캄보디아 프놈보콜국립공원의 Balanophora fungosa var. indica의 숙주식물에 대한 DNA barcoding 기법을 통한 동정 (Identification of host plant species of Balanophora fungosa var. indica from Phnom Bokor National Park of Cambodia using DNA barcoding technique)

  • 캄보디아 캄폿주 프놈보콜국립공원에 대한 식물상 조사 중 열대성 전기생식물인 B. fungosa var. indica를 발견하였다. 이들의 숙주를 확인하기 위해 숙주의 뿌리와 더불어 주변에 위치한 목본 식물을 채집하였으며, 이들을 DNA barcoding 방법을 사용하여 동정하였다. DNA barcode 마커로는 엽록체 rbcL 및 matK 유전자 구간을 적용하였으며, 15개의 숙주 뿌리와 7개의 주변 목본식물로부터 성공적으로 PCR 증폭 및 염기서열을 확보하였다. 숙주 뿌리로부터 얻어진 숙주의 염기서열은 앵초과, 노박덩굴과, 도금양과, 그리고 물푸레나무과로 식별되었으며, 주변의 목본식물은 물푸레나무과, 도금양과, 무환자나무과, 장미과, 물레나물과, 철쭉과와 녹나무과였다. 속 수준에서 앵초과는 Myrsine, 노박덩굴과는 Euonymus, 도금양과는 Syzygium, 물푸레나무과는 Olea 등으로 각각 식별되었으나, 종 수준에서의 동정은 불가능하였다. 앵초과 Myrsine와 물푸레나무과 Olea는 본 연구를 통해 최초로 B. fungosa var. indica의 숙주로 확인되었다. 추가적인 채집 조사 및 비교 연구, DNA barcoding을 통해 해당 지역의 생물다양성과 Balanophora속 식물의 숙주 식물 및 진화에 대해 좀더 명확하게 확인이 가능할 것으로 판단된다.

화장품 개발을 위한 배롱나무(Lagerstroemia indica Linnaeus) 가지 추출물의 생리활성에 관한 연구 (Study on Biological Activities of Extracts for Cosmeceutical Development from Lagerstroemia indica L. Branch)

  • 배롱나무 가지가 기능성화장품 소재 및 천연물 의약품으로써 활용 가능성이 있는지 검토해 보고자 그 생리활성을 확인하기 위해서 DPPH radical 소거능 측정, ABTS radical cation decolorization 활성, Nitrite 소거작용, collagenase 저해활성을 측정하였다. DPPH radical 소거능의 경우 배롱나무 가지의 아세톤 추출물은 50 ppm에서 73%의 항산화능을 나타내었으며 합성항산화제인 BHA는 50 ppm에서 90% 이상의 높은 항산화능을 보여준다. ABTS radical cation decolorization 활성은 50 ppm에서 78% 이상 높은 항산화활성을 보였으며 BHA의 경우 50 ppm에서 96% 이상 활성을 보여준다. Nitrite 소거작용 측정은 대조군인 합성항산화제 BHA와 비교하여 모든 농도에서 우수한 아질산염 소거능을 보여주었다. 배롱나무 가지 추출물은 50 ppm의 저농도에서 63% 이상의 활성을 보여주었으며 1000 ppm에서 73%의 활성을 보이는 것과 비교하여 저농도에서도 높은 활성을 보여주는 것을 알 수 있다. Collagenase 저해활성 측정 결과 대조군인 천연물 (-)-epigallo-catechin-3-gallate와 비교하여 모든 농도에서 우수한 collagenase 저해활성을 보여주었으며 50ppm의 저농도에서도 85% 이상의 높은 collagenase 저해활성을 보여주었다. 이와 같은 결과로 미루어 보아 대조군인 인공 합성 항산화제인 BHA와 비교하여 배롱나무 가지 추출물은 항산화 효과, 아질산염 소거작용 그리고 주름개선에 우수한 효과가 나타나는 것을 수 있다. 따라서 화장품 산업 및 천연물 의약품의 원료로 이용하기 위한 적합한 천연 물질인 것으로 판단된다.

NaCl 처리에 따른 멀꿀과 다정큼나무의 내염성 평가 (Salt Tolerance Assessment with NaCl of Stauntonia hexaphylla (Thunb.) Decene. and Raphiolepis indica var. umbellata (Thunb.) Ohashi)

  • 상록활엽 조경소재 연구개발의 일환으로써, 기후변화에 대응하여 내한성이 우수한 남부지역 상록활엽수 중에서 멀꿀과 다정큼나무에 대하여 가시적 피해, 엽록소 형광이미지, MDA(malondialdehyde) 농도 분석으로 내염성을 평가하였다. NaCl 농도가 증가함에 따라서 토양 pH는 감소하고 EC는 증가하였으며 멀꿀이 생육 중은 토양은 다정큼나무가 생육 중인 토양보다 더 강하게 영향을 받았다. 시각적 피해에서 멀꿀은 처리 후 20일에 200mM NaCl 농도 이상에서 고사하였다. 다정큼나무는 비록 400mM NaCl 농도에서 잎의 피해를 나타냈지만 실험기간 동안 모두 생존하였다. 엽록소 형광이미지에서 색상변화는 시각적 피해 결과와 강한 일관성을 보였다. NaCl 농도가 증가함에 따라서 멀꿀 잎의 적색은 유의성 있게 낮은 형광 값인 청색으로 변화하였고 변화는 가장자리에서 중앙으로 옮겨갔다. 다정큼나무의 형광이미지 반응은 NaCl 처리일이 증가할수록 잎의 가장자리에 변화가 나타났으나 여전히 실험기간 동안 적색을 나타냈다. 가장 높은 시각적 피해와 EC를 보였던 처리 후 5일에 멀꿀의 MDA 농도는 $4.56nmol{\cdot}g^{-1}$였다. 다정큼나무의 MDA 농도는 400mM NaCl 농도 처리구를 제외한 모든 처리구들에서 $1.5nmol{\cdot}g^{-1}$ 이하를 나타냈다.

  • In this study, uptake and translocation of 2,4,6-trinitrotoluene(TNT) by plant in a hydroponic culture was quantified with two indigenous plant species, Aeschynomene indica and Abutilon avicennae on various initial concentrations of TNT ranging from 20 to 80 mgTNT/L. Experiments were sterilized to exclude the activity of microorganisms and conducted in duplicate. Weight loss of two plant species in added TNT culture media was higher than in control. At over 2OmgTNTIL, there appeared to be phytotoxicity from TNT as indicated by severe yellow-chlorosis and increase of falling leaves. TNT removal rate normalized(K) to the plant fresh weight of Abutilon avicennae and Aeschynomene indica was that the higher TNT concentrations resulted in lower TNT removal rate normalized(K) to the plant fresh weight. Approximately 96% of the TNT in viable microflora-hydroponic culture was removed after 96h of the experiments.

Tissue culture of the plant Pluchea indica (L.) Less. and evaluation of diuretic potential of its leaves

  • The present study focused on the establishment of micropropagation protocol for the high value Pluchea (P.) indica (L.) Less., genotype, an important medicinal plant and evaluation of the diuretic activity of the leaf extract of the tissue cultured plant. Leaf explants, nodal segments and shoot tips were cultured in MS medium supplemented with auxin and cytokinin and their combinations. With the objective of inducing callus giving rise to new adult plants, naphthalene acetic acid was found to be most effective for (80%) for callus induction. The methanolic extract of leaves of the micropropagated P. indica was investigated for its diuretic activity in Wistar albino rats. Urinary excretion parameters were studied for evaluation of diuretic activity using Frusemide (20 mg/kg, p.o.) as standard. The extract showed significant diuretic activity at the doses of 100, 200 and 300 mg/kg. p.o. An oral acute toxicity study for the extract was carried out and the $LD_{50}$ value was found to be 2,825 mg/kg body weight.

Anti-inflammatory, anti-ulcer and hypoglycaemic activities of ethanolic and crude alkaloid extracts of Madhuca indica (Koenig) Gmelin seed cake

  • Madhuca indica has been used ethnomedically in Indian folks. In the present study we have investigated anti-inflammatory, anti-ulcer and hypoglycaemic effect of ethanolic extract (EE) and crude alkaloid extract of Madhuca indica seed cake on albino rats. The study showed that the EE had a significant, dose dependent anti-edematogenic, anti-ulcerogenic and hypoglycaemic activity, whereas the crude alkaloid extract exhibited a significant only. Both the extracts possess dose dependent inhibitory activity on carrageenan-induced edema, inhibiting prostaglandins or mediators involved in prostaglandin synthesis, the second phase of inflammation. The EE was significantly effective in protecting pylorus-ligation-induced gastric ulcers at a higher dose level. The active principle of EE seems to be a selective inhibitor of the COX II (prostaglandin synthesis) without important effect on COX I since, EE exhibited both anti-edematogenic and anti-ulcerogenic effect. The EE was effective in reducing the plasma glucose level in normal albino rats in a dose dependent manner, producing hypoglycaemic effect by stimulating the release of insulin from the ${\beta}-cells$ and/or increasing the uptake of glucose from the plasma.

Anti-inflammatory and Antinociceptive Activities of 'Coccinia indica W. &A.' Fruit Juice Powder in Animals

  • The fresh fruit juice powder of Coccinia indica W.&A., (Cucurbitaceae, CJP) was studied for the possible activities of antiinflammatory and antinociceptive to rationalize the folkloric use of the plant juice as rasayana. CJP at the doses of 50-200 mg/kg caused a significant (P<0.05 to P<0.001) inhibition of paw edema induced by ${\lambda}$ carrageenin (1%) and histamine $(10^{-3}\;g/ml,\;0.1\;ml)$ in rats. The effect was comparable to the standard cycloxygenase inhibitor brufen at 100 mg/kg and protective percentages were 63.41% and 65.78% respectively. Administration of CJP (50-200 mg/kg) exhibited a moderate increase of the pain threshold on analgesy-meter induced mechanical pain. However CJP significantly prevented the writhing induced by acetic acid in mice and the percentages of inhibitions were 16.98%-35.47%, which is equivalent to 36.67% produced by brufen. These data indicate that the fruit juice of Coccinia indica rationalizes the traditional system of medicine.

Gelatinization Characteristics of Glutinous Rice Varieties

  • Gelatinization characteristics of 111 glutinous rice varieties were evaluated by Rapid Visco Analyzer. Gelatinization viscosity of glutinous rice tested varied with ecotypes or varietal groups: indica, japonica, and Tongil type. Indica rice showed the highest average value of initial pasting temperature. The average values for peak, hot, and cool viscosities were highest in Tongil-type rice, and lowest in japonica rice. Japonica showed the lowest breakdown and consistency, but the highest setback value. Indica was lower in alkali digestion value (ADV), and shorter in gel length after gelatinization thanjaponica and Tongil-type. Glutinous rices tested could be divided into six groups by cluster analysis based on their gelatinization characteristics. Group I-A was mostly early maturing japonica varieties while I-B was mostly indica and Tongil-type rices. Groups II-A and II-B were consisted of very early maturingjaponica, and III-A and III-B included medium or medium late maturingjaponica varieties. Group III-A showed the lowest average values of peak, hot, cool, and consistency viscosities, and also in breakdown and setback ratios. Group I-B revealed the highest values in peak, hot, cool, breakdown, and consistency viscosities. ADV was low in groups I-A, I-B, and II-B, and gel consistency was not different among the six varietal groups. Principal component analysis using seven traits related with gelatinization produced four effective components, and the first and second components were highly correlated with all the gelatinization characters evaluated.

Antimicrobial activity and toxicity of Quisqualis indica

  • The plant Quisqualis indica (Compositae) has ehnopharmcological reputation of being used as a healing agent in Bangladesh. In this study, preliminary screenings were conducted to look at the antimicrobial susceptibility and cytotoxicity of the plant extract. The extractives of the plant were subjected to screening for inhibition of microbial growth by the disc diffusion method. The zones of inhibition demonstrated by the n-hexane, carbon tetrachloride, chloroform and aqueous soluble partitionates of the methanolic extract ranged from 8 - 15 mm, 8 - 18 mm, 12 - 20 mm and 10 - 16 mm, respectively at a concentration of 400 ${\mu}g$/disc. All the extractives were also subjected to brine shrimp lethality bioassay for primary cytotoxicity evaluation. Here, the carbon tetrachloride soluble materials demonstrated the highest cytotoxicity with $LC_{50}$ of 0.826 ${\mu}g$/ml, while n-hexane, chloroform and aqueous soluble partitionates of the methanolic extract revealed the $LC_{50}$ of 1.254, 3.866 and 5.366 ${\mu}g$/ml, respectively. This is the first report of the antimicrobial activity and cytotoxicity Q. indica.

벼 종자배양에서 Copper가 캘러스 형성 및 식물체 재분화에 미치는 영향 (Effect of Copper on Callus Formation and Plant Regeneration in Seed Culture of Rice)

  • 벼의 종자배양에서 식물체 재분화 능력을 향상시키기 위하여 2 mg/L의 2,4-D와 여러 가지 농도의 copper (0~5.0mg/L)가 첨가된 MS배지에 종자배양 한 바 캘러스 생장정도는 copper 농도에 따라 다르게 나타났는데 특히, 2.5 mg/L의 copper가 첨가된 배지에서 가장 양호하였다. 그리고 캘러스 형성률과 copper 농도 간에는 유의성 있는 차이가 인정되지 않았다. 식물체 재분화 배지에 2.5 mg/L의 copper의 첨가는 Indica 품종들의 식물체 재분화 효율을 크게 향상시켰다. Copper가 첨가되지 않은 N$_{6}$배지에서 Indica 6품종의 평균재분화율은 2.4%이었으나, copper가 첨가된 배지에서 27.4%로 높게 나타났다.
