생약복합조성물(HemoHIM)의 사람 비만세포주 활성 억제 효과 (Inhibitory Effects of a Herbal Composition (HemoHIM) on the Activation of Human Mast Cell Line (HMC-1))

  • 방사선에 대한 방호와 면역기능 조절을 목적으로 새로운 생약복합조성물인 HemoHIM을 개발하였다. 식품 원료로 사용 가능한 생약재 3종 당귀, 천궁, 백작약의 에탄올 분획을 열수추출물에 첨가하여 HemoHIM을 제조하였다. HemoHIM의 항알레르기 효과를 검증하기 위하여 사람 비만세포주 HMC-1을 사용해 compound 48/80으로 유도되는 히스타민 분비량과 PMA/A23187로 유도되는 염증성 사이토카인의 분비량을 측정하였다. 히스타민의 양은 형광분석법으로, 염증성 사이토카인 IL-6, IL-8, TNF-$\alpha$, GM-CSF의 양은 효소결합 면역측정법으로 측정하였다. 저농도의 HemoHIM에 의해 히스타민 분비량이 억제되었고, 모든 농도에서 IL-6, TNF-$\alpha$, GM-CSF의 분비량은 억제되었지만 IL-8은 고농도에서만 억제되었다. 사이토카인의 mRNA 발현량은 HemoHIM의 농도 의존적으로 억제되었다. 그리고 c-kit와 Fc$\varepsilon$RI의 mRNA 발현량도 모든 농도에서 억제되었지만, tryptase의 mRNA 발현량은 저 농도에서만 억제되었다. 이상의 결과로 HemoHIM이 비만세포의 활성을 억제하는 효과가 있다는 것을 알 수 있었으며, 상대적으로 독성이 적은 항알레르기 제재로 개발할 가능성을 제시한 것으로 생각된다.

  • 선천성 면역 반응을 위해 중요한 역할을 하는 TLRs가 외부 병원성 물질에 자극을 받게 되면 NF-${\kappa}$B를 활성화시키며, 그 결과로 염증을 유도하는 COX와 같은 유전자를 발현한다. 이번 연구에서, 옛날부터 지금까지 전통적인 약재로써 질병 치료에 다양하게 쓰이고 있는 감초의 뿌리에서 추출한 성분 중의 하나인 ILG가 NF-${\kappa}$B활성과 COX 발현을 어떻게 조절하여 항염증 효과를 가지고 있는지 알아보았다. ILG는 TLR agonists인 MALP-2, Poly[I:C], 그리고 LPS에 의해 유도된 NF-${\kappa}$B 활성화와 COX-2 발현을 억제시켰다. 또한 ILG는 리간드(ligand)에 독립적인 TLRs signaling downstream molecules인 MyD88, IKK ${\beta}$, 그리고 p65에 의해서 유도된 NF-${\kappa}$B 활성을 억제시켰다. 이러한 결과는 한약재로서 많이 이용되는 감초가 단지 한약의 쓴맛을 줄이기 위함이 아니라 TLRs 신호전달 체계를 조절하여 항염증 효과를 가지고 있다는 것을 보여주는 것이라 할 수 있겠다.

고농도 감마 토코페롤 보충식이가 흡연에 노출된 쥐의 혈액 및 조직 비타민 E와 대사산물 농도에 미치는 영향 (Effects of Gamma-Tocopherol (GT) Supplementation on Vitamin E Concentration in Cigarette Smoke (CS) Exposed Mice)

  • 흡연은 활성 산소/질소종의 생성을 증가시켜 체내 산화적 스트레스를 증가시키고 폐의 염증을 유발한다. 이는 흡연자들의 체내 항산화 영양소들의 감소와 밀접한 관계를 가지는 것으로 알려져 있다. 본 연구에서는 항산화, 항염증 기능을 가진 비타민 E 중 식이에 많이 포함되어 있는 GT를 이용하여 흡연에 의한 항산화, 항염증 작용을 알아보기 위한 선행 연구로 고농도의 GT 식이가 혈액과 간, 폐의 AT, GT농도와 이들의 대샤산물인 CEHC 농도에 미치는 영향에 대하여 알아보고자 실시하였다. 연구 결과 고농도 GT 식이는 체내 혈액과 간, 폐 조직에 GT를 축적시키고 G-CEHC의 배설을 증가시켰다. 흡연은 대조군의 혈액과 폐 AT 농도는 증가시켰지만, 고농도 GT군의 혈액과 폐의 GT농도와 간의 G-CEHC의 농도를 감소 시켰다. 이러한 변화는 흡연에 의한 산화적 스트레스 상태에서 각기 다른 기능을 가진 조직의 요구량에 따라, 폐의 이용률을 높이기 위해 혈액으로 운반되는 양을 증가시키고 폐로의 운반을 임시적으로 증가시켜 조직의 AT, GT 농도를 선택적으로 조절하고 GT의 생체 이용률의 증가 때문이라 사료된다. 하지만 정확한 기전에 대한 연구들이 부족한 실정이므로 고농도의 GT 식이가 흡연에 의해 유도된 체내 산화적 스트레스와 폐의 염증 반응에 긍정적인 효과를 알아보기 위한 후속 연구가 절설히 필요한 것으로 사료된다.

Stem Cells and Cell-Cell Communication in the Understanding of the Role of Diet and Nutrients in Human Diseases

  • The term, "food safety", has traditionally been viewed as a practical science aimed at assuring the prevention acute illnesses caused by biological microorganisms, and only to a minor extent, chronic diseases cause by chronic low level exposures to natural and synthetic chemicals or pollutants. "food safety" meant to prevent microbiological agents/toxins in/on foods, due to contamination any where from "farm to Fork", from causing acute health effects, especially to the young, immune-compromised, genetically-predisposed and elderly. However, today a broader view must also include the fact that diet, perse (nutrients, vitamins/minerals, calories), as well as low level toxins and pollutant or supplemented synthetic chemicals, can alter gene expressions of stem/progenitor/terminally-differentiated cells, leading to chronic inflammation and other mal-functions that could lead to diseases such as cancer, diabetes, atherogenesis and possibly reproductive and neurological disorders. Understanding of the mechanisms by which natural or synthetic chemical toxins/toxicants, in/on food, interact with the pathogenesis of acute and chronic diseases, should lead to a "systems" approach to "food safety". Clearly, the interactions of diet/food with the genetic background, gender, and developmental state of the individual, together with (a) interactions of other endogenous/exogenous chemicals/drugs; (b) the specific biology of the cells being affected; (c) the mechanisms by which the presence or absence of toxins/toxicants and nutrients work to cause toxicities; and (d) how those mechanisms affect the pathogenesis of acute and/or chronic diseases, must be integrated into a "system" approach. Mechanisms of how toxins/toxicants cause cellular toxicities, such as mutagenesis; cytotoxicity and altered gene expression, must take into account (a) irreversible or reversal changes caused by these toxins or toxicants; (b)concepts of thresholds or no-thresholds of action; and (c) concepts of differential effects on stem cells, progenitor cells and terminally differentiated cells in different organs. This brief Commentary tries to illustrate this complex interaction between what is on/in foods with one disease, namely cancer. Since the understanding of cancer, while still incomplete, can shed light on the multiple ways that toxins/toxicants, as well as dietary modulation of nutrients/vitamins/metals/ calories, can either enhance or reduce the risk to cancer. In particular, diets that alter the embryo-fetal micro-environment might dramatically alter disease formation later in life. In effect "food safety" can not be assessed without understanding how food could be 'toxic', or how that mechanism of toxicity interacts with the pathogenesis of any disease.

In vivo Pharmacokinetics, Activation of MAPK Signaling and Induction of Phase II/III Drug Metabolizing Enzymes/Transporters by Cancer Chemopreventive Compound BHA in the Mice

  • Phenolic antioxidant butylated hydroxyanisole (BHA) is a commonly used food preservative with broad biological activities, including protection against chemical-induced carcinogenesis, acute toxicity of chemicals, modulation of macromolecule synthesis and immune response, induction of phase II detoxifying enzymes, as well as its undesirable potential tumor-promoting activities. Understanding the molecular basis underlying these diverse biological actions of BHA is thus of great importance. Here we studied the pharmacokinetics, activation of signaling kinases and induction of phase II/III drug metabolizing enzymes/transporter gene expression by BHA in the mice. The peak plasma concentration of BHA achieved in our current study after oral administration of 200 mg/kg BHA was around $10\;{\mu}M$. This in vivo concentration might offer some insights for the many in vitro cell culture studies on signal transduction and induction of phase II genes using similar concentrations. The oral bioavailability (F) of BHA was about 43% in the mice. In the mouse liver, BHA induced the expression of phase II genes including NQO-1, HO-1, ${\gamma}-GCS$, GST-pi and UGT 1A6, as well as some of the phase III transporter genes, such as MRP1 and Slco1b2. In addition, BHA activated distinct mitogen-activated protein kinases (MAPKs), c-Jun N-terminal kinase (JNK), extracellular signal-regulated protein kinase (ERK), as well as p38, suggesting that the MAPK pathways may play an important role in early signaling events leading to the regulation of gene expression including phase II drug metabolizing and some phase III drug transporter genes. This is the first study to demonstrate the in vivo pharmacokinetics of BHA, the in vivo activation of MAPK signaling proteins, as well as the in vivo induction of Phase II/III drug metabolizing enzymes/transporters in the mouse livers.

Properties of hydrolyzed α-lactalbumin, β-lactoglobulin and bovine serum albumin by the alcalase and its immune-modulation activity in Raw 264.7 cell

  • This study investigated the effects of the proteolytic hydrolysates of α-lactalbumin (LA), β-lactoglobulin (LG) and bovine serum albumin (BSA) by alcalase on inflammatory cytokines. The proteolytic hydrolysates were separated into two fraction of peptides, ≤ 10,000 Da and > 10,000 Da, respectively, because various low molecular weight peptides were generated during the hydrolysis reaction time. Among the hydrolysate peptides, BSA (all types), β-LG (> 10,000 Da), and α-LA (> 10,000 Da) showed an inhibitory activity against thymic stromal lymphopoietin (TSLP) mRNA expression in lipopolysaccharide-induced RAW264.7 murine macrophages. α-LA (> 10,000 Da), β-LG (hydrolysates), and BSA (> 10,000 Da) showed an inhibitory activity against tumor necrosis factor (TNF)-α expression. α-LA (all types), β-LG (hydrolysates, > 10,000 Da), and BSA (> 10,000 Da) showed an inhibitory activity against interleukin-6 (IL-6) expression. α-LA (> 10,000 Da), β-LG (> 10,000 Da), and BSA (all types) showed an inhibitory activity against inducible nitric oxide synthase (iNOS) expression. α-LA (> 10,000 Da), β-LG (> 10,000 Da), and BSA (all types) showed an inhibitory activity against cyclooxygenase (COX)-2 expression. The lowest level of TNF-α production was measured with α-LA (> 10,000 Da) and β-LG (> 10,000 Da) for all types, and a similar low level was measured for all types of BSA. The highest level of IL- 6 production was measured with α-LA (≤ 10,000 Da) among α-LA, β-LG, and IL-6. The low level of NO production was similar with α-LA, β-LG, and BSA but not with α-LA (≤ 10,000 Da). These potential peptides from whey protein hydrolysates could be used for food, medicinal, and industrial applications.

Pilocarpine에 의한 경련중첩증 후 해마에서 Tbr2 발현에 관한 연구 (Expression of Tbr2 in the Hippocampus Following Pilocarpine-induced Status Epilepticus)

  • Tbr2는 T-box family 전사인자의 하나로써 뇌의 발달, 전구세포의 증식, 그리고 CD8+ T 세포와 자연살상세포의 분화와 기능에 중요한 역할을 하는 것으로 알려져 있다. 본 연구는 마우스에서 pilocarpine을 이용하여 경련중첩증을 유도한 후 나타나는 병리기전에 Tbr2의 연관성을 확인하였다. 경련중첩증은 해마의 CA3, hilus 그리고 조롱박피질 등에서 뚜렷한 신경세포의 손상을 유발하였다. 흥미롭게도 Tbr2를 이용한 조직 염색에서 경련중첩증 2일 후에 CA3와 조롱박피질에서 면역반응성이 뚜렷하게 증가하는 것을 관찰하였다. 또한 CA3와 조롱박피질에서 Tbr2를 발현하는 세포는 미세아교세포와 단핵구, CD8+ T세포 또는 자연살상세포 등 백혈구의 표지물질인 CD11b 와 이중염색되는 것을 발견하였다. Tbr2와 CD11b에 동시에 염색된 세포는 아메바 모양의 형태를 갖추고 있는 것을 발견하였다. 게다가 혈관 내피세포에서 발혈되는 platelet endothelial cell adhesion molecule-1(PECAM-1)과 이중 염색한 결과 Tbr2를 발현하는 세포가 CA3 지역의 혈관내에 다량 존재하는 것을 확인하였다. 이상의 결과를 종합할 때 Tbr2를 발현하는 세포는 뇌 조직으로 이주하는 백혈구일 가능성이 높음을 보여준다. 이러한 결과는 경련중첩증에 따른 신경병리기전에 Tbr2가 관여할 가능성이 높음을 처음으로 제시하였다.

연교(連翹)가 만성 비세균성 전립선영 Rat의 염증발현인자 및 세포조직 변화에 미치는 영향 (The Effects of Forsythiae Frucus on Inflammatory Genes and Cyto-pathological Alterations in Chronic Non-Bacterial Prostatitis Rat Model)

  • Objective : The etiology of chronic prostatitis is likely multifactorial, resulting from either a cascade of events after an initiating factor or from a variety of etiologic mechanisms. There is substantiating evidence to support the role of the inflammatory responses in its pathogenesis, and the clinical value in the evaluation of therapeutic efficacy. Forsythiae Frucus has been traditionally used in treatment of inflammatory diseases, including of prostatitis and urinary tract inflammation. In this study, we investigated the effects of Forsythiae Frucus on inflammatory cytokines and cyto-pathological alternation in the rat model of chronic non-bacterial prostatitis induced by castration and $17{\beta}$-estradiol treatment. Methods : Two-month-old rats were treated with $17{\beta}$-estradiol after castration for induction of experimental non-bacterial prostatitis. which is similar to human chronic prostatitis in histopathological profiles. Forsythiae Frucus as an experimental specimen, and testosterone as a positive control, were administered orally. The prostates were evaluated by histopathologlcal parameters including the epithelial score and epithelio-stromal ratio for glandular damage. and the expression of inflammatory cytokine genes including interleukin (IL)-$1{\beta}$, IL-5, IL-12, tumor necrosis factor (TNF)-$\alpha$. eotaxin, inducible nitric oxide synthase (iNOS) and cyclooxygenase-2(cox-2). Results : While prostates of control rats revealed severe acinar gland atrophy and stromal proliferation. the rats treated with Forsythiae Frucus showed a diminished range of tissue damage. Epithelial score was improved in the Forsythiae Frucus group over that of the control (P<0.05). The epithelia-stromal ratio was lower in the Forsythiae Frucus group when compared to that of the control (P<0.05). In the reverse transcription-polymerase chain reaction (RT-PCR) of inflammatory cytosine genes. Forsythiae Frucus inhibited the expression of IL-$1{\beta}$, TNF-$\alpha$, iNOS, cox-2 genes, while it modulated the expression of IL-5, which is an anti-inflammatory cytokine. Conclusions : These findings suggest that Forsythiae Frucus may protect the glandular epithelial cells and also inhibit stromal proliferation in association with the immune modulation including the suppression of inflammatory cytokines and increase of anti-inflammatory cytokines. From theses results. we suggest that Forsythiae Frucus could be a useful remedy agents for treating chronic non-bacterial prostatitis.

Adriamycin 유발 신병증에서 중간엽 줄기세포의 완화 효과 (Mesenchymal Stem Cells Ameliorate Adriamycin Induced Proteinuric Nephropathy)

  • 목 적 : 사구체신염은 흔히 단백뇨를 보이며 특이 치료법이 없고, 만성 신부전으로 발전하는 경우가 많다. 몇몇 연구에서 중간엽 줄기세포(Mesenchymal stem cell, MSC)를 실험적 사구체신염에 투여하여 단백뇨가 호전된 것을 보고한 바 있으나, 이는 신염을 일으키는 약제와 중간엽 줄기세포를 함께 투여하거나 신장에 직접 투여한 것이었다. 본 연구에서는 실험적 신병증에서 단백뇨가 발현된 시점에서 정주 요법으로 MSC를 투여함으로써 MSC의 임상적인 적용 가능성을 탐색하였다. 방 법 : 실험용 생쥐에 Adriamycin을 투여하여 신병증(ADR-GN)을 유발한 후, 2주 후에 대량의 단백뇨를 확인하고 MSC를 생쥐 꼬리의 정맥에 주사하였다. MSC에 의한 질병 완화의 기전을 확인하기 위한 in vitro 실험으로 mixed lymphocyte culture(MLC)에 MSC를 투여하였을 때의 염증 관련 cytokine인 IFN-$\gamma$ and IL-10의 변화를 측정하였다. 결 과 : 실험용 생쥐에 ADR-GN를 유발하고 단백뇨가 보일 때 MSC를 정주한 군에서는 단백뇨의 소실이 더 먼저 관찰되었다. 또한 MSC를 투여받은 군에서의 생존률이 더 나은 경향이 관찰되었다. MLC 에 MSC를 투여하였을 때, 염증을 유발하는 cytokine인 IFN-$\gamma$ 는 감소하고 염증을 억제하는 cytokine인 IL-10는 증가하였다. 결 론 : 이 연구는 이전의 보고들에서 관찰되었던 사구체신염에서의 MSC의 질병완화 효과가 좀더 임상적으로 적용 가능한 방법으로 투여된 경우, 즉 단백뇨가 있을 때 정주 요법으로 투여한 경우에도 관찰됨을 확인하였다. 이러한 효과의 기전과 임상적용에 요구되는 안전성 등에 대한 확인을 위해서는 추가 연구가 필요하겠다.

Association of Estrogen Receptor Alpha and Interleukin 6 Polymorphisms with Lymphovascular Invasion, Extranodal Extension, and Lower Disease-Free Survival in Thai Breast Cancer Patients

  • Breast cancer is the most frequent type of cancer diagnosed among women worldwide and also in Thailand. Estrogen and estrogen receptors exert important roles in its genesis and progression. Several cytokines have been reported to be involved in the microenvironment that promotes distant metastasis via modulation of immune and inflammatory responses to tumor cells. Estrogen receptor genetic polymorphisms and several cytokines have been reported to be associated with breast cancer susceptibility and aggressiveness. To investigate roles of genetic polymorphisms in estrogen receptor alpha (ESR1) and interleukin 6 (IL6), breast cancer patients and control subjects were recruited from the Division of Head, Neck and Breast Surgery (Siriraj Hospital, Bangkok, Thailand). Polymorphisms in ESR1 (rs3798577) and IL6 (rs1800795 and rs1800797) were evaluated by real-time PCR in 391 breast cancer patients and 79 healthy controls. Associations between genetic polymorphisms and clinicopathological data were determined. There was no association between genetic polymorphisms and breast cancer susceptibility. However the ESR1 rs3798577 CT genotype was associated with presence of lymphovascular invasion (OR=2.07, 95%CI 1.20-3.56, p=0.009) when compared to the TT genotype. IL6 rs1800795 CC genotype was associated with presence of extranodal extension (OR= 2.30, 95%CI 1.23-4.31, p=0.009) when compared to the GG genotype. Survival analysis showed that IL6 rs1800797 AG or AA genotypes were associated with lower disease-free survival. These findings indicate that polymorphisms in ESR1 and IL6 contribute to aggressiveness of breast cancer and may be used to identify high risk patients.
