The antimutagenic and antigenotoxic effects of ethanol, methanol, water and non-heating ethanol extract of Ligularia fischeri were investigated using Ames test and micronucleus test. Four solvent extracts by themseleves did not induce mutagenesis. The four extract of 200㎍/plate showed approximately 84.7%, 77.1%, 72.5% and 71% inhibitory effect on the mutagenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) and 67.9%, 66.8%, 64.6% and 56% inhibition on the mutagenesis by 4-nitroquinoline-1-oxide(4NQO) against TA100 strain, whereas 70.2%, 60.9%, 61.9% and 52.8% inhibitions were observed on the mutagenesis induced by 3-amino-1,4-dimethyl-5H-pyrido[4,3-b]indol(Trp-P-1) in the presence of 200㎍/plate. TA100 strain was more sensitive than TA98 strain by four kinds of extracts on antimutagenesis. The effects of Ligularia fischeri extracts on the frequencies of micronucleated poly chromatic erythrocytes(MNPECs) induced by MNNG were investigated in the bone marrow. Ten, 20, 40 and 80mg g/kg of each extract were administered to animals immediately after injection of MNNG and the exposure time was 36 hours. Inhibitory effects of Ligularia fischeri ethanol extracts were 12%, 35.3%, 58.8%, and 57%, in the presence of 20, 40, 60 and 80mg/kg, respectively whereas methanol extracts showed 15.5%, 32.7%, 50.8%, and 57.9% inhibitory effects, respectively. Both extracts showed enhanced antimutagenic and antigenotoxic effects. These results showed a good correlation between antimutagenic effects in in vitro and in in vitro assay.
The Saccharomyces0 cerevisiae KNU5377 strain, which was isolated from spoilage in nature, has the ability to convert biomass to alcohol at high temperatures and it can resist against various stresses [18, 19]. In order to understand the defense mechanisms of the KNU5377 strain under menadione (MD) as oxidative stress, we used several techniques for study: peptide mass fingerprinting (PMF) by matrix-assisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry (MS) followed by two-dimensional (2D) gel electrophoresis, liquid chromatography electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS), and surface-enhanced laser desorption ionization-time of flight (SELDI-TOF) technology. Among the 35 proteins identified by MALDI-TOF MS, 19 proteins including Sod1p, Sod2p, Tsa1p, and Ahp1p were induced under stress condition, while 16 proteins were augmented under normal condition. In particular, five proteins, Sod1p, Sod2p, Ahp1p, Rib3p, Yaf9p, and Mnt1p, were induced in only stressed cells. By LC-ESI-MS/MS analysis, 37 proteins were identified in normal cells and 49 proteins were confirmed in the stressed cells. Among the identified proteins, 32 proteins were found in both cells. Five proteins including Yel047cp and Met6p were only upregulated in the normal cells, whereas 17 proteins including Abp1P and Sam1p were elevated in the stressed cells. It was interesting that highly hypothetical proteins such as Ynl281wp, Ygr279cp, Ypl273wp, Ykl133cp, and Ykr074wp were only expressed in the stressed cells. SELDI-TOF analysis using the SAX2 and WCX2 chips showed that highly multiple-specific protein patterns were reproducibly detected in ranges from 2.9 to 27.0 kDa both under normal and stress conditions. Therefore, induction of antioxidant proteins, hypothetical proteins, and low molecular weight proteins were revealed by different proteomic techniques. These results suggest that comparative analyses using proteomics might contribute to elucidate the defense mechanisms of KNU5377 under MD stress.
A BOD sensor was prepared with S. marcescens LSY4 and was applied for measurement of BOD values of a solution containing the standard organic pollutants. The sensor sensitivity was nearly independent of the culture time in the range of 9-16 hours. It was also affected little by the cell mass in the range of 0.22-0.75 mg $cm^{-2}$. A cyclic change in the solution pH in the range of 4-9 was accompanied by a reversible variation in the sensor sensitivity. However, the reversibility was lost when the solution pH became more acidic or more basic. Heavy metal ions lowered the sensor sensitivity, which took place more precipitously in the presence of $Cu^{2+}$ and $Ag^+$ rather than in the presence of $Zn^{2+}$ and $Cd^{2+}$. The reduction of the sensor sensitivity was significantly attenuated by loading heavy metal ion tolerance induced strain. The $Cu^{2+}$tolerance induced strain was more efficient for the attenuation than $Zn^{2+}$ and $Cd^{2+}$ tolerance induced strain.

For the advanced high strength steels (AHSS), high-manganese TWIP (twinning induced plasticity) steels exhibit high tensile strength (800-1000 MPa) and high elongation (50-60%). However, the TWIP steels need to be understood of delayed fracture following the cup drawing test. Among the factors to cause delayed fracture, i.e, martensite transformation, hydrogen embrittlement and residual stress, the effects of martensite transformation (${\gamma}{\rightarrow}{\varepsilon}$ or ${\gamma}{\rightarrow}{\alpha}^{\prime}$) were investigated on the delayed fracture phenomenon. Microstructural phase analysis was conducted for cold rolled (20, 60, 80% reduction ratio) steels and tensile deformed (20, 40, 60% strain) steels. For the Al-added TWIP steels, no martensite phase was found in the cold rolled and tensile deformed specimen. But, the TWIP steels with no Al addition indicated the martensite transformation. The cup drawing specimens showed the martensite transformation irrespective of the Al-addition to the TWIP steel. However, the TWIP steel with no Al exhibited the larger amount of martensite than the case of the TWIP steel with Al addition. For the reason, it was possible to conclude that the Al addition suppressed the martensite transformation in TWIP steels, therefore preventing the delayed fracture effectively. However, it was interesting to note that the mechanism of delayed fracture should be incorporated with hydrogen embrittlement and/or residual stress as well as the martensite transformation.
Probiotics are microorganisms that can benefit host health when ingested in a live state, and lactic acid bacteria are the most common type. Among fungi, Saccharomyces boulardii (SB) is the only strain known to have a probiotic function with beneficial effects on colitis; however, information on other probiotic yeast strains is limited. Therefore, this study aimed to discover yeast strains expressing intestinal anti-inflammatory activities by exhibiting probiotic properties in dextran sodium sulfate (DSS)-induced colitis mice model. Nuruk (Korean traditional fermentation starter) containing various microbial strains was used as a source for yeast strains, and S. cerevisiae 28-7 (SC28-7) strain was selected with in vitro and in vivo characteristics to enable survival in the intestines. After 14 days of pretreatment with the yeast strains, DSS was co-administered for six days to induce colitis in mice. The results revealed that the disease activity index score was lowered by SC28-7 treatment compared to the DSS group, and the colon length and weight/length ratio were recovered in a pattern similar to that of the normal group. SC28-7 administration significantly reduced the secretion of pro-inflammatory cytokines in the serum and modified the mRNA expression of inflammatory cytokines (interleukin-1β, transforming growth factor-β, and interferon-γ) and proteins involved in gut barrier functions (mucin 2, mucin 3, zonula occludens-1, and occludin) in colon tissues. These results indicate that SC28-7 attenuates DSS-induced colon damage and inflammation, supporting its future use as a probiotic yeast for treating and preventing intestinal inflammatory diseases such as inflammatory bowel disease.

This paper introduces a newly developed high speed material testing apparatus for tensile tests at the strain rate up to 500/sec. The tensile properties of sheet metals are indispensable for the accurate crashworthiness analysis of auto-bodies since the local strain rate reaches to 500/sec in the car crash. An appropriate experimental method has to be developed to acquire the tensile properties at the intermediate strain rate ranged from 0.003/sec to 200/sec. Tensile tests of various different steel sheets for an auto-body were perform ed to obtain the dynamic properties with respect to the strain rate. The dimensions of specimens that can provide the reasonable results were determined by the finite element analysis. A special jig fixture of a load cell is designed to reduce the load ringing phenomenon induced by unstable stress propagation at the high strain rate. Stress-strain curves were acquired for each steel sheet from the dynamic tensile test and utilized to obtain the relationship of the stress to the strain rate.
A numerical model for the thermal response analysis of concrete structures is suggested. The model includes the stress-strain relationship, constitutive relationship, and multiaxial failure criteria at elevated temperature conditions. Modified Saenz's model was used to describe the stress-strain relationship at high temperatures. Concrete subjected to elevated temperatures undergoes rapid strain increase and dimensional instability. In order to explain those changes in mechanical properties, a constitutive model of concrete subjected to elevated temperature is proposed. The model consists of four strain components; free thermal creep strain, stress-induced (mechanical) strain, thermal creep strain, and transient strain due to moisture effects. The failure model employs modified Drucker-Prager model in order to describe the temperature dependent multiaxial failure criteria. Some numerical analyses are performed and compared with the experimental results to verify the proposed model. According to the comparison, the suggested material model gives reliable analytical results.
The objective of these experiments was to develop an attenuated thermostable Newcastle disease virus(NDV), CBP-1 strain isolated from infected pheasants. Safety, pathogenicity and protective effects against velogenic NDV were also investigated to evaluate if the attenuated NDV, CBP-1 strain could be a candidate for a new NDV vaccine strain. CBP-1 strain was passaged up to the 173 times by nine days old embryonated eggs and chicken embryo fibroblast(CEF) cell cultures. Its sensitivitly to lipid solvents and low pH, thermostability, mean death time(MDT), intracerebral pathogenicity index(ICPI) of one day old chicks and intravenous pathogenicity index(IVPI) of four weeks old chicks were examined. Safety, boosting and protective effects were tested by chicks mortality. CBP-1 NDV strain had significant thermostability at 56$\^{C}$ for 30 minutes. by hemagglutinin activity and egg infectivity test, but was not resistant to lipid solvent. It showed possibility to use as a feed or water vaccine because of the resistance to low pH. MDT, ICPI and IVPI of CBP-1 were attenuated from 51.5, 1.96, 2.60 to 112.4, 1.12, 1.45. These results implied that the 173rd passages in embryonated egg and CEF cell cultures induced a substantial attenuation of the pathogenicity of the parent virus, changing the virulence from velogenic to intermediate between mesogenic and lentogenic. After vaccination with CBP-1 at one day old by drinking water mortality was 17.5%. However, spray vaccination with B1 at one day old, CBP-1 at two weeks ild and challenge with velogenic Kyojeongwon strain at four weeks old showed 93.5% survival rate. Mortality of chicks, vaccination with 173rd passaged CBP-1 strain at one day old, two weeks old and challenge with Kyokeongwon strain at four weeks old, was 20.0%. The results of these studies indicated that partial attenuated CBP-1 strain tended to be a low safety for ND of broiler chicks and would need to be more successive attenuation.

To analyze the distribution of earthquake-induced strain data in rock masses, statistical analysis was performed on four-directional strain data obtained from a ground movement monitoring system installed in Korea. Strain data related to the 2011 Tohoku-oki earthquake and two aftershocks of >M7.0 in 2011 were used in x-MR control chart analysis, a type of univariate statistical analysis that can detect an abnormal distribution. The analysis revealed different dispersion times for each measurement orientation. In a more comprehensive analysis, the strain data were re-evaluated using multivariate statistical analysis (MSA) considering correlations among the various data from the different measurement orientations. $T_2$ and Q-statistics, based on principal component analysis, were used to analyze the time-series strain data in real-time. The procedures were performed with 99.9%, 99.0%, and 95.0% control limits. It is possible to use the MSA data to successfully detect an abnormal distribution caused by earthquakes because the dispersion time using the 99.9% control limit is concurrent with or earlier than that from the x-MR analysis. In addition, the dispersion using the 99.0% and 95.0% control limits detected an abnormal distribution in advance. This finding indicates the potential use of MSA for recognizing abnormal distributions of strain data.
In the course of screening for apoptotic substances that induce apoptosis in human leukemia U937 cells, a fungal strain, F000487, which exhibits potent inducible activity, was selected. The active compound was purified from an ethyl acetate extract of the microorganism by Sep-pak $C_{18}$ column chromatography and HPLC, and was identified as atromentin by spectroscopic methods. This compound induced caspase-3 processing in human leukemia U937 cells. The caspase-3 and poly (ADP-ribose) polymerase (PARP) were induced by atromentin in a dose-dependent manner. Furthermore, DNA fragmentation was also induced by this compound in a dose-dependent manner. These results show that atromentin potently induces apoptosis in U937 cells and that atromentin-induced apoptosis is related to the selective activation of caspases.
