영지버섯 다당체의 마우스 대식세포 면역증강 효과 (Studies on the Immunomodulating Effects of Polysaccharide Extracted from Ganoderma lucidum on Macrophage)

  • 본 연구는 전보의 영지버섯으로부터 분리 배양한 균 사체를 알카리추출하여 얻은 다당체의 일반성분과 당분석 및 항암작용에 관한 연구에 이어, 이들과 ATCC의 마우스 유래 탐식 세포주 TIB 71 및 BALB/c 마우스의 복강내 대식세포를 이용하여 영지버섯 다당체가 면역계 전체의 기능을 조정하는 대식세포의 면역기능에 미치는 영향을 연구하였다. 그 결과를 요약하면 다음과 같다 1. 대식세포의 환성에 영향을 미치는 영지 버섯 다당체의 효과를 알아보기 위하여 세포의 유사분 열능을 측정한 결과, 다당체의 처리 농도가 0.5mg 실험군에서부터 유의한 증가를 나타내었다(p<0.001). 2. 영지버섯 다당체가 대식 세포 활성에 미치는 효과를 알아보기 위해 대식세포의 표면에 존재하는 IL-2 수용체를 anti-mouse IL-2-FITC 단크론항체를 이용하여 측정한 결과는 PSG 0.05mg 실험군을 제외한 실험군 모두에서 대조군 37.6% 보다 현저한 발현증가(p<0.001)를 보였으며 PSG 0.1mg 실험군까지는 수용체가 증가 하나 그 이상 처리군 사이에서는 상호간 유의한 차이는 없었다. 3. 영지버섯 다당체는 대식 세포에서 유래하는 세포간물질 즉 IL-1, IL-6 및 TNF의 생성에 유의성 있는 관여는 하지 않는 것으로 생각되나 부분적으로 영지버섯 다당체와 BCG및 IFN-${\gamma}$를 모두 함께 병용 투여함으로써 각각 영지버섯 다당체와 BCG또는 영지버섯 다당체와 IFN-r을 투여하는 경우에 비하여 IL-1의 분비증가를 보였으며 TNF의 분비 역시 현저한 증가를 보여 영지버섯 다당체가 초기 감염시 숙주의 1차 방어기능에 관여함을 알 수 있었다.

목질진흙버섯 자실체와 배양 균사체 유래 ${\beta}-Glucan$성 다당류의 생리활성 (Biological Activities of Polysaccharide Extracted from the Fruit Body and Cultured Mycelia of Phellinus linteus IY001)

  • 목질진흙버섯 자실체와 배양 균사체의 열수추출 및 열수 추출물의 에탄올, UF처리 분획들에 대한 약리 효과를 조사하여 다음과 같은 결과를 얻었다. Sarcoma 180 암세포 증식 저지율은 자실체 열수 총 분획(Fr. I)보다 비교적 고분자인 다당류 분획(Fr. II, Fr. III, Fr. IV)에서 $80.4{\sim}85.9%$로 높게 나타났다. 면역계에 미치는 영향으로 보체계 활성화는 $10.1{\sim}13.6%$, 대식세포의 NO 생성능은 자실체 및 균사체 모든 분획에서 생성되었으며, $TNF-{\alpha}$생성능은 NO 생성능이 높았던 분획들에서 높은 생성능을 나타내었다. 효소적 및 비효소적 지질과산화 유발에 대한 영향은 목질진흙버섯 자실체와 배양 균사체의 모든 분획에서 지질 과산화 억제능을 나타내었고, 유리기 제거능에 있어서는 농도의존적으로 효과가 있는 것으로 나타났으며, DPPH에 대한 Fr. III의 $SC_{50}$$32.0\;{\mu}g/mL$로 조사되었다. 상기와 같은 결과들로부터 목질진흙버섯 자실체와 배양 균사체의 열수추출 및 열수 추출물의 에탄올, UF처리 분획들은 항암활성, 면역활성, 지질 과산화 억제효과 및 유리기 제거능이 있음을 확인할 수 있었다.

Atheroprotective nasal immunization with a heat shock protein 60 peptide from Porphyromonas gingivalis

  • Purpose: Immunization with Porphyromonas gingivalis heat shock protein 60 (PgHSP60) may have an immunoregulatory effect on atherogenesis. The aim of this study was to determine whether nasal immunization with a PgHSP60 peptide could reduce atherosclerotic plaque formation in apolipoprotein E knockout (ApoE KO) mice. Methods: Seven-week-old male ApoE KO mice were assigned to receive a normal diet, a Western diet, a Western diet and challenge with PgHSP60-derived peptide 14 (Pep14) or peptide 19 (Pep19), or a Western diet and immunization with Pep14 or Pep19 before challenge with Pep14 or Pep19. Results: Atherosclerotic plaques were significantly smaller in mice that received a Western diet with Pep14 nasal immunization than in mice that received a Western diet and no Pep14 immunization with or without Pep14 challenge. An immunoblot profile failed to detect serum reactivity to Pep14 in any of the study groups. Stimulation by either Pep14 or Pep19 strongly promoted the induction of CD4+CD25+ forkhead box P3 (FoxP3)+ human regulatory T cells (Tregs) in vitro. However, the expression of mouse splenic CD4+CD25+FoxP3+ Tregs was lower in the Pep14-immunized mice than in the Pep14-challenged or Pep19-immunized mice. Levels of serum interferon gamma (IFN-γ) and transforming growth factor beta were higher and levels of interleukin (IL) 10 were lower in the Pep14-immunized mice than in the other groups. Induction of CD25- IL-17+ T helper 17 (Th17) cells was attenuated in the Pep14-immunized mice. Conclusions: Nasal immunization with Pep14 may be a mechanism for attenuating atherogenesis by promoting the secretion of IFN-γ and/or suppressing Th17-mediated immunity.

  • Objective : Soybeans of Canavalia gladiata(CG) and root of Arctium lappa(AL) have been reported to have anti-inflammatory, antioxidant effect. However, the immunoregulatory mechanisms of its combinational prescription remain a matter of considerable debate. In the current study, we investigated whether CG and AL and its combinational prescription(CG+AL) regulate immune system using chronic immobilization-stress mouse model. Methods : C57BL/6J mice fixed for 2 hours into immobilization tube after CG, AL, CG+AL oral administration after 2 hours daily for 21 days. After every experiment has ended the C57BL/6J mice were sacrificed on 22 days. The production of Serotonin and Cortisol, lgA were observed by ELISA method, The proportion of immune cells such as T/B cell and macrophage, NK cell were measured by FACS. Then, Real-time PCR was performed to measure the mRNA expression of Inflammatory cytokines(IL-1beta, IL-6, TNF-a) and T cell activation cytokines(IL-2, IL-10, IFN-gamma, IL-12p35 / p40). Result : When chronic immobilization-stress mouse model were treated with CG+AL(1:4), the expression of mRNA were significantly decreased at the Inflammatory cytokines(IL-1beta, IL-6, TNF-a). While, the levels of mRNA were significantly increased at immune T cell activation cytokines. Additionally, CG+AL(1:4) combinational prescription group enhanced immune cells such as T/B cell and macrophage, NK cell. Furthermore, the Immuno-fluorescence result of brain tissue can confirm that CG+AL(1:4) group significantly increased the BDNF expression. Conclusion : These result suggest that CG+AL(1:4) combinational prescription has Immune System enhancement via stress-mediated immunocyte.

The activation of NLRP3 inflammasome potentiates the immunomodulatory abilities of mesenchymal stem cells in a murine colitis model

  • Inflammasomes are cytosolic, multiprotein complexes that act at the frontline of the immune responses by recognizing pathogen- or danger-associated molecular patterns or abnormal host molecules. Mesenchymal stem cells (MSCs) have been reported to possess multipotency to differentiate into various cell types and immunoregulatory effects. In this study, we investigated the expression and functional regulation of NLR Family Pyrin Domain Containing 3 (NLRP3) inflammasome in human umbilical cord blood-derived MSCs (hUCB-MSCs). hUCB-MSCs expressed inflammasome components that are necessary for its complex assembly. Interestingly, NLRP3 inflammasome activation suppressed the differentiation of hUCB-MSCs into osteoblasts, which was restored when the expression of adaptor proteins for inflammasome assembly was inhibited. Moreover, the suppressive effects of MSCs on T cell responses and the macrophage activation were augmented in response to NLRP3 activation. In vivo studies using colitic mice revealed that the protective abilities of hUCB-MSCs increased after NLRP3 stimulation. In conclusion, our findings suggest that the NLRP3 inflammasome components are expressed in hUCB-MSCs and its activation can regulate the differentiation capability and the immunomodulatory effects of hUCB-MSCs.

가미삼령백출산(加味蔘苓白朮散)의 B16 흑색종 암모델에 대한 항종양효과(抗腫瘍效果)와 면역증강효과(免疫增强效果)에 관한 연구(硏究) (The Effect of Gamisamryungbaekchul-san(加味蔘苓白朮散) on the Tumor and Immune Response in Mouse B16 Melanoma Tumor Model)

  • Background : Gamisamryungbaekchul-san(加味蔘?白朮散) is a herbal medicine which has been used for the traditional therapeutic agent of augmentation of the spleen and reinforcement of the Qi. Objective : This Study was performed to investigate the effect of Gamisamryungbaekchul-san on the tumor and immune response in the moose B16 melanoma tumor model. Materials and Methods : The tumor was induced by subcutaneous inoculation of B16BL6 melanoma cells in the shaved dorsal region of mice. Mice were orally administered with Gamisamryungbaekchul-san extract(26.3mg/mouse) for 14days after inoculation. For making examination of antitumor effect, the Increase of life span, Tumor growth inhibition rate, change of body weight were measured and evaluated. For the immune response increasing effect, the percentage of T lymphocyte and B Lymphocyte in the peripheral blood, the percentage of CD4+ T-cell, CD8+ T-cell and CD4+/CD8+ ratio in the peripheral blood and spleen, interleukin-2 productivity were measured and evaluated. Results : Gamisamryungbaekchul-san showed 16.59% increase of life span, 31.64% tumor growth inhibition rate and increase of body weight. Gamisamryungbaekchul-san increased the percentage of T lymphocyte in the peripheral blood, CD4+ T cell percentage of peripheral blood and spleen, and Interleukin-2 productivity as compared with the Control group. Whereas Gamisamryungbaekchul-san had no effect on the percentage of B lymphocyte in the peripheral blood, the percentage of CD8+ T cell, CD4+/CD8+ T-cell ratio in both of peripheral blood and spleen as compared with the Control group. Conclusion : This study shows that Gamisamryungbaekchul-san has anti-tumor effects and immunoregulatory effects on the B16 melanoma tumor model. It is suggested that Gamisarmyungbaekchul-san could be a useful immunomodulator and anti-tumor agent.

Feline adipose tissue-derived mesenchymal stem cells pretreated with IFN-γ enhance immunomodulatory effects through the PGE2 pathway

  • Background: Preconditioning with inflammatory stimuli is used to improve the secretion of anti-inflammatory agents in stem cells from variant species such as mouse, human, and dog. However, there are only few studies on feline stem cells. Objectives: This study aimed to evaluate the immune regulatory capacity of feline adipose tissue-derived (fAT) mesenchymal stem cells (MSCs) pretreated with interferon-gamma (IFN-γ). Methods: To assess the interaction of lymphocytes and macrophages with IFN-γ-pretreated fAT-MSCs, mouse splenocytes and RAW 264.7 cells were cultured with the conditioned media from IFN-γ-pretreated MSCs. Results: Pretreatment with IFN-γ increased the gene expression levels of cyclooxygenase-2, indoleamine 2,3-dioxygenase, hepatocyte growth factor, and transforming growth factor-beta 1 in the MSCs. The conditioned media from IFN-γ-pretreated MSCs increased the expression levels of M2 macrophage markers and regulatory T-cell markers compared to those in the conditioned media from naive MSCs. Further, prostaglandin E2 (PGE2) inhibitor NS-398 attenuated the immunoregulatory potential of MSCs, suggesting that the increased PGE2 levels induced by IFN-γ stimulation is a crucial factor in the immune regulatory capacity of MSCs pretreated with IFN-γ. Conclusions: IFN-γ pretreatment improves the immune regulatory profile of fAT-MSCs mainly via the secretion of PGE2, which induces macrophage polarization and increases regulatory T-cell numbers.

Recombinant Human Bone Morphogenetic Protein-2 Priming of Mesenchymal Stem Cells Ameliorate Acute Lung Injury by Inducing Regulatory T Cells

  • Mesenchymal stromal/stem cells (MSCs) possess immunoregulatory properties and their regulatory functions represent a potential therapy for acute lung injury (ALI). However, uncertainties remain with respect to defining MSCs-derived immunomodulatory pathways. Therefore, this study aimed to investigate the mechanism underlying the enhanced effect of human recombinant bone morphogenic protein-2 (rhBMP-2) primed ES-MSCs (MSCBMP2) in promoting Tregs in ALI mice. MSC were preconditioned with 100 ng/ml rhBMP-2 for 24 h, and then administrated to mice by intravenous injection after intratracheal injection of 1 mg/kg LPS. Treating MSCs with rhBMP-2 significantly increased cellular proliferation and migration, and cytokines array reveled that cytokines release by MSCBMP2 were associated with migration and growth. MSCBMP2 ameliorated LPS induced lung injury and reduced myeloperoxidase activity and permeability in mice exposed to LPS. Levels of inducible nitric oxide synthase were decreased while levels of total glutathione and superoxide dismutase activity were further increased via inhibition of phosphorylated STAT1 in ALI mice treated with MSCBMP2. MSCBMP2 treatment increased the protein level of IDO1, indicating an increase in Treg cells, and Foxp3+CD25+ Treg of CD4+ cells were further increased in ALI mice treated with MSCBMP2. In co-culture assays with MSCs and RAW264.7 cells, the protein level of IDO1 was further induced in MSCBMP2. Additionally, cytokine release of IL-10 was enhanced while both IL-6 and TNF-α were further inhibited. In conclusion, these findings suggest that MSCBMP2 has therapeutic potential to reduce massive inflammation of respiratory diseases by promoting Treg cells.

The role of RNA epigenetic modification-related genes in the immune response of cattle to mastitis induced by Staphylococcus aureus

  • Objective: RNA epigenetic modifications play an important role in regulating immune response of mammals. Bovine mastitis induced by Staphylococcus aureus (S. aureus) is a threat to the health of dairy cattle. There are numerous RNA modifications, and how these modification-associated enzymes systematically coordinate their immunomodulatory effects during bovine mastitis is not well reported. Therefore, the role of common RNA modification-related genes (RMRGs) in bovine S. aureus mastitis was investigated in this study. Methods: In total, 80 RMRGs were selected for this study. Four public RNA-seq data sets about bovine S. aureus mastitis were collected and one additional RNA-seq data set was generated by this study. Firstly, quantitative trait locus (QTL) database, transcriptome-wide association studies (TWAS) database and differential expression analyses were employed to characterize the potential functions of selected enzyme genes in bovine S. aureus mastitis. Correlation analysis and weighted gene co-expression network analysis (WGCNA) were used to further investigate the relationships of RMRGs from different types at the mRNA expression level. Interference experiments targeting the m6 A demethylase FTO and utilizing public MeRIP-seq dataset from bovine Mac-T cells were used to investigate the potential interaction mechanisms among various RNA modifications. Results: Bovine QTL and TWAS database in cattle revealed associations between RMRGs and immune-related complex traits. S. aureus challenged and control groups were effectively distinguished by principal component analysis based on the expression of selected RMRGs. WGCNA and correlation analysis identified modules grouping different RMRGs, with highly correlated mRNA expression. The m6 A modification gene FTO showed significant effects on the expression of m6 A and other RMRGs (such as NSUN2, CPSF2, and METTLE), indicating complex co-expression relationships among different RNA modifications in the regulation of bovine S. aureus mastitis. Conclusion: RNA epigenetic modification genes play important immunoregulatory roles in bovine S. aureus mastitis, and there are extensive interactions of mRNA expression among different RMRGs. It is necessary to investigate the interactions between RNA modification genes regulating complex traits in the future.

인체대장암세포주 SW480에 대한 재배 와송의 항암효과 연구 (Anti-cancer Effects of Cultivated Orostachys japonicus on Human Colon Cancer Cell Line SW480)

  • 와송(둥근바위솔, Orostachys japonicus, OJ)은 면역조절, 항노화, 항산화, 독성제거 등의 치료적 효과를 가진 약용식물로 알려져 있는데, 본 연구에서는 인체대장암세포에 대한 재배 와송의 항암효과를 규명하고자 하였다. 인체대장암세포주 SW480에 와송열수추출물을 72시간 동안 처리한 결과, 대장암세포의 생존율은 농도-의존적으로 유의하게 감소하였다. 또한, 와송은 SW480 세포의 증식과 이주를 억제하는 것으로 나타났는데, 대조군의 스크래치 gap은 24시간부터 유의하게 감소하는 반면, 와송열수추출물을 처리한 실험군 I과 II의 스크래치 gap은 48시간부터 감소하기 시작하였다. 특히 실험군 I의 스크래치 gap은 72시간까지 유의하게 유지되었다. 와송이 생체 내에서 종양의 형성에 어떠한 영향을 미치는지 알아보기 위하여 대장암세포 주사 전, 31일 동안 수컷 C57BL/6 마우스(4주령)에게 와송열수추출물을 경구로 자유 섭취하게 하였다. 이후 SW480 세포($1{\times}10^7cells/100{\mu}l$)를 피하로 주입한 후 7일, 14일 그리고 28일에서 종양의 형성을 관찰하였고, 각 실험동물을 희생하여 종양의 무게와 부피($mm^3$)를 측정하여 비교 분석하였다. 와송열수추출물을 섭취하는 동안 실험군 I, II의 체중은 대조군에 비해 지속적으로 유의하게 증가하였다. SW480 세포 주입 이후 모든 실험군의 체중은 감소하였으나, 세포 주입 후 14일부터 와송 섭취 실험군의 체중은 유의하게 증가하는 것으로 나타났다. 대장암세포 주입 후 7일과 14일에서 와송을 섭취한 실험군의 종양 무게와 부피는 대조군보다 높았으나, 28일에서는 대조군보다 낮게 나타났고, 특히 실험군 II에서 종양 무게와 부피는 유의하게 감소하는 것으로 확인되었다. 이상의 결과를 통해 와송이 인체 대장암세포의 성장, 증식 및 이주를 억제하며, 생체 내 종양형성(tumorigenesis)을 예방적으로 억제함을 알 수 있었다. 따라서, 재배 와송은 천연 항암제 소재로서 활용될 가능성을 가지는 것으로 보이며, 이에 대한 심층적인 연구가 필요할 것으로 사료된다.
