Antimicrobial Activity of N-Acetyl-Phenylalanine Produced from Streptomyces sp. G91353 (Streptomyces sp. G91353이 생산하는 N-Acetyl-Phenylalanine의 항균활성)

  • For screening of the compounds exhibiting antimicrobial activities against the D-alanyl-D-alanine of Gram positive bacteria, approximately 2,500 actinomycetes isolated from soil were examined far antimicrobial activity. In consequence, we recently isolated the Streptomyces sp. G91353 strain produced an active compound, A91353, that inhibits the growth of Gram positive bacteria. A91353 was identified as N-acetyl-phenylalanine by various spectroscopic methods. The minimum inhibitory concentration (MIC) values of N-acetyl-phenylalanine on Gram positive bacteria such as Streptococcus pyogenes 308A, Streptococcus pyogenes 77A were determined as $50{\mu}g/ml$, respectively, but did not effect on Gram negative strains. These results indicate that N-acetyl-phenylalanine have an antimicrobial activity, which may be caused by the disturbance of D-alanyl-D-alanine synthesis.

Antimicrobial and Antitumoral Activities of Candida kefyr TEP 7 Isolated from Tibetan Fermented Milk (티벳산 발효유로부터 분리한 Candida kefyr TFP 7의 항균활성 및 항암활성)

  • This study was carried out to investigate antimicrobial and antitumoral activities of Candida kefyr TFP7 isolated from Tibetan fermented milk Strains of TFP1∼10 were isolated from Tibetan fermented milk by agar diffusion method using potato dextrose agar(PDA). Antimicrobial activities were examined against 18 microorganisms of food-related bacteria, yeast, algae, fungi and actinomycetes isolated from soil. Antitumor activities were examined against 9 human tumor cell lines. Strains of TFP2∼10 showed strong antimicrobial activities against Micrococcus luteus ATCC l1880, and strains of TFP6∼10 to actinomycetes, Streptomyces murinus JCM 4333. In antitumor test, all isolated strains(TEP1∼10) showed the growth inhibition of SNU-5 and SW-534 by 60% and 70%, respectively. Among those, the strain TFP7 showed the most antitumor activity, which was 77.5% for SNU-5 and 76.5% for SW-534. The strain was identified as Candida kefyr by use of API 20C AUX kit and scanning electron micrograph.

Isolation and Characterization of MT2617-2B, a Phospholipase C Inhibitor Produced by an Actinomycetes Isolate (방선균 분리주가 생산하는 Phospholipase C 저해물질인 MT-2617-2B의 분리 및 특성)

  • A phospholipase C (PLC) inhibitor (MT267-2B) was isolated from the culture broth of actinomycetes isolate MT2617-2 by the extraction with n-butanol and column chromatographic techniques. The molecular weight of the inhibitor was 1057, by the spectroscopic analyses of IR, $^{13}C$-and $^{1}H$-NMR and ESI-MS. The chemical structure of MT2617-2B was found to be a macrolide compound consisted of a hemiketal ring, polyhydroxyl and polymethyl groups, which had a malonate and guanidine group as its side chain. MT2617-2B produced its two isomers having the same molecular weight by standing in methanol solution at room temperature. Therefore, MT2617-2B was identified as copiamycin and niphithricin A, macrolide antibiotics. The values of $IC_{50}$ against PLC-${\gamma}$1 and PLC-${\beta}$1 were 25 and 50${\mu}$g/ml, respectively. MT2617-2B had antimicrobial activities against Staphylococcus aureus and Candida albicans, but not against Escherichia coli.

  • Four actinomycetes which produces extracellular chitinase were isolated from soil and organic matter all over the Chonnam provincial area. The chemical composition, morphological, cultural and physiological properties of isolated strain S-25, S-42, S-172 and S-267 were studied in relation to the toxonomical properties. All of strains contained phospholipids such as phosphatidyl ethanolamine, phosphatidyl inositol and diphosphatidyl glycerol. The components of cell wall in all strains have L, L-DAP, Glutamic acid, Alanine, Glycine and Glucosamine. The surface of spore is smooth and colony is grey in all strains, Based on the results obtained in these experiments all of strains are identified as Streptomyces sp.S-25, Streptomyces sp.S-42, Streptomyces sp.S-l72 and Streptomyces sp. S-267.

  • The strain YSK-681 has been selected for antibiotic-producing strain against methicillin resistant Staphylococcus aureus(MRSA) form 1,000 strains of actinomycetes which had been isolated from soil. The strain YSK-681 has been identified on the point of morphological, cultural, physiological and chemical characteristics. Forty-one taxonomic unit characters were tested and the data were analysed numerically using the TAXON program. The isolate was classified into the major cluster 29 of Streptomyces and best-matched to Streptomyces lydicus. Therefore, it was concluded that the isolate was identified to be Streptomyces lydicus.

Screening and Identification of Actinomycetes Producing Novel Elastase Inhibitor (Elastase Inhibitor 생성 방선균의 탐색 및 동정)

  • Elastase inhibitor (EI) producing microorganisms were screened by microplate-assay of culture broth using Succinyl-$(Alanine)_3$-Nitroanilide as a chromogenic substrate. The portion of microorganisms showed EI activity above 90% was about 2% of all assayed. Isolated SMF-11 showed relatively high EI activity after filtration using membrane filter of NMWL 3,000. SMF-11 was identified as Streptomyces sp. according to results of morphological or physicochemical identification. SMF-11 has both rectiflexible and spiral spore chains with smooth spore surface and cell wall contained LL-DAP, iso- and anteiso-fatty acids. The amino acids in cell wall were alanine, glutamic acid and glycine. Fifty unit characters for major cluster were tested and the data were analyzed numerically using the TAXON program. The isolate SMF-11 was identified as a strain of Streptomyces lavendulae.

A report on five unrecorded bacterial species belonging to the phyla Actinomycetota, Bacillota and Pseudomonadota in Korea isolated in 2020

  • During an investigation into the indigenous prokaryotic species diversity in Korea, a total of five bacterial strains were isolated from various environments in Korea. The isolated bacterial strains were identified by analyzing their 16S rRNA gene sequences, and those with a minimum of 98.7% sequence similarity with known bacterial species but not reported in Korea were designated as unrecorded species. These isolates were assigned to three phyla, five orders, five families, and five different genera. The isolates were identified as Cumulibacter manganitolerans (99.1%) and Myolicibacterium tusciae (98.7%) of the class Actinomycetes; Bacillus marasmi (99.9%) of the class Bacilli; and Novosphingobium mathurense (100%) and Microvirga ossetica (98.8%) of the class Alphaproteobacteria. Gram reaction, colony and cellular morphology, basic biochemical characteristics, and phylogenetic position of theses isolates are also described.

Selection and Identification of a Strain KT-10 Producing the Cathepsin B Inhibitor

  • An actinomycetes, KT-10 isolated from ginseng field in Kyongpook, Korea was selected based on its ability to produce a lysosomal cathepsin B inhibitor. The inhibitor purified from the culture supernatant of the isolate KT-10 showed strong inhibitory effects against cathepsin B as well as against papain when the activities were measured using synthetic substrate, ${\alpha}$-N-benzyloxycarbonyl-L-Iysine p-nitrophenyl ester (CLN) or ${\alpha}$-N-benzoyl-D,L-arginine 2-naphthylamide (BANA). The isolate KT-10 was identified as a species of Streptomyces based on its morphological characteristics and chemotaxonomic data. The TAXON program of Ward was used to identify Streptomyces sp. KT-10 as a strain of Streptomyces luteogriseus belong to cluster 18 of the genus Streptomyces with a Willcox probability 0.999388. The cathepsin B inhibitor was presumed to a novel material composed of a polyhydroxylamine.

방선균이 생산하는 아미노산 대사길항물질, YS-460의 분리 정제 및 특성

  • An amino acid antimetabolite named YS-460 was isolated from the culture filtrate of a newly isolated Actinomycetes identified as Streptomyces sp. Fermentation was carried out in the synthetic medium at 30$\circ$C for 5 days. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 38 mg of pure active substance per liter of the broth. YS-460, an amino acid like substance, has the molecular formula of C$_{7}$H$_{11}$NO$_{3}$- Its structure determined to be furanomycin by spectral analysis. It is active against some bacteria on a chemically defined medium and reversed competitively by L-isoleucine and non-competitively by L-leucine and L-valine.

Identification of the $\alpha$-Amylase Inhibitor Producing Actinomycetes BY-445 ($\alpha$-아밀라제 저해물질을 생성하는 방선균 BY-445의 동정)

  • The strain BY-445 which produces new inhihitors of ${\alpha}$-amylase was isolated from a soil sample and identified. The aerial hyphae of this strain develope in the from of open spirals. The spore chain of BY-445 strain appears in spiral shape with spiny surface. Melanoid and soulble pigments were not observed. Gelatin was liquefied, and skin milk and starch was also hydrolyzed. The isolate contained LL-diaminopimelic acid in its cell wall hydrolysate. The content of fatty acid 16:iso, 15:0 anteiso and 16:0 was 25:30, 16.19 and 13.16%, respectively. BY-445 strain was closely related to Streptomyces violaceusinger but it was different from this strain in some cultural and physiological characteristics. This strain was, therefore, designated as Streptomyces sp. BY 445.
