Apoptosis signal-regulating kinase 1 (ASK1) is an upstream signaling molecule in oxidative stress-induced responses. Because oxidative stress is involved in asthma pathogenesis, ASK1 gene deficiency was investigated in animal models of allergic asthma. However, there is no study to investigate whether ASK1 inhibitors could be applied for asthma to date. Selonsertib, a potent and selective ASK1 inhibitor, was applied to BALB/c mice of an ovalbumin (OVA)-induced allergic asthma model. Selonsertib suppressed antigen-induced degranulation of RBL-2H3 mast cells in a concentration-dependent manner. The administration of selonsertib both before OVA sensitization and OVA challenge significantly reduced airway hyperresponsiveness, and suppressed eosinophil numbers and inflammatory cytokine levels in the bronchoalveolar lavage fluid. Histopathologic examination elucidated less inflammatory responses and reduced mucin-producing cells around the peribronchial regions of the lungs. Selonsertib also suppressed the IgE levels in serum and the protein levels of IL-13 in the bronchoalveolar lavage fluid. These results suggest that selonsertib may ameliorate allergic asthma by suppressing immune responses and be applicable to allergic asthma.
We have studied about the safety, immunity and protective potency in swine and experimental am mais of two inactivated vaccine produced with NYJ-1-87 strain of ADV that was isolated in Korea. Result obtained through the experiments were summarized as follows. 1. The safe potency of ADV antigens inactivated with BEI and formaline to mouse & guinea pig was on the whole good, but protective potency rates of those to challenge with ADV were 60-75% without the differences to two antigens. 2. Safety, immunity & protective potency of ADV antigens inactivated with BEI and formaline to swine were on the whole excellent, except for a mild increase of rectal temperature in some pigs after challenge with ADV. 3. When virus excretion of the experimental groups after challenge with ADV was examed by swabbing of nasal, all pigs of control gorup excreted virus from 2 days p.c., partially to 10 days p.c.. But in vaccinated groups, only 25-50% of all pigs of each group excreted virus during experimental periods. 4. Titers of antibodies in swine & quinea pig vaccinated with inactivated ADV antigens become increased after the 1 weeth p.i. showing the highest liters on the 4-5 weeths p.i.
The effects of Ephedrae herba on cellular and humoral immune responses were investigated in mice. Group A and B of mice received intraperitoneal injection of methanol extracts, ether fraction(E), petroleum ether fraction(PE), ethyl acetate fraction(EA) and water fraction(W) for 5 days or 10 days before sensitization. The other group received i.p. injection of ephedrine (Ep) or pseudoephedrine $({\psi}\;Ep)$ for 1 day prior to sensitization. The change of body and spleen weights showed a tendency of decreasing but that of thymus showed increasing. Ear swelling was maximum at 48 hours after challenge and was significantly decreased in the groups treated with EA, W, Ep, and ${\psi}\;Ep$. HA titer was enhanced in EA, W and EP treated groups, HE titer being significantly decreased.
Heat stress is a major challenge to animal production in tropical and subtropical climates. Rabbits suffer from heat stress more than farm animals because they have few sweat glands, and their bodies are covered with thick fur. Intensive farming relies on antibiotics as antimicrobials or growth promoters to increase animals' productivity and health. However, the European Union and many countries have banned or restricted the use of antibiotics in animal feed for human health concerns. Several studies have found that replacing antibiotics in rabbit feed with natural plants or feed additives increases productivity and improves immune capacity, especially under heat stress conditions. Growth performance, immune response, gut microflora, and carcass yield may be increased in rabbits fed a diet supplemented with some natural plants and/or propolis. In this review article, we discuss and summarize the effects of some herbs and plant extracts as alternative feed additives on rabbit productivity, especially for those raised under hot ambient temperatures.
The charcoal effects on the immune responses of flounder, Paralichthys olivaceus were studied. Fish were fed an experimental diet supplemented with 1%, 3% or 5% of charcoal to a commercial diet. Several factors such as weight gain, hematological parameters, nonspecific immune responses and resistance against Edwardsiella tarda were evaluated 4 and 8 weeks after the administraion. The changes of body weight and lysozyme activities were not signigicant among tested groups. But cholesterol and total protein concentration in the blood were significantly lower in the 5% supplemented group than in the control. The production of superoxide by macrophage from the head kidney, examined by NBT reduction, was significantly higher in the 3% charcoal supplemented groups than in the control. The survival rate after an artificial challenge with 1×10^exp(7) cells of E. tarda per fish was significantly the highest in the 3% charcoal supplemented group.
Cyclophosphamide, a cytotoxic anticancer agent, induces immunosuppression and has several adverse effects. N-acetylcysteine alleviates oxidative stress, liver injury, and intestinal tissue damage. The present study examined whether N-acetylcysteine modulates the adverse effects of cyclophosphamide in pigs. Miniature pigs (n = 15) were used as an experimental model to evaluate the effects of N-acetylcysteine treatment on immune reactions, liver injury, and oxidative stress after cyclophosphamide challenge. Corn-soybean meal based dietary treatments were as follows: control diet with either saline injection, cyclophosphamide injection, or 0.5% N-acetylcysteine and cyclophosphamide injection. N-acetylcysteine increased the number of immune cells and decreased TNF-α production after cyclophosphamide injection and decreased TNF-α, IFN-γ, NF-κB, and IL-8 expression and increased IL-10 expression in peripheral blood mononuclear cells. Serum levels of alanine transaminase and aspartate aminotransferase decreased, superoxide dismutase activity increased, and malondialdehyde activity decreased following N-acetylcysteine treatment after cyclophosphamide injection. N-acetylcysteine decreases immunosuppression, liver injury, and oxidative stress in cyclophosphamide-challenged miniature pigs. The present study suggests that N-acetylcysteine has therapeutic application in livestock for modulating immune reactions, liver injury, and oxidative stress.
Ginseng products on the market show high variability in their composition and overall quality. This becomes a challenge for both consumers and health-care professionals who are in search of high-quality, reliable ginseng products that have a proven safety and efficacy profile. The botanical extract standardization is of crucial importance in this context as it determines the reproducibility of the quality of the product that is essential for the evaluation of effectiveness and safety. This review focuses on the well-characterized and standardized ginseng extract, G115, which represents an excellent example of an herbal drug preparation with constant safety and efficacy within the herbal medicinal products. Over the many decades, extensive preclinical and clinical research has been conducted to evaluate the efficacy and safety of G115. In vitro and in vivo studies of G115 have shown pharmacological effects on physical performance, cognitive function, metabolism, and the immune system. Furthermore, a significant number of G115 clinical studies, most of them double-blind placebo-controlled, have reinforced the findings of preclinical evidence and proved the efficacy of this extract on blood glucose and lipid regulation, chronic obstructive pulmonary disease, energy, physical performance, and immune and cognitive functions. Clinical trials and 50 years of presence on the market are proof of a good safety profile of G115.

Cancer immunotherapy, in the form of vaccination, adoptive cellular transfer, or immune checkpoint inhibitors, has emerged as a promising practice within the field of oncology. However, despite the developing field's potential to revolutionize cancer treatment, the presence of immunotherapeutic-resistant tumor cells in many patients present a challenge and limitation to these immunotherapies. These cells not only indicate immunotherapeutic resistance, but also show multi-modal resistance to conventional therapies, abnormal metabolism, stemness, and metastasis. How can immunotherapeutic-resistant tumor cells render multi-malignant phenotypes? We reasoned that the immune-refractory phenotype could be associated with multi-malignant phenotypes and that these phenotypes are linked together by a factor that acts as the master regulator. In this review, we discussed the role of the embryonic transcription factor NANOG as a crucial master regulator we named "common factor" in multi-malignant phenotypes and presented strategies to overcome multi-malignancy in immunotherapeutic-resistant cancer by restraining the NANOG-mediated multi-malignant signaling axis. Strategies that blunt the NANOG axis could improve the clinical management of therapy-refractory cancer.
P. gingivalis를 단독면역하거나 또는 Fusobacterium nucleatum 선면역 후 P. gingivalis 항혈청을 각각 얻어냈다. 두 종류의 항혈청이 P. gingivalis biofilm을 침투해 들어가는 능력을 confocal laser scanning microscope를 이용하여 비교 감증하였다. 항혈청의 P. gingivalis에 대한 avidity index도 측정하였다. 결과적으로 F. nucleatum의 선면역은 P. gingivalis 특이 항혈청에 대해 세균성 biofilm의 침투능력을 저하시키고, 동일한 세균에 대한 avidity도 감소시켰다.
An expeliment was carried out to measure the cellular immune response in guinea pigs by sensitizing the animals with 2, 4-dinitrochlorobenzene(DNCB). The guinea pigs could be sensitized with one application of DNCB. The sensitizing and challenge dose was standardized. The histological response was characteristic of a delayed hypersensitivity reaction.
