Measurement of Hydroxyl Radical Density at Bio-Solutions Generated from the Atmospheric Pressure Non-Thermal Plasma Jet

  • Atmospheric pressure non-thermal plasma of the needle-typed interaction with aqueous solutions has received increasing attention for their biomedical applications [1]. In this context, surface discharges at bio-solutions were investigated experimentally. We have generated the non-thermal plasma jet bombarding the bio-solution surface by using an Ar gas flow and investigated the emission lines by OES (optical emission spectroscopy) [2]. Moreover, The non-thermal plasma interaction with bio-solutions has received increasing attention for their biomedical applications. So we researched, the OH radical density of various biological solutions in the surface by non-thermal plasma were investigated by Ar gases. The OH radical density of DI water; deionized water, DMEM Dulbecco's modified eagle medium, and PBS; 1x phosphate buffered saline by non-thermal plasma jet. It is noted that the OH radical density of DI water and DMEM are measured to be about $4.33{\times}1016cm-3$ and $2.18{\times}1016cm-3$, respectively, under Ar gas flow 250 sccm (standard cubic centimeter per minute) in this experiment. The OH radical density of buffer solution such as PBS has also been investigated and measured to be value of about $2.18{\times}1016cm-3$ by the ultraviolet optical absorption spectroscopy.

Physiological Properties of Jeju Traditional Doenjang (제주 재래식된장의 생리적 특성)

  • The antioxidant activities of water extracts from wild vegetables such as Ligularia fischeri (GC), Capsicum annuum L. (GCY), Aster scaber (CNM), Petasites japonicus S. et Z. Max (MYD), Ipomoea batatas L. (Lam) (GGM) were evaluated and compared with water extracts from freeze dried block. The antioxidant properties of water extracts from wild vegetables and their freeze dried block were evaluated using different antioxidant tests; 2.2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging, hydroxyl radical scavenging and nitrite scavenging activities. The water extracts from wild vegetables were found to have a higher total phenolic content than water extracts from freeze dried block. Total phenolic contents of water extracts from GC, GCY, CNM, MYD, and GGM were $471.66{\pm}3.52\;{\mu}g/mg,\;141.33{\pm}2.51\;{\mu}g/mg,\;177.33{\pm}2.88\;{\mu}g/mg,\;238.66{\pm}9.50\;{\mu}g/mg\;and\;122.67{\pm}3.51\;{\mu}g/mg$, respectively. At the concentrations of 1000 ppm, water extracts from GC, GCY, CNM, and GGM showed higher activities than water extracts from their freeze dried block on DPPH radical scavenger activity. The activity of water extracts from CNM, GC, GCY, MYD, and GGM were 90.9%, 89.9%, 76.6%, 71.1%, and 57.4%, respectively. When 10000 ppm of GC, GCY, CNM, MYD, and GGM water extracts tested for hydroxyl radical scavenging activity, activities were increased by 38.8%, 33.4%, 35.9%, 34.3%, and 33.8%, respectively and a similar effect was found with water extracts from GCY, CNM, and GGM freeze dried block at 10000 ppm concentration. However, the water extracts from GC and MYD was slightly more effective than freeze dried block extracts. The water extracts from wild vegetables and their freeze dried block had effective DPPH radical scavenger activity and hydroxyl radical scavenging activity at all tested concentrations. Nitrite scavenging activity of GC water extract significantly increased in a dose-dependent manner and the extract had higher nitrite scavenging activity than extracts freeze dried block extracts. We found that freeze dried block maintained antioxidant activities of the wild vegetables.

  • Several combinations of ozone based advanced oxidation processes were tested for the treatment of red water (RW) containing recalcitrant chemical pollutants produced from 2,4,6-trinitrotoluene (TNT) manufacturing process. $O_3$, $UV/O_3$, $UV/O_3/H_2O_2$, $UV/O_3/H_2O_2/Fe^{2+}$ processes were tested for the treatment of RW. The order of organic and color removal efficiency was found to be : $O_3{\leq}UV/O_3$ < $UV/O_3/H_2O_2$ < $UV/O_3/H_2O_2/Fe^{2+}$. The optimum conditions for the removal of organic and color in the $UV/O_3/H_2O_2/Fe^{2+}$ process were 0.053 g/min of ozone flow rate, 10 mM of $H_2O_2$ concentration and 0.1 mM of $FeSO_4$ concentration. Organic and color removal efficiencies were 96 and 100 % respectively in the $UV/O_3/H_2O_2/Fe^{2+}$ process. tert-butyl alcohol (t-buOH) was used as the hydroxyl radical scavenger. Enhancement of hydroxyl radical production was achieved by the combination of ozone with several oxidants such as UV, $H_2O_2$, $Fe^{2+}$.

The antioxidant and cytotoxic activities of Sonchus oleraceus L. extracts

  • This study investigated in vitro antioxidant activity of Sonchus oleraceus L. by extraction solvent, which were examined by reducing power, hydroxyl radical-scavenging activity(HRSA) and 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assays. 70% MeOH extract had the greatest reducing power while EtOH extract had the greatest HRSA. The antioxidant activity of S. oleraceus extracts was concentration dependent and its $IC_{50}$ values ranged from 47.1 to $210.5\;{\mu}g/ml$ and $IC_{50}$ of 70% MeOH, boiling water and 70% EtOH extracts were 47.1, 52.7 and $56.5\;{\mu}g/ml$, respectively. 70% MeOH extract of S. oleraceus contained the greatest amount of both phenolic and flavonoid contents. The extracts tested had greater nitrite scavenging effects at lower pH conditions. The cytotoxic activity showed that EtOH extract had the best activity against the growth of stomach cancer cell. These results suggest that S. oleraceus extract could be used as a potential source of natural antioxidants.

Antioxidant and $\alpha$-glucosidase Inhibition Activity from Different Extracts of Zanthoxylum schnifolium Fruits (산초 (Zanthoxylum schnifolium)열매 추출물의 항산화 및 $\alpha$-Glucosidase 저해 활성)

  • This study was performed to the activity of antioxidant and anti-diabetic from Zanthoxylum schnifolium fruit. The Zanthoxylum schnifolium fruits were extracted with water, 100% methanol, 100% ethanol, 70% methanol and 70% ethanol. The activities of each extracts were measured by antioxidant tests, such as total phenolic contents, total flavonoid contents, hydroxyl radical activity, DPPH radical scavenging activity, reducing power and $\alpha$-glucosidase inhibition activity assay. The water extract represented the highest activity in the antioxidants properties in vitro, including hydroxyl radical activity, DPPH radical scavenging activity, reducing power. Furthermore, water extract also showed significantly high total phenolic contents but the 100% methanol extract showed high total flavonoid contents estimated as $196.48{\mu}g/mg$. The $IC_{50}$ values for $\alpha$-glucosidase inhibition activity of the 100% ethanol and 100% methanol extracts were $275.66{\mu}g/mg$ and $261.44{\mu}g/mg$. Our results indicated that the Zanthoxylum schnifolium fruit expected to be useful as antioxidants.

  • 선박이 배출하는 안정수(ballast water)는 외부로부터 유해 생물들이 유입되어 전파해 오는 주요경로로써, 해양환경 변화를 초래하는 매우 중요하고 위험한 일중의 하나이다. 그러나 이에 대한 효과적인 처리방법은 아직까지도 미흡한 상태이다. 선박 안정수에 포함되어 타 지역으로부터 옮겨지는 외래침입생물 처리 방안의 하나로, 강 전리방전 기술을 적용하여 고 밀집 산소와 물분자로부터 고농도 수산자유기(OH: hydroxyl radical)를 전리, 발생시켜서 활성입자를 신속히 확산시켜 비교적 낮은 수산기농도 하에서 유해성침입생물을 소멸 처리하는 환경 친화적 녹색 청정처리방법을 제안하였다. 제안된 기술은 대상물의 처리 후 부수적으로 발생할 수 있는 처리잔류물이 전혀 발생하지 않으며, 인공적 화학성분의 약제를 사용하지 않는 저렴한 처리방법이므로 대ㆍ소형 원양선박의 안정수에 들어있는 외래침입생물의 타 지역 해양확산을 안전하게 처리할 수 있는 자연치유적 신기술이다.

Scavenging Effect of Extract from Perilla frutescens and Rosmarinic Acid from Free Radical and Lipid Peroxidation

  • The radical scavenging activity and inhibition effect from lipid peroxidation induced by peroxyl radical of methanol extract from Perilla frutescens and its active compound, rosmarinic acid (RA), were investigated in vitro. The treatment of extract and RA scavenged 1,1-diphenyl-2-picrylhydrazyl, hydroxyl radical (${\cdot}OH$) and nitric oxide in a concentration-dependent manner. In particular, the extract and RA showed strong radical scavenging activity against ${\cdot}OH$, the most toxic and reactive radical. In addition, Perilla frutescens and RA effectively inhibited lipid oxidation induced by sodium nitroprusside and 2,2'-azobis(2-aminopropane) dihydrochloride, determined by the ferric thiocyanate method. The present results suggest that Perilla frutescens and RA play a protective role against oxidative stress induced by free radical and lipid peroxidation.

Effects of Mulberry (Morus alba L.) Leaf Extract on Oxygen Radicals and Their Scavenger Enzymes in Liver of SD Rats (간장조직의 활성산소 및 그 제거효소에 미치는 뽕잎 추출물의 영향)

  • This study was designed to investigate the effects of mulberry (Morus alba L.) leaf extract (MLE) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (MLE-100 and MLE-300 groups) added 100 and 300 mg/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a significant decreases (15.2% and 18.1%, 5.6% and 8.0%, respectively) in liver mitochondria and microsomes could be not obtained. These are no significant differences in superoxide radical ($O_2$) levels of liver cytosol in MLE-100 and MLE-300 groups compared with control group. Lipid peroxide (LPO) levels were slightly decreased about 13.6% and 6.1% in liver mitochondria and microsomes of MLE-300 group compared with control group. Oxidized protein (OP) levels were remarkably decreased about 16.9% and 27.2% in liver microsomes only of MLE-100 and MLE-300 group compared with control group. Mn-SOD activities in liver mitochondria were remarkably increased (18.2% and 28.7%, respectively) in MLE-100 and MLE-300 groups, and Cu,Zn-SOD activities in liver cytosol were also significantly increased (11.3% and 20.2%, respectively) in MLE-100 and MLE-300 groups compared with control group. Mn-SOD activities in liver mitochondria were remarkably increased (18.2% and 28.7%, respectively) in MLE-100 and MLE-300 groups, and Cu,Zn-SOD activities in liver cytosol were also significantly increased (11.3% and 20.2%, respectively) in MLE-100 and MLE-300 groups compared with control group, but significant difference between GSHPx activities in liver cytosol could be not obtained. These results suggest that anti-aging effect of mulberry leaf extract (MLE) may play a pivotal role in attenuating a various age-related changes.

Effect of Silk Fibroin on Oxygen Radicals and Their Scavenger Enzymes in Liver of SD Rats (간장조직의 활성산소 및 그 제거효소에 미치는 실크 피브로인의 영향)

  • This study was designed to investigate the effect of silk fibroin (Mw 500)power (SFP) on oxygen radicals and their scavenger enzymes in liver membranes of rats. Sprague-Dawley (SD) male rats (160$\pm$10 g) were fed basic diet (control group), and experimental diets (SFP-2.5 and SFP-5.0 groups) added 2.5 and 5.0 g/kg BW/day for 6 weeks. Hydroxyl radical (.OH) levels resulted in a considerable decreases (5.8% and 8.4%, 3.7% and 11.1%, respectively) in liver motochondria and micorsomes of SFP-2.5 and SFP-5.0 groups compared with control group, and $O_2$radical level was remarkably decreased about 15% and 20% in liver cytosol of SFP-2.5 and SFP-5.0 groups compared with control group. Lipid peroxide (LPO) levels were significantly decreased (8.3% and 18.0%, 13.4% and 18.4%, respectively) in liver mitochondria and microsomes of SFP-2.5 and SFP-5.0 groups compared with control group. Oxidized protein (OP) levels were remarkably decreased about 11.6% in liver mitochondria of SFP-5.0 group compared with control group. Mn-SOD activities were remarkably in creased (17.6% and 28.8%, respectively) in mitochondria of SFP-2.5 and SFP-5.0 groups, and Cu/Zn-SOD activities were also effectively in creased (about 14.4%) in liver cytosol of SFP-5.0 groups, but significant difference between GSHPx activity in liver cytosol of these two groups could be not obtained. These results suggest that anti-aging effect of silk fibroin may play an effective anti-aging role in a aattenuating a oxidative stress and increasing a scavenger enzyme activity in liver membranes.
