  • Indirect photometric detection of aliphatic compounds such as alcohols, ketones and esters was investigated in high-performance liquid chromatography, These samples possessing nonchromophore or weak chromophore could be indirectly detected by a UV detector with addition of UV absorbing, p-chlorophenol or p-nitrophenol in mobile phase as detection reagent, and submicrogram detection limits were achieved. Some mixtures of samples were also able to be separated and quantitated with good resolution and comparatively high sensitivity under optimum conditions.

  • An ion chromatographic method based on indirect photometric detection of UV transparent anions was developed. Separation of anion was accomplished on strong anion exchange column (Waters SAX) using UV detector at 254 nm. Among examined UV-active additives (Dns-H, Dns-glu, Dnsser, Dns-val), Dns-glu showed the highest sensitivity. Studies on the effects of the pH and ionic strength of eluent revealed that the increase of pH and ionic strength of the eluent decreased capacity factor. The best eluent for the separation of acetate, fluoride, chloride, nitrate and bromide was $1\;{\times}\;10^{-4}M$ Dns-glu in 5 mM phosphate buffer (pH 6.30). The detection limit of chloride ion was 2.1 ng in this condition.

  • A simultaneous determination method of alkaline earth metals was attempted with the conventional high performance liquid chromatographic system. Four cations, namely, magnesium, calcium, strontium and barium ion, were injected directly as aqueous solution into an eluent containing copper chloride solution and and were successfully separated and determined on a separating column (Zipax SCX, 4.6 mm i.d. ${\times}25$ cm length, Du Pont, USA) by using a variable wavelength UV detector. The linear calibration curves were obtianed in the range from $1.0{\times}10^{-4}M$ to $5.0{\times}10^{-4}M$ and the correlation coefficient of the calibration curve for each metal of magnesium and calcium in tap water. Alkaline earth metals were determined with the conventional high performance liquid chromatographic system.

  • Nonchromophore compounds such as aliphatic acids, alcohols and tetraalkylammonium salts could be detected by indirect photometric detection on the revered-phase liquid chromatography. Benzyltriethylammonium bromide(BTEAB) was used as a detection reagent. Also, the retention mechanism and response of samples were investigated to the several factors such as pH, temperature, and concentration of MeOH as well as concentration of detection reagents in mobile phase. And some mixture of samples were able to be separated under optimum condition.

  • Single column anion chromatography by indirect UV detection was applied to the determination of 7 common anions($H_2PO_4^-$, $Cl^-$, $NO_2^-$, $NO_3^-$, $I^-$, $SO_4^{2-}$, $SCN^-$). A low capacity silica-based anion-exchange column(TSK guardgel QAE-SW; ca. 0.3 meq./g; 4.6 mm i.d${\times}$150 mm) was used. In separating common anions, we compared naphthalene trisulfonic acid(NTS) and naphthalene disulfonic acid(NDS) to find more sensitive, selective and rapid separation method. And cations and anions can be separated simultaneously in 0.20 mM NTS-5% acetonitrile-water.

Analysis of THMs and PCBs by Indirect Photometric Method in High Performance Liquid Chromatography (고성능 액체 크로마토그래피의 간접 분광광도법에 의한 THM과 PCB의 분석)

  • THMs and PCBs were separated and analysed with elution on Novapak ODS or $\mu-Bondapak$ phenyl column by an eluent containing p-nitrophenol (p-NP). THMs studied were CHCl3, CHBrCl2, CHBr2Cl, and CHBr3, and PCBs used were Aroclor 1221, 1242, 1248, $\alpha-$ and $\beta-BHC.$ It was thought that the retention on the stationary phase and sensitivities of the samples are related to the interaction between the sample and stationary phase or p-NP. THMs were separated completely on the ODS column by elution with MeOH-water (30 : 70) containing $1.0{\times}10^{-4}$ M p-NP and some of PCBs were separated on the phenyl column by elution with $CH_3CN$-water(50 : 50) containing $1.0{\times}10^{-4}$ M p-NP. Detection limits of THMs were from $1.0{\times}10^{-4}$ g to $1.0{\times}10^{-6}$ g. Aroclors were $2{\times}10^{-6}$ g, and $\alpha-$ and $\beta-BHC$ were $2{\times}10^{-4}$ g and $1.0{\times}10^{-4}$ g respectively.
