텍스처의 감성공학적 평가 기법의 개발 (A Development of Estimation Method for Texture in Human Sensibility Ergonomics)

  • The relationship between the product design and product image can be defined using human sensibility. If there is a way to grasp the product value or product design that people want most, it could be possible to make and sell user-oriented products. In human sensibility ergonomics, the categories of design factor can be broken into shape and color of the design. Although, the color is the major factor of the design, however, texture were is also an important factor for the shape. In this study, wall-papers that have various shapes of texture were used for the experiment. The purpose of this study was to conduct a sensibility estimation experiment for the product image, and to estimate the parameters of various textures for creating new textures that could be more suitable for human sensibility.

  • Using a methylotrophic yeast Hansenula polymorpha, a heterologous gene expression and secretion system was developed for the production of hEGF(human Epidermal Growth Factor) which has been shown to promote epithelial cell proliferation and to inhibit gastric acid secretion. The hEGF gene was chemically synthesized according to the preferred codon usage in H. polymor- pha and expressed under the control of the strong and inducible methanol oxidase(MOX) promoter. The mating factor $\alpha$ pre-pro leader sequence of Saccharomyces cerevisiae was employed for hEGF to be secreted into the extracellular medium. This expression cassette was stably integrated into the host chromosomal DNA. Mature hEGF was efficiently expressed and secreted into the extracel- lular medium. About 24 mg/l of hEGF was detected in the cuture supernatant of a transformant with pA-EGF3 under the suboptimal culture conditions.

  • 입체영상은 공간에 대한 정확한 깊이인식을 기본으로 하며, 공간 디자인 및 인지과학 등의 다양한 차원에서 접근해야 한다. 특히 휴먼 팩터(human factor)를 고려한 인간공학적 설계가 중요하다. 즉, 입체영상설계자는 양안시차를 결정할 때, 인간의 눈과 카메라 시점을 일치시켜야 한다. 이 과정에서 광학적(dichoptic) 오차에 의한 왜곡 등의 구조적인 문제가 발생하며, 이를 해결하기 위해 기하학의 재해석 및 인간공학적 설계를 기반으로 시각 피로를 최소화하는 3D 스위트닝(sweetening)과정이 요구된다. 본 연구는 입체영상의 근본적인 불일치 문제를 실험영상의 제작을 통해 분석하고, 생체 안정성과의 상관관계를 파악한다. 이를 통해 인간공학적 설계에 기초한 다양한 입체영상의 공간 연출 방법을 제안한다.

A Human Immunodeficiency Virus Type 1 (HIV-1) Tat Cofactor Absent in Rodent Cells is a TAR-associated Factor

  • Background: Although Tat plays a role as a potent transactivator in the viral gene expression from the Human Immunodeficiency Virus type 1 long terminal repeat (HIV-1 LTR), it does not function efficiently in rodent cells implying the absence of a human specific factor essential for Tat-medicated transactivation in rodent cells. In previous experiments, we demonstrated that one of chimeric forms of TAR (transacting responsive element) of HIV-1 LTR compensated the restriction in rodent cells. Methods: To characterize the nature of the compensation, we tested the effects of several upstream binding factors of HIV-1 LTR by simple substitution, and also examined the role of the configuration of the upstream binding factor(s) indirectly by constructing spacing mutants that contained insertions between Sp1 and TATA box on Tat-mediated transactivation. Results: Human Sp1 had no effect whereas its associated factors displayed differential effects in human and rodent cells. In addition, none of the spacing mutants tested overcame the restriction in rodent cells. Rather, when the secondary structure of the chimeric HIV-1 TAR construct was destroyed, the compensation in rodent cells was disappeared. Interestingly, the proper interaction between Sp1 and TATA box binding proteins, which is essential for Tat-dependent transcription, was dispensable in rodent cells. Conclusion: This result suggests that the human-specific Tat cofactor acts to allow Tat to interact effectively in a ribonucleoprotein complex that includes Tat, cellular factors, and TAR RNA, rather than be associated with the HIV-1 LTR upstream DNA binding factors.

운전자 설문을 통한 자동차 운전자의 실수 확률 추정 (Estimation of Car Driver Error Probabilities Through Driver Questionnaire)

  • Car crashes are the leading cause of death for persons of every age. Specially, human-related factor has been known to be the primary causal factor of such crashes than vehicle-and environmental-related factors. There are various studies to analyze driver's behavior and characteristics in driving for reducing the car crashes in many areas of car engineering, psychology, human factor, etc. However, there are almost no studies which analyze mainly the human errors in driving and estimate their probabilities in terms of human reliability analysis. This study estimates the probability of human error in driving, i.e. driver error probability. First, fifty driver errors are investigated through DBQ (Driver Behavior Questionnaire) revision and the error likelihoods in driving are collected which are judged by skillful drivers using revised DBQ. Next, these likelihoods are converted into driver error probabilities using the results that verbal probabilistic expressions are changed into quantitative probabilities. Using these probabilities we can improve the warning effects on drivers by indicating their driving error likelihoods quantitatively. We can also expect the reduction effects of car accident through controlling especially dangerous error groups which have higher probabilities. Like these, the results of this study can be used as the primary materials of safety education on drivers.

Kahweol from Coffee Induces Apoptosis by Upregulating Activating Transcription Factor 3 in Human Colorectal Cancer Cells

  • Kahweol as a coffee-specific diterpene has been reported to induce apoptosis in human cancer cells. Although some molecular targets for kahweol-mediated apoptosis have been elucidated, the further mechanism for apoptotic effect of kahweol is not known. Activating transcription factor 3 (ATF3) has been reported to be associated with apoptosis in colorectal cancer. The present study was performed to investigate the molecular mechanism by which kahweol stimulates ATF3 expression and apoptosis in human colorectal cancer cells. Kahweol increased apoptosis in human colorectal cancer cells. It also increased ATF3 expression through the transcriptional activity. The responsible cis-element for ATF3 transcriptional activation by kahweol was CREB located between -147 to -85 of ATF3 promoter. ATF3 overexpression increased kahweol-mediated cleaved PARP, while ATF3 knockdown attenuated the cleavage of PARP by kahweol. Inhibition of ERK1/2 and $GSK3{\beta}$ blocked kahweol-mediated ATF3 expression. The results suggest that kahweol induces apoptosis through ATF3-mediated pathway in human colorectal cancer cells.

TSG101 Physically Interacts with Linear Ubiquitin Chain Assembly Complex (LUBAC) and Upregulates the TNFα-Induced NF-κB Activation

  • Linear ubiquitin chain assembly complex (LUBAC) is a ubiquitin E3 ligase complex composed of HOIP, HOIL-1L, and SHARPIN that catalyzes the formation of linear/M1-linked ubiquitin chain. It has been shown to play a pivotal role in the nuclear factor (NF)-κB signaling induced by proinflammatory stimuli. Here, we found that tumor susceptibility gene (TSG101) physically interacts with HOIP, a catalytic component of LUBAC, and potentiates LUBAC activity. Depletion of TSG101 expression by RNA interference decreased TNFα-induced linear ubiquitination and the formation of TNFα receptor 1 signaling complex (TNF-RSC). Furthermore, TSG101 facilitated the TNFα-induced stimulation of the NF-κB pathway. Thus, we suggest that TSG101 functions as a positive modulator of HOIP that mediates TNFα-induced NF-κB signaling pathway.

3차원 인제 형상 데이터를 이용만 목밑둘레 유형화 연구 - 20대 여성을 중심으로 - (A Study on the Classification of Neck-Base Circumference by Three-Dimensional Automatic Measurements of the Human Body - With the Focus on Women in their 20's -)

  • The purposes of this study lied in the analysis and classification of neck-base circumference shapes of the women in their twenties, by the application of three-dimensional automatic measurement data of human body, and thereby in the understanding of neck-base circumference shapes by the classified type. The findings are as follows: 1. The comparison of three-dimensional human body measurement items relating to the neck-base circumference part of the women in their twenties indicated that the largest individual difference was found in cervicale-center-anterior neck radius than in other items. 2. The factor analysis, which was conducted to extract the factors constituting the neck-base circumference, showed the shape of cervicale(factor 1), the shape of section neck(factor 2), the thickness of neck(factor 3), the shape of anterior neck(factor 4), and the shape of side neck(factor 5). 3. The classification of the neck-base circumference shapes resulted in three types. Type 1 was the shape of a reverse triangle hanging forward, Type 2 was that of a circle, and Type 3 was that of an oval open to the sides.

Improvement of Virus Safety of an Antihemophilc Factor IX by Virus Filtration Process

  • Viral safety is an important prerequisite for clinical preparations of plasma-derived pharmaceuticals. One potential way to increase the safety of therapeutic biological products is the use of a virus-retentive filter. In order to increase the viral safety of human antihemophilic factor IX, particularly in regard to non-enveloped viruses, a virus removal process using a polyvinylidene fluoride membrane filter (Viresolve NFP) has been optimized. The most critical factor affecting the filtration efficiency was operating pH and the optimum pH was 6 or 7. Flow rate increased with increasing operating pressure and temperature. Recovery yield in the optimized production-scale process was 96%. No substantial changes were observed in the physical and biochemical characteristics of the filtered factor IX in comparison with those before filtration. A 47-mm disk membrane filter was used to simulate the process performance of the production-scale cartridges and to test if it could remove several experimental model viruses for human pathogenic viruses, including human hepatitis A virus (HAV), porcine parvovirus (PPV), murine encephalomyocarditis virus (EMCV), human immunodeficiency virus type 1 (HIV), bovine viral diarrhea virus (BVDV), and bovine herpes virus (BHV). Non-enveloped viruses (HAV, PPV, and EMCV) as well as enveloped viruses (HIV, BVDV, and BHV) were completely removed during filtration. The log reduction factors achieved were $\geq$6.12 for HAV, $\geq$4.28 for PPV, $\geq$5.33 for EMCV, $\geq$5.51 for HIV, $\geq$5.17 for BVDV, and $\geq$5.75 for BHV. These results indicate that the virus filtration process successfully improved the viral safety of factor IX.

CJ-50001 (recombinant human granulocyte-colony stimulating factor)의 흰쥐와 개에서의 약물동태학적 연구 (Pharmacokinetics of CJ-50001i Recombinant Human Granulocyte-Colony Stimulating Factor, in Rats and Dogs)

  • The pharmacokinetics of CJ-50001 (recombinant human granulocyte-colony stimulating factor, developed by R&D center of Cheil Jedang Corp.) were investigated in rats and dogs. The serum concentrations of CJ-50001 were measured by a sandwich enzyme immunoassay. After single intravenous (iv) administration of Cf-50001 to rats at a dose of 5 $\mu$g/kg, the mean terminal half-life and area under the concentration-time curve (AUC) were 0.96 h and 124.497g . h/ml, respectively. After single subcutaneous (sc) administration at the same dose, maximum serum concentration was observed at about 2 hours after administration, and the mean terminal half-life, AUC and the bioavailability were 1.11 h,63.58$\mu$g . h/ml and 51.07%, respectively. In repeated dosing studies, CJ-50001 was administered iv and sc to rats at a daily dose of 5$\mu$g/kg for 7 days. The pharmacokinetic parameters, such as mean AUC and terminal half-life, were no significantly different from those of single administration. Following single iv and sc administration of CJ-50001 to dogs at a dose of 5 $\mu$g/kg, mean AUCs were much higher than those of rats, due to the decreased clearence (CL). After sc administration to dogs, maximum serum concentration was observed at 2~4 hours after administration and the bioavailability was 54.60%.
