Effects of Polygonatum odoratum on In vivo Insulin Activity in Streptozotocin-Induced Diabetic Rats (둥글레 섭취가 Streptozotocin 유발 당뇨병 쥐의 In vivo 인슐린 작용에 미치는 영향)

  • This study investigated the in vivo insulin function of Polygonatum odoratum in normal and diabetic male Sprague-Dawley rats. Diabetes mellitus was induced by an i.p. injection of streptozotocin. Normal and diabetic rats were assigned to the diet groups of the control basal diet and Polygonatum odoratum diet. The animals were fed the diet and water ad libitum for 15 days. Initial and final body weights, total food intake and serum glucose and insulin levels were measured. An insulin suppression test was performed to elucidate the insulin function in the peripheral tissues. The results showed that the final serum glucose levels significantly decreased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The final serum insulin levels were increased in the diabetic rats on the Polygonatum odoratum diet compared with the diabetic rats on the control diet. The in vivo function of the insulin increased in the diabetic rats on the Polygonatum odoratum compared with the diabetic rats on the control diet. These data indicate that Polygonatum odoratum may be beneficial in improving the in vivo insulin function in streptozotocin-induced diabetic rats.

  • The reaction rates, duration times and neutralizing capacities of the antacids which are frequently used in Korean market and three different commercial combination products were evaluated in vitro by Fuchs method and Johnson-duncan method, respectively. In vivo tests of combination products were determined in the fasted state of rat by Aspiration method. Comparing the result of in vitro test with that of in vivo test, the maximal pH was lowered by 2-3 value and the durational time increased by two folds in vivo test. Each antacid composition and combination products from three phamaceutical companies (A, B, and C) were studied, respectively. The duration times measured by Fuchs method were double compared to those by Johnson-Duncan method. A and C preparation maintained the pH range from 3 to 7 for 60 min by Fuchs method. In vovo test, maximum pH of A, B and C preparation was 6.50, 3.65, 2.65 and duration time of those was 200, 500, 0 min, respectively.

Optimization of In-vivo Monitoring Program for Radiation Emergency Response

  • Background: In case of radiation emergencies, internal exposure monitoring for the members of public will be required to confirm internal contamination of each individual. In-vivo monitoring technique using portable gamma spectrometer can be easily applied for internal exposure monitoring in the vicinity of the on-site area. Materials and Methods: In this study, minimum detectable doses (MDDs) for $^{134}Cs$, $^{137}Cs$, and $^{131}I$ were calculated adjusting minimum detectable activities (MDAs) from 50 to 1,000 Bq to find out the optimal in-vivo counting condition. DCAL software was used to derive retention fraction of Cs and I isotopes in the whole body and thyroid, respectively. A minimum detect-able level was determined to set committed effective dose of 0.1 mSv for emergency response. Results and Discussion: We found that MDDs at each MDA increased along with the elapsed time. 1,000 Bq for $^{134}Cs$ and $^{137}Cs$, and 100 Bq for $^{131}I$ were suggested as optimal MDAs to provide in-vivo monitoring service in case of radiation emergencies. Conclusion: In-vivo monitoring program for emergency response should be designed to achieve the optimal MDA suggested from the present work. We expect that a reduction of counting time compared with routine monitoring program can achieve the high throughput system in case of radiation emergencies.

천연물 항암제의 유도체 개발

  • 본 연구소에서 분리하여 항암활성을 측정한바 있는 Sesquiterpene lactone계열의 KR-53170이 in vitro에 비해 in vivo에서 상대적으로 낮은 효력을 보이는데 착안 그 유도체들을 합성함으로써 in vivo stability 및 efficacy를 개선하고자 하였다. 합성된 물질의 in vivo activity는 human tumor cell중에서 A549(폐암), SK-OV-3(난소암1, SK-MEL-2(피부암), XF-498(중추신경계암), HCT-15(직장암)의 5개 암세포를 이용하여 ED$_{50}$($\mu\textrm{g}$/ml )를 측정하였으며 대조물질로는 KR-53170과 Cisplatin이 사용되었다. 그 결과 HR-53170의 terminal methyl ketone이 carboxylic acid로 변환된 CW-251001과 이것의 methyl ester인 CW-251002, ethyl ester인 CW-251003, 그리고 morpheline과 수용성으로 N-methylpiperazine, N-methyl-N'-aminopiperazine, 4-Piperidinopiperidine을 결합시켜서 합성한 CW-251011, CW-251012, CW-251013, CW-251014가 in vitro에서 어느 정도 활성을 유지하였다. 특히 이들중 CW-251001, CW-251012, CW-251013, CW-2s1014는 물에 대한 용해도가 상당히 개선되어 in vivo 활성검색을 위한 후보물질로 고려되었다. 하지만 이들중 CW-251001은 in vivo에서 낮은 농도에서는 활성이 거의 없었으며 놓은 농도에서는 독성을 나타내었다.

Novel Vectors for the Convenient Cloning and Expression of In Vivo Biotinylated Proteins in Escherichia coli

  • Biotinylation of recombinant proteins is a powerful tool for the detection and analysis of proteins of interest in a large variety of assay systems. The recent development of in vivo biotinylation techniques in E. coli has opened new possibilities for the production of site-specifically biotinylated proteins without the need for further manipulation after the isolation of the recombinantly expressed proteins. In the present study, a novel vector set was generated which allows the convenient cloning and expression of proteins of interest fused with an N-terminal in vivo biotinylated thioredoxin (TRX) protein. These vectors were derived from the previously reported pBIOTRX vector into which was incorporated part of the pBluescript II+phagemid multiple cloning site (MCS), amplified by PCR using a pair of sophisticated oligonucleotide primers. The functionality of these novel vectors was examined in this system by recombinant expression of rat transforming growth factor-$\beta$. Western-blot analysis using TRX-specific antibodies or peroxidase-conjugated streptavidin confirmed the successful induction of the fusion protein and the in vivo conjugation of biotin molecules, respectively. The convenience of molecular subcloning provided by the MCS and the effective in vivo biotinylation of proteins of interest makes this novel vector set an interesting alternative for the production of biotinylated proteins.

In-vivo Evaluation of Pneumatic Ventricular Assist Device and Durability Improvement (공압식 심실보조장치의 in-vivo 평가 및 내구성 개선)

  • In this paper, we described 23 cases of animal experiment with our pneumatic ventricular assist device and new durability-improvement method. The blood pump consists of blood housing, and back plate made by the injection molding of isoplast, and the diaphragm fabricated by dipping of polyurethane solution onto the aluminum mold. Its volume was 75 $m\ell$ and in-vitro test showed that maximum output was 4.5 $\ell$/min at the 100 mmHg. The adult female sheep with weight of 50 + 10 kg were employed for tile in-vivo experiments and the mean blood flow was sustained at 3.0 1/min. 4 animals survived more than 15 days and the longest survival time was 28 days. In the prior 10 cases, the major causes of death were the tearing of diaphragm at the diaphragm to blood housing junction. By the new mesh and alumina ball milling methods, the durability was enhanced, and its qualitative and quantitative improvement was proved via the in-vivo and in-vitro methods. Animal experiments demonstrated that all the physiologic parameters a ere maintained within the permissible ranges and no thrombus formation was observed through the visual and blood test. The in-vivo experiments demonstrated our pneumatic ventricular assist device to he one month's bridge to transplantation device.

Isolation and in vivo Activities of Antifungal Compounds from Myxococcus sp. JW154 (Myxobacteria). (점액세균 Myxococcus sp. JW154로 부터 항균물질의 분리 및 식물병원균에 대한 in vivo 활성)

  • Two bithiazole-type antibiotics were isolated from the culture broth of a Myxococcus species which isolated from the marine sediment off the coast of Cheju Island, Korea. The structures of these metabolites were determined as KR025 and melithiazole F, previously reported bithiazoles, using combined spectroscopic methods. Both compounds showed an antifungal activity. In in vivo tests, these compounds exhibited potent controlling activities against tomato late blight, wheat leaf rust, and barley powdery mildew with control values more than 80% at a concentraton of 20 $\mu\textrm{g}$/ml.

New record of five anaerobic ciliate species from South Korea

  • During a field survey to report unrecorded Korean ciliates, we collected five anaerobic ciliate species in two samples from freshwater and marine environments. These species belong to the classes Armophorea and Plagiopylea. The morphology of these species was studied based on live observations, protargol impregnation, and scanning electron microscope. Brachonella contorta is characterized by a size of 80-100×40-60 ㎛ in vivo, a bullet-shaped body, and about 60 oral polykinetids making 360° spiral around long axis. Brachonella pulchra is characterized by a size of 80-110×50-70 ㎛ in vivo and adoral zone composed of about 40 oral polykinetids with minimal spiralization. Metopus setosus is characterized by a size of 45-70×20-30 ㎛ in vivo, about 22 ciliary rows, very long caudal cilia, and 20 oral polykinetids. Plagiopyla nasuta is a freshwater species characterized by a size of 80-120×50-60 ㎛ in vivo, striated band present, straight extrusomes, and about 70 somatic ciliary rows. The marine Plagiopyla frontata is characterized by a size of 80-120×40-60 ㎛ in vivo, an oval to ellipsoid body shape, 50-60 somatic ciliary rows, and curved extrusomes.

  • Developing drugs targeting respiratory pathogen is essential to control respiratory diseases. Many experiments have been performed under in vivo situation. However, in vivo experiments have economical and ethical issues. The objective of this study was to determine the possibility of developing an ex vivo lung culture system with possible application for respiratory infection studies. After isolating lungs from naïve pigs, agarose-inflated lung tissues were prepared and sliced manually. These sliced lung tissues were then subsequently placed on 24-well plates. Eight different combinations of media were used to determine the optimum ex vivo lung culture condition. In addition, lung tissues were infected with porcine reproductive and respiratory syndrome (PRRS) virus at a titer of $1{\times}10^4\;TCID_{50}/mL$. Virus growth was confirmed by titration in MARC-145 cells at 2, 4, 6 days post infection (dpi). We found that ex vivo lung culture in physiological environment was not media specific based on histopathology and cytotoxicity. However, under virus-infected condition, thickened alveolar walls in the lung tissues and stable virus titers at 2, 4, 6 dpi were shown in F12K medium suggesting that it was useful for tissue maintenance and virus infection using PRRS virus infected lung tissues. The present study shows the possibility of using porcine ex vivo lung model for respiratory infection studies.

In vivo Evaluation of Flow Estimation Methods for 3D Color Doppler Imaging

  • In 3D ultrasound color Doppler imaging (CDI), 8-16 pulse transmissions (ensembles) per each scanline are used for effective clutter rejection and flow estimation, but it yields a low volume acquisition rate. In this paper, we have evaluated three flow estimation methods: autoregression (AR), eigendecomposition (ED), and autocorrelation combined with adaptive clutter rejection (AC-ACR) for a small ensemble size (E=4). The performance of AR, ED and AC-ACR methods was compared using 2D and 3D in vivo data acquired under different clutter conditions (common carotid artery, kidney and liver). To evaluate the effectiveness of three methods, receiver operating characteristic (ROC) curves were generated. For 2D kidney in vivo data, the AC-ACR method outperforms the AR and ED methods in terms of the area under the ROC curve (AUC) (0.852 vs. 0.793 and 0.813, respectively). Similarly, the AC-ACR method shows higher AUC values for 2D liver in vivo data compared to the AR and ED methods (0.855 vs. 0.807 and 0.823, respectively). For the common carotid artery data, the AR provides higher AUC values, but it suffers from biased estimates. For 3D in vivo data acquired from a kidney transplant patient, the AC-ACR with E=4 provides an AUC value of 0.799. These in vivo experiment results indicate that the AC-ACR method can provide more robust flow estimates compared to the AR and ED methods with a small ensemble size.
