A murine model immunized by systemic and mucosal delivery of plasmid DNA vaccine expressing glycoprotein B (pCIgB) of pseudorabies virus (PrV) was used to evaluate both the nature of the induced immunity and protection against a virulent virus. With regard to systemic delivery, the intramuscular (i.m.) immunization with pCIgB induced strong PrV-specific IgG responses in serum but was inefficient in generating a mucosal IgA response. Mucosal delivery through intranasal (i.n.) immunization of pCIgB induced both systemic and mucosal immunity at the distal mucosal site. However, the levels of systemic immunity induced by i.n. immunization were less than those induced by i.m. immunization. Moreover, i.n. genetic transfer of pCIgB appeared to induce Th2-biased immunity compared with systemic delivery, as judged by the ratio of PrV-specific IgG isotypes and Th1- and Th2-type cytokines produced by stimulated T cells. Moreover, the immunity induced by i.n. immunization did not provide effective protection against i.n. challenge of a virulent PrV strain, whereas i.m. immunization produced resistance to viral infection. Therefore, although i.n. immunization was a useful route for inducing mucosal immunity at the virus entry site, i.n. immunization did not provide effective protection against the lethal infection of PrV.

Root colonization of Arabidopsis thaliana with Pseudomonas chlororaphis O6 induces systemic tolerance against diverse pathogens, as well as drought and salt stresses. In this study, we demonstrated that 11 genes in the leaves were up-regulated, and 5 genes were down-regulated as the result of three- to five-days root colonization by P. chlororaphis O6. The identified priming genes were involved in cell signaling, transcription, protein synthesis, and degradation. In addition, expression of selected priming genes were induced in P. chlororaphis O6-colonized plants subjected to water withholding. Genes encoding defense proteins in signaling pathways regulated by jasmonic acid and ethylene, such as VSP1 and PDF1.2, were additional genes with enhanced expression in the P. chlororaphis O6-colonized plants. This study indicated that the expression of priming genes, as well as genes involved in jasmonic acid- and ethylene-regulated genes may play an important role in the systemic induction of both abiotic and biotic stress due to root colonization by P. chlororaphis O6.
The plant growth-promoting Pseudomonas strains, WR8-3 (Pseudomonas fluorescens), WR9-11 (Pseudomonas sp.) and WR9-16 (P.putida), which induced resistance systematically in watermelon to gummy stem rot were investigated on their induced systemic resistance(ISR)-related characteristics. The pyoverdine production was repressed in the standard succinate medium by increasing the concentration of $\textrm{FeCL}_3$. But the iron-binding ability on chrome azurol S agar media (CAS) was observed only in the strains, WR8-3 and WR9-16. When the two strains were mutated, the resulting iron-binding siderophore-negative mutants, WR8-3m and WR 9-16m, failed to promote the growth of watermelon and to induce resistance. The strains, WR8-3 and WR 9-16, slightly inhibited the growth of Didymella bryoniae at a low concentration of $\textrm{FeCL}_3$ on Kong's medium B, but not to exert control dffect. The strain WR9-11 showed antagonism in the concentration of $\textrm{FeCL}_3$ from 0 to $1,000\mu\textrm{M}$. When the crude lipoplysaccharide of each strain was treated in the rhizosphere of watermelon, mean lesion area was similar to that of the untreated control. The strains, WR9-11 and WR9-16 produced some level of hydrogen cyanide (HCN). Salicylic acid production was not detected in all of the strains.

In our previous study, we reported that a novel endophytic bacterium Bacillus oryzicola YC7007 has suppressed bacterial diseases of rice via induced systemic resistance and antibiotic production. This endophytic strain, B. oryzicola YC7007 was used as a biological control agent against bakanae disease of rice caused by Fusarium fujikuroi, and its mechanism of interaction with the pathogen and the rice was further elucidated. Root drenching with B. oryzicola YC7007 suspension reduced the disease severity of bakanae significantly when compared with the untreated controls. The treatments of B. oryzicola YC7007 suspension ($2.0{\times}10^7cfu/ml$) to the rice rhizosphere reduced bakanae severity by 46-78% in pots and nursery box tests containing autoclaved and non-autoclaved soils. Moreover, in the detached rice leaves bioassay, the development of necrotic lesion and mycelial expansion of F. fujikuroi were inhibited significantly by spraying the culture filtrate of B. oryzicola YC7007. Drenching of ethyl acetate extracts of the culture filtrate to the rhizosphere of rice seedlings also reduced the bakanae disease severity in the plant culture dish tests. With the root drenching of B. oryzicola YC7007 suspension, the accumulation of hydrogen peroxide was observed at an early stage of rice seedlings, and a hormonal defense was elicited with and without pathogen inoculation. Our results showed that the strain B. oryzicola YC7007 had a good biocontrol activity against the bakanae disease of rice by direct inhibition, and was also capable of inducing systemic resistance against the pathogen via primed induction of the jasmonic acid pathway.
Soil bacteria were screened for the ability to control cucumber anthracnose caused by Colletotrichum orbiculare through induced systemic resistance(ISR). Sixty-four bacterial strains having in vitro antifungal activity were used for selecting ISR-inducing strains in cucumber. Cucumber seeds(cv. Baeknokdadagi) were sown in potting mixtures incorporated with the soil bacteria, at a rate of ca. $10^8$ cells per gram of the mixture. Two week-old plants were then transplanted into the steam-sterilized soil. Three leaf-stage plants were inoculated with a conidial suspension($5{\times}10^5$ conidia/ml) of C. orbiculare. Diseased leaf area(%) and number of lesions per $cm^2$ leaf were evaluated on third leaves of the plants, $5{\sim}6$ days after inoculation. Among 64 strains tested, nine strains, GC-B19, GC-B35, GK-B18, MM-B22, PK-B14, RC-B41, RC-B64, RC-B65, and RC-B77 significantly(P=0.05) reduced anthracnose disease compared to the untreated control. In contrast, some bacterial strains promoted susceptibility of cucumber to the disease. From the repeated experiments using the nine bacterial strains, GC-B19, MM-B22, PK-B14, and RC-B65 significantly(P=0.05) reduced both diseased leaf area(%) and number of lesions per $cm^2$ leaf in at lease one experiment. These strains with control efficacy of $37{\sim}80%$ were determined to be effective ISR-inducing strains.

A phosphate-solubilizing bacterium, Enterobacter intermedium 60-2G, was examined for plant growth-promotion and induction of systemic resistance using a model system of cucumber and scab disease, caused by Cladosporium cucumerinum. Compared with a nonbacterized control, treatment of cucumber with E. intermedium significantly reduced the severity of scab disease after challenge-inoculation with C. cucumerinum. Treatment of cucumber with E. intermedium also enhanced cucumber growth. The 60-2G strain showed a strong antimicrobial activity against several plant pathogenic fungi including Fusarium soysporum and Magnaporthe grisea. These results suggest the E. intermedium 60-2G is a promising candidate as a biological control agent displaying multiple beneficial properties to promote plant health.

Biocontrol activity of five strains of selected rhizo-bacteria were tested in tomato against bacterial wilt caused by Ralstonia solanacearum. After root bacterization the plants were grown in a perlite-hydroponic system. Upon challenge inoculation with the pathogen, all of the rhizobacterial strains efficiently suppressed the bacterial wilt in tomato in various rates, at maximum by the strain, Bacillus vallismortis strain EXTN-1. While the percent of infected plants in the non-bacterized control plants were 95%, it was only 65% in plants pre-treated with EXTN-1. It was also demonstrated that the movement of R. solanacearum within the stem was significantly hampered when the plants were root bacterized. As EXTN-1 has no antagonistic properties against R. solanacearum, the bacterial wilt was probably suppressed by a mechanism other than antibiosis. Previously, the strain had been proven to produce an efficient elicitor for inducing systemic resistance in many crops. As the present study confirmed that EXTN-1 has the ability for reducing the pathogen spread in tomato, the strain could be effectively used as a potential biocontrol agent against bacterial wilt.
Plants have the ability to acquire an enhanced level of resistance to pathogen attack after being exposed to specific biotic stimuli. To obtain plant growth-promoting rhizobacteria inducing resistance against cucumber anthracnose by Colletotrichum orbiculare, more than 800 strains of rhizobacteria were screened in the greenhouse. Among these strains, Bacillus amyloliquefaciens solate EXTN-1 showed significant disease control efficacy on the plants. Induction of pathogenesis-related(PR-la) gene expression by EXTN-1 was assessed using tobacco plants transformed with PR-1a::$\beta$-glucuronidase(GUS) construct. GUS activities of tobacco treated with EXTN-1 and salicylic acid-treated transgenic tobacco were significantly higher than those of tobacco plants with other treatments. Gene expression analyses indicated that EXTN-1 induces the accumulation of defense-related genes of tobacco. The results showed that some defense genes are expressed by the treatment with EXTN-1 suggesting the similar resistance mechanism by salicylic acid.
Resistance to Tomato spotted wilt virus isolated from paprika (TSWV-Pap) was overcome at high temperatures ($30{\pm}2^{\circ}C$) in both accessions of Capsicum annuum S3669 (Hana Seed Company) and C. chinense PI15225 (AVRDC Vegetable Genetic Resources). S3669 and PI15225, which carrying the Tsw gene, were mechanically inoculated with TSWV-Pap, and then maintained in growth chambers at temperatures ranging from $15{\pm}2^{\circ}C$ to $30{\pm}2^{\circ}C$ (in $5^{\circ}C$ increments). Seven days post inoculation (dpi), a hypersensitivity reaction (HR) was induced in inoculated leaves of PI152225 and S3669 plants maintained at $25{\pm}2^{\circ}C$. Meanwhile, necrotic spots were formed in upper leaves of 33% of PI15225 plants maintained at $30{\pm}2^{\circ}C$, while systemic mottle symptoms developed in 50% of S3669 plants inoculated. By 15 dpi, 25% of S3669 plants had recovered from systemic mottling induced at $30{\pm}2^{\circ}C$. These results demonstrated that resistance to TSWV-Pap can be overcome at higher temperatures in both C. chinense and C. annuum. This is the first study reporting the determination of temperatures at which TSWV resistance is overcome in a C. annuum genetic resource expressing the Tsw gene. Our results indicated that TSWV resistance shown from pepper plants possess the Tsw gene could be overcome at high temperature. Thus, breeders should conduct evaluation of TSWV resistance in pepper cultivars at higher temperature than $30^{\circ}C$ (constant temperature).
