Thermophysiological Responses and Subjective Sensations when Wearing Clothing with Quickly Water-Absorbent and Dry Properties Under Exercise-Induced Heat Strain (운동에 의한 열 스트레스하에서 흡한속건성 소재 운동복 착용시의 온열생리적 반응 및 주관적 감각)

  • The purpose of this study was to compare the thermophysiological responses and subjective sensations of clothing materials with different water transfer property investigated in exercising and resting subjects at an ambient temperature of $20^{\circ}C$ and a relative humidity of 40%. Two kinds of clothing ensemble were tested: 100% cotton with highly water-absorbent but slowly dry properties(C) and 100% polyester with quickly water-absorbent and dry properties by four capillary channels(QADP). Seven apparently healthy male participants each undertook two series of experiments comprised 10-min of rest, 20-min of exercise with 70% of $VO_{2max}$ on a treadmill and 20-min of recovery. Mean skin temperature was significantly lower in QADP than in C during exercise and recovery. Clothing microclimate temperature was significantly lower in QADP during exercise and clothing surface temperature was also lower in QADP especially during recovery. Also, clothing surface humidity was significantly higher in QADP after the later half of exercise. The concentration of blood lactic acid tended to decrease to a lower level at recovery 3 minutes when wearing QADP rather than C clothing ensemble. Metabolic energy was marginally significantly less during the second half of exercise in QADP. Body mass loss tended to be greater in C than in QADP. The participants had better scores in thermal sensation, comfortable sensation and wetness in QADP during exercise and recovery. These results show that functional materials with quickly water-absorbent and dry properties can alleviate heat strain and induce more comfortable clothing microclimates and subjective sensations in the exercise-induced hyperthermia.

  • Mechanical stimulation is known to play a vital role on the differentiation of mesenchymal stem cells (MSCs) to pre-osteoblasts. In this research, we developed a tensile cell stimulator, composed of a DC motor-driven actuator and LVDT sensor for measuring linear displacement, to study the effects of tensile stress on osteogenic differentiation of MSCs. First, we demonstrated the reliability of this device by showing the uniform strain field in the silicon substrate. Secondly, we investigated the effects of tensile stretching on osteogenic differentiation. We imposed a pre-set cyclic strain at a fixed frequency on cell monolayer cultured on a flexible silicon substrate while varying its amplitude and duration. 60 min of resting period was allowed between 30 min of cyclic stretching and this cycle is repeated up to 7 days. Under the combined stimulation with osteogenic media and mechanical stretching, the osteogenic markers such as alkaline phosphatase (ALP), osterix, and osteopontin began to get expressed as early as 4 days of stimulation, which is much shorter than what is typically required for osteogenic media induced differentiation. Moreover, different markers were induced at different magnitudes of the applied strains. Lastly, for the case of ALP, we observed the antagonistic effects of osteogenic media when combined with mechanical stretching.

Experimental Study on the Hygrothermal Ageing Effect to the Strength of CFRP Materials for Marine Leisure Boat (열수노화 조건에서 레저선박용 탄소섬유강화플라스틱의 강도변화에 관한 실험적 연구)

  • This paper deals with the experimental study on the hygrothermal ageing effect to the strength of Carbon Fiber Reinforced Plastics (CFRP) materials for marine leisure boat manufactured by vacuum assisted resin infusion method. The experiments performed consist of tensile, flexural and shear tests according to American Society for Testing and Materials (ASTM) and Korean Industrial Standards (KS) test methods. Test coupons are varied from uni-directional(UD, $0^{\circ}$, $90^{\circ}$), Bi-Directional (BD), and Double-Bias (DB) carbon fiber fabrics in conjunction with epoxy resin. The results of tensile test show that tensile strength reduces significantly while not the same degree of reduction is observed for elasticity modulus with respect to the existence of hygrothermal ageing effect. This implies that the tensile strain induced from external load holds steady values but ultimate strength values change widely under hygrothermal ageing effect. In case of the flexural test, $0^{\circ}$ UD shows more strength reduction than $90^{\circ}$ UD while BD has reduced values in both flexural strength and elasticity modulus under hygrothermal ageing effect. It is learned that the bending strain induced from external load and ultimate strength values are reduced with respect to hygrothermal ageing effect. Shear test performed only on DB materials, and the results show marginal reduction in ultimate strength and moderate reduction in elasticity modulus. This means that the shear strain varies more than ultimate shear strength with respect to hygrothermal ageing effect. The experiment conducted in this paper clearly demonstrates the differences in material properties of the CFRP for the consideration of hygrothermal ageing effect. Findings obtained from this experimental study can serve as a fundamental input data for the realistic structural responses of marine leisure boat built in CFRP materials.

Deletion of xylR Gene Enhances Expression of Xylose Isomerase in Streptomyces lividans TK24

  • Glucose (xylose) isomerases from Streptomyces sp. have been used for the production of high fructose corn syrup for industrial purposes. An 11-kb DNA fragment containing the xyl gene cluster was isolated from Streptomyces lividans TK24 and its nucleotide sequences were analyzed. It was found that the xyl gene cluster contained a putative transcriptional repressor (xylR), xylulokinase (xylB), and xylose isomerase (xylA) genes. The transcriptional directions of the xylB and xylA genes were divergent, which is consistent to those found in other streptomycetes. A gene encoding XylR was located downstream of the xylB gene in the same direction, and its mutant strain produced xylose isomerase regardless of xylose in the media. The enzyme expression level in the mutant was 4.6 times higher than that in the parent strain under xylose-induced condition. Even in the absence of xylose, the mutant strain produce over 60% of enzyme compared with the xylose-induced condition. Gel mobility shift assay showed that XylR was able to bind to the putative xyl promoter, and its binding was inhibited by the addition of xylose in vitro. This result suggested that XylR acts as a repressor in the S. lividans xylose operon.

Fluorescent Pseudomonas Induced Systemic Resistance to Powdery Mildew in Mulberry (Morus spp.)

  • Native fluorescent pseudomonas bacteria were isolated from rhizosphere soil of mulberry and were evaluated against powdery mildew. In vitro conidial germination study showed significant (P<0.05) variation in conidial germination by bacterial strains Pf1 and Pf3. Mildew incidence was significantly varied due to treatment with various pseudomonas strains in vivo. Significantly (P<0.05) less mildew incidence was in plants treated with the bacterial strain Pf1 (9.11%) followed by Pf3 (13.48%) controlling 69.40% and 54.75% respectively compared with untreated control. Similarly, mildew severity was least (8.51%) in plants treated with strain Pf1 followed by Pf5 (9.23%) and Pf3 (9.72%) controlling the severity by 84.51%, 77.01% and 71.96% respectively compared with control. The bacterial strains significantly influenced biochemical constituents such as chlorophyll, protein and soluble sugar content of the mulberry leaf. Similarly, bacterial strains significantly increased the activity of the peroxidase (PO) and Polyphenol oxydase (PPO) activity from $7^{th}$ day up to the $28^{th}$ day after treatment. The strain Pf1, Pf3 and Pf5 exhibited a marked enhancement in the peroxidase at different periods of infection. Significant (P<0.01) negative correlation was found between powdery mildew severity with phenol content ($R^2=0.67$) as well as peroxidase ($R^2=0.92$) and polyphenol oxidase ($R^2=0.72$) activity thus confirms induction of systemic resistance in mulberry by pseudomonas bacteria. The study shows scope for exploration of rhizosphere fluorescent pseudomonas bacteria for induction of systemic resistance in mulberry to contain powdery mildew disease effectively.

Induced Tolerance to Salinity Stress by Halotolerant Bacteria Bacillus aryabhattai H19-1 and B. mesonae H20-5 in Tomato Plants

  • Salinity is one of the major abiotic stresses that cause reduction of plant growth and crop productivity. It has been reported that plant growth-promoting bacteria (PGPB) could confer abiotic stress tolerance to plants. In a previous study, we screened bacterial strains capable of enhancing plant health under abiotic stresses and identified these strains based on 16s rRNA sequencing analysis. In this study, we investigated the effects of two selected strains, Bacillus aryabhattai H19-1 and B. mesonae H20-5, on responses of tomato plants against salinity stress. As a result, they alleviated decrease in plant growth and chlorophyll content; only strain H19-1 increased carotenoid content compared to that in untreated plants under salinity stress. Strains H19-1 and H20-5 significantly decreased electrolyte leakage, whereas they increased $Ca^{2+}$ content compared to that in the untreated control. Our results also indicated that H20-5-treated plants accumulated significantly higher levels of proline, abscisic acid (ABA), and antioxidant enzyme activities compared to untreated and H19-1-treated plants during salinity stress. Moreover, strain H20-5 upregulated 9-cisepoxycarotenoid dioxygenase 1 (NCED1) and abscisic acid-response element-binding proteins 1 (AREB1) genes, otherwise strain H19-1 downregulated AREB1 in tomato plants after the salinity challenge. These findings demonstrated that strains H19-1 and H20-5 induced ABA-independent and -dependent salinity tolerance, respectively, in tomato plants, therefore these strains can be used as effective bio-fertilizers for sustainable agriculture.

  • We investigated the microstructure, crystal structure, dielectric, and elecromechanical strain properties of lead-free BaTiO3 (BT)-modified (Bi1/2Na1/2)TiO3-SrTiO3 (BNT-ST) piezoelectric ceramics. Samples were prepared by a conventional ceramic processing route. Temperature dependent dielectric properties confirmed that a phase transition from a nonergodic relaxor to an ergodic relaxor was induced when the BT concentration reached 1.5 mol%, interestingly, where the average grain size reached a maximum value of 4.5 ㎛. At the same time, enhanced electromechanical strain (Smax/Emax = 600 pm/V) was obtained. It is suggested that the induced ferroelectric-relaxor phase transition by the BT modification is responsible for the enhancement of electromechanical strain in 1.5 mol% BT-modified BNT-ST ceramics.

Lactobacillus sakei S1 Improves Colitis Induced by 2,4,6-Trinitrobenzene Sulfonic Acid by the Inhibition of NF-κB Signaling in Mice

  • Lactobacillus sakei S1 strongly inhibits the expression of interleukin (IL)-6 and IL-1β in lipopolysaccharide-induced peritoneal macrophages by a mechanism for which lactic acid bacteria from kimchi that inhibit tumor necrosis factor-alpha (TNF-α) were isolated. Therefore, we further evaluated the protective effect of this strain on the colitis mouse model induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS). TNBS significantly elevated myeloperoxidase (MPO) expression, macroscopic scores, and colon shortening. Oral L. sakei S1 administration resulted in reduction of TNBS-induced loss in body weight, colon shortening, MPO activity, expression of cyclooxygenase (COX)-2, inducible nitric oxide synthase (iNOS) and nuclear factor-kappa B (NF-κB). L. sakei S1 inhibited the expression of inflammatory cytokines IL-1β, IL-6 and TNF-α, induced by TNBS, but enhanced IL-10 expression. L. sakei S1 showed resistance to artificial digestive juices and adherence to intestinal epithelial Caco-2 cells. Thus, L. sakei S1 may inhibit the NF-κB pathway and be used in functional food to treat colitis.

Characterization of Prophages in Leuconostoc Derived from Kimchi and Genomic Analysis of the Induced Prophage in Leuconostoc lactis

  • Leuconostoc has been used as a principal starter in natural kimchi fermentation, but limited research has been conducted on its phages. In this study, prophage distribution and characterization in kimchi-derived Leuconostoc strains were investigated, and phage induction was performed. Except for one strain, 16 Leuconostoc strains had at least one prophage region with questionable and incomplete regions, which comprised 0.5-6.0% of the bacterial genome. Based on major capsid protein analysis, ten intact prophages and an induced incomplete prophage of Leu. lactis CBA3626 belonged to the Siphoviridae family and were similar to Lc-Nu-like, sha1-like, phiMH1-like, and TPA_asm groups. Bacterial immunology genes, such as superinfection exclusion proteins and methylase, were found on several prophages. One prophage of Leu. lactis CBA3626 was induced using mitomycin C and was confirmed as belonging to the Siphoviridae family. Homology of the induced prophage with 21 reported prophages was not high (< 4%), and 47% identity was confirmed only with TPA_asm from Siphoviridae sp. isolate ct3pk4. Therefore, it is suggested that Leuconostoc from kimchi had diverse prophages with less than 6% genome proportion and some immunological genes. Interestingly, the induced prophage was very different from the reported prophages of other Leuconostoc species.
