In immature seeds of Phaseolus vulgaris, the presence of threee new 2-deoxy type brassinosteroids (trihydroxybrassinosteroids) have been demonstrated. These less polar brassinosteroids have been demonstrated. These less polar brassinosteroids have been tentatively characterized to be (3ξ, 22ξ, 23ξ)-2-deoxy-25-methylodolicholide, (3ξ, 22ξ, 23ξ)-2-deoxoy-dolichosterone and (3ξ, 22ξ, 23ξ)-2-deoxy-24-ethylbrassinone by analysis of gas chromatography-mass spectrometry. Their less biological activity and oxidation state than those of tetrahydroxybrassinosteroids suggest that they are potent biosynthetic precusors of tetrahydroxybrassinosteroids.
The objective of this study is to establish an efficient celll culture system for somatic embryogenesis in Ostericum koreanum and Angelica purpuraefolia. The highest frequency of embryogenic callus on immature seeds of O. konanum and A, purpuraefolia. was obtained when seeds were cultured on MS medium containing 0.1 mg/L 2,4-D and 0.1 mg/L BA. However somatic embryos were formed directly from the edge of cotyledon and hypocotyl of plant which regenerated on medium supplemented with 0.1-3.0 mg/L NAA. Immature seed explane cultured at 25$\pm$2$^{\circ}C$ after 10 days treatment at 5$^{\circ}C$ produced embryogenic callus and somatic embryos, and these differentiated into whole plane. Addition of glutamine and coconut milk to media did not enhance the frequency of somatic embryogenesis in immature seed cultures of A. purpuraefolia. However in immature seed culture of O. koreanum, the frequency of somatic embryogenesis were increased on media supplemented with glutamine and 10% coconut milk. Especially addition of glutamine to the medium substituted effect of NH$_4$N0$_3$ in constast to coconut milk. The highest frequency of conversion somatic embryos into plantlet was 89.1% on MS basal medium Embryogenic calli were grown vigorously when maintained on medium with 0.01 mg/L 2,4-D and 0.01 mg/L BA.

This study was investigated the effect of immature rhizome production according to harvest times-based treatment method for seed production of Gastrodia elata. The results revealed that when the tuber weight of G. elata harvested in spring (GEHS) was ${\geq}100g$, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was 90.9%-94.8%, 3.1%-5.4%, and 10.1%-15.3%, respectively. When G. elata harvested in fall (GEHF) was treated at a low temperature for 4 weeks or more, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was 70.4%-87.6%, 2.2%-2.6%, 8.7%-9.5%, respectively. Therefore, to produce seeds and immature rhizomes, GEHS must have tubers of more than 100 g, whereas GEHF requires breaking dormancy by low-temperature treatment for 4 weeks or more. Compared with those of GEHS, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was lower in GEHF; however, it was higher than those in the natural germination state. Thus, it can be expected that G. elata can be produced throughout the year by ensuring that the seeds and immature rhizomes of G. elata are produced using a constant tuber weight and by breaking dormancy with low temperature treatment.

We investigated the effects of sodium hypochlorite (NaOCl) and culture medium on embryo swelling and germination of Calanthe discolor Lindl., and established a method for determining the swelling and protocorm formation of C. discolor seeds via in vitro examination of immature seeds. Treatment of immature seeds with NaOCl greatly enhanced the extent of embryo swelling and protocorm formation of immature zygote embryos compared to seeds without NaOCl treatment. The effects of the culture media were also evaluated with regard to embryo swelling and protocorm formation of in vitro cultured seeds with and without NaOCl treatment. Additionally, the effects of white fluorescent light and red and blue LEDs lights on seedling growth in in vitro culture were examined. The most suitable condition for seedling growth after 12 weeks of culture was the red LEDs light with POM medium. These results show effective asymbiotic germination and growth of C. discolor.
Structural changes of the endosperm of Panax ginseng C.A. Meyer from fertilization to germination were investigated by light microscope. The endosperm of the ginseng seed is cellular type. Since endosperm cells adjacent embryo continuously breakdown and disappear with the elongation of embryo, the real of endosperm is gradually decreased. As the anatropous ovules of immature seed with green seed coat developes more and more, ovary cells adjacent ovary cavity become abundant by the periclinal division, their size is decreased, hypotrophy of cell wall discern, and they are gradually differentiated in seed coat. Though embryo responds strongly to basic dye at the stage of completion of endosperm formation, tissue of endosperm responds to acidic dye positively Cell wall of embryo and endosperm are composed of primary cell wall not lignified. Endosperm cells adjacent embryo begin to breakdown in the endosperm tissue of indehiscent seed before the beginning of the after-ripening. Dehiscent seed of which seed coat is opened through after-ripening represent the form as a seedling in the result of embryo developments with the formation of organs; radicle, cotyledon, plumule. Umbilifom layer represents strong positive response to the toluidine blue and the basic function. Umbiliform layer that endosperm cells breakdown and disappear is observed clearly at the periphery of the embryo cotylemon, while slightly at the periphery of the radicle.
In this study, the dependence of local conspecific abundance on seed set and predation was examined, and determinates of population density of the temperate tree species Carpinus laxiflora (Betulaceae) in Inagi city, Tokyo, Japan were identified. During a good seed year (2013), seeds were sampled from 27 individuals and categorized as Sound, Empty, Predated, Immature, Decayed, or Broken. Empty seeds were identified as those that failed to fertilize and predation rates were defined as the proportion of seeds that were affected by predators. The proportion of the seed set that was fertilized was significantly positively correlated with local abundance of conspecific trees, while the proportion of seeds that escaped predation was significantly negatively correlated with local abundance of conspecific trees. Thereby, the production of Sound seeds was highest at sites with moderate local conspecific abundance. Although this phenomenon was only observed in a few cases, it clearly showed optimal reproductive success under conditions of moderate density of conspecific reproductive adults, suggesting a mechanism that controls the population density of this species.
Transformants of White Clover(Trifolium repens L.) were efficiently produced from immature seed derived callus cocultivated with Agrobacterium twnefaciens LBA4404 harboring plant binary vector. pBI121, using acetosyringone. The mean frequencies of transformants on the two kanamycin-containing media were 16 to 19% when the immature seed-derived calli were infected with bacteria cultured in the presence of 100$\mu$M acetosyringone compared with 7% in media without acetosyringone. Transgenic white clover was subject to molecular analysis for integration into plant nuclear genome and expression of $\beta$-glucuronidase(GUS) gene. PCR and Northern blot analyses demonstrated that GUS gene was integrated into white clover nuclear genome and expressed into its mRNA. The expression of GUS gene into its protein was confirmed by spectrophotometric assay of GUS activity.

Xanthoceras sorbifolium Bunge (yellowhorn) is a woody tree in the soapberry family, Sapindaceae, native to northern China. This species has been identified as a major woody bioenergy plant for bio-diesel production because of high oil content in seed. But the flowers do not bear fruit well while the many flowers blooming. This study was performed to regenerate in vitro plantlet using adventitious shoot formation. To establish the protocol of plant regeneration, adventitious shoots formation rate in the culture of cotyledon of immature zygotic embryos was 68.6% in 1/2 MS medium with 0.5 mg l-1 BA and 3% sucrose (w/v). In the culture of cotyledons of mature zygotic embryos, induction of adventitious shoots was needed to contain high sucrose in pre-culture medium and the frequency of shoot induction was 64.4%. Multiple shoots were induced in 0.5 mg l-1 TDZ, and rooting of shoot was induced 4.0 mg l-1 IBA. Flow cytometry analysis revealed that all the regenerated plantlets were diploid.

We use two different methods for laboratory propagation from seed of lady's slipper orchid(Cypripedium macranthos Sw.); immature seed which also called green capsule or fully mature seed about 120~130 days from pollination. In green capsule culture, the seed pods should be collected within precisely right time. The right time of seed collection could be diverse under the wether conditions or nutritional factors of the plants. In fully matured seed culture, the more complicated procedures are needed to break the dormancy of the seed; thermal or chemical treatment. The seedlings in this study were easily germinated from immature seeds in Harvais medium; 53 days after pollination(DAP) in Cypripedium pubescens, DAP 65 in C. parviflorum and C. macranthos. The germinated seedlings were transplanted to hormone free media immediately to avoid abnormal growth of seedlings. When the seedlings have roots with a minimum length of around 2-3cm and have visible dormant buds, the seedlings were removed from the flask and stored in refrigerator for vernalization. To examine the correlation of seedlings and maternal plants, the 125 seedlings of C. macranthos were transplanted in the soil bed at a distance of 20-100 cm from mother plants on April 20. The survival rate of seedlings were 92% in 20 cm distance from the ripe plants, and 56 % in 100 cm distance. The seedlings which were transplanted near mother plants showed vigorous growth in plant height, leaf width, and especially dormant buds. Considering the existence of mycorrhiza which is a symbiotic association between a fungus and the roots of a orchid vascular, the various fungus from mother plants could affect the growth of the seedlings. These results indicate the possibility of high and stable production and practical industrialization of endangered lady's slipper orchids.
From immature seed of Phaseolus vulgaris L., a novel phytosteryl glucoside was isolated. Strong ion peaks at m/z 613 $[M+Na}^{+},\;696\;[M+Matrix]^{+}$ in positive F AB- MS and at m/z 589 $[M-1]^{-}$ in negative F AB- MS indicated the molecular weight of the compound is 590. Four hundred MHz $^IH-NMR$ analysis revealed that the compound canys a 24-hydroxy-24-vinyl-cholesterol (saringosterol) as an aglycone and a ${\beta}-D-glucopyranose$. Four hundred MHz $^IH-NMR$ analysis of the acetate derivate of the compound revealed that hydroxyls at C-1' in glucose moeity and at C-3 in aglycone have been condensed. Therefore, the phytosteryl glucoside was characterized to be $3-0-{\beta}-D-glucopyranosyl-24-hydroxy-24-vinyl-cholesterol$ (saringosteryl glucoside). This is the first demonstration for the presence of saringosterol in higher plants. Also this is the first identification of saringosteryl glucoside in natural materials.erials.
