In Vivo Three-dimensional Motion Analysis of the Shoulder Joint During Internal and External Rotation at 90 Degrees of Abduction, using wide Gantry MRI.

  • Despite its importance for the understanding of joint kinematics in vivo, there has been few studies about shoulder joints. The purpose of this study is to analyze the glenohumeral joint during internal and external rotation at 90 degrees of abduction using in vivo noninvasive motion analysis system. MRI was performed for the following seven positions from maximum internal rotation to maximum external rotation at intervals of 30 degrees. We used 3D-gradient echo sequencing (TR: 12 ms, TE: 5.8 ms, 0.8 mm-slice thickness). Our method is based on matching three-dimensional MR images by the similarity of the image intensity. We analyzed the in vivo three-dimensional motions of the glenohumeral and scapulothoracic joint during this motion. In scapla plane, the mean rotation angle of the glenohumeral join was 105.5 degrees ($SD{\pm}39.0^{\circ}$). The mean rotation angle of the scapulothracic joint was 27.5 degrees ($SD\;{\pm}\;7.7^{\circ}$). The contribution ratio is almost 3.8:1 of glenohumeral and scapulothracic joint respectively.

Anticardiovascular Diseases Effects of Fermented Garlic and Fermented Chitosan

  • Garlic is a medicinal plant used throughout the world for its anti-inflammatory, antioxidant, and antiplatelet activities. Chitosan is a natural polysaccharide obtained from chitin, and derivatives of chitosan have been shown to inhibit platelet aggregation and adhesion. We hypothesized that fermented preparations of these products may possess stronger antiplatelet effects than the non-fermented forms owing to the increased bioavailability of the bioactive compounds produced during fermentation. Therefore, we compared these compounds via in vitro and ex vivo platelet aggregation assays by using standard light transmission aggregometry and ex vivo granule secretions from rat platelets. We found that fermented preparations exerted more potent and significant inhibition of platelet aggregation both in vitro and ex vivo. Likewise, ATP release from dense granules of platelets was also significantly inhibited in fermented preparation-treated rat platelets compared to that in non-fermented preparation-treated ones. We concluded that fermented preparations exerted more potent effects on platelet function both in vitro and ex vivo, possibly as a result of the increased bioavailability of active compounds produced during fermentation. We therefore suggest that fermented products may be potent therapeutics against platelet-related CVDs and can be used as antiplatelet and antithrombotic agents.

Inhibitory Effects of Phenolic Alkaloids of Menispermum Dauricum on Gastric Cancer in Vivo

  • The present study was conducted to investigate effects and mechanisms of action of phenolic alkaloids of Menispermum dauricum (PAMD) on gastric cancer in vivo. In vitro, cell apoptosis of human gastric cancer cell line SGC-7901 was observed using fluorescence staining. In vivo, a mice model was constructed to observe tumor growth with different doses. Cell apoptosis was examined using flow cytometry and K-RAS protein expression using Western blotting. The mRNA expression of P53, BCL-2, BAX, CASPASE-3, K-RAS was examined by real-time PCR. PAMD significantly suppressed tumor growth in the xenograft model of gastric cancer in a dose-dependent manner (p<0.01). Functionally, PAMD promoted cell apoptosis of the SGC-7901 cells and significantly increased the rate of cell apoptosis of gastric tumor cells (p<0.05). Mechanically, PAMD inhibited the expression of oncogenic K-RAS both at the mRNA and protein levels. In addition, PAMD affected the mRNA expression of the cell apoptosis-related genes (P53, BCL-2, BAX, CASPASE-3). PAMD could suppress gastric tumor growth in vivo, possibly through inhibiting oncogenic K-RAS, and induce cell apoptosis possibly by targeting the cell apoptosis-related genes of P53, BCL-2, BAX, CASPASE-3.

Texture Analyzer (TA)를 이용한 화장품 크림의 In Vivo 끈적임 평가법의 최적화 (Optimization of In Vivo Stickiness Evaluation for Cosmetic Creams Using Texture Analyzer)

  • 화장품의 사용감을 관계 있는 물성의 측정을 통해 정량화하려는 시도가 이어져오고 있다. 그 중 끈적임은 texture ananlyzer를 이용하여 수직 힘을 측정하는 방식이 대표적이며, 시간에 따른 수직 힘의 그래프에서 음의 면적인 area under curve (AUC)와 상관관계를 갖는 것으로 알려져 있다. 최근 노르망디 대학에서는 이러한 특성에 피부의 특성을 함께 고려하여 TA를 이용한 in vivo 끈적임 평가법을 개발하였다[8]. 본 연구에서는 이를 확장하여 화장품 크림의 in vivo 끈적임 평가법을 최적화하고자 하였다. 페이셜 크림 5 종을 대상으로 크림의 도포량 및 도포 횟수, 탐침의 모양과 소재를 바꾸어 보면서 실험을 진행하였고, 관능 평가 결과를 기준으로 가장 부합하는 조건을 최적의 평가법으로 설정하였다. 그 결과, 3.4 cm의 원 내부에 70 μL의 크림을 7 s 동안 10 회 문지르고 측정하는 방식이 가장 적합한 것으로 판단되었다. 탐침의 경우, 원기둥형보다 구형의 탐침이 재현성이 높게 나타나 구형의 금속 탐침을 택하였다. 최적의 평가법을 확보하여 10 인의 피험자를 대상으로 인체 평가를 진행한 결과, 사람에 따른 절대값에는 차이가 있으나 AUC의 순위는 모두 같게 얻어졌다. 마지막으로 AUC의 끈적임 표준화의 시도로 PVP를 표준 물질로 설정하여 농도 별로 AUC를 측정하고, 5종의 크림 별 끈적임 인지율을 확인하여 AUC와 끈적임의 상관관계에 대해 알아보았다.

Ce 검출기를위한 LLNL 팬텀의가슴벽두께측정 (Chest Wall Thickness Measurement of the LLNL Phantom for Ce Detectors)

  • 한국원자력연구소에서는 사람의 폐에 침착된 방사성핵종의 in vivo 측정을 위해 사용되고 있는 Ge 검출기의 교정을 위해 LLNL 팬텀을 구입하였다. 제작사는 팬팀 위에서 phoswich 검출기의 위치설정을 돕기 위해 팬텀의 몸통덮개판과 이의 오버레이판 위에 동심원을 그려 놓았으며, 그리고 동심원내에서 측정된 팬텀의 가습벽유효두께를 제공하였다. 그러나 Ge 검출기의 면적이 phoswich 검출기보다 작기때문에 phoswich 검출기를 위한 동심원은 한국원자력연구소에서 사용중인 Ge 검출기에는 적합하지 않는 것으로 나타났다. 따라서 본 연구에서는 제작사에 의해 제공된 팬텀의 가슴벽 유효두께가 Ge 점출기에 적합한지, 새로운 자료가 교정에 사용되어야 하는지를 결정하기 위해 팬텀의 가슴벽유효두께를 재평가하였다 그결과 한국원자력연구소에서 새로 설정한 Ge 검출기영역과 phoswich 검출기영역에서의 가습벽유효두께는 다소 차이가 있는 것으로 나타났다.

7, 12-dimethylbenz[a]anthracene(DMBA) 투여에 의한 랫드 유선암 모델에서 ex-vivo 면역치료 효과 (Effects of the ex-vivo Immunotherapy on the Mammary Gland Tumorigenesis Induced by 7, 12-dimethylbenz[a]anthracene(DMBA) in rats)

  • This study was examined on the effect of ex-vivo immunotherapy in 7, 12-dimethylbenz[a]anthracene (DMBA)-induced rat mammary carcinogenesis. Sprague-Dawley female 40 rats were divided into Jour groups. As a positive control, Group I was intubated with DMBA, 5 mg /100 g body weight and single dose, at experimental onset. Group II was treated ex-vivo immunotherapy with polyinosinic-polycytidylic acid (Poly I : C) and Group III was treated with Interleukin-2 (IL-2). Group IV was negative control. All rats were sacrificed at 16 weeks after DMBA intubation. Mammary gland wet weight, dry fat free tissue weight, incidence of tumor, and the number of lobules, alveolar buds, terminal end buds, and terminal ducts were examined. Morphological changes of the mammary gland after treated with DMBA were analyzed by whole mount and histopathological method. As results, the induced mammary tumors of Group I, II and III were 60%, 33% and 0%, respectively. Histopathological types of induced-mammary tumors were adenoma, adenocarcinoma and carcinosarcoma. In analysis of the whole mount method, the number of the terminal end buds, terminal ducts and lobules were significantly lower in Group II (p<0.01) and III (p<0.01) than DMBA alone treated Group I. In microscopic observation, hyperplastic alveolar nodules were significantly lower in Group III than Group I (p<0.01). In conclusion, IL-2 had strong inhibitory effect on the mammary gland tumorigenesis induced by DMBA in rats. Whole mount method may be a useful technique to assess the mammary carcinogenesis. Moreover, hyperplastic alveolar nodules were very sensitive parameter to assess the mammary carcinogenesis.

Identification of Enterococcus faecalis antigens specifically expressed in vivo

  • Objectives: Molecular mechanism of the pathogenicity of Enterococcus faecalis (E. faecalis), a suspected endodontic pathogen, has not yet been adequately elucidated due to limited information on its virulence factors. Here we report the identification of in vivo expressed antigens of E. faecalis by using a novel immunoscreening technique called change-mediated antigen technology (CMAT) and an experimental animal model of endodontic infection. Materials and Methods: Among 4,500 E. coli recombinant clones screened, 19 positive clones reacted reproducibly with hyperimmune sera obtained from rabbits immunized with E. faecalis cells isolated from an experimental endodontic infection. DNA sequences from 16 of these in vivo-induced (IVI) genes were determined. Results: Identified protein antigens of E. faecalis included enzymes involved in housekeeping functions, copper resistance protein, putative outer membrane proteins, and proteins of unknown function. Conclusions: In vivo expressed antigens of E. faecalis could be identified by using a novel immune-screening technique CMAT and an experimental animal model of endodontic infection. Detailed analysis of these IVI genes will lead to a better understanding of the molecular mechanisms involved in the endodontic infection of E. faecalis.

Utility of Structural Information to Predict Drug Clearance from in Vitro Data

  • In the present research, we assessed the utility of the structural information of drugs for predicting human in vivo intrinsic clearance from in vitro intrinsic clearance data obtained by human hepatic microsome experiment. To compare with the observed intrinsic clearance, human intrinsic clearance values for 51 drugs were estimated by the classical methods using in vivo-in vitro scale-up and by the new methods using the in vitro experimental data and selected molecular descriptors of drugs by the forward selection technique together. The results showed that taking consideration of molecular descriptors into prediction from in vitro experimental data could improve the prediction accuracy. The in vitro experiment is very useful when the data can estimate in vivo data accurately since it can reduce the cost of drug development. Improvement of prediction accuracy in the present approach can enhance the utility of in vitro data.

인삼 다당분획의 in vitro, in vivo에서 사염화탄소 유발 간독성에 대한 보호효과 (Protective Effect of Ginseng Polysaccharide Fraction on CCl4-induced Hepatotoxicity in vitvo ana in vivo)

  • Effect of ginseng polysaccharide fraction was examined for $CCl_4$-induced hepatotoxicity in vitro and in vivo. In $CCl_4$-injured primary cultured rat hepatocytes, treatment of the polysaccharide fraction (0.1, 0.3, 1.0 mg/ml) significantly Inhibited the release of LDH and GOT into the culture medium in a dose-dependent manner. Oral administration of the polysaccharide fraction (100, 200 mg/kg) inhibited the decrease of body weight and the increase of the ratio of liver to body weight in $CCl_4$-intoxicated rats. Elevation of GOT, GPT and ALP activity in the serum by $CCl_4$-induced hepatotoxicity was suppressed by administration of ginseng polysaccharide fraction. MDA levels increased in the serum as well as in the liver tissue by treatment with $CCl_4$ showed a tendency to be 연w in the rats given to the polysaccharide fraction. These results suggest that the polysaccharide fraction may be active substance responsible for antihepatotoxic effect of Panax ginseng.

안구 영상을 위한 OCT용 손잡이 형 프로브의 개발 (Development of Hand-held OCT probe for Ophthalmic Imaging)

  • 본 연구에서는 안구 영상을 위한 OCT용 손잡이 형 프로브를 개발하였다. 프로브는 휴대하기 간편한 손잡이 형태로 구현하였으며, 어댑터의 대물렌즈를 교체함으로써 각막(cornea)과 망막(retina) 모두 영상화 할 수 있도록 설계 하였다. 시스템의 성능검증을 위해 in vivo로 실험용 쥐의 눈 영상을 2D로 획득하였으며, 획득한 영상을 3D 영상으로 재구성하였다. 3D 영상은 쥐의 미세한 부분의 구조를 잘 나타내었으며, 움직임에 의한 흔들림 현상(Motion artifact)이 나타나지 않았으므로 개발된 OCT시스템의 이미지 디스플레이 속도는 생체 내 실험 (in vivo)에 적합함을 알 수 있었다.
