Effect of n-6, n-3 Fatty Acids and Vitamin A Levels on Humoral Immunity in Rats (n-6, n-3 지방산과 비타민 A 수준이 흰쥐의 체액면역에 미치는 영향)

  • This study was carried out to observe the effects of dietaryn-6, n-3 fatty acids and vitamin A levels on humoral immunity in rat. Sixty eight male Sprague-Dawley rats were fed 6 different experimental diets for 6 weeks. The diets were composed of 10% of either corn oil or fish oil with various levels of vitamin A ; deficient(12450 IU/kg diet) adequate(4000IU/kg diet) and excess(400,000 IU/kg diet) The weight of spleen from the excess vitamin A-fish oil group showed the lowest value of all the groups when spleen weight was expressed/100g body weight. The number of PFC to SRBC was not affected by dietary at type and vitamin A levels. Hemagglutination titers were significantly lower in fish oil groups compared to corn oil groups and the values of vitamin A deficient groups were lower than the ones of adequate and excess vitamin A groups. IgM contents is serum were significantly lower in fish oil groups than in corn oil groups. IgG contents were higher in fish oil groups than in corn oil groups and the highest levels was recorded in excess vitamin A-fish oil group which showed the smallest speen size. Light microscopical examination showed that spleen tissues of fish oil groups were well developed than those of the corn oil groups and vitamin A deficient and excessive groups showed poor development than the adequate groups. Therefore it is suggested that adequate amounts of vitamin A consumption is necessary for healthy individuals and fish oil intake along with excess vitamin A should be avoided in order to maintain immune function properly.

  • 청열조습(淸熱燥濕), 거풍살충(祛風殺蟲)의 효능이 있는 고삼(苦蔘)의 체내 면역활성에 미치는 영향에 대해 실험한 결과 아래와 같은 지견을 얻었다. ICR 생쥐에 고삼(苦蔘) 수침액을 투여한 결과 백혈구수 및 면역장기 변화는 없었다. in vitro실험에서 IgM PFCs에 유의성 있는 영향을 주지 못했고, BSA를 주사한 생쥐에서의 Arthus 반응에도 영향이 없었다. 그러나 지연형 과민반응을 억제하였으며, 비특이적 면역에 관여하는 macrophage의 탐식능을 약간 저하시켰다. 이상의 결과 고삼(苦蔘) 수침액(水浸液)은 세포성 면역을 억제하는 것으로 사료된다.

  • To clarify the activating effects of Buthus martensi Karsch on immunological function, its effect on primary and secondary antibodies production in mice of various ages was investigated. Buthus martensi Karsch increased the number of both antibody producing cells(anti-IgM and anti-IgG producing plaque forming cells, PFC) and phagocytic activity of peritoneal macrophage. Futhermore, these phenomena were significantly increased with aging in mice. Buthus martensi Karsch also increased natural killer cell activity concerning to cancer immunology. These results suggest that Buthus martensi Karsch markedly increases the reduced activity in the elderly and activates the immune response in senescence mice.

  • To clarify the activating effects of Scolopendrae corpus on immunological function, its effect on primary and secondary antibodies production in mice of various ages was investigated.   • This study was designed to investigated the effects of cadmium(Cd) feeding on the humoral immune responses such as PFC-responses and production of immunoglobulins in BALB/c mice. The results obtained were summarized as follows; 1. Total PFCs of direct IgM antibody response were significantly decreased in all Cd-fed goups, whereas total PFCs of IgG antibody response were slightly increased. 2. In secondary immunization, total HA titers were increased in all Cd groups as compared with control, especially in 100ppmm group and also IgG titers were slightly increased except for 50ppm group. 3. The levels of $IgG_1$ were increased to 5.5% 18.7%, 17.4% and 7.2% in 25, 50, 100 and 200ppm groups as compared with control, respectively. And also the levels of IgE were increased to 5.7%, 7.3%, 8.7% and 0.4% in those of Cd groups, in order. Conclusively, concentrations of $IgG_1$, and IgE were increased in all Cd groups. Based on the results of this study and previous report, it was shown that Cd might affect humoral immune responses by modifying the distribution and function of cells playing in the cellular immune response.

Oligopeptide derived from solid-state fermented cottonseed meal significantly affect the immunomodulatory in BALB/c mice treated with cyclophosphamide

  • In this study, the immunomodulatory activity of oligopeptide (CP) derived from solid-state fermented cottonseed meal were investigated in immunosuppressed BALB/c mice models by treatment with cyclophosphamide (CY). Results indicated that oligopeptide increased the thymus and spleen indices of CY-treated mice. The count of plague forming cells (PFC) and the content of half serum hemolysis ($HC_{50}$) in immunosuppressive mice were restored to the normal level in CP-10 and CP-20 groups while the cytokines interleukin (IL)-2, IL-6, and tumor necrosis factor alpha (TNF-${\alpha}$) were increased significantly in CP-20 group. Similar increasing the immunoglobulin of IgG and IgM content in the serum of CP-10 group were also observed. These findings indicated that oligopeptide derived from solid-state fermented cottonseed meal had a strong immune-enhancing activity as well as a protective effect against immunosuppression induced by cyclophosphamide in mice.

  • 산한지통(散寒止痛)의 효능(效能)이 있는 고량강(高良薑)이 체내 면역활성에 미치는 영향을 검토하기 위하여 인삼과 함께 실험을 행하여 아래와 같은 결과를 얻었다. 고량강(高良薑) 및 인삼 수침액을 Sarcoma-180 고형 종양 세포를 이식한 생쥐에 투여하여 생존기간을 연장시켰다. 이를 바탕으로 면역에 대한 효과를 검토한 결과, 정상 ICR 생쥐에서 고량강(高良薑) 수침액은 백혈구수, 체중에 대한 상대적 비장의 무게를 약간 감소시켰으나 통계적으로는 유의성이 없었고, 간에는 영향을 주지 않았다. in vitro 실험에서, splenic IgM PFCs을 유의성있게 증가시켰으며, BSA를 주사한 생쥐에서의 Arthus반응도 증가시켰다. 그러나, 지연형 과민반응에는 영향을 주지 않았으며, 비특이성 면역에 관여하는 Macrophage 등의 탐식 세포의 기능에도 영향을 주지 않았다. 이상의 결과, 고양강은 체액성 면역을 강화할 것으로 보여진다.

Effects of Fermented Milk Containing Herb Extract from Acanthopanax divaricatus var. albeofructus and Codonopsis Ianceolata on the Immune Status of Mouse (흰털오가피와 더덕 추출물을 첨가한 발효유 급여가 마우스의 면역기능에 미치는 영향)

  • We have investigated the immunomodulatory activity of water extracts of Acanthopanax divaricatus var. alveofructus in male ICR mice. Mice were administrated with fermented milk containing freeze-dried extract 3 mg/Kg (A), 9 mg/kg (B), 27 mg/Kg (C) per body weight with A. divaricatus var. alveofructus (loots : leaves : stem) : Codonopsis lanceolata = (5 : 2 : 1.5) : 1.5 for 7 and 10 weeks, respectively. Body weight, relative organ weight, cellularity of lymphoid organs, plaque- forming cell (PFC) assay, agglutination (AGG) test and lymphoproliferation were examined in various groups of animals. Any significant differences of body weight gain were not recorded in the tested ICR mice. There was significant different (p<0.05) in the spleen index in B group of 10 weeks and C group of 7 weeks fed mouse. The thymus gain weight was increased during administration of the extract, but there was no significant increase on other organs gain. Humoral immunity as measured by PFC showed more decreased PFC level in 10 weeks than in 7 weeks. In the HT, A. divaricatus var. albeofructus extract also showed a significant increase (p<0.05) in C group of 10 weeks. Administration of extracts from A. divaricatus var. albeofructus increased significantly in the production of IgG antibodies on the mice immunized with SRBC in B group of 7 and 10 weeks (p<0.05).
