녹차섭취와 운동에 의한 비만 청소년의 혈중 biochemical marker 함량 감소 (Green Tea Intake and Exercise Reduce Some Biochemical Markers of Obese Adolescents)

  • 녹차섭취와 유산소운동이 비만 남자중학생의 체중과 혈중 biochemical marker에 미치는 영향에 관하여 연구하였다. 체지방율이 25% 이상인 비만 남자중학생 36명을 대상으로 대조군(n=9), 녹차처리군(n=9), 운동군(n=9)과 녹차와 운동 병행군(n=9)으로 분류하여, 운동은 주 5회 60분씩 12주간 실시하였고, 녹차는 녹차열수출물을 음용(녹차건중 1.5-1.7 g/일) 하였다. 대조군은 녹차처리군이 섭취한 양의 물을 섭취하였다. 실험 시작전과 12주 후의 혈중 TNF-$\alpha$, IL-6, leptin, TG 및 HDL-C 함량을 측정하였다. 대조군을 포함한 모든 처리군에서 체중 변화는 없었으나 병행군은 복부지방률이 감소하였다. 대조처리는 TNF-$\alpha$, IL-6 및 leptin 함량에 아무런 영향을 미치지 않았다. 녹차처리는 leptin (p<0.05)을 감소시켰고, TG (p<0.01)는 증가시켰다. 운동은 TNF-$\alpha$ (p<0.05), IL-6 (p<0.01) 및 leptin (p<0.05) 함량을 감소시켰다. 또한 녹차처리와 운동 병행은 IL-6 (p<0.05), leptin (p<0.05) 및 HDL-C (p<0.01) 함량을 감소시켰다. 이 결과는 녹차섭취나 운동은 젊은 비만 중학생의 혈중 염증 cytokine인 TNF-$\alpha$α, IL-6 및 leptin 함량 감소에 효과가 있었으나, 녹차섭취와 운동의 병행에 의한 synergy 효과는 없었다.

돼지 말초혈액 다형핵 백혈구의 유주성에 있어서 conjugated linoleic acid의 면역증강효과 (Immunoenhancing Effects of Conjugated Linoleic Acid on Chemotactic Activity of Porcine Peripheral Blood Polymorphonuclear Cells)

  • 돼지 말초혈액 다형핵 백혈구(polymorphonuclear cell; PMN)의 유주성에 있어서 conjugated linoleic acid(CLA) 이성체(CLA mixture, 10t-12c CLA, 9c-11t CLA, 9c-11c CLA 및 9t-11t CLA)의 면역증강 효과를 검토하였다. PMN에 대한 유주성은 Boyden chamber 변법으로 측정하였다. CLA 이성체들을 고농도(50∼200μM)로 사용하였을 경우 말초혈액 단핵구 세포(mononuclear cell; MNC) 및 PMN의 cell viability가 감소되거나 세포가 사멸하였다. 따라서 cell viability가 높고 세포독성을 나타내지 않는 20μM 농도로 유주활성 실험을 하였다. CLA 이성체들은 돼지 말초혈액 PMN의 유주활성에 직접적인 효과는 없었다. CLA 이성체로 배양한 MNC의 배양상층액 중 CLA mixture, 10t-12c CLA 및 9c-11t CLA 처리군에서는 PMN의 유주활성이 현저하게 증강되었으나 9c-11c CLA 및 9t-11t CLA로 배양한 MNC 배양상층액에서는 PMN의 유주활성이 나타나지 않았다. 이러한 유주성 증강효과는 checkerboard assay를 실시한 결과 진성의 유주활성이었다. 유주성 인자인 porcine recombinant (pr) interleukin(IL)-8을 이용하여 돼지 PMN에 대한 유주성을 검토한 결과, pr IL-8에 의한 PMN의 유주활성은 CLA로 배양한 MNC 배양상층액에 의한 것과 동등한 활성을 보였다. 또한 CLA로 배양한 MNC 배양상층액의 PMN에 대한 유주성을 anti-pr IL-8pAb를 사용하여 중화반응을 실시한 결과, CLA mixture로 배양한 MNC 배양상층액에 의해 증가된 PMN의 유주활성은 anti-pr IL-8 pAb 첨가에 의해 억제되어, 본 유주활성은 MNC에서 분비되는 IL-8으로 인한 것임을 강하게 시사하였다. 이상의 결과로부터 CLA 중 CLA mixture, 10t-12c CLA 및 9c-11t CLA 이성체가 돼지 말초혈액 다형핵 백혈구의 유주활성에 증강효과를 가지고 있으며, 이러한 증강효과는 CLA로 자극된 MNC에 의해 생성되는 IL-8 인자에 의한 것임을 알 수 있었다.

Prolonged Exposure to Lipopolysaccharide Induces NLRP3-Independent Maturation and Secretion of Interleukin (IL)-1β in Macrophages

  • Upon sensing of microbial infections or endogenous danger signals in macrophages, inflammasome signaling plays a significant role in triggering inflammatory responses via producing interleukin (IL)-$1{\beta}$. Recent studies revealed that active caspase-1, a product of the inflammasome complex, causes maturation of inactive pro-IL-$1{\beta}$ into the active form. However, the underlying mechanism by which this leaderless cytokine is secreted into the extracellular space remains to be elucidated. In this study, we demonstrated that prolonged lipopolysaccharide (LPS) treatment to macrophages could trigger the unexpected maturation and extracellular release of IL-$1{\beta}$ through a nucleotide-binding oligomerization domain-like receptor family, pyrin domain-containing 3 (NLRP3)-independent manner. Short-term treatment (less than 6 h) of LPS induced robust production of the IL-$1{\beta}$ precursor form inside cells but did not promote the maturation and secretion of IL-$1{\beta}$ in bone marrow-derived macrophages or peritoneal macrophages. Instead, prolonged LPS treatment (more than 12 h) led to a significant release of matured IL-$1{\beta}$ with no robust indication of caspase-1 activation. Intriguingly, this LPS-triggered secretion of IL-$1{\beta}$ was also observed in NLRP3-deficient macrophages. In addition, this unexpected IL-$1{\beta}$ release was only partially impaired by a caspase-1 and NLRP3 inflammasome inhibitor. Collectively, our results propose that prolonged exposure to LPS is able to drive the maturation and secretion of IL-$1{\beta}$ in an NLRP3 inflammasome-independent manner.

Hydroquinone, a Reactive Metabolite of Benzene, Reduces Macrophage-mediated Immune Responses

  • Hydroquinone is a toxic compound and a major benzene metabolite. We report that it strongly inhibits the activation of macrophages and associated cells. Thus, it suppressed the production of proinflammatory cytokines [tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-$1{\beta}$, IL-3, IL-6, IL-10, IL-12p40, IL-23], secretion of toxic molecules [nitric oxide (NO) and reactive oxygen species (ROS)] and the activation and expression of CD29 as judged by cell-cell adhesion and surface staining experiments. The inhibition was due to the induction of heme oxygenase (HO)-1 in LPS-activated macrophages, since blocking HO-1 activity with ZnPP, an HO-1 specific inhibitor, abolished hydroquinone's NO inhibitory activity. In addition, hydroquinone and inhibitors (wortmannin and LY294002) of the phosphatidylinositol-3 kinase (PI3K)/Akt pathway had very similar inhibitory effects on LPS-induced and CD29-mediated macrophage responses, including the phoshorylation of Akt. Therefore, our data suggest that hydroquinone inhibits macrophage-mediated immune responses by modulating intracellular signaling and protective mechanisms.

Adenophorae Radix Attenuates Mast Cell-mediated Allergic Inflammation through Down-regulation of NF-κB/ Caspase-1 Activation

  • Adenophorae Radix (AR) has been used as a traditional medicine for various diseases. However, the regulatory mechanisms of AR in allergic inflammation are not yet understood. The present study was conducted to investigate the effect and mechanisms of AR on the mast cell-mediated allergic response. To determine the pharmacological mechanisms of AR in allergic inflammation, we evaluated the effects of AR on the production of tumor necrosis factor (TNF)-α, interleukin (IL)-6, IL-1β and IL-8 as well as the activation of nuclear factor-κB (NF-κB) and caspase-1 in phorbol 12-myristate 13-acetate plus calcium ionophore A23187 (PMACI)-stimulated human mast cells (HMC-1). Our results demonstrated that AR effectively attenuated the PMACI-induced production of TNF-α, IL-6, IL-1β and IL-8 in stimulated HMC-1. Additionally, we showed that the inhibitory effect of AR on inflammatory cytokines in PMACI-stimulated HMC-1 cells involved the suppression of the activation NF-kB/caspase-1 in PMACI-stimulated HMC-1. Collectively, these findings provide experimental evidence that AR may be a useful candidate for the treatment of allergic inflammation.

마황행인감초석고탕 추출물의 항염증 효과에 대한 연구 (Studies on Anti-inflammatory Effects of Mahwanghangingamchosukgo-tang Extracts)

  • Mahwanghangingamchosukgo-tang (MH) is recorded as a treatment to treat exterior-related respiratory diseases in the Korean medicine. In this study, we examined the anti-inflammatory effects of MH, using MH water extract and lipopolysaccharide (LPS)-induced RAW 264.7 cells. First of all, we measured the amount of nitric oxide (NO) and prostaglandin E2 (PGE2), the products of inflammatory metabolism. Also, we measured enzymes such as inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), as well as cytokines such as interleukin 6 (IL-6), interleukin 1 alpha (IL-1α), and interleukin 1 beta (IL-1β). MH suppressed the production of NO and PGE2 in a dose dependent manner and reduced the amount of protein and the mRNA expression of iNOS and COX-2. Also, MH reduced the mRNA expression of IL-6, IL-1α and IL-1β. In conclusion, MH decreased production of LPS-induced inflammatory factor, which could be a clinical basic subject for inflammatory diseases.

BLT 타겟제조 및 강유전 박막 특성에 관한 연구 (A Study on BLT Target Preparation and Ferroelectric Property)

  • In recent year, BLT($Bi_{3.25}La_{0.75}Ti_{3}O_{12}$) has been one of promising substitute materials at the ferroelectric random access memory applications. We manufactured $Bi_{3.25}La_{0.75}Ti_3O_{12}$ Target with a ceramic process. The BLT target was sintered at ${1100^{\circ}C}$ for 4 hours. Using RF magnetron sputtering, a deposited BLT thin films were estimated about ferroelectric properly as a functions of post annealing temperatures. The BLT thin films showed a promoted ferroelectric characteristics at the post annealied sample of ${700^{\circ}C}$. This sample exhibited the (117) preferred crystal orientation, current density of $3{\times}10^{-8}A/cm^2$, a remanent polarization of $8{\mu}C/cm^2$ and a coercive field of 42.1 KV/cm respectively.

A Study of the Yang-gyeong-gyu-il-ui (兩景揆日儀) in the Joseon Dynasty

  • The Yang-gyeong-gyu-il-ui (兩景揆日儀) is a kind of elevation sundial using three wooden plates. Sang-hyeok Lee (李尙爀, 1810~?) and Byeong-cheol Nam (南秉哲, 1817~1863) gave descriptions of this sundial and explained how to use it in their Gyu-il-go (揆日考) and Ui-gi-jip-seol (儀器輯說), respectively. According to Gyu-il-go (揆日考) there are two horizontal plates and two vertical plates that have lines of season and time. Subseasonal (節候) lines are engraved between seasonal (節氣) lines, subdividing the interval into three equal lines of Cho-hu (初候, early subseason), Jung-hu (中候, mid subseason) and Mal-hu (末候, late subseason); there are 13 seasonal lines for a year, thus resulting in 37 subseasonal lines; also, there are 12 double-hour (時辰) lines for a day engraved on these plates. The only remaining artifact of Yang-gyeong-gyu-il-ui was made in 1849 (the $15^{th}$ year of Heon-jong) and is kept at the Korea University Museum. We have compared and analyzed Yang-gyeong-gyu-il-ui and similar western sundials. Also, we have reviewed the scientific aspect of this artifact and built a replica. Yang-gyeong-gyu-il-ui is a new model enhanced from the miniaturization development in the early Joseon Dynasty and can be applied to the southern part of the tropic line through a structure change.

Interferon-γ and Interleukin-10 Gene Polymorphisms are not Predictors of Chronic Hepatitis C (Genotype-4) Disease Progression

  • Immunoregulatory cytokines have an influence on hepatitis C virus (HCV) infection outcome. This study aimed to determine whether single nucleotide polymorphisms (SNP) in IFN- ${\gamma}$ and IL-10 genes are associated with susceptibility and/or are markers of prognosis regarding chronic hepatitis C outcomes. IFN ${\gamma}$ (+874T/A) and IL-10 (-1082G/A) genotypes were determined in 75 HCV genotype 4 patients with different disease severities (chronic hepatitis, n=25, liver cirrhosis and hepatocellular carcinoma (HCC) on top of liver cirrhosis, n=50) and 25 healthy participants using allele-specific polymerase chain reaction. No statistical differences in allele or genotype distributions of IFN ${\gamma}$ and IL-10 genes were detected between patients and controls or between patientgroups. No significant difference in the frequency of IL-10 SNP at position -1082 or IFN-${\gamma}$ at position +874T/A was found between chronic HCV genotype 4 and with progression of disease severity in liver cirrhosis or HCC. In conclusion; interferon-${\gamma}$ and interleukin-10 gene polymorphisms are not predictors of disease progression in patients with chronic hepatitis C (Genotype-4).

Synergistic Effect of Interleukin-18 on the Expression of Lipopolysaccharide-Induced IP-10 (CXCL-10) mRNA in Mouse Peritoneal Macrophages

  • Interleukin (IL)-18, a member of the family of IL-l cytokine, is one of the principal inducers of $interferon-{\gamma}(IFN-{\gamma})$ in T lymphocytes and natural killer cells. The objective of the present study was to evaluate the effect of IL-18 on the expression of chemokine IP-10 (CXCL-10) mRNA in mouse peritoneal macrophages. IL-18 had very weak direct effect or synergistic effect with IL-12 on the expression of IP-10 mRNA in C57BL/6 mouse peritoneal macrophages. However, IL-18 pretreatment was found to playa cooperative role in the expression of lipopolysaccharide (LPS)-induced IP-10 mRNA. For the expression of LPS-induced IP-10 mRNA, the synergistic effect was detected after 16 h of IL-18 pretreatment prior to LPS stimulation. The expression level of CD14 in cells stimulated with LPS was not changed by IL-18 pretreatment, and the level of $IFN-{\gamma}$ production during IL-18 pretreatment plus LPS stimulation was barely discernible ($0.36{\pm}0.31pg/ml$). Namely, the synergistic effect of IL-18 pretreatment was not related to a change of LPS receptor, CD14 expression, and the production of $IFN-{\gamma}$ by the interaction between IL-18 and LPS. The synergistic effect of IL-18 pretreatment on the expression of LPS-induced IP-10 was related to not NF-kB but AP-1 activation, and associated with the extracellular signal-regulated kinase (ERK) pathway, one of the mitogen-activated protein kinase signaling pathways. These results provide useful information that may elucidate the mechanisms underlying the effect of IL-18 on the expression of IP-10 mRNA.
