A finite element-based, integrated process model has been developed and applied to predict the detailed, three-dimensional aspects of the thermo-mechanical behavior occurring in the slab caster considering inclusion removal from molten steel by argon bubble flotation. Gas bubbles are simulated using the dispersion model calculating the volume fraction, and the bubble capture effect is included by a source term in the transport equation for particles. The process model is applied to the investigation of the effect of various process parameters on the inclusion removal in the molten steel.
A tube-type filter dam(or baffle with holes) for ferrous melt refining is applied to the tundish operation for molten steel having low carbon and ultra-low carbon. The changes in the total oxygen content insoluble aluminum content and the distribution of inclusion size in molten steel during tundish operation were in-vestigated at the pouring part strand and mold of tundish. Removal mechanism of inclusions is considered to be caused by buoyancy action of the filter dam better than filtration action and the size range of in-clusions filtrated by the filter dam was for 30-50$\mu\textrm{m}$. Decrease in deviation of inclusion content in molten steel was confirmed for appling the filter dam in comparison with using conventional dam and weir. Also the filter dam had the advantage of baffle with holes at lower part in the efficiency of inclusions removal.
In this study soft computing techniques including, Artificial Neural Network (ANN) and Adaptive Neuro-Fuzzy Inference System (ANFIS) were investigated for the prediction of Cr(VI) transport efficiency by novel Polymer Inclusion Membranes (PIMs). Transport experiments carried out by varying parameters such as time, film thickness, carrier type, carier rate, plasticizer type, and plasticizer rate. The predictive performance of ANN and ANFIS model was evaluated by using statistical performance criteria such as Root Mean Standard Error (RMSE), Mean Absolute Error (MAE), and Coefficient of Determination (R2). Moreover, Sensitivity Analysis (SA) was carried out to investigate the effect of each input on PIMs Cr(VI) removal efficiency. The proposed ANN model presented reliable and valid results, followed by ANFIS model results. RMSE and MAE values were 0.00556, 0.00163 for ANN and 0.00924, 0.00493 for ANFIS model in the prediction of Cr(VI) removal efficiency on testing data sets. The R2 values were 0.973 and 0.867 on testing data sets by ANN and ANFIS, respectively. Results show that the ANN-based prediction model performed better than ANFIS. SA demonstrated that time; film thickness; carrier type and plasticizer type are major operating parameters having 33.61%, 26.85%, 21.07% and 8.917% contribution, respectively.

Nitrate removal rate in three cattail wetland cells was investigated. They were a part of a pond-wetland system for stream water treatment demonstration. The system was composed of two ponds and six wetland cells. The acreage of each cell was approximately $150m^2$. The earth works for the system were finished from April 2000 to May 2000 and cattails were planted in the three cells in June 2000. Waters of Sinyang Stream flowing into Kohung Estuarine Lake were pumped into a primary pond, whose effluent was discharged into a secondary pond. The reservoir was formed by a tidal marsh reclamation project and located in southern coastal area of Korean Peninsula. Effluents from the secondary pond were funneled into the three cells. Volumes and water quality of inflow and outflow were analyzed from July 2000 through January 2001. Inflow and outflow averaged $20.2m^3/day$ and $19.8m^3/day$, respectively. Hydraulic retention time was about 1.6 days. Average influent and effluent nitrate concentration was $1.98mg/{\ell}$, $1.38mg/{\ell}$, respectively. Nitrate removal rate averaged $82.6mg\;m^{-2}\;day^{-1}$. Seasonal changes of nitrate retention rates were closely related to those of wetland cell temperatures. The average nitrate removal rate in the cells was a little lower, compared with that of $125.0mg\;m^{-2}\;day^{-1}$ for the wetlands operating in North America. This could be attributed to the initial stage of the cells and inclusion of three cold months into the seven-month study period. Root rhizosphere in wetland soils and litter-soil layers on cell bottoms could not developed. Increase of standing density of cattails within a few years will establish both root zones suitable for the nitrification of ammonia to nitrates and substrates beneficial to the denitrification of nitrates into nitrogen gases, which may lead to increase of the nitrate retention rate.

$NO^3$-N removal was examined from July 2002 to December 2002 of a surface-flow constructed treatment wetland cell, which was a part of a treatment wetland system composed of four wetland cells and one distribution pond. The system was established on rice paddy near the Kohung Estuarine Lake located at the southern part of the Korean Peninsula. The lake and the paddy were formed by a salt marsh reclamation project. Effluent from a secondary-level treatment plant was funneled into the system. The investigated cell was created in June 2002. Its dimensions were 87 m in length and 14 m in width. It had an open water zone at its center, which was equivalent to 10 percent of its total area. Reeds(Phragmites australis) were transplanted from natural wetlands into the cell and their stems were cut at about 40 cm height from their bottom ends. Average 25 $m^3$/day of effluent from the plant was funneled into the cell by gravity flow and average 24.2$m^3$/day of its treated effluent was discharged into the Sinyang Stream flowing into the lake. Its water depth was maintained about 0.2 m and its hydraulic detention time averaged 5.2 days. The average height of the reed stems was 45.2 cm in July 2002 and 80.5 cm in September 2002. The number of stems averaged 40.3 stems/$m^2$ in July 2002 and 74.5 stems/$m^2$ in September 2002. The reeds were established initially well. $NO_3$-N loading rate of influent and effluent averaged 173.7 and $93.5mg/m2{\cdot}day$, respectively. Removal of $NO_3$-N averaged $80.2mg/m2{\cdot}day$ and its removal rate by mass was about 50 %. Considering the initial operation of the cell and the inclusion of the cold months of November and December in the analysis period, the $NO_3$-N removal rate was good.

Expressed protein ligation (EPL) technique, joining recombinantly expressed proteins to polypeptides, has been widely adopted for addressing various biological questions and for drug discovery. However, joining two recombinant proteins together is sometimes difficult when proteins are expressed insoluble and unrefoldable, because ligation-active proteins via intein-fusion are obtainable when they are folded correctly. We overcame this limitation coexpressing target protein with additional methionine aminopeptidase (MAP) which enhances removal of the initiation methionine of recombinantly expressed protein. Our approach demonstrated that two domains of 46 kDa 5-Enolpyruvylshikimate-3-phosphate (EPSP) synthase, a target of herbicide glyphosate, were successfully joined by native chemical ligation, although its C-terminal domain was expressed as an inclusion body. The intein-fused N-terminal fragment of EPSP synthase (EPSPSN, residues 1-237) was expressed and the ligation-active thioester tagged N-terminal fragment (EPSPSN-thioester) was purified using a chitin affinity chromatography and mercapto-ethanesulphonate (MESNA) as intein thiolysis reagent. Its Cterminal fragment (EPSPSC, residues Met237-238CYS-427), expressed as an inclusion body, was prepared from an additional MAP-expressing strain. Protein ligation was initiated by mixing ~1 mM of EPSPSN-thioester with ~2 mM of EPSPSCCYS (residues 238CYS-427). Also we found that addition of 2% thiophenol increased the ligation efficiency via thiol exchange. The ligation efficiency was ~85%. The ligated full-length EPSP synthase was dissolved in 6 M GdHCl and refolded. Circular dichroism (CD) and enzyme activity assay of the purified protein showed that the ligated enzyme has distinct secondary structure and ~115% specific activity compared to those of wild-type EPSP synthase. This work demonstrates rare example of EPL between two recombinantly expressed proteins and also provides hands-on protein engineering protocol for large proteins.
Phosphorous removal rate and emergent plant growth were examined of a surface-flow constructed treatment wetland system, whose dimensions were 31 meter in length and 12 meter in width. The system was established on floodplain in the down reach of the Kwangju Stream in Korea in one and half months from May to June 2001. Cattails(Typha angustiflora) were transplanted in the system. They were dug out of natural wetlands and stems were cut at about 40 cm height from their bottom ends. Water of the Kwangju Stream were funneled into it via a pipe by gravity flow and its effluent were discharged back into it. The stems of cattails grew from 45.2 cm in July 2001 up to 186 cm in September 2001 and the number of cattail stems per square meter increased from 22 in July 2001 to 53 in September 2001. The early establishment of cattails was good. Volume and water quality of inflow and outflow were analyzed from July 2001 through December 2001. Inflow averaged $40\;m^3/day$ and hydraulic retention time was about 1.5 days. The concentration of total phosphorous in influent and effluent was 0.85 mg/L, 0.41 mg/L, respectively. The average removal rate of total phosphorous in the system was about 52%. The retention efficiency was slightly lower, compared with that in surface-flow wetlands operating in North America, whose retention efficiency was reported to be about 57%. The lower abatement rate could result from the initial stage of the system and inclusion of two cold months into the six-month monitoring period. Root rhizosphere in wetland soils and litter-soil layers on bottoms were not properly developed. Increase of standing density of cattails within a few years will establish both root zones and substrates beneficial to the removal of phosphorous, which may lead to increase of the phosphorous retention rate. The system was submerged one time by heavy storm during the monitoring period. The inundation, however, scarcely disturb its environment.

Gas hydrate (GH)-based desalination process have a potential as a novel unit desalination process. GHs are nonstoichiometric crystalline inclusion compounds formed at low temperature and a high pressure condition by water and a number of guest gas molecules. After formation, pure GHs are separated from the remaining concentrated seawater and they are dissociated into guest gas and pure water in a low temperature and a high pressure condition. The condition of GH formation is different depending on the type of guest gas. This is the reason why the guest gas is a key to success of GH desalination process. The salt rejection of GH based desalination process appeared 60.5-93%, post treatment process is needed to finally meet the product water quality. This study adopted reverse osmosis (RO) as a post treatment. However, the test about gas rejection by RO process have to be performed because the guest gas will be dissolved in a GH product (RO feed). In this research, removal potential of dissolved gas by RO process is performed using lab-scale RO system and GC/MS analysis. The relation between RO membrane characteristics and gas removal rate were analyzed based on the GC/MS measurement.

Electrolytic polishing is the anodic dissolution process in the transpassive state. It removes non-metallic inclusion and improves mechanical and corrosion resistance of stainless steel. If there is a Bailby layer, it will be removed and the true structure of the surface will be restored. Electrolytic polishing is normally used to remove a very thin layer of material from the surface of metal object. A new electrolyte composed of phosphoric, sulfuric and distilled water has been developed in this study. Two current density, high & low current density regions, have been applied in this study. In this study, In the region of high current density, there is no plateau region but excellent electrolytic polishing effect can be accomplished in short machining time because material removel process and leveling process occur simultaneously. In the low current density region, there can be found plateau region. The material removal process and leveling process occur successively. The aim of this work is to determine electrolytic polishing for stainless steel in terms of high & low current density and workpiece surface roughness.
Bacillus thuringiensis Cyt2Aa toxin is a mosquito-larvicidal and cytolytic $\delta$-endotoxin, which is synthesized as a protoxin and forms crystalline inclusions within the cell. These inclusions are solubilized under alkaline conditions and are activated by proteases within the larval gut. In order to assess the functions of the N-and C-terminal regions of the protoxin, several N- and C-terminal truncated forms of Cyt2Aa were constructed. It was determined that amino acid removal at the N-terminal, which disrupts the $\beta$1 structure, might critically influence toxin production and inclusion formation. The deletion of 22 amino acids from the C-terminus reduced the production and solubility of the toxin. However, the removal of more than 22 amino acids from the C-terminus or the addition of a bulky group to this region could result in the inability of the protein to adopt the proper folding. These findings directly demonstrated the critical roles of N- and C-terminal amino acids on the production and folding of the B. thuringiensis cytolytic $\delta$-endotoxin.
