The inhibitory effects of 25 polyphenols against in vitro allergic reactions were compared using biochemical and cell assays. Three polyphenols including curcumin, gallic acid, and quercetin suppressed the release of $\beta$-hexosaminidase from ionophore A23187-stimulated RBL-2H3 cells more effectively (>50% inhibition at $100{\mu}M$ concentration). They were found to have potencies in suppressing the release of histamine not only from ionophore A23187-, but also from immunoglobulin E (IgE)-stimulated RBL-2H3 cells. Moreover, such suppressive effects of the three polyphenols were also observed in A23187 plus PMA-costimulated rat peritoneal mast cells. The extent of inhibition were quantified as the respective polyphenol concentration that inhibit 50% ($IC_{50}$) of $\beta$-hexosaminidase or histamine release, showing an inhibition tendency with decreasing order of curcumin>gallic acid>quercetin. Down-regulation of $Ca^{2+}$ influx was suggested as the cause of the inhibition of $\beta$-hexosaminidase and histamine releases in these cells. The immune process inhibition was confirmed by the observed reduction in the gene expressions and release of pro-inflammatory cytokine tumor necrosis factor (TNF)-$\alpha$, interleukin (IL)-$1\beta$, and IL-4, due probably to antioxidant activity of the polyphenols. These findings illustrate that curcumin, gallic acid, and quercetin may be beneficial against allergic inflammatory diseases.
본 논문에서는 직사각형 및 삼각형 기생 패치를 이용한 광대역 마이크로스트립 패치 안테나를 제안하였다. 직사각형 마이크로스트립 패치 위에 직사각형 및 삼각형 모양의 기생 패치들을 적층하여 860MHz대역에서 광대역 특성을 얻었다. 주 방사부인 마이크로스트립안테나와 기생 패치와의 효과적인 결합은 이들 사이에 두꺼운 공기층을 두어 구현하였다. 또한, 이들 공기층 두께와 기생 패치의 위치는 광대역 정합에 중요한 요소임을 알 수 있었다. 제안된 안테나는 향후 소형 트랜시버에 적용하기 위해 $119mm{\times}109mm$ 크기의 소형 접지면 위에 설계 및 제작되었다. FR4기판에 제작된 안테나의 임피던스 대역은 818~919MHz(11.7%)이다. 방사패턴은 기존 마이크로스트립 패치 안테나와 유사했으며, 최대 이득은 주파수 824MHz에서 2.11dBi로 측정되었다.
The seroprevalence of human toxoplasmosis has been increasing in Korea, and it is controversial whether cats are an important infection source or not. This study was performed to evaluate the seroprevalence of Toxoplasma gondii infection in a high risk group (cat sitters) and to determine the possible importance of cats as an infection source in Korea. Risk factors, including the age, sex, and diet of cat sitters, their contact experience and contact frequency with stray cats, and origin, number, and outdoor activity of their pet cats, were analyzed using structured questionnaires. A total of 673 serum samples from people who have frequent contact with cats (high risk group) and 1,114 samples from general people (low risk group) were examined for specific IgG antibodies against T. gondii by ELISA. The results revealed that the overall seroprevalence of T. gondii infection was 7.4% (n=1,787). The seroprevalence among low risk group was 8.0% (89/1,114), whereas that among high risk group was rather lower 6.4% (43/673), though this difference was statistically not significant (P=0.211). Among the risk factors, only the outdoor activity of pet cats was important; people having cats with outdoor activities revealed 2 times higher seroprevalence than people having cats with only indoor activities (P=0.027). In conclusion, the seroprevalence of T. gondii was not significantly different between the high risk group and low risk group, and the importance of cats as a source of infection in Korea is questionable.
Black-red ginseng (BRGP) exhibits more potent biological activities than white and red ginseng. However, the effect of BRGP on allergic inflammation has not been studied extensively. In this study, we attempted to evaluate the effects of BRGP on allergic inflammation in vitro and in vivo. The effects of BRGP on pro-inflammatory mediators in phorbol myristate acetate (PMA) and A23187-stimulated human mast cells (HMC-1) and on trimellitic anhydride (TMA)-induced atopic dermatitis-like skin lesions in BALB/c mice were evaluated. BRGP suppressed the expression of $TNF-{\alpha}$, IL-6 and IL-8 from HMC-1 stimulated with PMA and A23187. Furthermore, the oral administration of BRGP markedly significantly reduced apparent severity of atopic dermatitis-like skin lesions, ear swelling, lymph node weight gains and serum IgE levels induced by TMA in mice. BRGP treatment also ameliorated epidermal hyperplasia and infiltration of inflammatory cells including mast cells. Taken together, BRGP possesses significant anti-atopic efficacy, suggesting that it could be used as a potential therapeutic agent for allergic inflammatory diseases, including atopic dermatitis.
To investigate the effects of anion replacement on NO3- concentration in Italian ryegrass. Plants weregrown hydroponically to the full vegetative stage. NO3 supply(control) was replaced with SO1- (Tl), C1-(T2) and water (T3) to during 14 days. The determination of inorganic nutrient uptake and quantification ofprincipal metabolites (nitrate. protein and sugar) followed. Relatively high uptake of ~ a a'n d Ca" for controlplants, K+ and POJ- for T2 plants, and C 1 for TI plants was observed, respectively. Proteins in shoot andstubble were relatively higher in control and TI plants, which coupled N source. In root proteins largelydecreased (especially in T3 plants) during experimental period. Sugars in shoot of all four treatments tendedto decrease during the first 7 days and recovered afterward. Sugars in stubble also markedly decreased duringthe first 7 days, while those in root was much less varied during experimental period. After 14 days oftreatment, nitrate concentration in shoot of control plants was 13mgIg FW. Comparing to control: nitrate inshoot reduced by 27%, 46% and 50% in TI, T2 and T3 plants, respectively. Objectives: Whitmania pigra whitman (WP) has been used in herbal medicine to treat various conditions, such as eczema, skin burns and frostbites in herbal medicine. The purpose of this study is was to investigate the effect of WP on anti-allergy mechanism. Methods: To clarify the mechanism, the effect of WP on vascular permeability of rat cutaneous tissue and histamine and cytokines (IL-6, IL-8, $TNF-{\alpha}$) released from mast cells were observed. Results: The results were 1. the pretreatment with WP significantly decreased the compound 48/80-induced degranulation and histamine release from RPMC 2. WP did not inhibit the anti-DNP IgE-induced increment of vascular permeability of rat cutaneous tissue 3. WP significantly reduced the PMA plus A23187-induced increment of expression of IL-6, IL-8, and $TNF-{\alpha}$ in HMC-1 cells. Conclusions: The present study providesevidence that WP acid inhibits mast cell-derived inflammatory allergic reactions by blocking histamine release and pro-inflammatory cytokine expression, and suggest the mechanisms of action. Furthermore, in vivo and in vitro anti-allergic effect of WP suggests a possible therapeutic application of this agent in inflammatory allergic diseases.
Purpose: B cell-activating factor (BAFF) is a tumor-necrosis factor (TNF) superfamily member best known for its role in the survival and maturation of B cells. BAFF activity is observed in naive cells as well as in effector/memory T cells. We aimed to explore whether BAFF in sputum is expressed at elevated levels in asthmatic airways and associated with eosinophilic inflammation, pulmonary function, and bronchial hyperresponsiveness in children. Methods: One hundred and fifty-four asthmatic children and 98 healthy children were enrolled in the study. Sputum supernatants were collected and sputum BAFF and eosinophil cationic protein (ECP) levels were measured. We performed pulmonary function tests and methacholine challenge tests, while measuring total eosinophil count, total serum IgE, and serum ECP in all subjects. Results: Asthmatic children had significantly higher levels of BAFF in induced sputum [26.50 (10.50-100.27) pg/mL] compared to healthy children [18.32 (7.68-44.63) pg/mL; $P$=0.011]. Sputum BAFF positively correlated with sputum eosinophils (${\gamma}$=0.406, $P$<0.001) and sputum ECP (${\gamma}$=0.789, $P$<0.001). Significant negative correlations were found between sputum BAFF and FEV1 (${\gamma}$=-0.291, $P$<0.001) or post-bronchodilator FEV1 (${\gamma}$=-0.334, $P$<0.001), whereas nonsignificant correlations were found between sputum BAFF and bronchial hyperresponsiveness, serum eosinophil count, and serum ECP. Conclusion: These findings suggest that BAFF may play a role in childhood asthma, and BAFF levels in sputum could be a supportive marker that represents airway inflammation, especially eosinophilic inflammation.

Atopic dermatitis (AD) is an inflammatory skin condition accompanied by symptoms such as edema and hemorrhage. Kimchi is a traditional fermented Korean dish consisting of various probiotics. In this study, the therapeutic effect of Lactobacillus plantarum CJLP55 isolated from Kimchi was studied in AD-induced mice. Orally administered Lactobacillus strain, CJLP55, suppressed AD symptoms and high serum IgE levels. CJLP55 administration reduced the thickness of the epidermis, infiltration of mast cells and eosinophils into the skin lesion, enlargement of axillary lymph nodes, and increase in cell population in axillary lymph nodes. CJLP55 treatment decreased the production of type 2 cytokines, such as interleukin (IL)-4, IL-5, IL-10, IL-12, interferon (IFN)-${\gamma}$, and IL-6,which were stimulated by house dust mite extracts, in the axillary lymph node cells. Orally administered CJLP55 exhibited a therapeutic effect on house dust mite-induced AD in NC/Nga mice after onset of the disease by altering immune cell activation. The Lactobacillus strain, CJLP55, isolated from Kimchi, suppressed AD. Our results suggest its possible use as a potential candidate for management of AD.
This study was performed to investigate the effect of dietary iron intake on the immune status of male college students. Twenty healthy male university students participated in the study. The mean age of the subjects was 22.6 years old, mean height was 173.3 cm and mean body weight was 68.4 kg. The mean daily iron intake of the subjects was 19.9 mg, 158.1% of the Korean recommended dietary allowances (RDA). The blood iron status and immune responses of the subjects were analyzed and compared between the high dietary iron group consuming more than 100% of the RDA of iron (Hi-Fe) and the low dietary iron group consuming less than 100% of the RDA of iron (Low-Fe). The serum iron concentration and percent saturation of transferrin were within the normal range in both groups. However, the Hi-fe group had higher serum iron and percent saturation of transferrin than the Low-fe group (p<0.05). When differential white blood cell counts were compared, the Low-Fe group had a lower percentage of neutrophils than the Hi-Fe group (p <0.1). The plasma IL-2 concentration, immunoglobulin levels and lymphocyte subsets were not affected significantly by the differences in iron intake as shown in this study. Serum iron had a positive correlation with monocyte percentage but had a negative correlation with IgM concentration. The results of this study suggest that slightly-low dietary iron intake without anemia has no effects on the cell-mediated and humoral immunities of healthy male university students. However, natural defenses, such as neutrophils and monocytes, seem to be more sensitively affected by changes in dietary iron intake.
Aim: The risk factors mostly strongly associated with gastric cancer are gastric bacteria Helicobacter pylori and diet. Using a case-control study among residents in Jinan, we examined the association between the salt taste and gastric cancer according to H. pylori infection, smoking and histological type as well as tumor site. Methods: This population-based case-control study included 207 cases and 410 controls. Data on potential risk factors of gastric cancer were obtained by interview of cases and controls with a questionnaire, salt taste preference was measured for all subjects, and IgG antibodies to H. pylori were applied to assess infection. Risk measures were determined using unconditional logistic regression. Results: The proportions of salt taste at intervals of 1.8-7.2 g/L and ${\geq}7.2$ g/L were significantly higher in cases than controls, with ORs of 1.56 (1.23-3.64) and 2.03 (2.12-4.11), respectively, subjects with high salt intake having an elevated risk for gastric cancer when infected with H. pylori. Significant modification by smoking and tumor site was observed across the different measures of salt intake, the highest salt taste showed higher cancer risk in ever smokers or with non-cardia cancers. Conclusion: Our study supports the view that high intake of sodium is an important dietary risk factor for gastric cancer, with a synergistic effect found between salt and H.pylori and smoking, dependent on the tumor site.
