Influence of Respiration on Photosynthetic Electron Transport in psaB Mutants from Cyanobacterium Synechocystis sp. PCC6803 (Cyanobacterium Synechocystis sp.PCC6803 psaB 돌연변이주의 광합성 전자전달에 미치는 호흡의 영향)

  • The influence of respiration on photosythetic electron transport were investigated in the Wid type and psaB mutants from Syneehocystis sp. PCC6803. The amount of glucose uptake in the wild type was proportional to the glucose concentration added in wild type and less than that of psaB mutants in the dark. It was suggested that psaB mutants more depend on the glucose than the wild type. It was investigated how the activities of isocitrate dehydrogenase(IDH) and glucose-6-phos-phate dehydrogenase(G6PDH) were changed. The activities of IDH were very low. While, the ac-tivities of G6PDH were much higher than that of IDH. These results agree to the reports that ex-ogenous glucose was dismilated aerobically via Oxidative Pentose Phosphate Pathway in heterotrophic cyanobacteria. PsaB mutants showed high G6PDH activity in the presence of glucose as well as in the dark and high respiratory activities especially in the dark. It was also investigated how photosynthetic electron transport activities were changed. PsaB mutants showed higher photosynthetic electron tranasport activities than wild type in the presence of glucose as well as in the dark. In the results, it was proposed that photosynthetic electron transport between PS I and PS U was complemented by respiratory electron transport through the NADPH generated by Dxidative Pentose Phophate Pathway in psaB mutant from Synechocystis sp. Cloning and Characterization of the $_L$-Lactate Dehydrogenase Gene (IdhL) from Lactobacillus reuteri ATCC 55739

  • The ldhL gene encoding the $_L$-(+) lactate dehydrogenase was cloned from Lactobacillus reuteri ATCC 55739 chromosomal DNA and characterized. An internal 750-bp fiagment of ldhL gene was amplified by PCR using primers based on the conserved region of lactobacilli ldhL genes. A genomic library off. reuteri ATCC 55739 was constructed using pBR322, and colony hybridization experiments were performed using the 750-bp fragment as aprobe. One clone harboring a 4.0-kb PstI fragment was identified, and nucleotide sequencing confirmed it as an open reading frame of 972 bp in size in the middle. In addition to IdhL gene, an ORF homologous to Streptococcus pneumoniae TIGR4 hydrolase gene and 3' part of phosphomevalonate kinase gene (mvaK2) were also found on the 4 kb fragment. $_L$-LDH of L. reuteri ATCC 55739 showed the highest degree of homology with the $_L$-LDH of Pediococcus acidilactici (62.4%), fullowed by the $_L$-LDH of Lactobacillus pentosus (58.7%). The size of IdhL transcript determined by Northern blot was 1 kb, indicating the monocistronic nature of IdhL.

Isozyme Patterns at Five Loci Salmonids and Their Hybrid(I) (연어류에서의 종간교배 및 Isozyme Pattern에 관한 연구)

  • 연어류의 종간교배 실험과 LDH, MDH, IDH, $\alpha$GODH, ME 등 다섯가지 isozyme에 대한 genetic marker로서의 활용 가능성을 알아보고자 1차적으로 연어, 산천어 및 무지개송어를 이용하여 종간교배를 실시하였고 이들 세종의 isozyme pattern을 비교하였다. 교배실험은 종간교배, 및 allotriploid 구간 등 12구간으로 나누어 실시하였으며 연어 암컷과 산천어 수컷의 교배결과가 초기성장 단계적에서 가장 우수하게 나타났고이들의 allotriploid도 부활율이 28.1%로 가장 우수하였다. Genetic identification을 위한 isozyme loci 분석결과 연어와 산천어는 대부분의 loci에서 거의 찾아볼 수 없었고 무지개송어는 MDH-B와 IDH에서 다형현상을 확인할 수 있었다. 특히 MDH-B loci는 b 유전자의 출현 빈도에서 세 종간의 식별이 가능하였으며, IDH pattern을 산천어와 무지개송어의 비교에 유효한 것으로 나타났다. 이들 두 loci는 hybrid의 genotype 분석시 유용한 marker로 활용할 수 있는 가능성을 보였고, 앞으로의 어류 육종에 좋은 기초 자료로 이용될 것으로 사료된다.

  • Changes in the activities of isocitrate dehydrogenase (IDH) and glutamate dehydrogenase (GDH) and changes in free amino acids in the cytoplasm of Saccharomyces cerevisiae have been studied under heat shock condition. Heat shock conditions led to a significant decrease of NAD-IDH and NAD-GDH, It was shown appeared that the meaningful patterns of increase of NADP-IDH and NADP-GDH. It suggested that heat shock in yeast leads to a splitting of the TCA cycle and that glutamate synthesis takes place through the coupling of the NADP-linked isocirate and glutamate dehydrogenase. It was shown that about 14% of total free amino acids of yeast cells was decreased by heat shock. Especially heat shock condition resulted in the marked decreases of serine family amino acids such as serine, glycine and cysteine, and in the considerable increases of the rates of methionine, alanine, glutamin.

Comparisons of Isozyme Patterns in Pythium Species and Application to Pythium Systematics (Isozyme을 이용한 Pythium species의 비교 및 Pythium systematics에의 이용)

  • For the enzymes AAT, GmDH, ME, GPI, LDH and IDH, nine, seven, four, nine, seven, and four different phenotypes, respectively, were observed. All six isolates of an unidentified sterile Pythium sp. isolated from field soil showed the same band positions for all six enzymes compared. These phenotypes were not similar to any of the known Pythium species. Two isolates of unknown Pythium species (145 and 299) showed the same band positions for all six enzymes. The phenotypes for all three unknown Pythium spp. were different from the other species in the experiment. Five isolates of P. heterothallicum showed the same band positions for all enzymes compared except one enzyme, lDH. Two isolates of P. torulosum showed the same band petitions for enzymes AAT, GmDH and ME, and three isolates of P. totulosum showed the same positions for enzymes GPI, LDH, and IDH. Single isolates of P. spinosum and P. irregulare showed the same band positions for enzymes AAT, GmDH and GPI. In conclusion, sterile types of Pythium species showed 100% similarities among themselves but did not show any similarity with all isolates of P. heterothallicum and P. spinosum isolate, and showed very low similarities with other isolates in general except with unknown Pythium isolate 306. Similarity levels between different species were low in general with few exceptions.

Comparison of Genetic Profiles and Prognosis of High-Grade Gliomas Using Quantitative and Qualitative MRI Features: A Focus on G3 Gliomas

  • Objective: To evaluate the association of MRI features with the major genomic profiles and prognosis of World Health Organization grade III (G3) gliomas compared with those of glioblastomas (GBMs). Materials and Methods: We enrolled 76 G3 glioma and 155 GBM patients with pathologically confirmed disease who had pretreatment brain MRI and major genetic information of tumors. Qualitative and quantitative imaging features, including volumetrics and histogram parameters, such as normalized cerebral blood volume (nCBV), cerebral blood flow (nCBF), and apparent diffusion coefficient (nADC) were evaluated. The G3 gliomas were divided into three groups for the analysis: with this isocitrate dehydrogenase (IDH)-mutation, IDH mutation and a chromosome arm 1p/19q-codeleted (IDHmut1p/19qdel), IDH mutation, 1p/19q-nondeleted (IDHmut1p/19qnondel), and IDH wildtype (IDHwt). A prediction model for the genetic profiles of G3 gliomas was developed and validated on a separate cohort. Both the quantitative and qualitative imaging parameters and progression-free survival (PFS) of G3 gliomas were compared and survival analysis was performed. Moreover, the imaging parameters and PFS between IDHwt G3 gliomas and GBMs were compared. Results: IDHmut G3 gliomas showed a larger volume (p = 0.017), lower nCBF (p = 0.048), and higher nADC (p = 0.007) than IDHwt. Between the IDHmut tumors, IDHmut1p/19qdel G3 gliomas had higher nCBV (p = 0.024) and lower nADC (p = 0.002) than IDHmut1p/19qnondel G3 gliomas. Moreover, IDHmut1p/19qdel tumors had the best prognosis and IDHwt tumors had the worst prognosis among G3 gliomas (p < 0.001). PFS was significantly associated with the 95th percentile values of nCBV and nCBF in G3 gliomas. There was no significant difference in neither PFS nor imaging features between IDHwt G3 gliomas and IDHwt GBMs. Conclusion: We found significant differences in MRI features, including volumetrics, CBV, and ADC, in G3 gliomas, according to IDH mutation and 1p/19q codeletion status, which can be utilized for the prediction of genomic profiles and the prognosis of G3 glioma patients. The MRI signatures and prognosis of IDHwt G3 gliomas tend to follow those of IDHwt GBMs.

Genetic Variation in Natural Populations of the Cat-Snake (Elaphe dome) (누룩뱀(Elaphe diane) 자연집단의 체전적 변리에 관하여)

  • Starch gel electrophoresis was used to estimate the degree of genic variation and genetic relatedness among twelve populations of the Cat-Sanke(Elaphe diene) in Korea. Out of the 17 loci examined 9 loci (Mdh-2, Ldh-1, Ldh-2, Idh-1, Idh-2, Adh, Aco-3, ep, Got-1) were monomorphic with identical mobility in all populations, 4 loci (Pgm, Mpi, Pept-1, Got-2) were nearly identical with minor allele frequency differences, and 4 loci (Mdh-1, Fgi, Est-2, 6psd) showed moderate variation. Levels of genic variation were similar to those found for other reptilian species. The mean genetic similarity value(S) among twelve populations is 0.966. Chechu population was slightly remote genetically from the mainland populations.

  • For the purpose of identification of inheritance in allotriploid between rainbow trout (Oncorhynchus mykiss) and coho salmon (O. kisutch), five isozymes, lactate dehydrogenase (LDH), malate dehydrogenase (MDH), isocitrate dehydrogenase (IDH), phosphoglucose isomerase (PGI) and phosphoglucomutase (PGM) from skeletal muscle in two species and their allotriploid were analyzed. All of these loci showed differences between two species and their allotriploid except PGI. Generally, coho salmon was more monomorphic in these isozyme loci than rainbow trout. Their allotriploids showed intermediate patterns between the parental species in those isozyme loci except PGI. As a result of this study, LDH, MDH, IDH and PSM may be used as useful genetic markers in these two species, and they also be of use in studying hybrid and allotriploid in salmonids.

The oncometabolite d-2-hydroxyglutarate induces angiogenic activity through the vascular endothelial growth factor receptor 2 signaling pathway

  • The mutation of isocitrate dehydrogenase (IDH)1 (R132H) and IDH2 (R172K) and the induction of hypoxia in various solid tumors results in alterations in metabolic profiles, including the production of the d- or l-forms of 2-hydroxyglutarate (2HG) from α-ketoglutarate in aerobic metabolism in the tricarboxylic acid (TCA) cycle. However, it is unclear whether the oncometabolite d-2HG increases angiogenesis in endothelial cells. Therefore, in this study, we analyzed the levels of various metabolites, including d-2HG, under hypoxic conditions and in IDH2R172K mutant breast cancer cells by mass spectrometry. We then further evaluated the effects of this metabolite on angiogenesis in breast cancer cells. The results revealed that treatment with d-2HG increased the levels of secreted vascular endothelial growth factor (VEGF) in cancer cells and enhanced endothelial cell proliferation in a concentration-dependent manner. Wound healing and cell migration (examined by Transwell assay) were significantly increased by d-2HG to a level similar to that induced by VEGF. Tube formation was significantly stimulated by d-2HG, and chick chorioallantoic membrane angiogenesis was also enhanced by d-2HG. d-2HG activated VEGF receptor (VEGFR)2 and VEGFR2 downstream signaling, extracellular signal-regulated kinase 1/2, focal adhesion kinase, AKT and matrix metalloproteinase (MMP)2. Taken together, the findings of this study suggested that d-2HG induced angiogenic activity via VEGFR2 signaling and increased MMP2 activity.

Qualitative and Quantitative Magnetic Resonance Imaging Phenotypes May Predict CDKN2A/B Homozygous Deletion Status in Isocitrate Dehydrogenase-Mutant Astrocytomas: A Multicenter Study

  • Objective: Cyclin-dependent kinase inhibitor (CDKN)2A/B homozygous deletion is a key molecular marker of isocitrate dehydrogenase (IDH)-mutant astrocytomas in the 2021 World Health Organization. We aimed to investigate whether qualitative and quantitative MRI parameters can predict CDKN2A/B homozygous deletion status in IDH-mutant astrocytomas. Materials and Methods: Preoperative MRI data of 88 patients (mean age ± standard deviation, 42.0 ± 11.9 years; 40 females and 48 males) with IDH-mutant astrocytomas (76 without and 12 with CDKN2A/B homozygous deletion) from two institutions were included. A qualitative imaging assessment was performed. Mean apparent diffusion coefficient (ADC), 5th percentile of ADC, mean normalized cerebral blood volume (nCBV), and 95th percentile of nCBV were assessed via automatic tumor segmentation. Logistic regression was performed to determine the factors associated with CDKN2A/B homozygous deletion in all 88 patients and a subgroup of 47 patients with histological grades 3 and 4. The discrimination performance of the logistic regression models was evaluated using the area under the receiver operating characteristic curve (AUC). Results: In multivariable analysis of all patients, infiltrative pattern (odds ratio [OR] = 4.25, p = 0.034), maximal diameter (OR = 1.07, p = 0.013), and 95th percentile of nCBV (OR = 1.34, p = 0.049) were independent predictors of CDKN2A/B homozygous deletion. The AUC, accuracy, sensitivity, and specificity of the corresponding model were 0.83 (95% confidence interval [CI], 0.72-0.91), 90.4%, 83.3%, and 75.0%, respectively. On multivariable analysis of the subgroup with histological grades 3 and 4, infiltrative pattern (OR = 10.39, p = 0.012) and 95th percentile of nCBV (OR = 1.24, p = 0.047) were independent predictors of CDKN2A/B homozygous deletion, with an AUC accuracy, sensitivity, and specificity of the corresponding model of 0.76 (95% CI, 0.60-0.88), 87.8%, 80.0%, and 58.1%, respectively. Conclusion: The presence of an infiltrative pattern, larger maximal diameter, and higher 95th percentile of the nCBV may be useful MRI biomarkers for CDKN2A/B homozygous deletion in IDH-mutant astrocytomas.
