Induction of cell cycle arrest and apoptosis by an indirubin analog, a CDK inhibitor, in human lung cancer cells

  • Cyclin-dependent kinases (CDKs) regulate the cell division cycle, apoptosis, transcription and differentiation. Inhibition of CDK is a promising target in development of anti-cancer agents. An indirubin analog (AGM01l), a CDK inhibitor, is a synthetic compound that inhibits human cancer cell growth in vitro. AGM01l showed a potent cytotoxicity in cultured human cancer cell lines (IC$\sub$50/ = 5.43 ${\mu}$M for A549, human colon cancer cell; IC$\sub$50/ = 1.21 ${\mu}$M for SNU-638, human stomach cancer cell; IC$\sub$50/ 9.23 ${\mu}$M for HL-60, human leukemia cell).   • To elucidate the mechanism of cyclic nucleotides, such as adenosine 3',5'-cyclic monophosphate (cAMP) and guanosine 3',5'-cyclic monophosphate (cGMP), in the regulation of human gastric motility, we examined the effects of forskolin (FSK), isoproterenol (ISO) and sodium nitroprusside (SNP) on the spontaneous, high $K^+$ and acetylcholine (ACh)-induced contractions of corporal circular smooth muscle in human stomach. Gastric circular smooth muscle showed regular spontaneous contraction, and FSK, ISO and SNP inhibited its phasic contraction and basal tone in a concentration-dependent manner. High $K^+$ (50 mM) produced sustained tonic contraction, and ACh $(10\;{\mu}M)$ produced initial transient contraction followed by later sustained tonic contraction with superimposed phasic contractions. FSK, ISO and SNP inhibited high $K^+$-induced tonic contraction and also ACh-induced phasic and tonic contraction in a reversible manner. Nifedipine $(1\;{\mu}M)$, inhibitor of voltage-dependent L-type calcium current $(VDCC_L)$, almost abolished ACh-induced phasic contractions. These findings suggest that FSK, ISO and SNP, which are known cyclic nucleotide stimulators, inhibit smooth muscle contraction in human stomach partly via inhibition of $VDCC_L$.

Comparison of histopathological and molecular diagnostic methods in Helicobacter suis infection in Korea (국내 도축돈의 위병변과 Helicobacter suis의 검출과의 상관관계)

  • Helicobacter suis is a gram negative bacterium and colonizes in porcine stomach. It causes gastric diseases in the stomach and plays a significant role in daily weight gains in pigs. Recent studies about one of potential sources of human gastric diseases. Therefore, this study was conducted to compare histopathological lesions and molecular detection of Helicobacter suis in the pyloric mucosa of porcine stomachs transferred from slaughterhouses, based on gross and histological examinations and a PCR assay. A total 90 stomach samples were investigated to record gastric lesion scores by characteristic gastric lesions, followed by routine H & E and Warthin-Starry silver staining to detect Helicobacter-like organisms. For PCR assay, H. suis specific primers and conditions are used. Sixty-one samples (67.8%) showed gross gastric lesions, of which 38 samples (40.2%) presented grade 1, 12 samples (13.3%) presented grade 2, and 11 samples (12.2%) presented grade 3, respectively. In Warthin-Starry silver stain, Helicobacter-like organisms were detected from 11 samples (12.2%) with 4 samples (4.4%) for grade 0, 5 samples (5.6%) for grade 1, 1 sample (1.1%) for grade 2 and 1 sample (1.1%) for grade 3, respectively. The PCR resulted positive in 37 samples (41.1%) with 14 samples (15.6%) for grade 0, 14 samples (15.6%) for grade 1, 3 samples (3.3%) for grade 2 and 6 samples (6.7%) for grade 3, respectively. Positive samples for both examinations were 5 samples (5.6%). The result suggested that it should be considered as one of factors causing a gastric disease in pigs. Also, it could be acknowledged to research fundamental aspects of Helicobacter-induced gastritis in human as an animal model.

Novel Clean End-to-End Anastomosis Method, Without Opening the Stomach Lumen, in Totally Laparoscopic or Robotic Pylorus-Preserving Gastrectomy

  • Purpose: Intra-abdominal infection is a common postoperative complication of laparoscopic pylorus-preserving gastrectomies (PPGs). Many studies have reported that intra-abdominal infectious complications after gastrectomy adversely affect patient survival outcomes. To prevent gastric fluid leakage into the abdominal cavity, we developed a novel anastomosis method in which the stomach lumen is not opened (termed the non-opened clean end-to-end anastomosis method [NoCEAM]) and evaluated its feasibility. Materials and Methods: Subsequent to lymphadenectomy, the oral and anal resection lines were sutured using an intraoperative endoscope. After closing the stomach circumferentially with clips, the specimen was rolled outward like a "donut." We resected the specimen circumferentially using a linear stapler, and anastomosis was completed simultaneously. We examined the feasibility of this procedure ex vivo, using three porcine stomachs, and in vivo, using one pig. Subsequently, we applied the procedure to 13 consecutive patients with middle-third early gastric cancer utilizing laparotomic, laparoscopic, and robotic PPG. Results: NoCEAM was completed in all porcine models and human cases. In the human cases, the mean operation time (±standard deviation) was 279±51 minutes, and mean blood loss volume was 22±45 mL. The mean number of linear staples used was 5.06±0.76. None of the patients had complications, and all were discharged on the eighth postoperative. The serum total protein, serum albumin, and hemoglobin levels did not change significantly after surgery. Conclusions: NoCEAM is feasible and safe for performing totally laparoscopic or robotic PPG. It may reduce postoperative complications, such as intra-abdominal infections.

Cancer Mortality and Incidence in Cement Industry Workers in Korea

  • Objectives: Cement contains hexavalent chromium, which is a human carcinogen. However, its effect on cancer seems inconclusive in epidemiologic studies. The aim of this retrospective cohort study was to elucidate the association between dust exposure in the cement industry and cancer occurrence. Methods: The cohorts consisted of male workers in 6 Portland cement factories in Korea. Study subjects were classified into five groups by job: quarry, production, maintenance, laboratory, and office work. Cancer mortality and incidence in workers were observed from 1992 to 2007 and 1997-2005, respectively. Standardized mortality ratios and standardized incidence ratios were calculated according to the five job classifications. Results: There was an increased standardized incidence ratio for stomach cancer of 1.56 (27/17.36, 95% confidence interval: 1.02-2.26) in production workers. The standardized mortality ratio for lung cancer increased in production workers. However, was not statistically significant. Conclusion: Our result suggests a potential association between cement exposure and stomach cancer. Hexavalent chromium contained in cement might be a causative carcinogen.

Synergistic Anti-tumor Effect of KLF4 and Curcumin in Human Gastric Carcinoma Cell Line

  • Kr$\ddot{u}$ppel-like factor 4 is a transcription factor which plays an important role in development and progression of various carcinomas. Curcumin characterized by excellent anti-cancer properties is regarded as a serviceable natural compound used in carcinoma therapy. This study aimed at exploring the impact of KLF4 overexpression in cooperation with curcumin on the proliferation, apoptosis and invasion of human gastric carcinoma BGC-823 cells. Flow cytometry analysis, CCK-8 assays, transwell assays and Western blot results showed that KLF4 overexpression combined with curcumin had significant anti-proliferation, pro-apoptosis and anti-invasion effects on BGC-823 cells. We also found that KLF4 had synergistic effects with curcumin, better promoting apoptosis and inhibiting proliferation and invasion of gastric carcinona cells. These results indicate that KLF4 could be used as a potential therapeutic target; curcumin could act as an auxiliary and provide a promising therapeutic strategy in stomach cancer.

Comparative susceptibility of different cell lines for culture of Toxoplasma gondii in vitro (톡소플라스마 곤디의 세포내 배양에 있어서 세포 주에 따른 감수성 비교)

  • In order to establish a useful cell culture system for T gondii we compared the degree of proliferation of T gondii tachyzoites among 8 different cell lines: 2 kinds of normal animal cells (MDCK-canine kidney cells; Vero-monkey kidney cells) and 6 kinds of human tumor cells (A 549, PC 14-lung cancer cells; SNU 1, SNU 16. Mlm 45-stomach cancer cells; HL-60-promyelocytic leukemia cells), through morphological observation and 3H-uracil uptake assay. The degree of susceptibility to infection with T gondii tachyzoites was highest in A 549 and PC 14 cells, medium in Vero, HL-60, MDCK and SNU 1, and lowest in SNU 16 and MBm 45 cells. The kinetics of T gondii multiplication during the post-Infection 60 hours were higllly dependent upon the dose of tachyzoites administered and the duration among the 8 tested fur the growth and multiplication of T gondii in vitro.

  • I studied connection between meridians and Dianxian(癲癎, Epilepsy). Dianxian is caused by Feun(風) based on the Blood-Deficiency(血虛) which is induced by Heart-deficiency(心虛). Therefore, when Heart and blood are in deficient condition(心血虛), Heart meridian(心經) and Pericardium meridian(心包經) can be easily affected by Feng(風), which is main cause of Dianxian. Furthermore, Liver heat(肝熱) is another main cause of Dianxian. It shows along Liver meridian(肝經) and affects Pericardium Meridian(心包經). Heart, Pericardium and Liver are main organs of human psychoconscious activities, and also are main pathological organs of Dianxian. Dianxian progresses in several ways and simtoms of Dianxian are variant according to the deficiency-exess state(虛實) of Stomach and Spleen meridians(脾胃經). So, I concluded that controling Heart, Pericardium, Liver, Stomach and Spleen meridians can make therapeutical effects on Dianxian. So I report it for the better treatment, and it shoud need further study.
