Melanin Synthesis Inhibition and Radical Scavenging Activities of Compounds Isolated from the Aerial Part of Lespedeza cyrtobotrya

  • The EtOAc fraction of Lespedeza cyrtobotrya showed mushroom tyrosinase inhibitory and radical scavenging activities. Four active compounds were isolated based on Sephadex LH-20 chromatography and HPLC, and the structures were elucidated, on the basis of their LC-MS and NMR spectral data, as 2-(2,4-dihydroxyphenyl)-6-hydroxybenzofuran (1), eriodictyol-7-O-glucopyranoside (2), haginin A (3), and dalbergioidin (4), respectively. Compound (1) showed mushroom tyrosinase inhibitory activity with an $IC_{50}$ value of $5.2\;{\mu}M$ and acted as a competitive inhibitor. Furthermore, $37.3\;{\mu}M$ of compound 1 reduced 50% of the melanin content on human melanoma (MNT-1) cells. The radical scavenging activities of compounds 1, 2, 3, and 4 were shown to have $IC_{50}$ values of 11.0, 24.5, 9.0, and $36.5\;{\mu}M$, respectively, in an ABTS system and $IC_{50}$ values of 42.7, 36.0, 37.7, and $61.7\;{\mu}M$, respectively, in a DPPH system. The mushroom tyrosinase inhibitory activity of the EtOAc fraction of Lespedeza cyrtobotrya was contributed by compounds 1, 3, and 4, and its radical scavenging activity was contributed by compounds 1-4.

  • A cast stainless steel may experience an embrittlement when it is exposed to approximately 30$0^{\circ}C$ for long period. In the present investigation, The three classes of the thermally aged CF8M specimie n are prepared using an artificially accelerated aging method. Namely, after the specimen are held for 300, 1800 and 3600hrs. at 43$0^{\circ}C$ respectively, the specimens are quenched in water to room temperature. Load versus load line displacement curves and J-R curves are obtained using the unloading compliance method. $J_{IC}$ values are obtained following ASTM E 813-87 and ASTM E 813-81 methods. In addition to these methods, JIC values are obtained using SZW(stretch zone width) method described in JSME S 001-1981. The results of the unloading compliance method are $J_Q$=485.7 kJ/m$^2$ for virgin material, $J_{IC}$ of the degraded materials associated with 300, 1800 and 3600hrs are obtained 369.25 kJ/m$^2$, 311.02 kJ/m$^2$, 276.7 kJ/m$^2$, respectively. The results of SZW method are similar to those of the unloading compliance method. Through the elastic-plastic fracture toughness test, it is found that the value of $J_{IC}$ is decreased with increasing of the aging time. The results obtained through the investigation can provide reference data for a leak before break(LBB) of reactor coolant system of nuclear power plants.

Biological Activities of Sesquiterpene Lactones isolated from Several Compositae Plants Part 3 -Inhibitory Activity on Nitric Oxide Release and ACAT- (수종의 국화과 식물에서 분리한 Sesquiterpene Lactone들의 생리활성(제3보) -Nitric oxide 방출 및 ACAT 저해활성-)

  • Nine sesquiterpene lactones, which were isolated from Hemisteptia lyrata Bunge, Chrysanthemum zawadskii Herbich var. latilobum Kitamura and Chrysanthemum boreale Makino were evaluated for the inhibition upon the nitric oxide release from the Raw cell and for the ACAT (acyl Co A: cholesterol acyltransferase) inhibitory assay. In the nitric oxide release inhibitory experiment, hemistepsin B $(IC_{50}\;=\;0.05\;{\mu}g/ml,\;0.15\;{\mu}M),$ cumambrin B, tulipinolide and costunolide exhibited strong inhibition with $IC_{50}$ values $0.05\;{\mu}g/ml\;(0.15\;{\mu}M),\;0.1\;{\mu}g/ml\;(0.38\;{\mu}M),\;0.2\;{\mu}g/ml\;(0.69\;{\mu}M)$ and $0.2\;{\mu}g/ml(0.86\;{\mu}M),$ respectively. In the ACAT inhibitory assay, angeloylcumambrin B, tigloylcumambrin B, tulipinolide and costunolide exhibited strong inhibition with $IC_{50}$ values $33\;{\mu}g/ml\;(95.4\;{\mu}M),\;22\;{\mu}g/ml\;(63.6\;{\mu}M),\;38\;{\mu}g/ml\;(151\;{\mu}M)$ and $17\;{\mu}g/ml(73.3\;{\mu}M),$ respectively, whereas other compounds did not exhibit significant activity against ACAT above $100\; {\mu}g/ml.$.

  • A diverse panel of human tumor cell lines and a mouse melanoma cell line (B16-F1) were used for the cytotoxicity test of the nine sesquiterpene lactones with ${\beta}-methylene-{\gamma}-lactone$ group isolated from Hemisteptia lyrata, Chrysanthemum zawadskii and Chrysanthemum boreale. In the cell adhesion inhibitory activity test against B16-F1 mouse melanoma cell, hemistepcin B, cumambrin B, costunolide and tulipinolide were shown significant activities with $IC_{50}$ range of 2.2, 4.1, 0.9 and $0.3\;{\mu}g/ml$, respectively. In the cytotoxicity test against human tumor cells, the most active compound was costunolide having $IC_{50}$ values of below $0.3\;{\mu}g/ml$ against all the tested cell lines except for UACC62. Cumambrin A, hendelin and costunolide exhibited more strong activity against HCT15 and UO-31 cell lines than a positive control, adriamycin. All tested compounds showed an $IC_{50}$ values of below $5.0\;{\mu}g/ml$ against all the tested cell lines.

Melanin-concentrating Hormone-1 Receptor (MCH-1) Antagonism of the Leaves Extract from Morus alba

  • The present study was performed to investigate the binding affinity of the ethanol extract from the leaves of Morus alba (EMA) and some EMA related plant materials (EMA-D, EMA-DM) for melanin-concentrating hormone-1 receptor (MCH-1) and also to examine the antagonistic effect of them for the recombinant MCH-1 receptor expressed in CHO cells. EMA, dichloromethane fraction (EMA-D) and EMA-DM exhibited high affinity for mammalian MCH receptor in receptor binding assays ($IC_{50}$ value: 2.3, 1.6 and $1.0{\mu}g/ml$, respectively). Other plant materials (MMA-D, MMA-DM) obtained from methanol extracts from the leaves of Morus alba (MMA) also exhibited high affinity for mammalian MCH receptor, even though the $IC_{50}$ values of them were lower than those of EMA-D and EMA-DM. In Chinese hamster ovary (CHO) cells expressing human MCH-1, EMA-DM and EMA-D significantly inhibited MCH-induced intracellular $Ca^{2+}$ increase ($IC_{50}$ values: 16.5 and $22.7{\mu}g/ml$, respectively). These results clearly indicate that the ethanol extract from the leaves of Morus alba (EMA) and some EMA related plant materials (EMA-D, EMA-DM) are novel selective MCH-1 receptor antagonist, respectively.

Evaluation of Cytotoxicity, Carbohydrate, and Lipid Inhibitory Activity of Codonopsis lanceolata using Different Solvent Fractions

  • This study was conducted to evaluate the cytotoxicity and ${\alpha}-Amylase$, ${\alpha}-Glucosidase$, pancreatic lipase inhibition in vitro by different solvent fractions from the roots of Codonopsis lanceolata. The values of $IC_{50}$ against Calu-6 cell showed a high effect in n-hexane fraction ($10.13{\mu}g/mL$) whereas DW fraction exhibited the weakest inhibition on cell viability, having an $IC_{50}$ value of over $1,000{\mu}g/mL$. The values of $IC_{50}$ against HCT-116 cell showed the highest activity in n-BuOH fraction ($102.01{\mu}g/mL$), followed by n-hexane fraction ($145.85{\mu}g/mL$), methylene chloride fraction ($332.02{\mu}g/mL$), ethyl acetate fraction ($462.93{\mu}g/mL$) and DW fracion ($>1,000{\mu}g/mL$). ${\alpha}-Amylase$ inhibitory activity in methylene chloride fraction and ethyl acetate fraction was found to have a higher inhibitory effect with 24.5% and 25.6% than the other fractions. The highest ${\alpha}-Glucosidase$ inhibitory activity was observed from the ethyl acetate fraction extract, while the extract of DW fraction showed the lowest level of inhibitory activity at given experiment concentration. The pancreatic lipase inhibitory activity of C. lanceolata was found to have a higher the effect in ethyl acetate fraction. Inhibition of lipase activity of the ethyl acetate fraction and n-hexane fraction showed a relatively high, while the extract of DW fraction showed the lowest level at given experiment concentration. These results suggested that the roots of C. lanceolata may assist in the potential biological activity on carbohydrate, lipid Inhibitory activity and anticancer activity.

Enhancement of Interfacial Adhesion of Epoxy/Red Mud Nanocomposites Produced by Acidic Surface Treatment on Red Mud (Red Mud의 산처리에 의한 에폭시/Red Mud 나노복합재료의 계면 결합력 향상)

  • In this work, red mud (RM) was chemically modified by 0.1, 1, and 5 M H3PO4 solution to prepare epoxy/RM nanocomposites. The effect of chemical treatment on pH, acid-base values, specific surface area, and porosity of RM surface was analyzed. To estimate the mechanical interfacial properties of epoxy/RM nanocomposites, the critical stress intensity factor (K$_{IC}$) was measured. From the experimental results, it was clearly revealed that the porosity, specific surface area, and acid values of RM surface were developed as the increase of the treatment concentration due to the increase of acidic functional group, including hydroxyl group on RM surface. The mechanical interfacial properties of epoxy/treated-RM nanocomposites were higher than those of epoxy/RM as-received due to an improvement of interfacial bonding between basic matrix and RM surface.

Effects of Methyl Gallate on Arachidonic Acid Metabolizing Enzymes: Cyclooxygenase-2 and 5-Lipoxygenase in Mouse Bone Marrow-Derived Mast Cells

  • Methyl gallate (MG) is a medicinal herbal product that is isolated from Paeonia lactiflora that inhibits cyclooxygenase-2 (COX-2) dependent phases of prostaglandin $D_2\;(PGD_2)$ generation in bone marrow-derived mast cells (BMMC) in a concentration-dependent manner with an $IC_{50}$ values of $17.0\;{\mu}M$. This compound also found inhibited the COX-2-dependent conversion of the exogenous arachidonic acid to $PGD_2$ in a dose-dependent manner with an $IC_{50}$ values of $190\;{\mu}M$, using a COX enzyme assay kit. However, at concentrations up to $80\;{\mu}M$, MG did not inhibit COX-2 protein expression in BMMC, indicating that MG inhibits COX-2 activity directly. Furthermore, MG consistently inhibited the production of leukotriene $C_4\;(LTC_4)$ in a dose dependent manner, with an $IC_{50}$ value of $5.3\;{\mu}M$. These results demonstrate that MG has a dual cyclooxygenase-2/5-lipoxygenase inhibitory activity, which might provide the basis for novel anti-inflammatory drugs.

Evaluation of Abelmoschus Esculentus Seed for Antioxidant and Phytochemical Analysis Using In vitro Assays

  • The aim of this study was to assess the in vitro potential of methanolic seed extract of Abelmoschus esculentus as a natural antioxidant. The DPPH activity of the Ethyl acetate soluble fraction (10, 20, 40, 80, and $160{\mu}g/mL$) was increased in a dose dependent manner, which was found in the range of 18.97-90.47% as compared to ascorbic acid 26.44-93.71%. The $IC_{50}$ values of Ethyl acetate soluble fraction (EAES) and ascorbicacid in DPPH radical scavenging assay were obtained to be 28.12 and $18.43{\mu}g/mL$, respectively. Measurement of polyphenol content of the EAES of A. esculentus seed was achieved using Folin-Ciocalteau reagent containing 53.80 mg/g of total phenolic content, which was found signicantly higher when compared to reference standard gallic acid. Similarly total flavonoids and proabthocyanidis of EAES and chloroform soluble fraction (CAES) were found significantly 147 mg/g and 14.24 mg/g respectively when both compared to reference standard quercetin. EAES exhibited high significant lipid peroxidation inhibition effects in a dose-dependent manner, with $IC_{50}$ values of $38.08{\mu}g/mL$, whereas, standard quercetin, with $IC_{50}$ value of $36.67{\mu}g/mL$. All extract/fractions showed dose dependent reducing power ability and these differences were statistically significant (p<0.001). The results obtained in this study clearly indicate that A. esculentus seed has a signicant potential to use as a natural antioxidant agent.

Effect of specimen size on fracture toughness of reduced activation ferritic steel (JLF-l) (저방사화 철강재 (JLF-1)의 파괴인성에 미치는 시험편 크기의 영향)

  • Reduced activation ferritic (JLF-1) steel is leading candidates for blanket/first-wall structures of the D-T fusion reactor. In fusion application, structural materials will suffer effects of repeated changes of temperature. Therefore, the data base of tensile strength and fracture toughness at operated temperature $400^{\circ}C$ are very important. Fracture toughness ($J_{IC}$) and tensile tests were carried out at room temperature and elevated temperature ($400^{\circ}C$). Fracture toughness tests were performed with two type size to investigate the relationship between the constraint effect of a size and the fracture toughness resistance curve. As the results, the tensile strength and the fracture toughness values of the JLF-1 steel are slightly decreased with increasing temperature. The fracture resistance curve increased with increasing plane size and decreased with increasing thickness. The fracture toughness values of JLF-1 steel at room temperature and at $400^{\circ}C$ shows an excellent fracture toughness ($J_{IC}$) of about $530kJ/m^2\;and\;340kJ/m^2$, respectively.
