Preparation of Silica Coated Zinc Oxide and UV Protection Effect (이산화규소가 코팅된 산화아연의 제조와 자외선 차단 특성)

  • Due to the high UV light scattering effect of Zinc oxide (ZnO), it is frequently used in sunscreen skincare products. Recently ZnO coated with silica has been used in cosmetics to improve UV protection, texture, decreased photocatalytic activity, dispersibility and stability of the skin care product. In this study, we developed a ZnO composite powder coated with silica for the future application to skincare products to block UV rays that could cause photoaging. To improve consumer's satisfaction rating, we used ZnO microparticles which are widely used in the cosmetics industry. The silica was coated using hydrothermal method with sodium silicate and acid hydrolysis. UV protection of the composite powder was analyzed by UV-Vis and in-vitro test and the advantages for practical use of this powder as a skincare product were determined.

Effects of Amomi, Semen Extract on Synthesis of Insulin-like Growth Factor-1 and Anti-wrinkle in Skin (사인추출물의 인슐린 유사 성장인자-3의 합성과 피부 노화 개선에 대한 효과)

  • We screened several materials to stimulate IGF-1 promoter activity using luciferase reporter assay and found that Amomi Semen extract (ASE) among them is the most powerful stimulator We also studied about the anti-wrinkle effect of ethanolic extract of Amoni Semen in vitro and in vivo. Semi-quantitative RT-PCR showed that the extract elevated the presence level of IGF-1 mRNA. And $[^3H]$ proline incorporation and semi-quantitative RT-PCR showed that the extract increased the expression of type-I collagen compared with vehicle in vitro and in vivo, respectively. Significant inhibition of MMP-1 expression was determined by ELISA and Western blot. Finally, topical treatment of the extract on hairless mouse's dorsal skin expanded the volume of collagen and dermal thickness. These results suggest that Amomi Semen may be a good candidate for improving extracellular matrix through the increase of collagen expression and inhibition of MMP-1 expression. Moreover, this study enables us to guess that IGF-1 stimulated by the extract may be involved in the mechanism of anti-wrinkle effect of it.

Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs (한약재 복합 추출물이 항산화 및 항노화에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

Effect of ethanol extract from mixture including Angelicae Dahuricae Radix on Dermal Anti-aging and Whitening (백지를 포함하는 한약재 복합 에탄올 추출물이 피부 항노화 및 미백에 미치는 영향)

  • Objective : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix, and Angelicae Dahuricae Radix. Each of these herbal medicines has studied on anti-aging effect in vitro. So this study was conducted to investigate efficacy and potency of JMB extract on dermal anti-aging and whitening. Methods : The JMB was extracted at room temperature by 80% ethanol. Collagenase and elastase inhibition activity in JMB ethanol extract were determined at 10, 50, 100, 500, 1000 mg/ml concentrations by colorimetric method. The toxic range of JMB ethanol extract was evaluated using MTT assay. Also, The inhibitory effect of JMB ethanol extract on tyrosinase activity and melanin contents in mouse melanoma cell line (B16F10 cell) was identified at 50, 100, 200 ㎍/㎖ levels by spectrometric assay. In each analysis, EGCG (epigallocatechin gallate) and Kojic acid were used as positive controls, respectively. Results : The elastase and collagenase inhibitory activity of JMB ethanol extract increased dose dependently. Also, The MTT assay showed that JMB, up to 400 ㎍/㎖ concentration, exhibited no toxic effect to the B16F10 cell. And following the JMB ethanol extract treat, cellular melanin contents and tyrosinase activity were dose-dependently decreased compared to those of control. Conclusion : These results suggest that JMB ethanol extract has effects to inhibitory activity on dermal wrinkle enzyme and melanogenesis. Therefore, JMB has applicable benefits for development of materials or products to have whitening and anti-aging functions on skin.

Preparation and Characteristics of Multilayer Lamellar Vesicle Using Phosphate Ester Surfactant (Phosphate ester 계면활성제를 이용한 다중층 라멜라 베시클의 제조 및 특성)

  • Various vehicles have been studied in order to protect skin ageing and sustain constantly moisturization. Recently, in relation to maintain of moisture, absorption and penetration of active materials, there has been introducing many preparing methods such as liposome, liquid crystal and multilamellar emulsion. We developed multilayer lamellar vesicle using cetearyl alcohol/ceteth-20 phosphate/dicetyl phosphate as analogy of phospholipid according to variation of shear rate and pH. These multilayer lamellar vesicles were confirmed by cross polarizing microscope. As results, morphologies of lamellar vesicle were not uniformed at low shear rate and pH. Also, stabilities for encapsulation of retinol were observed at $42^{\circ}C$ during two months. As a result, quantitative content of retinol decreased at low pH. Multilayer lamellar vesicle decreased 14% of transepidermal water loss compared with O/W emulsion. We compared multilayer lamellar sun cream to O/W sun cream using in vitro SPF test of water resistance and concluded that multilayer lamellar sun cream is similar to O/W sun cream in water resistance.

Development of Peptides from the Germinated Black Rice and Applications as Cosmetics Ingredients (발아 흑미 유래 펩타이드의 개발과 화장품 응용에 대한 연구)

  • To develop novel anti-aging peptides from the germinated black rice, we treated with bromelain, papain and Pronase E. And we investigated the effects of the germinated black rice peptide (GBRP) as anti-aging cosmetic ingredients, and compared with the non-germinated black rice protein (NBRP). We investigated the effects on in vitro inhibition of matrix-metalloprotease (MMP), proliferation of human skin fibroblasts, stimulation of collagen synthesis and expression of UVA-induced MMPs in human skin fibroblasts, UVA induced MMP-1 expression and collagen contents in human skin fibroblasts were analyzed by enzyme-linked immunosorbent assay (ELISA). As a result, the molecular weight distributions of GBRP and NBRP were determined by gel permeation chromatography to be approximately 900 and 10,000 daltons. GBRP increased skin cell proliferation about 40% and reduced UVA-induced MMP-1 expression about 50%. Also the collagen protein level of cells, which were cultured with GBRP, was increased about 25%. These results suggest that the geminated plant seed peptides can be novel anti-aging ingredients for cosmetics.

In Vitro Cytotoxicity, Skin Regeneration, Anti-wrinkle, Whitening and In Vivo Skin Moisturizing Effects of Oncheongeum (온청음 물 추출물의 세포독성, 피부재생, 주름개선, 미백 및 보습 효과)

  • Objectives: The objective of this study was to evaluate the effects of cytotoxicity, skin regeneration, anti-wrinkle, whitening and skin moisturizing of Oncheongeum (OCE).Methods: The cytotoxicity of OCE lyophilized aqueous extracts (yield=13.82%) was observed against human normal fibroblast cells and B16/F10 murine melanoma cells by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium Bromide (MTT) assay, and skin regeneration and anti-wrinkle effects were also evaluated through the assay of collagen type I synthesis compared to the transformation of the growth factor (TGF)-β1, hyaluronidase, collagenase and matrix metalloproteinase (MMP)-1 inhibitory assays compared to oleanolic acid (OA), and elastase inhibitory effects compared to phosphoramidon disodium salt (PP). In addition, OCE’s whitening effects were measured by a tyrosinase inhibitory assay and melanin formation test in B16/F10 murine melanoma cells compared to arbutin, and skin moisturizing effects were observed through a mouse skin water content test, respectively. Results: No OCE treatment-related cytotoxic effects appeared on human normal fibroblasts and B16/F10 murine melanoma cells. OCE concentration-dependently increased the collagen Type I synthesis on human normal fibroblast cells, and also effectively inhibited hyaluronidase, elastase, collagenase and MMP-1 activities. In addition, OCE inhibited melanin production of B16/F10 murine melanoma cells and activity of tyrosinase. And significant and dose-dependent increases of skin water content were detected in OCE-treated mouse skin compared to vehicle control skins. Conclusions: OCE showed favorable and sufficient effects in skin regeneration, anti-wrinkle, whitening and skin moisturizing in this experiment. But more detail mechanisms and studies on the skin protective efficiency of in vivo are needed with the screening of active biological compounds in individual OCE herbs.

Stimulation of the Extracellular Matrix Production in Dermal Fibroblasts by Areca catechu Extract (진피섬유모세포에서 대복피추출물의 세포외기질 합성 촉진 효과)

  • Dermal fibroblasts produce the many components of the extracellular matrix (ECM) that are needed to maintain connective tissue integrity and repair tissue injuries. This study investigated the effects of Areca catechu extract (ACE) on dermal fibroblast cell activation. Cultured human dermal fibroblasts were treated with ACE, and then ECM production was determined by ELISA, Western blot and RT-PCR. ACE significantly accelerated the production of type 1 collagen, fibronectin, and transforming growth factor (TGF)-${\beta}1$ by ELISA and type 1 collagen by Western blot assay. ACE also increased the gene expression of COL1A1, TGF-${\beta}1$, keratinocyte growth factor (KGF) and insulin growth factor (IGF)-1. These results suggest that ACE has the potential to stimulate ECM production and that it might be suitable for maintaining skin texture.

Fermented Jeju Soybean Extract Promotes Hair Growth in Human Hair Follicle Organ Culture and Clinical Trial (모낭기관배양과 임상에서 제주푸른콩발효추출물의 육모 효능에 미치는 영향)

  • Soybean extract is known to play an important role in preventing and treating diseases associated with aging, cancer, obesity, and cardiovascular disease. A recent has revealed that soybean extract has a potent effect on hair growth in in vitro, in vivo, and clinical studies. Recently, it has been reported that their fermented extracts exhibit numerous and high efficacy, as compared to general extracts. However, the underlying mechanisms that induce hair growth after using fermented soybean extract are not well understood. The present study aimed to determine the effects of fermented Jeju soybean (FJS) extract on hair growth, with a focus on the underlying mechanisms similar to those of minoxidil. We conducted in vitro and ex vivo investigations and clinical studies. FJS extract enhanced dermal papilla cell proliferation, VEGF levels, and potassium channel opening. Moreover, it promoted human hair follicle elongation. These effects were comprehensively demonstrated in the clinical results, in which FJS extract-containing shampoo improved hair density after 24 weeks of utilization. Collectively, the results of this study demonstrate that FJS extract promotes hair growth and inhibits hair loss through a mechanism similar to that of minoxidil in hair follicles.

Inhibitory Effects of Schisandrae Fructus Ethanol Extract on the Production of Matrix Metalloproteinases in in vitro and in vivo Osteoarthritis Models (In vitro 및 in vivo 퇴행성관절염 모델에서 오미자 에탄올 추출물에 의한 matrix metalloproteinases의 생성 억제)

  • Schisandrae Fructus (SF), the fruit of Schisandra chinensis (Turcz.) Baill., is widely used in traditional medicine for the treatment of a number of chronic diseases. SF extracts have been recently reported to attenuate the inflammatory responses in SW1353 human chondrocyte cells in in vitro and monosodium iodoacetate (MIA)-induced cartilage degradation in in vivo osteoarthritis (OA) models. However, their protective and therapeutic potentials against OA in primary culture chondrocytes and animal models remain unclear. Therefore, we investigated the effects of the ethanol extract of SF on the activity of matrix metalloproteinases (MMPs), biomarkers for diagnosis of OA, on interleukin $(IL)-1{\beta}-induced$ primary cultured rat cartilage chondrocytes and MIA-induced osteoarthritis in a rat model. Our data indicated that SF treatment significantly reduced the mRNA expression and enzyme activity of MMP-1, -3 and -13 in $IL-1{\beta}-induced$ primary cultured rat cartilage chondrocytes. The chondro-protective effects of SF were then analyzed in a rat OA model using a single intra-articular injection of MIA in the right knee joint. According to our results, the elevated levels of MMP-1 and -3 were markedly ameliorated by SF administration. Collectively, these findings indicate that SF could be a candidate for the treatment of OA.
