치담(治痰) 한약의 항알츠하이머 효능 비교 연구 (Comparative study on anti-Alzheimer's effects of herbal medicines treating phlegm)

  • Objectives : It has been known to be correlated between phlegm and dementia from the perspective of oriental medicine, but it is unexplored whether herbal medicines to treat phlegm have pharmacological actions on Alzheimer's disease (AD). The aim of this study was to evaluate and to compare effects of herbal medicines to treat phlegm against AD in vitro. Methods : We selected 11 herbal medicines which treat phlegm and obtained each extract by boiling in 10-fold distilled water for 2 h. And we performed the assay of acetylcholinesterase (AChE) inhibitory effects of 11 herbal extracts. Next, we evaluated neuroprotective effects of them against amyloid $beta_{25-35}$ ($A{\beta}_{25-35}$) plaque-induced toxicity in HT22 mouse hippocampal neuronal cells using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. To investigate whether they show the anti-inflammatory effects against lipopolysaccharide (LPS), we also measured the levels of nitric oxide (NO) in BV2 microglia cells using griess reagent assay. Results : We found that Gamiyeongsin-hwan (GYH) and Cheonghunhwadam-tang (CHT) exhibited remarkable AChE inhibitory effects. In HT22 cells, Arisaematis Rhizoma, Trichosanthis Semen and Fritillariae Thunbergii Bulbus suppressed $A{\beta}_{25-35}$ plaque-induced neuronal cell death. In BV2 cells, Cheongung-hwan significantly inhibited the increase of NO contents induced by LPS and GYH and CHT showed a tendency to inhibit LPS-induced NO generation. Conclusions : These results suggest that several herbal medicines to treat phlegm showed the significant effects on AChE inhibition, neuroprotection against $A{\beta}_{25-35}$ plaque-induced toxicity, and inhibition of NO generation. Therefore, we demonstrate the possibility that herbal medicines with treating phlegm has effects against AD.

잘피 추출물의 UVB로 손상을 유도한 각질형성세포에 대한 항염 효능 (Anti-inflammation effect of extract from Zostera marina using UVB-induced damage on keratinocytes)

  • Objectives : In order to confirm whether extracts of different parts of Zostera marina (ZM), a marine flowering plant, can be used as cosmetic ingredients, this study evaluated their cytotoxicity and cytoprotective effects against ultraviolet B (UVB). Inflammatory responses induced by UV stimuli are also associated with the aging of the skin.Methods : We investigated the effects of ZM extracts on cells through the water soluble tetrazolium salt-1(WST-1) assay for cell viability. In order to investigate the anti-inflammatory effects, we evaluated the suppression of Cyclooxygenase-2 (COX-2) expression by ZM extracts in HaCaT cells with UVB-induced damages, and also evaluated the production of Prostaglandin E2 (PGE2) in RAW 264.7 cells with LPS-induced damages.Results : High cell viabilities above 90% were observed in all types of ZM extracts, except for whole ZM extract at 0.5 mg/ml; in keratinocytes with UVB-induced damages, the cell viabilities were above 80% when treated with all types of ZM extracts. We confirmed their anti-inflammatory effects by investigating the suppression of inflammatory mediators. In keratinocytes with UVB-induced damages, COX-2 expression decreased in the experimental group treated with ZM extract. Similarly, in RAW 264.7 cells where inflammation was induced with LPS, the biosynthesis of PGE2 was inhibited.Conclusion : These results suggest that ethanol extracts from Zostera marina may have value as the potential anti-inflammatory medicinal plant. Also based on the abovementioned results, ZM extract protects skin cells from UV-induced damages, and thus can be used in topically applied products for skin protection.

보기제통탕(補氣除痛湯)이 Streptozotocin으로 유발된 당뇨병성 말초신경병증 동물 모델에 미치는 영향 (Effects of Bogijetong-Tang on Diabetic-peripheral Neuropathy Induced by Streptozotocin in the Mouse)

  • Objectives: Previous reports have shown that Bogijetong-Tang (BJT) is effective in peripheral neuropathy induced by taxol and crush injury. In this study, we researched the effects of BJT on diabetic neuropathy induced by STZ in the mouse. Methods: We performed both in vitro and in vivo experiments to verify the effects of BJT on diabetic neuropathy induced by STZ in mice. Changes in axonal recovery were observed with immunofluorescence staining using NF-200, Hoechst33258, $S100{\beta}$, caspase 3 and anti-cdc2. Proliferation and degeneration of Schwann cells were investigated by immunofluorescence staining and western blot analyses. Results: BJT showed considerable effects on neurite outgrowth and axonal regeneration in diabetic neuropathy. BJT contributed to the creation of NF-200, GAP-43, Cdc2, phospho-vimentin, ${\beta}1$, active ${\beta}1$, ${\beta}3$ integrin, phospho-Erk1/2 protein. Conclusions: Through this study, we found that BJT is effective for enhanced axonal regeneration via dynamic regulation of regeneration-associated proteins. Therefore, BJT had a pharmaceutical property enhancing recovery of peripheral nerves induced by diabetic neuropathy and could be a candidate for drug development after more research.

Bergenin 및 Acetylbergenin의 사염화탄소 유발 간독성에 대한 치료효과I (Therapeutic Effects of Bergenin and Acetylbergenin on Carbon Tetrachloride-Induced Hepatotoxicity in Rats)

  • Bergenin is a C-glucoside of 4-O-methyl gallic acid that has been isolated from the cortex of Mallotus japonicus (Euphorbiaceae). Acetylbergenin was synthesized by acetylation from bergenin to increase lipophilic and physiological activities. The therapeutic effects of bergenin and acetylbergenin were evaluated against carbon tetrachloride ($CCl_4$)-induced hepatotoxicity in rats. Bergenin and acetylbergenin were administered orally once daily for successive 5 days, after the intraperitoneal injection of a mixture 0.5 m1/kg of $CCl_4$ in olive oil (1:1). The substantially elevated serum enzymatic activities of alanine/aspartate aminotransferase, sorbitol dehydrogenase and ${\gamma}$-glutamyltransferase induced by $CCl_4$ were restored towards normalization by posttreatment with bergenin and acetylbergenin. Bergenin and acetylbergenin also significantly prevented the elevation of hepatic malondialdehyde formation and depletion of glutathione content induced by $CCl_4$ in a dose dependent fashion. In addition, the decreased activities of glutathione S-transferase and glutathione reductase were restored towards normalization. These results suggest that therapeutic effects of bergenin and acetylbergenin may be related complex mechanisms that involve prevention of lipid peroxidation and preservation of hepatic GSH. The results of this study clearly indicate that bergenin and acetylbergenin have potent hepatothrapeutic action against $CCl_4$-induced hepatotoxicity in rats. In addition, acetylbergenin 50 mgHg showed almost the same levels of hepatoprotective activity as those of bergenin 100 mgAg, indicating the fact that lipophilic acetylbergenin is more effective in the hepatoprotective action against $CCl_4$ than bergenin.

사군자탕(四君子湯)이 $CCl_4$에 의한 생쥐의 간 조직 손상에 미치는 영향 (The Effects of Sagunja-tang on Liver Injury of Mice Induced by $CCl_4$)

  • Objectives : This study was carried out to research the protective effects of Sagunja-Tang(SA) through in vivo experiments, and tried to investigate the relation between oxidation of liver tissues and deficiency of Qi. Methods : Acute liver injury which initiated from free radical induced by $CCl_4$, were applied to mice and metabolic data were obtained. In order to measure the degree of liver injury, serum level of alanine aminotransferase(AST), aspartate aminotransferase(ALT), creatinine, blood urea nitrogen(BUN), total protein(TP) and glucose were measured. Lipid peroxidation of liver slice was examined by measuring malondialdehyde(MDA), a product of lipid peroxidation. Results : SA had protective effects on $CCl_4$ induced acute liver injury by decreasing serum level of ALT. Kidney injury was induced by injection of $CCl_4$ too, and SA protected kidney injury by decreasing serum level of creatinine and BUN. Conclusions : Through this study, we found that SA have healing effects on liver and kidney injury of $CCl_4$ induced oxidative stress that is similar to deficiency of Qi. And further studies have to be followed to certify the mechanisms.

Activating transcription factor-3 induction is involved in the anti-inflammatory action of berberine in RAW264.7 murine macrophages

  • Berberine is an isoquinoline alkaloid found in Rhizoma coptidis, and elicits anti-inflammatory effects through diverse mechanisms. Based on previous reports that activating transcription factor-3 (ATF-3) acts as a negative regulator of LPS signaling, the authors investigated the possible involvement of ATF-3 in the anti-inflammatory effects of berberine. It was found berberine concentration-dependently induced the expressions of ATF-3 at the mRNA and protein levels and concomitantly suppressed the LPS-induced productions of proinflammatory cytokines ($TNF-{\alpha}$, IL-6, and $IL-1{\beta}$). In addition, ATF-3 knockdown abolished the inhibitory effects of berberine on LPS-induced proinflammatory cytokine production, and prevented the berberine-induced suppression of MAPK phosphorylation, but had little effect on AMPK phosphorylation. On the other hand, the effects of berberine, that is, ATF-3 induction, proinflammatory cytokine inhibition, and MAPK inactivation, were prevented by AMPK knockdown, suggesting ATF-3 induction occurs downstream of AMPK activation. The in vivo administration of berberine to mice with LPS-induced endotoxemia increased ATF-3 expression and AMPK phosphorylation in spleen and lung tissues, and concomitantly reduced the plasma and tissue levels of proinflammatory cytokines. These results suggest berberine has an anti-inflammatory effect on macrophages and that this effect is attributable, at least in part, to pathways involving AMPK activation and ATF-3 induction.

Hypoglycemic and Hypolipidemic Effects of Tectorigenin and Kaikasaponin III in the Streptozotocin-Induced Diabetic Rat and their Antioxidant Activity in vitro

  • Tectorigenin and kaikasaponin III from the flowers of Pueraria thunbergiana showed potent hypoglycemic and hypolipidemic effects in the streptozotocin-induced diabetic rats. Intraperitoneal administration of these two compounds with 5 and 10 mg/kg, respectively, for seven days to streptozotocin-induced rats significantly reduced the blood glucose, total cholesterol, LDL- and VLDI-cholesterol and triglyceride levels when compared with those of control group. Glycitein in which 5-OH is unlinked and tectoridin (7-O-glycoside of tectorigenin) isolated from the flowers of P. thunbergiana did not improve hyperglycemia and hyperlipidemia. In addition, tectorigenin showed in vitro antioxidant effects on 1,1-diphenyl-B-pirylhydrazyl (DPPH) radical, xanthine-xanthine oxidase superoxide anion radical, and lipid peroxidation in rat microsomes induced by enzymatic and non-enzymatic methods. We further found that tectorigenin and kaikasaponin III protected the Vero cell line(normal monkey kidney) from injury by hydrogen peroxide. From these findings, it seems likely that the antioxidant action of tectorigenin and kaikasaponin III may alleviate the streptozotocin-induced toxicity and contribute to hypoglycemic and hypolipidemic effects.

항우울제의 부작용과 대처 방안(1) - 성기능 장애를 중심으로 - (Antidepressant-Induced Adverse Effects and Management Strategy - Focused on Sexual Dysfunction -)

  • Sexual dysfunction is a relatively common adverse effect in the use of antidepressants. The sexual side effects may result in a lack of compliance with the prescribed antidepressants. The author reviewed the prevalence and updated treatment for the antidepressant-induced adverse effects focusing on sexual dysfunction. The incidence of sexual dysfunction is reported to exceed more than 50% especially with SSRIs. In order to obtain a quantified baseline and as an ongoing evaluation tool, clinicians may use some of the established questionnaires and validated instruments such as the Arizona Sexual Experience scale and Changes in Sexual Functioning Questionnaire. Clinicians should be aware that delayed ejaculation and orgasm, symptoms most frequently associated with antidepressants, are not usually associated with depression itself. Although many antidotes have been proposed, few have been subjected to double-blind trials. Some evidences have suggested that bupropion and buspiron may be the effective antidotes for SSRI induced sexual dysfunction. Additional trials will be requied to define what role, if any, bupropion and buspiron might have in the treatment of SSRI-induced sexual side effects. The available evidence is rather limited, with only small number of trials assessing each strategy. While further randomized data is awaited, for men with antidepressant induced erectile dysfunction, the addition of sidenafil or tadalafil may appear to be an effective strategy.

Cyclopiazonic acid 및 aflatoxin B1이 토끼의 혈소판 응집 및 ATP 방출에 미치는 영향 (Effects of cyclopiazonic acid and aflatoxin B1 on rabbit platelet aggregation and ATP release)

  • Cyclopiazonic acid(CPA) known as stimulating the release of intracellular calcium, aflatoxin $B_1(AFB_1)$ causing gastrointestinal hemorrhage frequently were used as model toxic mycotoxins in these studies. First of all, the effects of various mycotoxins on the platelet aggregation response were determined. The effects of mycotoxins on the ATP release from platelet by aggregating factors were investigated. The results and conclusions obtained from these studies are : 1) CPA promoted ADP, collagen, thrombin, A.A. and PAF-induced rabbit platelet aggregation. $AFB_1$ inhibited collagen, A.A. and PAF-induced rabbit platelet aggregation only. 2) CPA increased both aggregation and disaggregation time, whereas $AFB_1$ decreased in a dose dependent manner. 3) CPA increased ADP, thrombin, A.A. and PAF-induced ATP release. $AFB_1$ increased A.A.-induced ATP release and decreased PAF-induced release in a dose dependent manner. In conclusion, CPA promoted platelet aggregation by the increase of ATP. Antiaggregating effects of AFB1 may be due to decreases of ATP. These data provide the basis for the future study of roles of ATP release in platelet aggregation.

원적외선 전신조사가 류마티스 관절염 유발 흰쥐에 미치는 영향 (The Effects of Far-Infrared on Rheumatoid Arthritis Induced Rats)

  • The purpose of this study was to observe the effects of far-infrared on rheumatoid arthritis induced rats by type ⅡI collagen with the change of arthritis index, hind paw volume, TNF-α and histopathologic findings. Thirty-six Sprauge-Dawley rats which were divided into four group. Group Ⅰ : Normal group, Group Ⅱ : Collagen-Induced Arthritis group, Group Ⅲ : Far-Infrared 25㎝ radiation group, Group Ⅳ : Far-Infrared 35㎝ radiation group. The results of this study were as follows: In arthritis index, far-infrared radiation group(Ⅲ, Ⅳ) are more decreased than collagen-induced arthritis group(Ⅱ). Group Ⅲ and Ⅳ showed the most significant effect at 14 day(p<0.05). In hind paw, far-infrared 25㎝ radiation group(Ⅲ) significantly decreased more than only collagen-induced arthritis group on day 14(p<0.05). Far-infrared 35㎝ radiation group(Ⅳ) significantly decreased more than collagen-induced arthritis group on day 3, 7 and 14(p<0.05). In the ELISA study of TNF-α concentration, collagen-induced arthritis group significantly increased in the concentration more than normal group. Far-infrared radiation group(Ⅲ, Ⅳ) decreased in TNF-α concentration more than collagen-induced arthritis group(Ⅱ) on day 14. In histopathologic findings, collagen-induced arthritis(Ⅱ) and far-infrared 25㎝ radiation group(Ⅲ) increased in synovial membrane thickness on 3 day. Far-infrared 35 em radiation group(Ⅳ) decreased in the concentration more than collagen-induced arthritis group on day 14. The above results suggest that far-infrared radiation effectively reduced in rheumatoid arthritis. It would be considered that far-infrared has an effects on relieving rheumatoid arthritis.
