Purpose: To investigate temporal changes in IL-1${\beta}$ mRNA expression in spinal dorsal horn (DH) and dorsal root ganglion (DRG) in a rat lumbar disc herniation (LDH) model. Methods: Autologous nucleus pulposus, harvested from the tail disc between the second and third coccygeal vertebrae (Co2-3), was implanted next to the left L5 nerve root just proximal to the DRG after partial laminectomy. IL-1${\beta}$ mRNA expression was investigated in DRG and DH in our LDH model. Real-time PCR assays were done using a 7500 Real Time PCR system (Applied Biosystems, USA). Results: Expression of IL-1${\beta}$ in DRG and DH was observed for 30 days postoperatively. Expression of IL-1${\beta}$ mRNA in the ipsilateral DRG of the LDH group gradually increased from 5 to 30 days after surgery. The amount of IL-1${\beta}$ in the contralateral DRG peaked 10 days after surgery and then gradually decreased. However, there was no difference in IL-1${\beta}$ mRNA expression in spinal DH between the LDH group and the sham-operated group. Conclusion: Long-term expression of IL-1${\beta}$ in the LDH model may worsen the chronic pain state. Future studies on inhibition of IL-1${\beta}$ expression in the LDH model will be needed to develop selective treatment strategies for patients with LDH.
Transferrin ($T_{f}$) has been used as a targeting ligand for delivering liposome/interleukin-12 (IL-12) pDNA complexes to cancer cells mostly due to the greater number of transferrin receptors ($T_{f}R$) found on tumor cells than on normal cells. $T_{f}$ was conjugated to liposomes via the reaction of MPB-PE with thiol groups of $T_{f}$ introduced by a heterobifunctional cross-linking agent, N-succinimidyl-3-(2-pyridyldithio)propionate (SPDP). Four days after C26 inoculation when the tumor volume reached ${\sim}100mm^{3}$, tumor-bearing Balb/c mice were injected intravenously with $T_{f}-liposome/IL-12 pDNA$complexes twice a week for 3 weeks. Significant suppression of tumor growth was achieved in the group treated with the $T_{f}-liposome/IL-12 pDNA$ complexes, with a dose of $10{\mu}g$ of IL-12 pDNA showing the highest suppression effect among the tested doses. Similar results were obtained when the therapy was initiated one day after tumor inoculation, although in this case $30{\mu}g$ IL-12 pDNA/$T_{f}-liposome$ complexes showed a significant suppression of tumor growth between 19 and 23 days after tumor inoculation. This result indicates that the transferrin receptor-targeted liposomal system is an efficient delivery agent of therapeutic genes, such as IL-12, in mice and that its potential clinical use warrants further research investigation.
Purpose: Alterations in the structure and function of vascular smooth muscle cells (VSMCs) are important in cardiovascular disease and maintaining chronic hypertension. Chronic hypertension is associated with changes in vascular smooth muscle tone. The spontaneous or myogenic tone of a blood vessel reflects the ability to adapt smooth muscle tone to changes in transmural pressure. However, the intracellular signaling mechanisms involved in myogenic tone are not fully understood. Methods: Here, we investigated the relationship between mitogen-activated protein kinases (MAPKs) and phosphatidylinositol-3 kinase (PI3K) in isometric contraction and enzymatic activity using muscle strips from rats made hypertensive with aldosterone-analogue deoxycorticosterone acetate (DOCA) salts. Results: Changes in myogenic tone and intracellular $Ca^{2+}$ concentration ($[Ca^{2+}]_i$) were different after physiological salt solution (PSS) in normotensive and hypertensive rats. The myogenic tone and quiescent phosphorylation induced by the PSS treatment were inhibited by 10 ${\mu}$M PD098059, an extracellular-regulated protein kinase 1/2 (ERK1/2) inhibitor, and 10 ${\mu}$M wortmannin, an inhibitor of PI3K, in hypertensive rats. Conclusion: The development of DOCA-induced hypertension is associated with altered isometric contractions and $[Ca^{2+}]_i$ via changes in activation of ERK1/2 and PI3K after DOCA-salt treatment. Therefore, ERK1/2 and PI3K activity affect hypertension and may be suitable targets for physical therapy in cardiovascular disease.
Objectives : This study was conducted to investigate the effects Innotus obliquus on the cytokine expression in Raw 264.7 cells. Methods : In Raw 264.7 cells stimulated with lipopolysaccaride(LPS) to Mimic inflammation, Innotus obliquus ethanol extract inhibited nitric oxide production in a dose-dependent manner and abrogated inducible nitric oxide sythase(iNOS) and cyclooxigenase (COX-2). The levels of cytokines were analyzed by sandwich immuno assays. Results and Conclusions : Results provided evidence that Innotus obliquus inhibited the production of IL-b, and IL-6 in Raw 264.7 cells activated with lipopolysaccharide (LPS). These finding suggested that Innotus obliquus can produce anti-inflammatory effect, which may playa role in adjunctive therapy in Gram-negative bacterial infections.
Flavonoids are well-known anti-inflammatory agents that exert their effects via a variety of mechanisms including antioxidative action, inhibition of eicosanoid metabolizing enzymes and regulation of theexpression of proinflammatory molecules. In this review, synthetic approaches to obtain more useful flavonoid derivatives are summarized. Human clinical trials of flavonoid therapy are discussed. Through continual investigation to identify more potent and comparable flavonoids, new anti-inflammatory flavonoid therapy will be successfully launched, especially for the treatment of chronic inflammatory disorders.
Equisetum hyemale Linne. (EH) (Equisetaceae) has been used for the treatment of eye and skin disease, chronic eczema, pneumoconiosis and asthma in Korea and China. Human leukemic mast cells are widely distributed in the connective tissues of mammals and other vertebrates. Phorbol 12-myristrate 13-acetate (PMA) and calcium ionophore A23187 stimulated Human leukaemic mast cell line-1 (HMC-1) can produce a variety of inflammatory mediators and several pro-inflammatory and chemotactic cytokines such as TNF-$\alpha$, IL-6 and IL-8. Since TNF-$\alpha$, IL-6 and IL-8 are major factors during the inflammatory process, we studied the effects of EH on TNF-$\alpha$, IL-6 and IL-8 release in HMC-1 stimulated with PMA and A23187. The result of this study indicate that EH inhibits TNF-$\alpha$, IL-6 and IL-8 in activated HMC-1 cells via $I{\kappa}B$/Nuclear factor-${\kappa}B$ pathway. Therefore, EH might contribute significantly to the prevention or treatment of mast-cell mediated inflammatory diseases and EH has potential use in the therapy of chronic allergic inflammation.

The purpose of this study were to examine the effect of high intensity, low frequency transcutaneous electrical nerve stimulation at auricular acupuncture points on experimental pain threshold measured at the wrist and to determine the changes in effect over time. Forty-two healthy adult men and women were assigned randomly to one of three treatment groups. Group 1(n=15) received TENS to appropriate auricular points for wrist, Group 2(n=12) received TENS to wrist, and Group 3(n=15) received no TENS. We measured experimental pain threshold at the wrist after an electrical stimulus during one pre-treatment and three post-treatment time periods. Group 1 and 2 showed stastically significant increase(p<0.05) in pain threshold after treatment whereas the Group 3 did not. Group 1 showed a significant increase in pain threshold than Group 2. These results suggest that high intensity, low frequency TENS applied to appropriate auricular acupuncture points can increase pain threshold.

Forteen cases were observed among the Temporomandibular Disorder who were outpatients at the Department of Acupuncture & Moxibustion Oriental Medical Hospital, Dae Jeon University. Objective : To improve the therapeutic rate of Temporomandibular Disorder by treatment of acupuncture, chiropractic therapy and exercise therapy. Methods : Forteen outpatients suffering from Temporomandibular Disorder were treated by acupuncture, chiropractic therapy and exercise therapy. Acupuncture therapy was taken on Waiguan(TE5,外關), Zulingi(G41, 足臨泣), Sidu(TE9, 四瀆), Yanglingquan(G34, 陽陸泉), Qiuxu(G40, 丘墟), Xiaguan(S7, 下關). Chiropractic therapy was taken, when the subluxation of outpatients's Cervical spine was observed. Results : The syndrome of TMD, that is the Pain, the movement disorder and the click sound, was disappeared by 2~3 times acupuncture, chiropractic and exercise therapy.
Objective : This study shows the clinical effects on Bee-venom acupuncture and Burning acupuncture therapy for a patient who was diagnosed as Rheumatoid arthritis with ankle pain. Methods : This study was carried out a patient who was diagnosed as Rheumatoid arthritis with ankle pain. The patient was treated by Bee-venom acupuncture, Burning acupuncture therapy. To evaluate the effect of treatment, measured grade of pain by Visual Analogue Scale(VAS). Result and Conclusion: The ankle pain was reduced from VAS 10 to VAS 2 and the patient could walk on foot. In this study, Bee-venom acupuncture, Burning acupuncture therapy was effective in arthralgia of Rheumatoid arthritis.
