Isolation and Characterization of a Lytic and Highly Specific Phage against Yersinia enterocolitica as a Novel Biocontrol Agent

  • The aim of this study was to isolate and characterize a lytic Yersinia enterocolitica-specific phage (KFS-YE) as a biocontrol agent. KFS-YE was isolated and purified with the final concentration of ($11.72{\pm}0.03$) log PFU/ml from poultry. As observed by transmission electron microscopy, KFS-YE consisted of an icosahedral head and a contractile tail, and was classified in the Myoviridae family. KFS-YE showed excellent narrow specificity against Y. enterocolitica only. Its lytic activity was stable at wide ranges of pH (4-11) and temperature ($4-50^{\circ}C$). The latent period and burst size of KFS-YE were determined to be 45 min and 38 PFU/cell, respectively. KFS-YE showed relatively robust storage stability at -20, 4, and $22^{\circ}C$ for 40 weeks. KFS-YE demonstrated a bactericidal effect in vitro against Y. enterocolitica and provided excellent efficiency with a multiplicity of infection as low as 0.01. This study demonstrated the excellent specificity, stability, and efficacy of KFS-YE as a novel biocontrol agent. KFS-YE may be employed as a practical and promising biocontrol agent against Y. enterocolitica in food.

Isolation and Characterization of Chlorella Virus from Fresh Water in Korea and Application in Chlorella Transformation System

  • Chlorella viruses are large icosahedral, plaque-forming, dsDNA viruses that infect certain unicellular, chlorellalike green algae. The genomic DNA of over 300 kb contains many useful genes and promoters. Over 40 chlorella viruses have been isolated from fresh water in Korea since 1998. The viruses were amplified initially in chlorella strain NC64A, and pure isolates were obtained by repeated plaque isolation. SDS-PAGE analysis revealed similar but distinct protein patterns, both among the group of purified viruses and in comparison with the prototype chlorella virus PBCV-1. Digestions of the 330- to 350-kb genomic DNAs with 10 restriction enzymes revealed different restriction fragment patterns among the isolates. The tRNA-coding regions of 8 chlorella viruses were cloned and sequenced. These viruses contain 14-16 tRNA genes within a 1.2- to 2-kb region, except for the SS-1 isolate, which has a 1039-bp spacer in a cluster of 11 tRNA genes. Promoter regions of several early genes were isolated and their activities were analyzed in transformed chlorella. Some promoters showed stronger activity than commonly used CaMV 35S promoter and chlorella transformation vectors for heterologous protein are beings constructed using these promoters.

A Cytoplasmic Polyhedrosis Virus Isolated from the Pine Processionary Caterpillar, Thaumetopoea pityocampa

  • A cytoplasmic polyhedrosis virus (CPV) was isolated from the larvae of Thaumetopoea pityocampa and shown to cause an infection of midgut cells. This viral infection revealed several important diagnostic symptoms, including discoloration of the posterior midgut, reduced feeding, and extended development time of the larvae. The virus infection is lethal to Thaumetopoea pityocampa, and with the increasing doses kills the larvae within 4-5 days post infection. Electron microscopy studies showed typical cytoplasmic polyhedral inclusion bodies that are icosahedral, and ranged from 2.4 to $5.3{\mu}m$ in diameter. Electrophoretic analysis of the RNA genome showed that the virus has a genome composed of 10 equimolar RNA segments with the sizes of 3,907, 3,716, 3,628, 3,249, 2,726, 1,914, 1,815, 1,256, 1,058, and 899 bp, respectively. Based on morphology and nucleic acid analysis, this virus was named Thaumetopoea pityocampa cytoplasmic polyhedrosis virus (TpCPV), and belongs to the genus Cypovirus, family Reoviridae.

Ab initio DFT studies on dimerization and one-dimensional (1-D) polymerization of M@$Au_{12}$ (M = W, Mo) clusters

  • Using ab initio density functional theory, we investigate the dimerization and one-dimensional (1-D) polymerization of metal-encapsulated gold nanoclusters, M@$Au_{12}$ (M=W, Mo) and their structural and electronic properties. M@$Au_{12}$ clusters with a magic number 13 can form icosahedral and cuboctahedral structures. We consider various dimer configurations with different compounds and symmetries to find the most stable dimer structure in each case. Au atoms in the one cluster, which participate directly in dimerization, tend to form triangular bonds together with counterpart Au atoms in the other. It is found that both M@$Au_{12}$ and M@$Au_{12}$ clusters are stabilized by about 3 eV due to dimerization. We also calculate and compare the electronic and magnetic properties of different dimerized clusters. Based on our investigation on dimerization, we further study on 1-D polymerization of M@$Au_{12}$ with different compounds and symmetries. We will also discuss their formation energies as well as their electronic and magnetic properties.

Isolation and Characterization of a Bacteriophage Preying an Antifungal Bacterium

  • Several Bacillus species were isolated from rice field soils, and 16S rRNA gene sequence analysis showed that Bacillus cereus was the most abundant. A strain named BC1 showed antifungal activity against Rhizoctonia solani. Bacteriophages infecting strain BC1 were isolated from the same soil sample. The isolated phage PK16 had an icosahedral head of $100{\pm}5nm$ and tail of $200{\pm}5nm$, indicating that it belonged to the family Myoviridae. Analysis of the complete linear dsDNA genome revealed a 158,127-bp genome with G + C content of 39.9% comprising 235 open reading frames as well as 19 tRNA genes (including 1 pseudogene). Blastp analysis showed that the proteins encoded by the PK16 genome had the closest hits to proteins of seven different bacteriophages. A neighbor-joining phylogenetic tree based on the major capsid protein showed a robust clustering of phage PK16 with phage JBP901 and BCP8-2 isolated from Korean fermented food.

Outbreak of Canine Parvoviral Enteritis in Korea (개 파보바이러스성(性) 장염(腸炎)의 국내발생(國內發生))

  • The disease syndrome characterized by the acute vomiting and diarrhea with high mortality had been greatly epidemic in Korea since June, 1931 and it was followed serologically and electron microscopically for the clarification of the agent. The agent present in feces of dogs associated with this syndrome had characteristic feature in agglutinating pig red blood cells that was specifically inhibited by anti-CPV reference dog serum. This also showed the serological identity with the reference CPV antigen in immuno-diffusion. Electron micrograph of the material revealed parvovirus particles with size of 20nm and icosahedral structure. These results clearly indicated that CPV was the primary cause of canine epidemic prevailing in 1981 in Korea.

  • In the present study, the possibility of Mg-Zn-Y alloys as high temperature casting alloys has been investigated. The fluidity of alloys containing yttrium were better than that of commercial AZ91 alloy because the oxide layer on the surface reduced the reaction between melt, and air and mold, which would reduce the resistance during the process of filling the mold. However, this oxide film reduced the hot-tearing resistance. In the case of ZAW942, this alloy exhibited fluidity and hot-tearing resistance better than AZ91 alloy. Because of thermally stable quasicrystal and other phases obstructed the movement of grains, the creep resistance of alloys containing rare earth elements more than 2 wt% was better than that of AZ91 alloy.

Effect of calcium addition on creep properties in Mg-Zn-Y alloys (Mg-Zn-Y 합금의 크리프 저항성에 미치는 칼슘의 영향)

  • In the present study, the high temperature mechanical properties and creep resistance of Mg-Zn-Y-Ca alloys has been investigated. The Mg-4Zn-0.8Y alloy consists of ${\alpha}$-Mg matrix and icosahedral quasicrystalline phase. Calcium addition into Mg-4n-0.8Y based alloy results in the formation of ${\tau}(Ca_{2}Mg_{6}Zn_{3})$ and $Mg_{2}Ca$ as the second solidification phases. Creep properties of the Mg-Zn-Y and Mg-Zn-Ca based alloys measured at applied stresses between 65 MPa and 85 MPa are significantly improved with adding calcium and yttrium, respectively. The improved creep resistance is due to the formation of thermally stable $Mg_{2}Ca$ phase.

Extrusion Behavior of Gas Atomized Mg Alloy Powders (가스분무 Mg-Zn-Y 합금분말의 압출거동)

  • This work is to report not only the effect of rapid solidification of $MgZn_{4.3}Y_{0.7}$ alloys on the micro-structure, but also the extrusion behavior on the materials properties. The average grain size of the atomized powders was about $3-4{\mu}m$. The alloy powders of $Mg_{97}Zn_{4.3}Y_{0.7}$, consisted of I-Phase (Icosahedral, $Mg_{3}Zn_{6}Y_{1}$) as well as Cubic structured W-Phase ($Mg_{3}Zn_{3}Y_{2}$), which was finely distributed within ${\alpha}-Mg$ matrix. The oxide layer formed along the Mg surface was about 48 nm in thickness. In order to study the consolidation behavior of Mg alloy powders, extrusion was carried out with the area reduction ratio of 10:1 to 20:1. As the ratio increased, fully deformed and homogeneous microstructure could be obtained, and the mechanical properties such as tensile strength and elongation were simultaneously increased.

Isolation and Characterization of Temperate Phages in Enterococcus faecium from Sprouts (새싹채소 유래 Enterococcus faecium으로부터 Temperate Phage의 분리와 특성)

  • To analyze the characteristics of bacteriophages in Enterococcus faecium, D-19 and F6 phages were induced from five E. faecium isolated from sprouts by the treatment with mitomycin C. The bacteriophages of D-19 and F-6 had long, non-contractile tails and icosahedral heads, and were members of Siphoviridae family. As the host spectrum, D-19 phage lysed five out of 55 strains of E. faecium, whereas F6 phage lysed only three strains. Both D-19 and F6 phages displayed similar and high stabilities against ethanol and pH capable of resisting the exposure to 100% ethanol and pH 4.
