Effects of Dietary Combinations of Vitamin A, E and Methionine on Growth Performance, Meat Quality and Immunity in Commercial Broilers

  • The experiment was conducted to study the effect of dietary combinations of vitamin A (VA), vitamin E (VE) and methionine (Met) on growth performance, meat quality and immunity in commercial broilers. Ross chicks (n=3,630) were allocated to five experimental treatments with three replicates per diet. The dietary treatments were: VA 8,000 IU, VE 10 IU (diet 1); VA 12,000 IU, VE 10 IU (diet 2); VA 8,000 IU, VE 100 IU (diet 3); VA 12,000 IU, VE 100 IU (diet 4) and; VA 12,000 IU, VE 100 IU/kg diet and 20% Met higher than other groups (diet 5). The Met content in diet 1 to diet 4 were as per the requirement suggested by NRC. Separate vitamin premixes were prepared for each treatment diet as per the requirement of study. The 35 d study revealed significantly (p<0.0001) higher weight gains in broilers fed diet 3 and diet 5, than in the rest of the groups during starter phase (0-3 weeks) only. The feed intake did not vary significantly at all phases of study, but feed efficiency was significantly (p<0.05) lower in diet 1 during starter and overall phase (4-5 weeks). The bone strength and bone composition, except bone calcium, remained unaffected due to experimental diets studied after 35 d of experimental feeding. The thio-barbituric acid reactive substances (TBARS) were significantly (p=0.0013) lower in the breast meat in group 5, followed by group 3, than in the rest of the groups. The immune studies conducted, antibody titers to sheep red blood cells, thickness index to phytohaemagglutinin-P, and heterophil: lymphocyte ratio, did not show any significant difference among treatments. It could be concluded that supplementation of VA, VE and Met at higher levels could be beneficial to broilers only during the starter phase.

  • It is noted that Dioscorea quinqueloba is a medicinal herb that is widely used to treat cardiovascular disease and is assessed as useful to treat other various medical conditions. The immunopotentiating effects of the protein extract (DQP-1) from Dioscorea quinqueloba were thus formally investigated in vivo under incident of cold stress. In this case study, the spleen and thymus weight in mice was shown to have decreased after a measured exposure to cold stress, while the adrenal gland weight in the mice was shown to have increased. The systematic oral administration of DQP-1 significantly recovered the weight loss of the spleen and suppressed the adrenal gland hypertrophy during the association with cold stress. Additionally, the DQP-1 also restored the ascorbic acid level in the adrenal gland reduced after cold stress. The cold stress exposure lowered the percentage of $CD4^+$ and $CD8^+$ cells in the mouse thymus as determined by the flow cytometric analysis, as well as the levels of some serum immunological cytokines(interleukin-2, interleukin-12, and interferon-${\gamma}$) in the studied mice. The resulting identified weakened immunity caused by cold stress was also recovered by a treatment with DQP-1. The DQP-1 significantly suppressed the formation of serum enzymes of alanine aminotransferase, aspartate aminotransferase, and lactate dehydrogenase, which were systematically elevated during the cold stress episode. These results indicate that DQP-1 can improve immunity in mice that are characteristically weakened under stress.

손 반사요법이 말기 신부전증 환자의 생기와 면역 반응에 미치는 영향 (The Effects of Hand Reflexology on Saeng-Chi and Immunity in ESRD Patients)

  • The purpose of this study was to identify the Effect of Hand Reflexology on Saeng-chi of physiologic, emotional & motivational responses and Immunity in ESRD patients who received hemodialysis in two general hospital from June to September. 2001 A two group quasi-experimental research with pre and post test design was used. The number of participants in the experimental group was 23, and in the control group, 20 The Hand Reflexology Intervention was developed by the researcher based on hand reflexology by Carter & Weber and Chi-massage by Chia. The Hand Reflexology was applied to both hands for 10 minutes per day. and 5 days by 5 times. To evaluate the effects of the program, Pre and Post evaluations were done. In the physiologic response, the PR was decreased at the 1st times post treatment and at the 5th. In addition BP was decreased at the 1st time, but not the 5th. After 5 treatments, there were significant increase in Hb and significant decreases in the BUN and Cr. levels in the experimental group. In the emotional and motivational responses, there were significant increases in vigor, mood, uplifts and self care agency scores in the experimental group, but there was no significant difference in the Rosenberg's self esteem score. In the experimental group, significantly increased CD4, and h/s ratios were found, also NK cells were significantly decreased, and there was a decrease in the CD8. However, no significant differences between groups were observed. There were significant increases in CD32, CD33, CD34 in the experimental group. The self care agency score correlated negatively with the CD8. From the above results, Hand Reflexology is shown to be an effective mind-body nursing intervention for enhancing Saeng-chi responses and affecting some of the immune responses. However, Immune cell activation and differentiation with hand reflexology will be achieved with future study.

Effects of Cochlodinium polykrikoides on Oxidative Status and Immune Parameters in the Marine Medaka Oryzias javanicus

  • In this study, the marine medaka Oryzias javanicus was exposed to two concentrations of non-toxin-producing red tide dinoflagellate C. polykrikoides (1,000 and 2,000 cells ml-1) for 96 h, and the time-course biochemical responses of antioxidant and immunity parameters were analyzed in the liver tissue. Significant ichthyotoxicity with increasing cell concentrations of C. polykrikoides and exposure period was observed for 96 h. Opercular respiratory rate was lowered in marine medaka exposed to 2,000 cells ml-1 of C. polykrikoides. Intracellular malondialdehyde (MDA) content significantly elevated in response to both cell concentrations. In the case of glutathione (GSH) content, the levels were significantly elevated by 1,000 cells ml-1 of C. polykrikoides, but the contents significantly depleted upon exposure to 2,000 cells ml-1 of C. polykrikoides. Similarly, enzymatic activities of catalase (CAT) and superoxide dismutase (SOD) were increased by 1,000 cells ml-1 of C. polykrikoides, whereas their activities were lowered by 2,000 cells ml-1 of C. polykrikoides. Analysis of the two immunity parameters, alternative complement pathway and lysozyme, showed significantly lowered activities in 2,000 cells ml-1 of C. polykrikoides-exposed liver tissue. These biochemical effects of C. polykrikoides on marine medaka would be helpful for understanding its acute effects in marine fish.

흰다리새우(Penaeus vannamei) 사료 내 괭생이모자반(Sargassum horneri) Celluclast 가수분해물 이용성 평가 (Evaluation of Dietary Supplementation of Sargassum horneri Celluclast Hydrolysate for Pacific White Shrimp Penaeus vannamei)

  • This study investigated the effects of dietary supplementation with Sargassum horneri celluclast hydrolysate (SHCH) on the growth performance, feed utilization, non-specific immunity and antioxidant capacity of the Pacific white shrimp Penaeus vannamei. Six diets were prepared by SHCH supplemention levels of 0, 5, 10, 20, 40 and 80 g/kg (designated as Con, S5, S10, S20, S40 and S80, respectively). A total of 540 shrimp were randomly distributed in triplicate across 18 tanks and fed one of the diets for five weeks. Weight gain did not differ significantly among the groups except for S80. The feed conversion ratio was significantly lower in S10 than in S80. Phenoloxidase and nitro-blue tetrazolium activities were significantly higher in S5 than in Con. The prophenoloxidase gene was significantly upregulated in S10 compared to that in Con. The gene expression of crustin, glutathione peroxidase, lipopolysaccharide and β-1,3-glucan-binding protein were significantly upregulated in S5 compared to Con. The superoxide dismutase gene was significantly upregulated in S20 and S80 compared to Con. Supplementing shrimp diets with up to 8% of SHCH did not impair non-specific immunity or antioxidant capacity. However, based on growth results, supplementation up to 4% was considered appropriate.

Immunity of the Buffalo Mammary Gland during Different Physiological Stages

  • To study the immunity of the buffalo mammary gland during involution and around parturition and compare it with the mastitic mammary gland, milk samples were collected from 9 Murrah buffaloes during the above critical periods. SCC of buffalo milk increased significantly (p<0.01) by day 21 of involution and one week prepartum. SCC was significantly higher around parturition but became normal at 14 days postpartum. Phagocytic activity (PA) and phagocytic index (PI) of the buffalo milk neutrophils decreased as the duration of the dry period increased. Elevated levels of immunoglobulins at calving improved the PA and PI, but the lowest PA of 18.8% and PI of 1.75 were recorded at 7 days postpartum. Buffaloes suffering from clinical mastitis had PA of 12.3% and PI of 1.46 that increased significantly (p<0.01) on the third day of treatment. Distance of teat from ground level was found to be minimum at one week before parturition. The investigation showed that in vitro phagocytic activity of buffalo neutrophils was weakest at one week postpartum.

사료 중 크릴 분말이 살모넬라 LPS로 자극한 브로일러의 비장세포와 PBMC 증식에 미치는 영향

  • 본 연구는 사료 중 크릴밀이 브로일러의 비장세포와 PBMC의 증식도에 미치는 영향을 조사하였다. 2주령 비장세포 증식도는 사료 중 크릴의 함량에 따라 증가하나, LPS주입으로 감소하였다. 콘카나발린 A(Con A) 첨가는 급여사료나 LPS 자극에 관계없이 증식도를 증가시켰다. 21일령 브로일러 비장세포의 증식도는 사료 중 크릴의 함량에 따라 높아졌다. LPS 스트레스로부터 회복중인 브로일러 비장세포의 증식도는 낮았다. 21일령에 PBMC의 증식도는 사료 중 크릴의 영향이 없었으나, LPS 자극에서 회복중인 브로일러의 증식도는 높아졌다. Con A 첨가는 급여사료나 LPS 자극에 관계없이 PBMC의 증식도를 높였다. 본 성적은 사료 중 크릴밀 첨가가 브로일러 비장세포 및 PBMC 증식에 영향을 미친다는 것을 나타낸다.

CaO (oxide calcium)가 첨가된 사료 공급이 넙치(Paralichthys olivaceus)에 미치는 생리적 변화에 대한 연구 (The Physiological Changes in the Olive Flounder Paralichthys olivaceus Caused by a Diet Containing CaO)

  • This study investigated the physiological effects of a diet containing calcium oxide (CaO) on the olive flounder, Paralichthys olivaceus. The results indicate that the amount of calcium in the blood was higher in the calcium group compared with the controls. The aspartate amino transferase (AST) and alanine amino transferase (ALT) levels were lower in the calcium group, while lysozyme activity was higher in the calcium group. Histologically, fish in the calcium group had more hepatocytes in the liver and more intact scales. In addition, an attack experiment using Vibrio anguillarum (KCTC-2711) and Edwardsiella tarda (KCTC-3657) showed that the survival was higher in the calcium group compared with the controls. These results suggest that feeding fish CaO when they require it, such as during periods of gonadal maturation and rapid growth, will increase their immunity and resistance to pathogens.

Gamma Irradiation-reduced IFN-γ Expression, STAT1 Signals, and Cell-mediated Immunity

  • The signal transducer and activator of transcription (STAT)1 is a cytoplasmic-transcription factor that is phosphorylated by Janus kinases (Jak) in response to interferon $\gamma$ (IFN-$\gamma$). The phosphorylated STAT1 translocates to the nucleus, where it turns on specific sets of IFN-$\gamma$-inducible genes, such as the interferon regulatory factor (IRF)-1. We show here that gamma irradiation reduces the IFN-$\gamma$ mRNA expression. The inhibition of the STAT1 phosphorylation and the IRF-1 expression by gamma irradiation was also observed. In contrast, the mRNA levels of IL-5 and transcription factor GATA-3 were slightly induced by gamma irradiation when compared to the non-irradiated sample. Furthermore, we detected the inhibition of cell-mediated immunity by gamma irradiation in the allogenic-mixed lymphocytes' reaction (MLR). These results postulate that gamma irradiation induces the polarized-Th2 response and interferes with STAT1 signals, thereby causing the immunosuppression of the Th1 response.

  • The task force of the Korean Society for Reproductive Immunology recommends intravenous immunoglobulin G treatment in women with reproductive failure, including recurrent pregnancy loss and/or repeated implantation failure, who show cellular immune factors such as abnormal natural killer cell levels, natural killer cell cytotoxicity, and/or type 1 T helper immunity.
