The leakage characteristics of electrical conductivity, inorganic constituents-K, Ca, Mg and Na, total sugar and total amino acid from the exudates of some vegetable seeds of viable and non-viable(artificially aged) were quantified to get basic information about the detection of the non-viable seeds. The crops studied were radish, cabbage, broccoli, onion and carrot. The time course electrolyte leakage was different from viable and non-viable seed of cruciferae but not sensitive in onion and carrot seed In inorganic constituents, potassium leakage was the greatest amount and difference between viable and non-viable seeds, but Ca, Mg and Na leakages were not as much differences as potassium. Total sugar as glucose and total amino acid as glycin leaked a lot more in aged radish, cabbage and broccoli seed than non-aged seed and the large differences were appeared after 4 hour imbibition. As a results, in general the leakages from the aged seeds were greater than from the non-aged seeds in most components tested but they were varied depending on species or varieties and components.

Primed seeds germinate faster and uniformly, since these have already started imbibition and metabolism requiring to complete germination. Several factors such as solution composition, osmotic potential, and treatment duration affect seed priming response. Water potentials of priming solution and germination response of primed seed were investigated to clarify the effects different inorganic salt types and the duration. Pepper seeds were primed in osmotic solutions that were osmotic potential ranged -1.54 to -0.45 MPa in an aerated solution of polyethylene glycol 8000 (PEG; 17%, 22%, 27%), and inorganic salt solution of $KNO_3$, $K_2SO_4$, and $Na_2SO_4$ (100, 200, 300mM) in aerated condition. The seeds were treated at $20^{\circ}C$ for 4, 6 day (D) and stepwise duration combined concentration of 100mM (2 or 4 days) and 300mM(2 or 4days). After soaking treatment, seeds were washed with distilled water and then were dried to approximately 6% moisture content in dry chamber at $25^{\circ}C$. The germination of seed was characterized by ISAT rules at $20^{\circ}C/30^{\circ}C$ and $15^{\circ}C$. Total germination percentage (GP), mean germination time (MGT), germination uniformity (GU), germination rate (GR), and health seedling percent (HS) were calculated on the germinated seed in a 14-day period. Seed water potential (${\psi}$) was correlated with water potential of priming solution ($r^2=0.84$). The effect of seed priming on germination varies with inorganic salt and organic agents (PEG). Germination percentage (GP, 77 and 73%), GR (21.3 and $19.4 %{\cdot}day^{-1}$), and HS (58 and 52 %) was greatest and lowest MGT (4.2 and 4.8 day) when they were primed in 100mM $K_2SO_4$ (${\psi}=-0.45MPa$), and 100mM $KNO_3$ (${\psi}=-0.52MPa$) for 6 days compared to untreated control (67% GP, $19.7%{\cdot}day^{-1}\;GR$, 18% HS, and 10.8 day MGT) in $15^{\circ}C$ chamber. Water potential less then -1.2MPa of osmotic solution resulted lower GP, GR and HS and in $K_2SO_4$ and $KNO_3$. However, stepwise treatment of water potential that changed solution as 100mM for 4 D + 300mM for 2 D or 300mM for 4 D + 100mM for 2 D) were not better than single concentration alone in germination characteristics.
Seed germination is the important stage to express many genes for regulation of energy metabolism, starch degradation and cell division from seed dormancy state. For the functional analysis of seed germination mechanisms, we were analyzed the rice cDNA clones (Oryzasativa cultivar Ilpum) obtained from seed imbibition during 48 hours. Total number of 18,101 Expressed Sequence Tags (ESTs) were clustered using SeqMan program. Among them, 8,836 clones were identified as unique clones. We identified the chitinase gene specifically expressed in seed germination and amylase gene involved to starch degradation from the full length cDNA analysis, and several genes were registered to NCBI GeneBank. To analyzed the commonly expressed genes between inmature seed and germinated seed, 25,66 inmature ESTs and 18,101 germinated ESTs were clustered using SeqMan program and identified 2,514 clones as commonly expressed unigene. Among them, alpha-glubulin and alcohol dehydrogenase I were supposed to LEA genes only expressed in the immature and germinated seed stages. For the clustering of orthologous group genes, we further analyzed the 8,836 EST clones from germinating seeds using NCBI clusters of orthologous groups database. Among the clones, 5,076 clones were categorized into information storage and processing, cellular processes and signaling, metabolism and poorly characterized genes, proportioning 783 (14.29%), 1,484 (27%), 1,363 (24.8%) and 1,869 (34%) clones to the previous four categories, respectively.
Decay during soybean sprout culture detracts their quality as well as increases their production costs. This study was done to determine the effects of acetic and propionic acids on growth and morphological characters of the sprouts. The soybean seeds of 3 cultivars (cv. Eunhakong, Pungsannamulkong and Orialtae) imbibed for 2 minutes at their different concentrations (0, 0.1 and 0.2%) were soaked for 6 hours in 4 ppm BA solution after the first 5.5 hour water imbibition and 0.5 hour aeration, and cultured at $20^{\circ}C$. On the 6th day, harvested soybean sprouts were classified into 4 categories on the base of hypocotyl length;>7cm, 4 to 7cm,<4cm and not germinated, and their morphological characters, fresh weights, lesion spots on cotyledons were measured or analyzed. The stronger concentration the higher rate of longer than 4cm although there was no significant difference between the two acids. Pungsannamulkong showed the longest hypocotyls but Eunhakong did the thickest ones, and Orialtae did the greatest total fresh weight, in which all the characters were not influenced by the two acids and their concentrations. Lesion spots on the cotyledons were equal to the two acids although less in their treatments than in no treatment. Utilization of propionic acid was more desirable Dan acetic acid through their treatment cost analysis.
Seed dormancy and germination responses to light and gases were determined for Echinochloa colona (L.) Link. E. colons seeds did not require a period of after-ripening for breaking dormancy. Water movement occurred readily across the seed coat. Repeated cycles of hydration and dehydration reduced viability and thence germination. Water imbibition for 24 h increased seed moisture by 21%; seeds returned to their original weight after drying at room temperature for 13 h. Removal of seed-coats increased germination in the dark. Light stimulated germination. Germination at a daylight intensity of 51.9 $Wm^{-2}$ or less was significantly reduced. Germination of seeds which were exposed to light for 1 h each day was significantly less than that of seeds exposed for longer than 2 h a day. Seeds subjected to blue light had delayed and decreased germination compared to seeds exposed to red light. Ethylene or carbon dioxide exogenenously added in the presence of light stimulated germination. The addition of the two gases together had a synergistic effect. In the dark, however, the two gases did not increase germination.

This study was conducted to investigate seed dormancy type and germination characteristics of Lonicera chrysantha. In imbibition test, the seeds imbibed water readily. Therefore, the seeds did not have physical dormancy (PY). The seeds have undeveloped embryo at seed dispersal and, then the E:S ratio (embryo/seed ratio) increased more than about 46% just before germination. In temperature experiments (5, 15, 20 and $25^{\circ}C$), the seeds did not germinate during incubation at $5^{\circ}C$. At $20^{\circ}C$, the seeds started to germinate from a week after sowing and the final germination was 86.7% at 2 weeks after sowing. At $15^{\circ}C$, the seeds started to germinate at 2 weeks and the final germination rate was 75.0% at 4 weeks after sowing. At $25^{\circ}C$, the seeds started to germinate at a week and the final germination rate was 48.3% at 19 weeks after sowing. Consequently, Optimum temperature of germination is $20^{\circ}C$ that shown highest final germination rate and shortest mean germination time. The seeds germinated all temperature treatments within 4 weeks except to $5^{\circ}C$. Therefore, it is concluded that seeds of L. chrysantha have only morphological dormancy (MD).
Recently, as consumers' interest and importance in health care have significantly increased, they prefer natural and organic foods that do not use chemical pesticides. Since sprout vegetables effectively promote health and prevent diseases such as cancer and cardiovascular disease, the consumption of sprout vegetables, a highly functional and safe food, has been increased significantly. This study aimed to investigate the effect of light on the growth and bioactive compounds of seven different sprout vegetables. After sowing the seeds of various sprout vegetables (kale, Chinese kale, broccoli, red cabbage, alfalfa, red radish, and radish), the sprouts were cultivated under light conditions (20℃, RGB 6:1:3, 130 μmol·m-2·s-1, 12 hours photoperiod) and dark condition for 7 days. Sprouts samples were taken at 1-day intervals from 4 to 7 days after treatment. The fresh weight, dry weight, plant height, total phenol content, and antioxidant capacity were measured. Brassica species (kale, Chinese kale, broccoli, red cabbage) and Medicago species (alfalfa) had significantly higher fresh weight values under dark conditions, while the content of bioactive compounds was increased considerably under light conditions. In contrast, the fresh weight of Raphanus genus (red radish, radish) significantly increased under the light condition, but the antioxidant phenolic compounds were significantly higher under the dark state. A negative correlation was observed between the growth and secondary metabolites in various sprout vegetables. This study confirmed the effect of light and dark conditions on different sprout vegetables' growth and nutritional value and emphasizes the importance of harvest time in producing high-quality sprout vegetables.
Soybean seed coat consisted of three layers, and the aleurone layer was attached to the innermost part of seed coat. It showed the different morphological characteristics with single cell layer compared with many cell layers in barley aleurone layer. The structural difference in aleurone cell among varieties was not detected in this experiment. The hole of middle part of hilum consisted of net formed material in order to pass water and gas. In the experiment, it was not studied whether the varieties with hard seed consist of the same structure or not, but the detailed study on this would be necessary. The activity of acid phosphatase showed a tendency to increase in process of imbibition in distilled water. There was no significant difference in the enzyme activity among the varieties tested, but the enzyme activity of Danyoupkong was slightly higher than that of Hwanggeumkong. In germinability, Danyoupkong is higher than Hwangkeumkong, so it might be attributed to the higher activity. There was no difference in acid phosphatase activity between released from the aleurone cell and accumulated in the aleurone cell from 6 to 12 hours incubation of the medium in the absence of GA$_3$, but the difference was detected after 12 hours incubation. And enzyme activity was the highest in the 18 hours incubation. The increase in the release of acid phosphatase from the aleurone cell would be considered as passive diffusive effect due to the increase in turgo pressure of aleurone cell. The acid phosphatase released from aleurone layer increased greatly after 24 hours incubation of the medium in the presence of GA$_3$ and the accumulation within the aleurone cell decreased linearly after 18 hours incubation. The result indicates that GA$_3$ enhance the rate of enzyme release from aleurone layer, suggests that the aleurone cell wall be digested by the introduction of GA and the digested wall act as the channels for enzyme release.
Seed germination test done in laboratory does not coincide with field emergence in general. The experiments were carried out to examine the effect of priming and $GA_3$, treatment to seeds of Platycodon grandiflorum; Codonopsis lanceolata and C. pilosula on lapsed time to first seedling emergence, seedling emergence, morphological characters and growth and the cause of poor emergence of C. pilosula. No-treatment as Control (water), priming or $GA_3$ treatment was done with only distilled water for 2 days, $CA(NO_3)_2$ 150 mM for 2 days or $GA_3$ 0.1 mM for 3 days, respectively. Seedling emergence rate was counted every 2 days but morphological characters and dry weight of shoot and root were measured on 38 days after sowing. Their internal seed structures were examined with Scanning Electron Microscope. C. pilosula had poorer seedling emergence rate than P grandiflorum and C. lanceolata showing nearly same rate: Compared to the other treatment (s) P. grandiflorum displayed higher rate in priming and $GA_3$, treatments but C. lanceolata or C. pilosula did the greatest rate in only $GA_3$ or priming treatment, respectively. $GA_3$ treatment to seeds of P. grandiflorum and C. lanceolata shortened the lapsed time to seedling emergence in comparison with Control, 2-days water imbibition before sowing. In all the species plant height and number of leaves per seedling became shorter and less in priming treatment than the other treatments except plant height of C. Pilosula while their hypocotyl length was nearly same in all treatments. Although priming treatment had nearly similar effect to morphological characters, $GA_3$ treatment forced greater shoot, root and aftermath total dry weight per seedling. Poor seedling emergence of C. pilosula was caused by its seed defect like cleavage or lack of embryo, poor development of embryo and endosperm or their separation.
Rice seed maturation and germination involve drastic changes in water and nutrient transport, in which tonoplast aquaporins may play an important role. In the present study, gene expression profiles of 10 tonoplast intrinsic proteins (TIP) from rice were investigated by RT-PCR during seed development and germination. OsTIP3;1 and OsTIP3;2 were specifically expressed in mature seeds. Their transcript level rapidly decreased after onset of seed germination and gene expression was induced by ABA treatment. In contrast, expression of OsTIP2;1 and OsTIP4;3 was not seed specific as transcripts were found in vegetative tissues as well. Their respective transcript levels decreased at an early stage of seed development, whereas they increased at a later stage of seed germination and elongation of embryonic roots and shoots. When seed germination was inhibited by various stress conditions and ABA, expression of OsTIP2;1 and OsTIP4;3 was completely suppressed. In contrast, the expression level of OsTIP2;2 rapidly increased after seed imbibition and the transcript level was maintained under conditions inhibiting seed germination. These results implicate that tissue specific and developmental transcriptional regulation of OsTIPs in rice seeds depends on their specific function. In addition, OsTIPs can be discriminated by different potential phosphorylation and methylation sites in their protein structures. OsTIP3;1 and OsTIP3;2 possess unique phosphorylation signatures at their N-terminal domain, loop B and loop E, respectively. OsTIP2;1 and OsTIP4;3 have a potential methylation site at their Nterminal domain. This suggests that activity of specific tonoplast aquaporins may be regulated by post-translational modification as well as by transcriptional control.
