Ichthyotoxic Cochlodinium polykrikoides red tides offshore in the South Sea, Korea in 2014: I. Temporal variations in three-dimensional distributions of red-tide organisms and environmental factors

  • The ichthyotoxic Cochlodinium polykrikoides red tides have caused great economic losses in the aquaculture industry in the waters of Korea and other countries. Predicting outbreak of C. polykrikoides red tides 1-2 weeks in advance is a critical step in minimizing losses. In the South Sea of Korea, large C. polykrikoides red tide patches have often been recorded offshore and transported to nearshore waters. To explore the processes of offshore C. polykrikoides red tides, temporal variations in 3-dimensional (3-D) distributions of red tide organisms and environmental parameters were investigated by analyzing 4,432 water samples collected from 2-5 depths of 60 stations in the South Sea, Korea 16 times from May to Nov, 2014. In the study area, the vegetative cells of C. polykrikoides were found as early as May 7, but C. polykrikoides red tide patches were observed from Aug 21 until Oct 9. Cochlodinium red tides occurred in both inner and outer stations. Prior to the occurrence of large C. polykrikoides red tides, the phototrophic dinoflagellates Prorocentrum donghaiense (Jun 12 to Jul 11), Ceratium furca (Jul 11 to Aug 21), and Alexandrium fraterculus (Aug 21) formed red tides in sequence, and diatom red tides formed 2-3 times without a certain distinct pattern. The temperature for the optimal growth of these four red tide dinoflagellates is known to be similar. Thus, the sequence of the maximum growth rates of P. donghaiense > C. furca > A. fraterculus > C. polykrikoides may be partially responsible for this sequence of red tides in the inner stations following high nutrients input in the surface waters because of heavy rains. Furthermore, Cochlodinium red tides formed and persisted at the outer stations when $NO_3$ concentrations of the surface waters were < $2{\mu}M$ and thermocline depths were >20 m with the retreat of deep cold waters, and the abundance of the competing red-tide species was relatively low. The sequence of the maximum swimming speeds and thus potential reachable depths of C. polykrikoides > A. fraterculus > C. furca > P. donghaiense may be responsible for the large C. polykrikoides red tides after the small blooms of the other dinoflagellates. Thus, C. polykrikoides is likely to outgrow over the competitors at the outer stations by descending to depths >20 m and taking nutrients up from deep cold waters. Thus, to predict the process of Cochlodinium red tides in the study area, temporal variations in 3-D distributions of red tide organisms and environmental parameters showing major nutrient sources, formation and depth of thermoclines, intrusion and retreat of deep cold waters, and the abundance of competing red tide species should be well understood.

Classification of Seismic Stations Based on the Simultaneous Inversion Result of the Ground-motion Model Parameters (지진동모델 파라미터 동시역산을 이용한 지진관측소 분류)

  • The site effects of seismic stations were evaluated by conducting a simultaneous inversion of the stochastic point-source ground-motion model (STGM model; Boore, 2003) parameters based on the accumulated dataset of horizontal shear-wave Fourier spectra. A model parameter $K_0$ and frequency-dependent site amplification function A(f) were used to express the site effects. Once after a H/V ratio of the Fourier spectra was used as an initial estimate of A(f) for the inversion, the final A(f) which is considered to be the result of combined effect of the crustal amplification and loca lsite effects was calculated by averaging the log residuals at the site from the inversion and adding the mean log residual to the H/V ratio. The seismic stations were classified into five classes according to $logA_{1-10}^{max}$(f), the maximum level of the site amplification function in the range of 1 Hz < f < 10 Hz, i.e., A: $logA_{1-10}^{max}$(f) < 0.2, B: 0.2 $\leq$ $logA_{1-10}^{max}$(f) < 0.4, C: 0.4 $\leq$ $logA_{1-10}^{max}$(f) < 0.6, D: 0.6 $\leq$ $logA_{1-10}^{max}$(f) < 0.8, E: 0.8 $\leq$ $logA_{1-10}^{max}$(f). Implication of the classified result was supported by observing a shift of the dominant frequency of average A(f) for each classified stations as the class changes. Change of site classes after moving seismic stations to a better site condition was successfully described by the result of the station classification. In addition, the observed PGA (Peak Ground Acceleration)-values for two recent moderate earthquakes were well classified according to the proposed station classes.

Effects of Chronic Treatments with 5-HT Uptake Inhibitors on the [$^3H$]Imipraine and [$^3H$]Paroxetine Binding, [$^3H$]5-HT Uptake, and 5-HT Content of the Rabbit Platelet (5-HT 흡수억제성 항우울제들이 가토혈소판의 [$^3H$]Imipramine과 [$^3H$]Paroxetine Binding, [$^3H$]5-HT 흡수, 및 5-HT함량에 미치는 영향)

  • Many evidences are compatible with the correlation between the inhibition of [$^3H$] imipramine([$^3H$]IMI) and [$^3H$]paroxetine([$^3H$]PAT) binding to the 5-hydroxytryptamine(5-HT) transporter complex and the 5-HT uptake of 5-HT neurons and platelets, and most antidepressants have been shown to inhibit the [$^3H$]IMI and [$^3H$]PAT binding and the neuronal 5-HT uptake. However, several paradoxical research findings led to doubt about the pharmacological significance of the [$^3H$]IMI and [$^3H$]PAT binding sites. This study was carried to clarify the correlation between the [$^3H$]IMI and [$^3H$]PAT binding parameters and the tissue 5-HT content or/and [$^3H$]5-HT uptake in the rabbit platelet, which contains 40 times ad much 5-HT as that of human platelet and shows the 10 fold higher $B_{max}$ of the 5-HT transporter binding to a 5-HT uptake inhibitor. The rabbits were treated for 28 days with amitriptyline(4mg/kg/day : AP), fluoxetine(0.5mg/kg/day : FO), and sertraline(0.5mg/kg/day : SA) via an Alzet osmotic pump implanted for constant infusion. The [$^3H$]IMI binding $B_{max}$ and $K_d$ of the rabbit platelets were $6.4{\pm}1.2$pmol/mg protein and $10.9{\pm}2.1$nM and those in the [$^3H$]PAT binding were $8.6{\pm}1.1$pmol/mg protein and $1.6{\pm}0.3$nM, respectively. AP slightly increased $B_{max}$ of [$^3H$]IMI binding and both [$^3H$]IMI binding and [$^3H$]PAT binding $K_d$, and i contrast, it slightly decreased $B_{max}$ of [$^3H$]PAT binding. FO Slightly increased $K_d$ of both and [$^3H$]IMI and [$^3H$]PAT binding and slightly decreased $B_{max}$ of [$^3H$]IMI and [$^3H$]PAT binding. SA produced the significant increase of [$^3H$]PAT binding $B_{max}$ and the slight increase of both [$^3H$]IMI and [$^3H$]PAT binding $K_d$ and in contrast, it slightly decreased $B_{max}$ and of [$^3H$]IMI binding. And, the $V_{max}$ and $K_m$ of platelet [$^3H$]5-HT uptake were $24.2{\pm}2.4$pmol/$10^8$ platelets/min and $3.3{\pm}0.3$nM, respectively. The $V_{max}$ was little affected by AP, FO, or SA, but the [$^3H$]5-HT uptake $K_m$ value was moderately increased by FO. However, the platelet 5-HT content was moderately decreased by all of the 5-HT uptake inhibitors used in this study. These results seem to be consistent with the allosterical and competitive interaction of 5-HT uptake inhibiting antidepressants with each other as well as 5-HT in the 5-HT transporter binding, and provide no support for the view that the potencies of 5-HT uptake inhibitors to inhibit the [$^3H$]IMI or [$^3H$]PAT binding with 5-HT transporter complex correlate with their antidepressant potencies.

Springtime Distribution of Inorganic Nutrients in the Yellow Sea: Its Relation to Water Mass (수괴특성에 따른 춘계 황해의 영양염 분포 특성)

  • Inorganic nutrient concentrations in relation to springtime physical parameters of the Yellow Sea were investigated during April 1996. Three major water masses, i.e., the Yellow Sea Warm Current Water (YSWC), Coastal Current Water (CCW) and Changjiang River Diluted Water (CRDW), prevailed in the study area. Water masses were vertically wel1 mixed throughout the study area, and nutrients were supplied adequately from bottom to surface layer. As result of ample nutrients supplied by vertical mixing together with progressed daylight condition, springtime phytoplankton blooms were observed, which was responsible for the depletion of inorganic nutrients in surface water column. Low nutrients concentration in bottom water of the central Yellow Sea (Stn. D9; nitrate: <2 ${\mu}$M, phosphate: <0.3 ${\mu}$) was associated with the entrance of YSWC which is characterized by high temperature and salinity. Influenced by runoff and vertical tidal mixing, CCW with high nutrient concentrations probably associated with China and Korea coastal waters with high nutrients concentration. For the local scale of inorganic nutrient distribution, nutrient transfers from coast to central areas were limited due to restriction imposed by tidal fronts (Stn. D6) and thus affected the horizontal nutrient profiles. Relatively high phytoplankton biomass was observed in the tidal front (Chl-${\alpha}$=12.38 ${\mu}$gL$^{-1}$) during the study period. Overall, the springtime nutrient distribution patterns in the Yellow Sea appeared to be affected by: (1) Large-scale influx of YSWC with low nutrient concentrations and CCW with high nutrient concentrations influenced by Korea and China coastal waters; (2) vertical mixing of water mass and phytoplankton distribution; and (3) local-scale tidal front as well as phytoplankton blooms alongthe tidal front.

Effects of Herbal Product (Miracle®) on the Performance, Nutrient Digestibility, Fecal Microflora and Immunoresponse in Weanling Pigs (생약제제(미라클®)첨가가 이유 자돈의 성장률, 영양소 이용율, 분내 미생물균총 및 면역기능에 미치는 영향)

  • An experiment was conducted to investigate the effects of dietary herbal product(Miracle$^{\circledR}$) supplementation on the performance, nutrient digestibility, fecal microflora and blood parameters in pigs. Forty eight cross bred(Y${\times}$L${\times}$D) weanling pigs (average initial body weight 7.84kg${\pm}$0.17, 28 d old) were used in 35 d feeding trial. Pigs were allotted to 16 raised floor cages(W 35.5cm${\times}$L45cm${\times}$H 55cm). Each treatment had four replications of 3 pigs each. Treatments were: control (T1), CTC 100ppm (T2), Miracle$^{\circledR}$ 0.15% (T3) and CTC 100ppm + Miracle$^{\circledR}$ 0.15% (T4). The herbal product supplementation(T3) tended to improve the weight gain, feed intake, feed/gain but there were no significant differences among treatments. Nutrient digestibility was significantly(P〈0.05) affected by dietary treatments. The pigs fed T3 diet showed the lowest digestibility of DM, crude protein, NFE, P, and amino acids among treatments. Serum cholesterol level of pigs fed T3 diet was lowest and significantly(P〈0.05) lower than those fed antibiotic supplemented diet(T2). Triglyceride and HDL level of the pigs fed T3 diet were also lower than those of others but the differences were not significant. The level of serum IgG was significantly(P〈0.05) different among treatments. The level of serum IgG was highest in T4, followed by T1, T2 and T3. Serum AST(Aspartate aminotransferase) level of the control(T1) was lower than other treatments. Levels of serum total protein, albumin and ALT(Alanine aminotransferase) were not significantly different among treatments. The colony forming units (CFU) of Lactobacillus, Cl. perfringens and E.coli in feces were not significantly different among treatments but those of Cl. perfringens of T2 (CTC 100ppm) showed the lowest count at both 3rd and 5th wk. It is concluded that herbal product Miracle$^{\circledR}$ does not significantly affect growth performance of weanling pigs but it influenced the digestibility, serum IgG, cholesterol and AST level. The combination of the herbal product Miracle$^{\circledR}$ with antibiotic(CTC) showed no synergistic effects.

Development of a Window Program for Searching CpG Island (CpG Island 검색용 윈도우 프로그램 개발)

  • A CpG island is a short stretch of DNA in which the frequency of the CG dinucleotide is higher than other regions. CpG islands are present in the promoters and exonic regions of approximately $30{\sim}60$% of mammalian genes so they are useful markers for genes in organisms containing 5-methylcytosine in their genomes. Recent evidence supports the notion that the hypermethylation of CpG island, by silencing tumor suppressor genes, plays a major causal role in cancer, which has been described in almost every tumor types. In this respect, CpG island search by computational methods is very helpful for cancer research and computational promoter and gene predictions. I therefore developed a window program (called CpGi) on the basis of CpG island criteria defined by D. Takai and P. A. Jones. The program 'CpGi' was implemented in Visual C++ 6.0 and can determine the locations of CpG islands using diverse parameters (%GC, Obs (CpG)/Exp (CpG), window size, step size, gap value, # of CpG, length) specified by user. The analysis result of CpGi provides a graphical map of CpG islands and G+C% plot, where more detailed information on CpG island can be obtained through pop-up window. Two human contigs, i.e. AP00524 (from chromosome 22) and NT_029490.3 (from chromosome 21), were used to compare the performance of CpGi and two other public programs for the accuracy of search results. The two other programs used in the performance comparison are Emboss-CpGPlot and CpG Island Searcher that are web-based public CpG island search programs. The comparison result showed that CpGi is on a level with or outperforms Emboss-CpGPlot and CpG Island Searcher. Having a simple and easy-to-use user interface, CpGi would be a very useful tool for genome analysis and CpG island research. Distribution of Nutrients and Chlorophyll α in the Surface Water of the East Sea (동해 표층수 중 영양염과 Chlorophyll α의 분포 특성)

  • During the period between July 3 and 27 of 2009, water samples were collected from the Russian coast at a depth of 30m from 26 stations (including Ulleung and Japan basins) onboard the Russian survey vessel R/V Lavrentyev following 4 lines (D, R, E, and A). The samples were analyzed for nutrients and chlorophyll a contents. All parameters exhibited higher values in warm waters than in cold waters ($NH_4:1.8-fold$, $PO_4:1.8-fold$, $SiO_2:1.2-fold$, and chlorophyll-${\alpha}$:1.9-fold), except nitrates, which was 1.4-fold higher in cold waters than in warm waters. The horizontal distribution of ammonia, phosphate, and chlorophyll-${\alpha}$ was very similar to each other and showed the highest values in the waters near Russia, where a upwelling influence of cold current and bottom water prevails, while relatively low distribution was observed at the Ulleung Basin. On the other hand, nitrates showed the highest concentration at the Ulleung Basin, which is under the direct influence of the Tsushima warm water, and showed a gradual decrease northward. The N/P ratio showed the highest value in the Tsushima middle water, rather than in the North Korean Cold Water, the Tsushima Warm Water was the primary source of nitrate flow into the East Sea. However, the average concentration of phosphate in the warm waters was < $0.2{\mu}M$, thereby limiting phytoplankton growth, while a high concentration of phosphate in cold waters showed a direct correlation with chlorophyll-${\alpha}$. The results of principal component analysis for the identification of primary factors that influence the marine environment showed that principal component I was water temperature and principal component II was influenced chlorophyll-${\alpha}$ and nutrients. Therefore, Study area has greatest influenced by water temperature, and clearly distinct cold and warm water regions were observed in the East Sea.

Substituting oat hay or maize silage for portion of alfalfa hay affects growth performance, ruminal fermentation, and nutrient digestibility of weaned calves

  • Objective: The impact of forage feeding strategy on growth performance, ruminal fermentation and nutrient digestibility in post-weaning calves was investigated. Methods: Forty-five female Holstein calves (body weight [BW] = $79.79{\pm}0.38kg$) were enrolled in the 35-d study at one week after weaning and randomly assigned to one of three dietary treatments. All diets were fed as total mixed ration containing 60% (dry matter [DM] basis) of basal starter feed and 40% (DM basis) of forage, but varied in composition of forage source including i) alfalfa (40% DM, AH); ii) alfalfa hay (26.7% DM)+oat hay (13.3% DM; OH); iii) alfalfa hay (26.7% DM)+corn silage (13.3% DM; WS). Results: Dry matter intake was not different among treatment groups (p>0.05). However, BW (p<0.05) and average daily gain (p<0.05) of calves fed AH and OH were greater than WS-fed calves, whereas heart girth was greater in OH-fed calves than those fed AH and WS (p<0.05). Ruminal fermentation parameters including proportion of butyric acid, acetated-to-propionate ratio, concentration of total volatile fatty acid, protozoal protein, bacterial protein, and microbial protein in rumen were the highest in OH (p<0.05) and the lowest in WS. Compared with the AH and WS, feeding oat hay to postweaning calves increased crude protein digestibility (p<0.05), and decreased duration of diarrhea (p<0.05) and fecal index (p<0.05). Conclusion: Our results suggested that partially replacing alfalfa hay with oat hay improved ruminal fermentation, nitrogen utilization, and reduced incidence of diarrhea in post-weaning dairy calves.

Comparison of Rabbit Caecal Content and Rabbit Hard Faeces as Source of Inoculum for the In vitro Gas Production Technique

  • In order to find an alternative source of inoculum to caecal content for studying the fermentation activity of rabbit hindgut, caecal content and faeces of 25 hybrid Hyla rabbits were used as inocula for an in vitro gas production trial. About 1 g of three substrates (dehydrated alfalfa meal, dehydrated beet pulp, barley) was weighed, in quadruplicate per inoculum, in 120 ml bottles; 75 ml of anaerobic medium and 4 ml of reducing solution were added and bottles were placed at $39^{\circ}C$. Caecal content and faeces were diluted respectively 1:2 (CI) and 1:8 (FI) with anaerobic medium and were introduced in the respective bottles (10 ml). Gas production was recorded 20 times at 2-24 h intervals throughout fermentation (96 h). The fermentation characteristics (i.e. degraded organic matter, OMd; potential gas production, A; fermentation rate, Rmax; time at which it is reached, Tmax; pH, volatile fatty acid, VFA) were studied by inoculum and feedstuffs. The feedstuffs, according to their chemical composition, showed very different fermentation characteristics. In particular, OMd, A and Rmax allowed feedstuff classification as follows: barley>beet pulp>alfalfa. The inocula differ (p<0.05) in Tmax, were higher for CI (15.53 vs. 11.96 h) and in VFA production. In particular, CI produced higher levels of acetate (38.9 vs. 33.4 mM/g OM incubated, p<0.01) and isobutyrate (0.72 vs. 0.42, p<0.01) but less propionate (7.1 vs. 10.3, p<0.01) and butyrate (11.3 vs. 14.0, p<0.01). However, the trend of gas production, similar for the inocula according to the fermented substrate, and the good regression equation to estimate some caecal fermentation parameters from faeces suggest that, after standardisation, the faeces could be used as an alternative inoculum for gas tests in rabbit.

Evaluation of Impact Factor in Composite Cable-Stayed Bridges under Reliability-based Live Load Model (신뢰도 기반 활하중모델에 의한 강합성 사장교의 충격계수 평가)

  • AASHTO LRFD and Korean Bridge Design Code (Limit State Design) specify to consider Truck and Lane load simultaneously determined from reliability-based live load model, and impact shall be applied to the truck load while it shall not be applied to the lane load. In this paper, vehicle-bridge interaction analysis under moving truck and lane loads were performed to estimate impact factor of the cables and girders for the selected multi-cable-stayed composite bridges with 230m, 400m and 540m main span. A 6-d.o.f. vehicle was used for truck load and a series of single-axle vehicles was applied to simulate equivalent lane load. The effect of damping ratio on the impact factor was estimated and then the essential parameters to impact factor, i.e., road surface roughness and vehicle speed were considered. The road surface roughness was randomly generated based on ISO 8608 and it was applied to the truck load only in the vehicle-bridge interaction analysis. The impact factors evaluated from dynamic interaction analysis were also compared with those by the influence line method that is currently used in design practice to estimate impact factor in cable-stayed bridge.
