Nonvolatile Flavor Components in Chinese Quince Fruits, Chaenomeles sinensis koehne (모과의 비휘발성 Flavor 성분에 관한 연구)

  • This study was performed to identify detailed informations on the nonvolatile flavor of Chinese quince fruits, Chaenomeles sinensis koehne. About 72% of the free amino acids were shown to be valine, asparagine, ${\gamma}-aminobutyric\;acid$, aspartic acid and serine. Arginine, tyrosine, methionine and tryptophan were not present. Glutamic acid and glutamine as a amino acid for peptides were the major components, whereas cysteic acid, methionine sulfone and tryptophan were not detected. The nucleotides attained were composed of cytosine, uridine-5'-monophosphate and cytidine-5'-monophosphate, and these were proved to be a very small quantity. Guanosine-5'-monophosphate, inosine-5'-monophosphate and adenosine-5'-monophosphate were not present. The major sugars were shown to be glucose, sorbose, sucrose and fructose. Fructose was the most abundant one among them. A total of 11 organic acids were identified by capillary gas chromatography and capillary gas chromatography-mass spectrometry. The major components identified were tartaric acid and α-ketoglutaric acid. The total content of vitamin C determined was 386.6mg%, and those of ascorbic, dehydroascorbic, and 2, 3-diketo-L-gulonic acid were 28.8mg%, 154.5mg% and 197.3mg%, respectively. Calcium and phosphorus were the major components, while heavy metals such as cadmium, copper and lead were determined to be a small amount. In the result of organoleptic test on the natural and synthetic extract of Chinese quince fruits, the principal taste components consisted of free amino acids, sugars, organic acids, vitamin C and minerals. Five groups mentioned would have a favorable influence upon the taste of fresh Chinese quince fruits.

  • RNA degrading bacteria were isolated from soil of Korea. One strain (no. JSC-114), having strong 5'-phosphodiesterase activity, was identified as belonging to the genus Streptomyces on the basis of taxonomic characteristics. The optimum conditions of 5'-phophosdiesterase production were found at $30^{\circ}C$ for 4 day in a medium containing 4.5% of soluble starch, 0.15% of peptone, 0.6% of yeast extract, 0.1% of $MgSO_4{\cdot}7H_2O$, 0.01% of $CaCl_2{\cdot}2H_2O$, 0.25% of $KNO_3$, and 0.5% of $KH_2PO_4$(pH 7.0). The maximum production rate of 5'-nucleotides from yeast RNA was 95% at $40-45^{\circ}C$ for 4hrs, and the products were identified as 5'-IMP, 5'-GMP, 5'-CMP and 5'-UMP(5.5 : 5.0 : 4.9 : 5.0).

  • Carp, Cypinus carpio, has been esteemed as one of the most tasty fresh water fishes in Korea. But little study on its food has been reported. The composition of fatty acids, amino acids, nucleotides and their related compounds as chemical component of carp muscle were analyzed by gas-liauid chromatography, amino acid autoanalyzer and high speed liauid chromatography. The fatty acids of the carp muscle lipid consisted of the large amount of $C_{18}\;:\;2,$ $C_{18}\;:\;1,$ $C_{18}\;:\;3,$ $C_{16}\;:\;0,$ $C_{16}\;:\;1,$ acids, small amount of $C_{16}\;:\;2,$ $C_{18}\;:\;0,$ $C_{18}\;:\;4,$ $C_{14}\;:\;0,$ $C_{14}\;:\;1,$ $C_{12}\;:\;0,$ acids, and $C_{13}\;:\;1,$ $C_{13}\;:\;0,$ $C_{20}\;:\;1,$ $C_{17}\;:\;0,$ $C_{15}\;:\;1,$ $C_{17}\;:\;1,$ $C_{22}\;:\;1$ acids were smaller. In fresh carp, the results showed that inosine $17.25{\mu}mole/g$, hypoxanthine $10.06{\mu}mole/g$ were dominant and the content of GMP, IMP, AMP, UMP, CMP were 1.56, 1.36, 0.92, 0.49, $0.34\:{\mu}mole/g$ on dry base, respectively. Histidine and lysine were dominant amino acids in carp extract, having 49.4% (214.6mg%), 33.9% (147.1mg%) of total free amino acid contents, respectively, but the content of glycine, serine, alanine and glutamic acid were low, and arginine, aspartic acid, threonine, proline, valine, methionine, isoleucine, phenylalanine were detected in trace amount. The content of amino acid composition showed that glutamic acid, aspartic acid, lysine, leucine, alanine, valine and arginine were the most abundant amino acids, while such amino acids as glycine, isoleucine, phenylalanine, threonine, histidine, methionine, serine and tyrosine were low, and proline was detected in trace amount.

The Porcine FoxO1, FoxO3a and FoxO4 Genes: Cloning, Mapping, Expression and Association Analysis with Meat Production Traits

  • FoxO1, FoxO3a and FoxO4 belong to the FoxO gene family, which play important roles in the PI3K/PKB pathway. In this study, we cloned the porcine FoxO1, FoxO3a and FoxO4 sequences and assigned them to SSC11p11-15, SSC1p13 and SSC xq13 using somatic cell hybrid panel (SCHP) and radiation hybrid panel (IMpRH). RT-PCR results showed that these three genes are expressed in multiple tissues. Sequencing of PCR products from different breeds identified a synonymous T/C polymorphism in exon 2 of FoxO3a. This FoxO3a single nucleotide polymorphism (SNP) can be detected by AvaII restriction enzyme. The allele frequencies of this SNP were investigated in Dahuabai, Meishan, Tongcheng, Yushan, Large White, and Duroc pigs. Association of the genotypes with growth and carcass traits showed that different genotypes of FoxO3a were associated with carcass length and backfat thickness between 6th and 7th ribs (BTR) and drip loss (p<0.05).

Growth and Lodging of Rice as Affected by Growth Regulators under Different Midsummer Drainage Times in Puddled-soil Drill Seeding (벼 무논골뿌림 재배시 중간낙수 및 생장조절제 처리가 생육 및 도복에 미치는 영향)

  • This experiment was carried out to elucidate the effect of the midsummer drainage times and some growth regulators on lodging characters, lodging and yield in puddled-soil drill seeding in rice. Dongjinbyeo, the mid-late maturing rice variety was seeded at May 11 by seeding machine with 4cm of furrow depth. Experimental plots were divided two main treatment without midsummer drain and two time drains (30 and 50 days after seeding), Inabenfide(IBF) was applied 40days before heading(DBH) and IBP was applied at 30DBH, respectively. Culm length was shorten, the wall of N$_4$ was thicken, and the breaking weight was increased at two time drainage and growth regulators applied in order of Inabenfide, IBP, and Control. Lodging wasn't occurred at two time drainage but it was occurred at none drainage in the order of Control, IBP, and Inabenfide applied. Yield was higher at two time drainage compared with none drainage and higher in order of Inabenfide, IBP and Control in none drainage but wasn't significantly different among growth regulators applied in two time drainage. Therefore, two times midsummer soil drying is recommendable management method for puddled -soil drill seeding of rice. Mapping, Tissue Distribution and Polymorphism of Porcine Retinol Binding Protein Genes (RBP5 and RBP7)

  • The retinoids (vitamin A and its derivatives) play a critical role in vision, growth, reproduction, cell differentiation and embryonic development. Using the IMpRH panel, porcine cellular retinol binding protein genes 5 and 7 (RBP5 and RBP7) were assigned to porcine chromosomes 5 and 6, respectively. The complete coding sequences (CDS) of the RBP5 and RBP7 genes were amplified using the reverse transcriptase polymerase chain reaction (RT-PCR) method, and the deduced amino acid sequences of both genes were compared to human corresponding proteins. The mRNA distributions of the two genes in adult Wuzhishan pig tissues (lung, skeletal muscle, spleen, heart, stomach, large intestine, lymph node, small intestine, liver, brain, kidney and fat) were examined. A total of nine single nucleotide polymorphisms (SNPs) were identified in two genes. Three of these SNPs were analyzed using the polymerase chain reaction-restriction-fragment length polymorphism (PCR-RFLP) method in Laiwu, Wuzhishan, Guizhou, Bama, Tongcheng, Yorkshire and Landrace pig breeds. Association analysis of genotypes of these SNP loci with economic traits was done in our experimental populations. Significant associations of different genotypes of $RBP5-A/G^{63}$, $RBP5-A/G^{517}$ and $RPB5-T/C^{intron1-90}$ loci with traits including maximum carcass length (LM), minimum carcass length (LN), marbling score (MS), back fat thickness at shoulder (SBF), meat color score (MCS) and hematocrit (HCT) were detected. These SNPs may be useful as genetic markers in genetic improvement for porcine production.

  • This study developed a natural seasoning (NS) and characterized its food components. Hydrolysate from Alaska Pollock Theragra chalcogramma heads and sea tangle Laminaria japonica byproduct were obtained by incubating them with Neutrase for 4 h. NS was prepared by mixing sorbitol 2%, salt 2%, ginger powder 0.04%, garlic powder 0.2%, onion powder 0.2% and inosine monophosphate (IMP) 0.1% based on concentrated hydrolysates from Alaska pollock head and sea tangle byproduct before vaccum filtering. The proximate composition of NS was 82.7% moisture, 9.0% crude protein, and 5.1% ash. It had a higher crude protein content than commercial anchovy sauce (CS), it was lower in moisture and ash. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and angiotensin-I converting enzyme (ACE) inhibiting activity of NS were 90.1% and 88.9%, respectively, which were superior to those of CS. The free amino acid content and total taste value of NS were 1,626.0 mg/100 mL and 165.86, respectively, which were higher than those of CS. According to the results of taste value, the major free amino acids were glutamic acid and aspartic acid. In the sensory evaluation, the color and taste of NS were superior to those of CS. No difference in fish odor between NS and CS was found.

Study on the Taste Characteristics of the Chemical Seasoning (MSG) Mixed with the Various Contents of Nucleotides (핵산함유 화학조미료의 맛특성에 관한 연구)

  • This study was to investigate the synergistic taste effect between monosodium glutamate(MSG) and 5'-ribonucleotides consisted of disodium 5'-inosinate (IMP) and 5'-guanylate (GMP) as 1:1 ratio. Solvent was distilled water. Sensory evaluation with 10 panelists was performed by using ratio scaling method (magnitude estimation). The results were as follows: 1) Taste intensities were increased, as nucleotides content to MSG increased. 2) Multiple regression analyses were carried out with the taste intensity data as a function of nucleotides content at three concentrations of seasonings, 0.025%, 0.05% and 0.1%. 3) Predicted taste intensities ($Y_P$) were calculated from the regression equation. Also taste intensity ratios ($Y_{TR}$)-$Y_{TR}$=$Y_P$/taste intensity of MSG only-were calculated. 4) The taste intensity ratios ($Y_{TR}$) at three concentrations of seasonings in the same nucleotides contents showed about the same. Therefore, instead of above regression equations, only one multiple regression equation expressing $Y_P$ of nucleotides seasonings could be determined, as functions of nucleotides content and seasoning concentration.

Quality Properties of Fermented Squid Seasoning Manufactured with Fermentation Accelerator (발효촉진제를 첨가하여 제조한 오징어 조미료의 품질특성)

  • Squid was fermented with fermentation accelerators to develop a natural complex seasoning. The quality properties of fermented squid were determined at different fermentation periods. Squid fermented with 10% Aspergillus oryzae koji for 10 days had the highest amino-N, acidity, and total viable cell content during fermentation periods, whereas volatile basic nitrogen content and pH were the lowest. Based on the amino-N content, squid with 10% koji fermented for 10 days was selected for further analyses. The inosine and glutamic acid contents of the fermented squid were highest innucleotide composition, their related compounds, and free amino acids, respectively. The $IC_{50}$ values of the fermented squid on DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging and ${\alpha}$-glucosidase inhibitory activities were 6.20 and 4.41 mg/mL, respectively. Based on the results of a sensory evaluation, the fermented squid seasoning was similar to other natural complex seasonings such as anchovy, cowmeat, and fisheries seasonings.

Extractive Nitrogenous Constituents of Anchovy Sauce and their Quality Standardization (멸치액젓의 맛성분조성(成分組成) 및 품질표준화(品質標準化)에 관(關)한 연구(硏究))

  • Extractive nitrogen, free amino acids, oligopeptides, nucleotides and related compounds, quaternary ammonium bases, and guanidino compounds were analyzed to evaluate quality of anchovy sauce. The commercial products contained low proximate composition, extractive nitrogen and other extractive components, than the experimentally prepared anchovy sauce. Both samples, commercial products and experimentally prepared anchovy sauce, were rich in free amino acids, such as glutamic acid, leucine, alanine, lysine, and aspartic acid. The extractive nitrogenous components which consist of total nucleotides and related compounds, total free amino acids, methionine, isoleucine, valine, taurine, tyrosine, histidine, leucine, aspartic acid, cystine, and lysine, showed significant correlation(p<0.01) with extractive nitrogen. Possibly, seven kinds of free amino acids such as methionine, isoleucine, valine, taurine, tyrosine, histidine, and leucine, might be recommend as quality indices of standardization for anchovy sauce.
