증가하나 MCPL투여에 의해 발현의 감소를 볼 수 있었다. 이상의 결과로 보아 MCPL은 알코올에 의한 간 손상에 대해 보호 기능을 가짐을 알 수 있다.

Ardisia arborescens 에탄올 추출물의 항산화 및 항염증 활성 (Antioxidative and Anti-inflammatory Activities of Ardisia arborescens Ethanol Extract)

  • 본 연구에서는 Ardisia arborescens 에탄올 추출물(AAEE)의 항산화능과 항염증 생리활성을 in vitro assay system 및 cell culture model system을 이용하여 분석하였다. 먼저 AAEE의 항산화능을 DPPH radical 소거능으로 분석한 결과 양성 대조군으로 사용한 ascorbic acid와 유사한 정도의 높은 활성을 보여 매우 강한 항산화능을 보유함을 확인하였다. 또한 RAW 264.7 세포주를 이용한 $H_2O_2$ 및 LPS의 유도에 의해 생성된 ROS에 대한 소거능을 분석한 결과에서도 농도의존적인 강한 소거능을 보였다. 뿐만 아니라 대표적인 항산화효소로 천연물에 의한 항산화능 활성에 의해 발현이 유도되는 HO-1, TrxR1 및 그 전사 인자인 Nrf2의 단백질 발현이 AAEE의 처리에 의해 농도의존적으로 증가됨을 보였으며 이러한 HO-1, TrxR1 및 Nrf2의 발현 변화는 상위신호전달계인 MAPKs에 의해 조절될 가능성을 보였다. 한편 AAEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 세포독성 없이 농도의존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이와 같은 AAEE의 NO 생성 억제 효과는 염증 상위신호전달계인 NF-${\kappa}B$ 및 AP-1의 조절을 통해 일어날 가능성을 보였다. 이러한 결과를 통해 AAEE의 높은 항산화능과 항염증 활성을 처음으로 확인하였으며 향후 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.

대식세포 내에서의 홍화자 추출물의 항염증 활성 (Anti-Inflammatory Activity of Carthamus tinctorious Seed Extracts in Raw 264.7 cells)

  • 본 연구에서는 홍화자 추출물이 항염증에 대한 실험연구가 이루어져 있지 않은 것에 착안하여 LPS에 의해 활성화된 대식세포로부터 유도되는 염증반응에 대한 억제효과를 조사하였다. 홍화자 추출물을 이용하여 피부 염증에 대하여 연구를 하였다. 산화질소와 cytokine의 생산은 면역세포의 대표적인 염증인자이다. 세포는 LPS 처리 후 한 시간 뒤에 홍화자 추출물을 처리를 하였다. 세포 독성이 나타나지 않는 농도인 5, 10, 25 및 50 ${\mu}g/ml$를 사용하였다. 홍화자의 에틸아세테이트 분획물은 NO, $PGE_2$, TNF-${\alpha}$, IL-$1{\beta}$, IL-6, iNOS, COX-2의 생성을 저해 시켰다. $PGE_2$는 50 ${\mu}g/ml$의 농도에서 60%에 가까운 저해율을 나타내었다. iNOS와 COX-2 역시 ${\mu}g/ml$의 농도에서 각각 54%, 65%가 저해가 되었다. 게다가 홍화자 에틸아세테이트 분획물은 염증성 사이토카인인 TNF-${\alpha}$, IL-$1{\beta}$, IL-6의 생성을 감소 시켰다. 이러한 결과로 홍화자 추출물은 염증 예방과 치료에 효과적임을 확인 할 수 있었다.

치자(梔子) 약침(藥鍼)이 백서(白鼠) 모델 족과 염좌(捻挫) 통증(痛症)에 미치는 영향(影響) (Effect of Frutus gardeniae herbal acupuncture on the rat model of ankle sprain pain)

  • Objective : Frutus gardeniae, seed of Gardenia jasminoides Ellis is one of the crude drugs used for the treatment of inflammatory condition in oriental medicine. Methodes : The present study aimed to examine the analgesic effect and anti-inflammatory effect of Frutus gardeniae extract (FGE) on a rat model of ankle sprain pain, and the relations between FGE-induced effect and endogenous nitric oxide (NO) and inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord. As a chronic pain model, ankle sprain pain model was used to test the effect of FCE injection applied to acupuncture point. After the induction of ankle sprain, rats subsequently showed a reduced stepping force of the affected limb for at least the next 4 days. The reduced stepping force of the limb was presumably due to a painful knee. FGE dissolved in normal saline was injected several acupoints. Results : After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 8 hours. FGE produced significant improvement of stepping force of the hindlimb affected by the ankle sprain lasting at least 4 hours. FGE produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. In addition, FGE injection showed inhibitory effect on the paw edema induced by ankle sprain. Both NO production and iNOS, COX-2 protein expression increased by ankle sprain were suppressed by FGE. FGE on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the ankle sprain than either FGE or EA did. The present study suggest that FGE produces a potent analgesic effect on the ankle sprain pain model of the rat and that FGE-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.

Protective effect of Korean Red Ginseng extract against Helicobacter pylori-induced gastric inflammation in Mongolian gerbils

  • Helicobacter pylori-induced gastric inflammation includes induction of inflammatory mediators interleukin (IL)-8 and inducible nitric oxide synthase (iNOS), which are mediated by oxidant-sensitive transcription factor NF-${\kappa}B$. High levels of lipid peroxide (LPO) and increased activity of myeloperoxidase (MPO), a biomarker of neutrophil infiltration, are observed in H. pylori-infected gastric mucosa. Panax ginseng Meyer, a Korean herb medicine, is widely used in Asian countries for its biological activities including anti-inflammatory efficacy. The present study aims to investigate whether Korean Red Ginseng extract (RGE) inhibits H. pylori-induced gastric inflammation in Mongolian gerbils. One wk after intragastric inoculation with H. pylori, Mongolian gerbils were fed with either the control diet or the diet containing RGE (200 mg RGE/gerbil) for 6 wk. The following were determined in gastric mucosa: the number of viable H. pylori in stomach; MPO activity; LPO level; mRNA and protein levels of keratinocyte chemoattractant factor (KC, a rodent IL-8 homolog), IL-$1{\beta}$, and iNOS; protein level of phospho-$I{\kappa}B{\alpha}$(which reflects the activation of NF-${\kappa}B$); and histology. As a result, RGE suppressed H. pylori-induced mRNA and protein levels of KC, IL-$1{\beta}$, and iNOS in gastric mucosa. RGE also inhibited H. pylori-induced phosphorylation of $I{\kappa}B{\alpha}$ and increases in LPO level and MPO activity of gastric mucosa. RGE did not affect viable H. pylori colonization in the stomach, but improved the histological grade of infiltration of poly-morphonuclear neutrophils, intestinal metaplasia, and hyperplasia. In conclusion, RGE inhibits H. pyloriinduced gastric inflammation by suppressing induction of inflammatory mediators (KC, IL-$1{\beta}$, iNOS), MPO activity, and LPO level in H. pylori-infected gastric mucosa.

In vitro and in vivo anti-inflammatory activities of Korean Red Ginseng-derived components

  • Background: Although Korean Red Ginseng (KRG) has been traditionally used for a long time, its anti-inflammatory role and underlying molecular and cellular mechanisms have been poorly understood. In this study, the anti-inflammatory roles of KRG-derived components, namely, water extract (KRG-WE), saponin fraction (KRG-SF), and nonsaponin fraction (KRG-NSF), were investigated. Methods: To check saponin levels in the test fractions, KRG-WE, KRG-NSF, and KRG-SF were analyzed using high-performance liquid chromatography. The anti-inflammatory roles and underlying cellular and molecular mechanisms of these components were investigated using a macrophage-like cell line (RAW264.7 cells) and an acute gastritis model in mice. Results: Of the tested fractions, KGR-SF (but not KRG-NSF and KRG-WE) markedly inhibited the viability of RAW264.7 cells, and splenocytes at more than 500 mg/mL significantly suppressed NO production at $100{\mu}g/mL$, diminished mRNA expression of inflammatory genes such as inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis factor-${\alpha}$, and interferon-${\beta}$ at $200{\mu}g/mL$, and completely blocked phagocytic uptake by RAW264.7 cells. All three fractions suppressed luciferase activity triggered by interferon regulatory factor 3 (IRF3), but not that triggered by activator protein-1 and nuclear factor-kappa B. Phospho-IRF3 and phospho-TBK1 were simultaneously decreased in KRG-SF. Interestingly, all these fractions, when orally administered, clearly ameliorated the symptoms of gastric ulcer in HCl/ethanol-induced gastritis mice. Conclusion: These results suggest that KRG-WE, KRG-NSF, and KRG-SF might have anti-inflammatory properties, mostly because of the suppression of the IRF3 pathway.
