Protective Effects of $\beta$-Immunan Isolated from the Mycelium of Ganoderma lucidum IY009 against Cisplatin-induced Nephrotoxicity (영지버섯 균사체(Ganoderma lucidum IY009)로부터 추출한 $\beta$-Immunan의 시스플라틴 유발 신독성 보호효과)

  • $\beta$-Immunan was proteoglycan obtained from mycelium of Ganoderma lucidum IY009. In this study, the protective effects of $\beta$-Immunan, against the CDDP induced in vitro cytotoxicity and in vivo renal toxicity, was measured. Concentration dependent cytotoxicities of CDDP in normal kidney cells (Vero, TCMK-l) were reduced by $\beta$-Immunan treatment. Increased renal toxicity factors, such as elevation of blood urea nitrogen (BUN) and serum creatinine, reduction of kidney weight and malonidialdehyde (MDA), by intraperitoneal administration of CDDP in rats was improved. These results indicated that $\beta$-Immunan have a protective effects against the CDDP induced renal toxicity, however, it needed to confirm the detailed mechanism for therapeutic effects.

Studies on the effect of Kamikuibitang on the Gastric Ulcer in Rats (가미귀비탕(加味歸脾湯)이 흰쥐의 위궤양(胃潰瘍)에 미치는 영향(影響))

  • This study was aimed to evaluate the anti-pain effect of Kamikuibitang in acetic acid method and the anti-ulceration effect of Kamikuibitang in indomethacin, aspirin and immobilization stress method in rats. The results were follows; 1. The anti-pain effects of Kuibitang and Kamikuibitang were decreased compared with those of control group. 2. In indomethacin and aspirin method, the anti-ulcerative effects of experimental groups were shown compared with those of control group. 3. In immobilization stress method, the anti-ulcerative effect of experimental groups was significantly shown compared with that of control group. 4. The serum gastrin levels of Kuibitang groups showed very significant decrease in indomethacin-induced and immobilization stress-induced ulcers. The serum gastrin levels of Kamikuibitang groups showed very significant decrease in indomethacin-induced, aspirin- induced and immobilization stress-induced ulcers. 5. The serum $V_{B12}$ levels of Kuibitang groups showed very significant increase in both indomethacin-induced and immobilization stress-induced ulcers. The serum $V_{B12}$ levels of Kamikuibitang groups showed significant increase in aspirin-induced and immobilization stress-induced ulcers whereas very significant increase in indomethacin-induced ulcer. According to the above results, it was concluded that Kamikuibitang had very significant anti-ulceration effect as well as anti-pain effect on gastric ulcer in rats.

Scavenging Effects of Hydroxycinnamic Acids on Paraquat Induced Pulmonary Toxicity (III) (Paraquat 유도 페독성에 대한 Hydroxycinnamic Acid계 화합물의 독성 경감 효과 (III))

  • The scavenging effects of two hyaroxycinnamic acids such as caffeic acid and chlorogenic acid on paraquat induced pulmonary toxicity were investigated. The results are summerized as follows: 1. In the 5-lipoxygenase assay, caffeic acid and chlorogenic acid inhibited the enzyme activities whose inhibition concentration (IC$\_$50/) were 4.1 and 9.6 ${\mu}$M respectively. 2. To evaluate the antiinflammatory effects on mediator related to the mechanism of inflammation, ADP-induced platelet aggregation assay and histamine degranulation assay were used. Caffeic acid and chlorogenic acid inhibited on ADP-induced platelet aggregation and histamine release at a concentration dependent manners. 3. Arachidonic acid-induced ear edema were inhibited by administration of caffeic acid and chlorogenic acid. 4. Cytologicad analysis of branchoalveolar lavage fluid (BALF) which was the useful tool for detection of an inflammatory response in the lungs of animals intoxicated with chemicals were used. Alveolar macrophages and neutrophils in BALF, as well as the protein content and the LDH activity in BALF supernatant increased by intoxication of paraquat, but decreased by administration of caffeic acid and chlorogenic acid. Therefore, two hydroxyeinnamic acids tested were the useful candidates for scavenger and antiinflammatory agents on paraquat induced pulmonary toxicity.

Effects of Small Molecular Antioxidants on Cerulein-induced Acute Pancreatitis in Rat

  • It has been suggested that oxygen free radicals are involved in the initiation process of acute pancreatitis, although its pathogenesis is not clear. This study evaluates the roles of oxygen radicals and the effects of small molecular antioxidants (rebamipide, N-acetyl-cysteine, allopurinol, ${\beta}-carotene)$ on the development of cerulein-induced acute pancreatitis. Acute edematous pancreatitis was induced by the intravenous infusion of cerulein at supramaximal dose of 10 ${\mu}g/kg/hour$ for 3.5 hours. The effects of antioxidants, rebamipide (100 mg/kg, i.p.), N-acetyl-cysteine (200 mg/kg, i.v.), allopurinol (20 mg/kg/hour), ${\beta}-carotene$ (50 mg/kg, i.p.), were examined. Cerulein administration resulted in a significant increase in serum amylase activity and pancreatic malondialdehyde (MDA), but not glutathione peroxidase (GSHpx). The glutathione (GSH) content in pancreatic tissue decreased dramatically. Pretreatment of N-acetyl-cysteine significantly decreased the cerulein-induced hyperamylasemia and maintained GSH content in pancreas, but MDA was slightly decreased. In addition, N-acetyl-cysteine ameliorated histological damage. Allopurinol and ${\beta}-carotene$ attenuated cerulein-induced hyperamylasemia, but histologically there was no difference from control. These results indicate that oxygen free radicals play an important role in the initiation of experimental acute pancreatitis. N-acetyl-cysteine is an effective antioxidant that ameliorates the cerulein-induced acute pancreatitis, and the possible therapeutic application of antioxidants against acute pancreatitis needs a further evaluation.

Association of Dexamethasone-induced Apoptosis and $G_l-Arrest$ of Human Leukemic CEM Cells with Polyamine Deficit

  • The effects of DFMO or/and putrescine on the dexamethasone-induced apoptosis of CEM cells were studied to investigate the role of polyamines in anti-leukemic glucocorticoid action. Dexamethasone- induced apoptosis was preceded by significant decreases of cellular polyamine contents and putrescine uptake activity. But DFMO produced decreases of putrescine and spermidine contents and marked increase of putrescine uptake activity, but did not induce apoptosis. However, dexamethasone and DFMO, respectively, induced $G_1-arrest$ in cell cycle and hypophosphorylation of pRb, resulting in the increase of $G_1$ to S ratio and decrease of CEM cell count. DFMO enhanced the dexamethasone-induced apoptosis and $G_1-arrest$. On the other hand, putrescine little affected the apoptotic and $G_1-arresting$ activities of dexamethasone, but almost suppress the effects of DFMO and also the DFMO-dependent enhancement of dexamethasone effects. These results suggested that the dexamethasone-induced apoptosis to be associated with pRb hypophosphorylation and $G_1-arrest$ in CEM cells might be ascribed to the concomitant decreases of cellular polyamine contents and putrescine uptake activity.

Effects of Yanggyuksanhwa-tang on Diabetic Rats Induced by Streptozoticin (양격산화탕(凉膈散火湯)이 실험적 당뇨에 미치는 영향)

  • This study evaluated effects of Yanggyuksanhwa-tang(YST) on diabetic rats. Diabetic condition in rats was induced by streptozotocin injection. Then control effect of YST was observed with changes of serum glucose, insulin, and triglyceride levels and hepatic glucokinse activity. YST treatment was resulted significant decrease of high serum glucose level of diabetic rats induced by streptozotocin at 5th day of YST treatment. YST treatment was resulted increase of low serum insulin level of diabetic rats induced by streptozotocin, but it was not significant statistically. YST treatment was resulted significant decrease of high serum triglyceride level of diabetic rats induced by streptozotocin at 3rd and 5th of YST treatment. YST treatment was resulted significant increase of low hepatic glucokinase activity of diabetic rats induced by streptozotocin at 5th day of YST treatment. These results suggest that YST has effects on serum glucose control against cerebral inchemic damage under diabetic condition.

Evodiamine Reduces Caffeine-Induced Sleep Disturbances and Excitation in Mice

  • Worldwide, caffeine is among the most commonly used stimulatory substances. Unfortunately, significant caffeine consumption is associated with several adverse effects, ranging from sleep disturbances (including insomnia) to cardiovascular problems. This study investigates whether treatment with the Evodia rutaecarpa aqueous extract (ERAE) from berries and its major molecular component, evodiamine, can reduce the adverse caffeine-induced sleep-related and excitation effects. We combined measurements from the pentobarbital-induced sleep test, the open field test, and the locomotor activity test in mice that had been dosed with caffeine. We found that ERAE and evodiamine administration reduced the degree of caffeine-induced sleep disruption during the sleep test. Additionally, we found that evodiamine significantly inhibits caffeine-induced excitation during the open field test, as well as decreasing hyperlocomotion in the locomotor activity test. Additional in vitro experiments showed that caffeine administration decreased the expression of ${\gamma}$-aminobutyric acid $(GABA)_A$ receptor subunits in the mouse hypothalamus. However, evodiamine treatment significantly reversed this expression reduction. Taken together, our results demonstrate that ERAE and its major compound, evodiamine, provide an excellent candidate for the treatment or prevention of caffeine-induced sleep disturbances and excitatory states, and that the mechanism of these beneficial effects acts, at least in part, through the $GABA_A$-ergic system.

Effects of Ethylacetate Fraction of Persimmon Leaves on Experimentally-induced Gastric Mucosal Damage and Gastric Ulcers in Rats

  • The protective effects of the ethylacetate fraction of persimmon leaves(PEF) against experimentally induced gastric mucosal damage and gastric ulcers were evaluated in ratss. In prophylatic study, 100 mg/kg ethylacetate fraction of persimmon leaves (PEFH) exhibited a total protection of 73.8% and 65.7% against HCl-ethanol and 0.2N NaOH-induced gastric mucosal membrane lesions, respectively, which was superior to cimetidine 50 mg/kg, a commonly used anti-ulcer drug. PEFH showed excellent anti-ulcer effects against pylorus ligation induced gastric ulcers, compared to the control group, however, 50 mg/kg ethylacetate fraction of persimmon leaves (PEFL) and PEFH did not affect ulcers induced by water immersion stress, and that is inferior to cimetidine 50 mg/kg. In conclusion, the results suggest that the ethylacetate fraction of persimmon leaves can be used both in prevention and treatment of experimentally induced gastric mucosal damage and ulcers.

The Ameliorating Effect of Hoveniae Semen Seu Fructus Extract against Ethanol-induced Psychomotor Alterations in Rats (흰쥐에서 지구자 추출물의 에탄올 유도 숙취 행동 개선 효능)

  • Ethanol consumption causes psychomotor alterations. Hovenia Semen seu Fructus (HS), widely distributed in Korea, China, and Japan, has been reported to have beneficial effects on acute alcohol-induced liver injury. The present study sought to assess the effects of HS extract on ethanol-induced psychomotor alterations in rats. Sprague-Dawley rats were orally (p.o.) given ethanol (4 g/kg) (ethanol group) to induce psychomotor alterations. A separate group (HS-treated groups), were treated with different dosages of HS (50, 100, and 200 mg/kg, p.o.), 30 minutes before ethanol treatment. The control group received only the vehicle (saline). Ethanol-induced psychomotor alterations were evaluated in the open-field, rota-rod, hanging wire, and cold swimming test. In addition, blood ethanol and acetaldehyde concentrations were also measured. Behavioral evaluations and blood analysis were carried out 0.5, 1, 2, 4, and 8 hours after ethanol administration. Pre-treatment of HS ameliorated ethanol-induced alterations in the open-field, rota-rod, and cold swimming test, significantly evident in 2 and 4 hours after ethanol treatment. These improvements coincided with decrease in blood ethanol and acetaldehyde concentration. Based on these results, the present study suggests that HS may have ameliorating effects against ethanol-induced psychomotor alterations.

Effect of Yukgunja-tang on Glutamate-induced Apoptosis in C6 Glial Cells (육군자탕(六君子湯)이 Glutamate에 의한 C6 신경교세포의 Apoptosis에 미치는 영향)

  • Objective : The water extract of Yukgunja-tang(YGJT) has been traditionally used in treatment of qi deficiency and phlegm in Oriental medicine. However, little is known about the mechanism by which YGJT protects neuronal cells from injury damages. Therefore, this study was designed to evaluate the protective effects of YGJT on C6 glial cells by glutamate-induced cell death. Methods : The present study describes glutamate, which is known as an excitatory neurotransmitter, related with oxidative damages, and YGJT, which shows protective effects against glutamate-induced C6 glial cell death. One of the main mediators of glutamate-induced cytotoxicity was known on the generation of reactive oxygen species(ROS) via activation of NADPH oxidase (NOX). The protective effects of antioxidant(NAC) and NOX inhibitor(apocynin) on the glutamate-induced C6 glial cells were determined by a MTT reduction assay. Result : YGJT inhibited glutamate-induced ROS generation via inhibition of NOX expression on glutamate-stimulated C6 glial cells. Furthermore, YGJT attenuated glutamate-induced caspase activation. These results suggest that YGJT could be a new potential candidate against glutamate-induced oxidative stress and cell death. Conclusion : These findings indicate that in C6 glial cells, ROS plays an important role of glutamate-induced cell death and that YGJT may prevent cell death from glutamate-induced cell death by inhibiting the ROS generation.
