Antibody Responses in Sera of Different Mouse Strains Experimentally Infected with Neodiplostomum seoulense

  • To examine humoral immune responses in the host, we measured serum antibody levels in different strains of mice (ICR, BALB/c, and C3H) experimentally infected with Neodiplostomum seoulense. Specific IgG antibody levels were increased remarkably with little difference among 3 strains of mice infected with N. seoulense from day 7 to 35 post-infection. More target proteins of adult parasites reacted with IgG at the time when the worm recovery decreased compared with other times. More than 20 protein bands, from 14 kDa to 94 kDa in size, were separated from the crude antigen of N. seoulense adults by SDS-PAGE, and among them 26, 30, 35, 43, 54, 67, and 94 kDa proteins were the major antigenic proteins. The results suggest that significant IgG antibody responses occur against N. seoulense in mice and this may be related with expulsion of worms.

  • Two proteases were partially characterized from culture filtrate of Mycobacterium, tuberculosis KIT110. Their molecular weights were approximately 200 and 180 kDa, respectively and they exhibited similar enzymatic characteristics. These enzymes were inhibited significantly by EDTA and to some extent by EGTA. Their activity was enhanced by $Ca^{2+}$ and $Mg^{2+}$ to some degree. However, $Cu^{2+}$ and $Ag^{2+}$ completely inhibited the enzyme activity at the concentration of 2.5 and 5 mM, respectively. The optimal pH was 7.0 and optimal temperature was around $40^{\circ}C$. These enzymes were rapidly inactivated at $80^{\circ}C$. Therefore, they were heat-labile, neutral metalloproteases. These enzymes exhibited antigenicity shown by their reacting with sera from the partients with pulmonary tuberculosis. These enzymes were able to degrade serum proteins including hemoglobin, bovine serum albumin, lysozyme and immunoglobulin G and structural matrix protein such as type I collagen. Therefore, these enzymes may be thought to contribute to tissue necrosis and pathogenesis during infection.

Production and characterization of monoclonal antibodies (MAb) against flounder serum immunoglobulin (Ig)

  • Specific polyclonal and/or monoclonal antibodies (MAbs) to immunoglobulins (Igs) and their subunits have proved to be valuable tools in immunological research and in immunological assays. In this study, we developed and characterized MAbs against flounder serum Igs. To obtain the pure flounder serum Igs, mouse IgG (mIgG) was immunized to flounder. Flounder Igs were purified by using mIgG-agarose affinity column chromatography. The structure of purified flounder Ig was observed, on denatured SDS-PAGE, to be composed of two heavy chains (77 and 72 kd) and two light chains (28 and 26 kd). MAbs were produced by fusion of myeloma cells (SP2/0) with Balb/c mouse spleen cells previously primed with the flounder Igs. Finally, three hybridoma clones, FIM 511, FIM 519 and FIM 562 were established to recognize both 2 heavy chains, 26 kd of light chain and 28 kd of light chain, respectively. On the other hand, the flounder immune sera collected on the weekly basis were tested on ELISA and immunoblot analysis whether boosting effect is present in flounder humoral immune system. As a result, the secondary immune response in flounder was ascertained on ELISA, but not on immunoblot analysis. Further, we observed an alteration of serum protein levels following immunization. Our MAbs and basic information on flounder humoral immune system obtained in this study will be helpful to control and monitor the efficiency of fish vaccines and therapeutic process of flounder diseases.

Intracellular Responses of Antibody-Producing H69K-NGD Transfectoma Subjected to Hyperosmotic Pressure

  • When subjected to hyperosmotic pressure by NaCl addition, H69K-NGD transfectoma, like KR12H-2 transfectoma, displayed decreased specific growth rate (${\mu}$) and increased specific antibody productivity ($q_{Ab}$): Elevation of medium osmolality from 280 mOsm/kg to 415 mOsm/kg decreased ${\mu}$ by $79\%$ in batch cultures of H69K-NGD transfectoma, while it increased $q_{Ab}$ by $103\%$. However, unlike KR12H-2 tranfectoma, enhanced $q_{Ab}$ of H69K-NGD transfectoma at hyperosmolalities was not due to elevated levels of Ig mRNAs. In hyperosmotic cultures of H69K-NGD transfectoma, heavy-chain mRNA per cell was not enhanced with increasing osmolality. Hyperosmotic pressure was found to preferentially enhance immunoglobulin (Ig) translation rates of H69K-NGD transfectoma. However, under hyperosmotic pressure, the translation rate of Ig polypeptides was not enhanced as much as $q_{Ab}$. This result suggests that hyperosmotic pressure also influences the post-translational process. Taken together, the results obtained show that intracellular response of transfectomas to hyperosmotic pressure, in regard to the main intracellular steps of the antibody secretory pathway, is cell-line dependent.

Analysis of Biological Experiment on Socheongryong-tang (Xiaoqinglong-tang) (소청룡탕 효능에 관한 기초 실험 연구 문헌 분석)

  • Objectives: To investigate the scientific evidence of Korean Medicine (KM), papers on Socheongryong-tang (Xiaoqinglong-tang), which is frequently used in medical clinics or hospitals of Korean medicine, were collected and analyzed. Methods: Papers were classified by the registration of domestic or international journals, the year of publication, experimental models and the subjects of biological activities. The mechanisms of biological activity in accordance with therapeutic effects of Socheongryong-tang (Xiaoqinglong-tang) were noted. Results: Among 98 papers included, 21 were published in domestic journals whereas 35 were in Chinese journals and 43 in Japanese journals. Most reported biological activities were amelioration of asthma. Socheongryong-tang (Xiaoqinglong-tang) regulated interleukin and interferon and immunoglobulin, inhibited the production of nerve growth factor, endotheliln-1, nitric oxide, toll-like receptor-4, p-Akt and increased extracellular signal regulated kinase and cyclin D1, which led to decreased bronchi inflammation and bronchoconstriction, and inhibited the proliferation of airway smooth muscle cells, mucus secretion and airway hyperresponsiveness. In addition, Socheongryong-tang (Xiaoqinglong-tang) also restored tissues injured by asthma so that respiratory function recovered. Conclusions: Amelioration of asthma by Socheongryong-tang (Xiaoqinglong-tang) is supported by objective and scientific evidence.

Vaccines against periodontitis: a forward-looking review

  • Periodontal disease, as a polymicrobial disease, is globally endemic as well as being a global epidemic. It is the leading cause for tooth loss in the adult population and has been positively related to life-threatening systemic diseases such as atherosclerosis and diabetes. As a result, it is clear that more sophisticated therapeutic modalities need to be developed, which may include vaccines. Up to now, however, no periodontal vaccine trial has been successful in satisfying all the requirements; to prevent the colonization of a multiple pathogenic biofilm in the subgingival area, to elicit a high level of effector molecules such as immunoglobulin sufficient to opsonize and phagocytose the invading organisms, to suppress the induced alveolar bone loss, or to stimulate helper T-cell polarization that exerts cytokine functions optimal for protection against bacteria and tissue destruction. This article reviews all the vaccine trials so as to construct a more sophisticated strategy which may be relevant in the future. As an innovative strategy to circumvent these barriers, vaccine trials to stimulate antigen-specific T-cells polarized toward helper T-cells with a regulatory phenotype (Tregs, $CD_{4+}$, $CD_{25+}$, $FoxP_{3+}$) have also been introduced. Targeting not only a single pathogen, but polymicrobial organisms, and targeting not only periodontal disease, but also periodontal disease-triggered systemic disease could be a feasible goal.

Effects of Socheongryong-Tang on Immunoglobulin Production in Asthmatic Mice (소청용탕(小靑龍湯)이 천식(喘息)이 유발된 생쥐의 면역 글로불린 분비에 미치는 영향)

  • Objectives : Socheongryong-Tang(小靑龍湯, SCRT), a herbal remedy, has been widely used to treat respiratory disease such as cough and asthma in Oriental countries. Recent years SCRT was known as anti-allergic agent. However, its therapeutic mechanisms including immunoglobuline such as IgE, IgG1, IgG2a productions are unclear. Methods : We investigated the effects of SCRT on levels of antigen specific total antibody, IgE, IgG1, IgG2a using ELISA method in serum from allergen-induced asthma mice. Results : SCRT decreased level of antigen specific IgE significantly. And SCRT treated mice showed downward tendency of IgG1, a Th1 relative antibody, level. But, SCRT did not affect levels of antigen specific total antibody and IgG2a, a Th1 relative antibody. Conclusions : we demonstrated the strong possibility of SCRT as a complementary or alternative drug to western drug also demonstrated that regulation of Th1/Th2 imbalance may be one of mechanism contributed to treatment for respiratory disease by SCRT.

Association of the 94 KDa Glucose-regulated Protein with Immunoglobulin Heavv Chain Binding Protein (BiP) (94 KDa Glucose-regulated Protein의 BiP과의 결합)

  • The 94 KDa glucose-resulsted Protein (SH 94), one of stress Proteins, is a Ca2+-binding protein in the endoplasmic reticulum (ER). In this study, the possible effect of Ca2+ on the native conformation of grp 94 was examined. When the purified grp 94 was analyzed by Sel filtration in the presence of either EGTA or CaCl2, it was eluted with apparent molecular weight (MW) of 100 KDa in both cases. When similarly analyzed with microtome or cell Ivsate, however, srp 94 was eluted with apparent IW of 200 KDa in the presence of E6TA, while with apparent MW of 100 KDa in the presence of CaCl2, indicating possible association of grp 94 with one or more other proteins in the absence of CaCl2. Consequently, immunoprecipitation with anti-grp 94 was carried out to determine which proteins specifically interact with grp 94. It is sho%un that srp 94 may interact, in a Ca2+_dependent manner. with other proteins including BiP (grp 78) which is also a stress protein in the ER.

Scutellaria baicalensis Modulates Cytokine Production, T Cell Population and Immunoglobulin Level by Mesenteric Lymph Node Lymphocytes in Experimental Mice with Colitis (한약재인 황금의 궤양성 실험동물에 대한 장간막 임파절 임파구의 면역글로블린 수준, T세포집단, 사이토카인 생성의 조절작용)

  • We previously examined extracts, isolated from Scutellaria baicalensis (SB), chemical mediators, and IgE by mesenteric lymph node (MLN) lymphocytes in rats. The present study was to evaluate the effects of extracts of SB on the MLN lymphocytes function of mice given orally by 20 mg/kg for 2 weeks with dextran sulfate sodium (DS)-induced colitis. Results show that IgE levels in MLN lymphocytes was low, while IgA was high, in mice given SB compared to that fed water. Concentrations of $Inteferon-{\gamma}$ and interleukin (IL)-2 of T cells by concanavalin A treatment was significantly higher in the SB fed group than the normal group. Activation-induced IL-4 and IL-10 secretion was lower in SB fed mice compared control mice after DS-induced colitis. These results suggested that SB suppresses the inflammation in DS-induced colitis through the modulation of Th1/Th2 balance to down-regulate $Th_2$ response in MLN lymphocytes.

A Study on the Effect of Conjugated Linoleic Acid on Immunosuppressed Mice with Corticoids

  • This study was performed to investigate the immunopotentiative effects of conjugated linoleic acid on mice immunosuppressed by administratin corticoids. Mice were divided into four groups of 8 mice. Two groups (C, CP) were given diet supplemented with 1% linoleic acid (CLA) and the other two groups (L, LP) were given diet supplemented with 1% linoleic acid (LA) instead of CLA. Prednison was administered to two groups (CP, LP) for immune depression. After feeding diets for 3 weeks containing PDS injection for last 1 week. Serum and gut lumen lavage were taken. Measurement of total Ig were executed using sandwich ELISA. Serum levels of IgA, IgG, and IgM showed some trend which groups fed with CLA were higher than groups fed with LA while IgE was reduced in those fed the CLA intake, and groups administered with PDS were lower than groups administered with saline. However, no significant differences were seen in the proportion of total immunoglobulin in serum. In case of secretory IgA, Group C and CP were significantly higher than group L and LP. Especially between CP and LP, it can be seen effects of CLA. In addition that the CLA treated group weighted a significantly lower level than the one's that have not been treated with CLA. These result support the view that CLA potentiate the immune response and prevent immune depression caused by administrating of corticoids. In conclusion, CLA produced a situation favorable for immunopotentiative effects. Thus, the clinical application of CLA is warranted.
