A study on the determination of lead in whole blood by ICP/MS (ICP/MS에 의한 전혈 중 납의 분석방법 연구)

  • An accurate analysis method for the determination of lead in whole blood by ICP/MS was developed. Whole blood samples were decomposed in microwave digestion system without any contamination and loss of lead. The 96 samples were analyzed by ICP/MS using mass$^{208}$ isotope of lead. Lead concentrations of human whole blood were ranged of $2.50{\sim}22.8{\mu}g/dL$. The accuracy of this analysis method was verified by analyzing of NIST SRM 955a series(lead in blood).

  • In the cardiovascular system, the waveform of the pulsatory blood pressure appears variously due to the cardiac impulse and compliance of blood vessels and arm tissue. We have constructed a blood pressure simulator to investigate effects of mechanical properties of artery walls and tissue on blood pressure measurements. The blood pressure simulator is designed to reproduce wave forms of blood pressure in human arteries. To minimize tracking error, we use a linear control valve, and adapt a hybrid control scheme which consists of a feedback controller and a feedforward controller. Any form of the pressure wave can be reproduced, changing function of the wave form in the computer connected to the simulator for control. From experiments, it has been shown that the simulator reproduces wave forms very well, and that the hybrid scheme adapted is superior to the feedback controller.

  • Characteristics of the pulsatile flow in a 3-dimensional elastic blood vessel are investigated to understand the blood flow phenomena in the human body arteries. In this study, a model for the elastic blood vessel is proposed. The finite volume prediction is used to analyse the pulsatile flow in the elastic blood vessel. Variations of the pressure, velocity and wall shear stress of the pulsatile flow in the elastic blood vessel are obtained. The magnitudes of the velocity waveforms in the elastic blood vessel model are larger than those in the rigid blood vessel model. The wall shear stresses on the elastic vessel vary with the blood vessel motions. Amplitude indices of the wall shear stress for blood in the elastic blood vessel are $4\sim5$ times larger than those of the Newtonian fluid. As the phase angle increased, point of the phase angle is are moved forward and the wall shear stresses are increased for blood and the Newtonian fluid.

Effects of obesity adult participation in boxing diets on weight loss, blood pressure and blood sugar improvements

  • This research is about effects of obesity adults participating in 12weeks of boxing diets program on weight loss, blood pressure and blood sugar improvements. The exercise program was conducted three times a week with a maximum heart rate of 50 to 65 percent for first to forth weeks and 65 to 85 percent for fifth to twelfth. The study included 59 obese people in their 20s and 50s, with a BMI of 25kg/m2 or higher, blood pressure of 140/90mmHg, and high blood pressure and diabetes patients of 125 mg/dl per fasting blood sugar. The following results were obtained by observing the results before and after the boxing exercise program. High blood pressure, diabetes, high blood pressure + diabetes after 12 weeks showed significant weight, BMI and body fat reduction, as well as significant decrease in blood pressure and fasting blood sugar. Consequently, through this study, obese adults can lower their weight, blood pressure, and blasting fasting blood sugar through a 12-week high-strength boxing exercise program and it is believed to help improve and prevent obesity, cardiovascular disease and metabolic diseases.

Systematic review of the effect of omega-3 fatty acids on improvement of blood flow while focused on evaluation of claims for health functional food (건강기능식품의 기능성을 중심으로 한 오메가-3 지방산 함유유지의혈행개선 효과에 대한 체계적 고찰)

  • Omega-3 polyunsaturated fatty acids are essential fatty acids because humans cannot synthesize them de novo and must obtain them in their diet. Fish and fish oil are rich sources of omega-3 fatty acids, including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA). Significant evidence of the beneficial role of dietary intake of omega-3 fatty acids in blood flow has been reported and putative mechanisms for improvement of blood flow include anti-thrombotic effects, lowered blood pressure, improved endothelial function, and anti-atherogenic effects. Edible oils containing omega-3 fatty acids were registered as functional ingredients in the Korea Health Functional Food Code. Although omega-3 fatty acids have been evaluated by the Korea Food and Drug Administration (KFDA) based on scientific evidence, periodic re-evaluation may be needed because emerging data related to omega-3 fatty acids have accumulated. Therefore, in this study, we re-evaluated scientific evidence for the effect of omega-3 fatty acids as a functional ingredient in health functional food on improvement of blood flow. A comprehensive literature search was conducted for collection of relevant human studies using the Medline and Cochrane, KISS, and IBIDS databases for the years 1955-2012. Search keywords were used by combination of terms related to omega-3 fatty acids and blood flow. The search was limited to human studies published in Korean, English, and Japanese. Using the KFDA's evidence based evaluation system for scientific evaluation of health claims, 112 human studies were identified and reviewed in order to evaluate the strength of the evidence supporting a relation between omega-3 fatty acids and blood flow. Among 112 studies, significant effects on improvement of blood flow were reported in 84 studies and the daily intake amount was ranged from 0.1 to 15 g. According to this methodology of systematic review, we concluded that there was possible evidence to support a relation between omega-3 fatty acid intake and blood flow. However, because inconsistent results have recently been reported, future studies should be monitored.

Genoprotective Effect of Melatonin Against to the Genotoxicity of Glyphosate on Human Blood Lymphocytes (글라이포세이트의 유전자 독성에 대한 멜라토닌의 유전자 보호 효과)

  • Purpose: Glyphosate is a widely used non-selective herbicide. Previous studies have shown that glyphosate has genotoxicity, and that even low-doses of glyphosate can cause DNA damage. Melatonin is a hormone produced and secreted by the pineal gland that is known to be a potent anti-carcinogen, anti-oxidant, and genetic protector. This study was conducted to investigate the genoprotective effect of melatonin against glyphosate in human blood lymphocytes. Methods: Human peripheral blood was obtained from 15 young, healthy volunteers and cultured under four different toxicologic conditions. The four groups consisted of a control group, glyphosate only group (300 ng/mL), glyphosate with low level of melatonin group ($50{\mu}M$), and glyphosate with high level of melatonin group ($200{\mu}M$). The mean Sister Chromatid Exchange (SCE) frequency of each group was then analyzed. Results: Glyphosate exposed groups had a higher mean SCE frequency ($10.33{\pm}2.50$) than the control group ($6.78{\pm}2.31$, p<0.001). Interestingly, the group that received a low-level of melatonin had a lower mean SCE frequency ($8.67{\pm}2.58$) than the glyphosate-only group, while the group that received a high level of melatonin had a much lower mean SCE frequency ($8.06{\pm}2.50$) than the glyphosate-only group. There was statistical significance. Conclusion: Melatonin exerted a potent gene protective effect against the genotoxicity of glyphosate on human blood lymphocytes in a dose-dependent fashion.

Determination of Cd and Pb in Human Blood by Isotope Dilution Inductively Coupled Plasma Mass Spectrometry : International Comparison (동위원소희석 질량분석법에 의한 혈액 중 Cd, Pb 성분의 측정 : 국제공동분석)

  • Inorganic analytical laboratory of Korea Research Institute of Standards and Science participated in an interlaboratory comparison program operated by Quebec Toxicology Centre of Canada in 1994 and again in 1995. The objective of this program is to enable participating laboratories to assess reproducibility and accuracy of their analytical results for trace toxic elements in human biological fluids. This laboratory determined Cd and Pb concentrations in 3 levels of human blood samples by isotope dilution inductively coupled plasma mass spectrometry. 0.5mL of blood sample is added to the digestion bomb together with 2mL of nitric acid and enriched spike isotopes and then decomposed in the microwave digestion system. The decomposed sample is diluted to 10mL and nebulized into ICP-MS. The Cd and Pb values reported by all participating laboratories are presented and compared. The values reported by this laboratory are within the acceptable range of target values.

  • This study was carried out to identify the components of the human heart meridian muscle, the regional muscle group being divided into outer, middle, and inner layers. The inner parts of the body surface were opened widely to demonstrate muscles, nerves, blood vessels and to expose the inner structure of the heart meridian muscle in the order of layers. Different Gene Expression on Human Blood by Administration of OLT-2 (OLT-2의 복용에 의한 인간 혈중 유전자 발현 변화)

  • This study was performed to investigate genes which are differently expressed in human blood by administrating of OLT-2. OLT-2 was medical precipitation composed of three medicinal herbs, Ginseng Radix, Astragali Radix, Glycyrrhizae Radix, and anti-leukemia effect of it was evaluated from Byung Hun Jeon of Wonkwang University this study was approved by Institutional Review Board of Korea Institute of Oriental Medicine (Taejeon, Korea) and four male subjects participated in this study. Gene expressions were evaluated by cDNA chip, in which 24,000 genes were spotted. Hierarchical cluster and biological process against the genes, which expression changes were more than 1.6 fold, were constructed by cluster 3.0 providing Stanford University and EASE(http://apps1 .maid.nih.gov/DAVID). Five groups were clustered according to their expression patterns. Group A contained gene decreased by OLT-2 and increased genes by OLT-2 were involved in Group B, C, D. In biological process, expression of genes involved in cytokine or cell calcium signaling, such as interleukin 18 and G-protein beta 4 were increased, but protein tyrosine phosphatase receptor type c, which function is cell adhesion between antigen-presenting cell and T or B-cell, was decreased by OLT-2. This study provides the most comprehensive available survey of gene expression changes in response to anti-leukemia effect of OLT-2 in human blood.

Effects of scalp treatment using combinational massage technique on human physiology

  • The purposes of this study are to measure physiological reactions of human body according to the scalp treatment, a popularized service in beauty care industry and propose efficient ways of scalp treatment. To meet the goals, total 30 applicants without any medical history(5 males and 5 females in 20's, 30's and 40's respectively) were informed on the purpose of experiment hereof and were investigated and received a 30-minute scalp treatment, which combines standardized scalp treatment massage technique proposed by KAT and ITF with another massage technique operated in the beauty salon run by the author of this paper. 5ml of blood samples were taken from each subject before and after the scalp treatment respectively and the blood sample was divided into 3 different tubes for analysis: 1) 2 ml for blood cell analysis, 2) 2ml for enzyme activity measurement, 3) 1ml for hormone level reading. In order to determine effects of scalp treatment on ALP, GOT, GPT, ${\gamma}-GTP$, WBC, RBC, Hb, Hct, Platelet, MCV, MCH and MCHC, all collected data were used for measuring respective levels of these blood substances by means of enzyme reaction measurement, enzyme activity measurement and automated hematology analyzer. Then, all measured data were analyzed through paired t-test using SPSS WIN 11.5. As a result, the scalp treatment is associated with improving hepatic function, facilitating blood circulation and helping blood coagulation and hemostasis in a effective way. Therefore, it would be necessary to conduct further studies on this subject related to anemia in the future.
