Background: It has been found that Helper T cells in the peripheral blood are decreased in the cell mediated immunity in the pulmonary tuberculosis. But it has not been confirmed yet that only decrease in number of cells which has phenotype in the peripheral blood is defined to decrease in cell mediated immunity. The immunocytochemical study was performed to observe the change of the percentage of T-lymphocytes with their subsets and activated T cells in the peripheral blood of pulmonary tuberculosis and to know how many T cells would be activated, relative to resting cells in the peripheral blood. Methods: The peripheral blood obtained from twenty two patients and ten healthy controls were smeared on the gelatin coated slide glass prepared for of mononuclear cells. The double bridge technique of alkaline phosphatase-antialkaline phosphatase(APAAP) method was used. As the primary antibodies, $T_1$(anti-human T cell), $T_4$(anti-human helper/inducer T cells) and $T_8$(anti-human supressor/cytotoxic T cell) antibodies and interleukin-2 receptor (for early activated T cell), very late activation antigen (for activated cytotoxic T cell), T cell lineage specific activation antigen monoclonal actibodies were used. Results: 1) There were significantly decrease in the absolute number of $T_4$(+) cells but significantly increase of $T_8$(+) cells in the peripheral blood of pulmonary tuberculosis (p<0.05). 2) The percentage of $T_4$(+) cells showed significantly decrease in pulmonary tuberculosis but $T_8$(+)cells significantly increase(p<0.05). $T_4(+)/T_8(+)$ ratio showed significantly decrease in the peripheral blood of the pulmonary tuberculosis(p<0.05). 3) There were significantly increase in the absolute number of variable stages of activated T cells in the peripheral blood of the pulmonary tuberculosis(p<0.05). 4) The percentage of IL-2R, VLA-1, TLiSA were 6.45+1.56%, $7.64+1.34^*$, 10.45+1.16% in order which showed significantly increase in the peripheral blood of the pulmonary tuberculosis(p<0.05). Conclusion: We speculate that only a few percentage of T lymphocyte is activated in cell mediated immunity in pulmonary tuberculosis.
A rapid, selective and sensitive reverse-phase HPLC methods for the determination of atenolol and chlorthalidone in human serum and whole blood were validated, and applied to the pharmacokinetic study of atenolol and chlorthalidone combination therapy. Atenolol and an internal standard, pindolol, were extracted from human serum by liquid-liquid extraction, and analyzed on a $\mu$-Bondapak C18 $10-{\mu}$ column in a mobile phase of methanol-0.01 M potassium dihydrogenphosphate(30:70, v/v, adjusted to pH 3.5) and fluorescence detection(emission: 300 nm, excitation: 224 nm). Chlorthalidone and an internal standard, probenecid, were extracted form human whole blood by liquid-liquid extraction, and analyzed on a Luna C18 $5-{\mu}$ column in a mobile phase of acetonitrile containing 77% 0.01 M sodium acetate and UV detection at 214 nm. These analysis were performed at three different laboratories using the same quality control(QC) samples. The chromatograms showed good resolution, sensitivity, and no interference by human serum and whole blood, respectively. The methods showed linear responses over a concentration range of 10-1,000 ng/mL for atenolol and 0.05-20 ${\mu}g/mL$ for chlorthalidone, with correlation coefficients of greater than 0.999 at all the three laboratories. Intra- and inter-day assay precision and accuracy fulfilled international requirements. Stability studies(freeze-thaw, short-, long-term, extracted sample and stock solution) showed that atenolol and chlorthalidone were stable. The lower limit of quantitation of atenolol and chlorthalidone were 10 ng/mL and 0.05 ${\mu}g/mL$, respectively, which was sensitive enough for pharmacokinetic studies. These methods were applied to the pharmacokinetic study of atenolol and chlorthalidone in human volunteers following a single oral administration of Hyundai $Tenoretic^{\circledR}$ tablet(atenolol 50 mg and chlorthalidone 12.5 mg) at three different laboratories.

National reference standards (NRSs) for biologics are established through potency estimation by a multi-center joint study of standard materials used in the approval process for national lot release and quality control of vaccines, blood products, and other biologics. In this study, a stability evaluation was conducted to determine whether the potency of NRSs for six blood products was being maintained at a consistent level in Korea. The present study conducted real-time stability tests via in-vivo/in-vitro bioassay on NRSs for blood coagulation factor VIII concentrate (2nd standard), antithrombin concentrate, prekallikrein activator, anti-hepatitis B immunoglobulin, blood coagulation factor IX concentrate, and anti-tetanus human immunoglobulin, as well as a trend analysis using cumulative annual results. The real-time stability test results showed that the mean potency of six NRSs was all within the control limit. In the trend analysis, the potency of NRS for blood coagulation factor VIII concentrate (2nd standard) showed a decreasing trend, while the potency of all other products had been stably maintained. The present study confirmed that the mean potency of NRSs for six blood products had been stably maintained in Korea. The findings of the present study establish a foundation that can ensure the quality of NRSs for biologics in Korea, and it is expected to make a major contribution to the supply of high-quality biologics.

Objective: To evaluate the effects of contamination by either blood or a hemostatic agent on the shear bond strength (SBS) of orthodontic buttons. Methods: We used 45 freshly extracted, non-carious, impacted third molars that were divided into 3 groups of 15. Each tooth was etched with 37% phosphoric acid gel for 30 s. Human blood or the blood stopper agent was applied to the tooth surface in groups I and II, respectively. Group III teeth were untreated (controls). Orthodontic buttons were bonded to the teeth using light-curing composite resin. After bonding, the SBS of the button was determined using a Universal testing machine. Any adhesive remaining after debonding was assessed and scored according to the modified adhesive remnant index (ARI). ANOVA with post-hoc Tukey's test was used to determine significant differences in SBS and Fisher's exact test, to determine significant differences in ARI scores among groups. Results: ANOVA indicated a significant difference between groups (p < 0.001). The highest SBS values were measured in group III ($10.73{\pm}0.96$ MPa). The SBS values for teeth in groups I and II were significantly lower than that of group III (p < 0.001). The lowest SBS values were observed in group I teeth ($4.17{\pm}1.11$ MPa) (p < 0.001). Conclusions: Contamination of tooth surfaces with either blood or hemostatic agent significantly decreased the SBS of orthodontic buttons. When the contamination risk is high, it is recommended to use the blood stopper agent when bonding orthodontic buttons on impacted teeth.
The human physiological systems are so complex and irregular dynamics. Dynamics of peripheral blood vessel, in particular, have quite sensitive and complex. Before, the linear analytic method have been used to analyze the system. But, the method have many problems to predict the following results. In the other hand, the nonlinear analytic method, chaotic time series analysis method, is suitable for measuring complex, vary system. In this study, the scalar data of the blood flow of peripheral blood vessel of rabbits, in accordance with injection of glucose, was obtained and redefined as multi-dimensional vectors, with time-series analytic methods. This study also intended to confirm that the peripheral blood flow is chaotic dynamics and evaluate the availability of non-linear analytic method. As a result, the existing FFT, and mean could show the difference of blood flow of peripheral blood vessel by injection of glucose, but the nonlinear analytic method could show the definite difference. The hemodynamics is a chaotic phenomenon.
The causes and treatment of alopecia are concluded based on the records found successive oriental medicine in order to persuade new treatments for alopecia as following : 1. In the context of modern medical science male-dominated alopecia was referred as dok-rak(禿落), cho-dok(早禿), chon-dok(全禿), pal-ju-tal-bal, chu-bal-sun and pal-sun(髮癬) alternatively, while alopecia areata was refereed as yu-pung(油風), pan-dok(斑禿), kwi-ji-du and kwi-che-du(鬼剃頭). 2. The causes of alopecia is related with condition of Gi(vital energy) and Hyul(blood) in the twelve channels, such as weakening of vital energy in the conduits, condition of internal organs as heat in the lung, weak kidney or liver vitality, and eating habits such as severe ingestion of bitter taste or sweet taste food. Other distinct reasons are penetration of wind and dryness into a head due to weakening Gi and Hyul inside human body, flaring up of the asthenic fire due to excessive eatings, hard labour or psychological sufferings deficiency of blood, deficiency of vital essence of kidney, phlegm caused by dampness and heat, and stagnated blood. 3. According to demonstration of alopecia, the causes of alopecia areata are listed as internal wind due to heat of blood, deficiency of liver and kidney, blookage of channels and collaterals by stagnated blood, and causes of male-dominated alopecia are listed as wind dryness caused by heat of blood, dampness and heat, and heat, wind and dryness due to deficiency of blood.
Objectives: This study was designed to assess the level of physical exposure to heavy metals among residents who live around a Chungcheongnam-do Province industrial complex and to provide baseline data on the effects and harms of heavy metals on the human body by comparing their exposure levels to those of people from control regions. Methods: We measured blood lead and cadmium levels and urine mercury and chromium levels and conducted a survey among 559 residents from the affected area and 347 residents of other areas. Results: Blood lead and cadmium levels and urine mercury levels were significantly higher in the case region than among those in the control region (p=0.013, p<0.001, p<0.001, respectively). In the thermoelectric power plant area, blood cadmium and urine mercury levels were significantly higher than in the control region (p<0.001, p<0.001, respectively). In the steel mill and petrochemical industry areas, blood cadmium level was significantly higher than that in the control region (p<0.001). Dividing groups by the reference level of blood cadmium ($2{\mu}g/L$), the odds ratios between the case and control regions were 2.56 (95% CI=1.83-3.58), 3.11 (95% CI=2.06-4.71) for the thermoelectric power plant area, 1.78 (95% CI=1.19-2.65) for the steel mill area and 4.07 (95% CI=2.40-6.89) for petrochemical industry area. Conclusion: This study showed that the levels of exposure to heavy metals among residents living near a Chungcheongnam-do Province industrial complex were significantly higher than those in the control region. This seems to be attributable to exposure to heavy metals emissions from the industrial complex. Further research and safety measures are required to protect residents' health.
Vitamin B(sub)12(cobalamin) is an essential nutrient in human and it is particularly important during pregnancy. Nevertheless very few studies have reported, concerning vitamin B(sub)12 in relation with reproduction. This study was conducted to evaluate the vitamin B(sub)12 nutrition status of Korean pregnant women and to investigate the relationship between serum vitamin B(sub)12 levels of maternal-umbilical cord blood and pregnancy outcomes. Dietary vitamin B(sub)12 intakes of the pregnants were estimated by semiquantitative frequency questionnaire. Serum vitamin B(sub)12 levels in both maternal blood and umbilical cord blood of 30 pregnant women at delivery were measured by radioimmunoassay. Mean vitamin B(sub)12 intake was 3.3$\pm$1.4$\mu\textrm{g}$/d which was 125.8% of the Korean RDA(2.6$\mu\textrm{g}$) for vitamin B(sub)12 level of umbilical cord blood was 607.8$\pm$282.9pg/ml, more than two fold of maternal vitamin B(sub)12 level 268.6$\pm$97.8pg/ml. This finding indicates that fetal uptake of vitamin B(sub)12 in the fetus may be due to an active transport mchanism across the placenta. Umbilical cord blood vitamin B(sub)12 levels were highly correlated with maternal levels($r^2$=0.548, p<0.001), showing that fetal vitamin B(sub)12 level is affected by maternal status. However there was no significant correlation between the serum vitamin B(sub)12 levels in maternal-umbilical cord blood and the pregnancy outcomes except for the birth weight. Maternal-umbilical serum vitamin B(sub)12 levels were the highest in the group of birth weight 3.0-3.5kg, and the lowest in the group of birthweight below 3.0kg. Objectives: Bee venom (BV) is a complex mixture of proteins and contains proteins such as phospholipase and melittin, which have an effect on blood clotting and blood clots. The mechanism of action of honey bee venom (HBV, Apis mellifera) on human plasma proteins and its anti-thrombotic effect were studied. The purpose of this study was to investigate the anti-coagulation effect of BV and its effects on blood coagulation and purification. Methods: Crude venom obtained from Apis mellifera was selected. The anti-coagulation factor of the crude venom from this species was purified by using gel filtration chromatography (sephadex G-50), and the molecular weights of the anti-coagulants in this venom estimated by using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). Blood samples were obtained from 10 rabbits, and the prothrombin time (PT) and the partial thromboplastin time (PTT) tests were conducted. The approximate lethal dose (LD) values of BV were determined. Results: Crude BV increased the blood clotting time. For BV concentrations from 1 to 4 mg/mL, clotting was not observed even at more than 300 seconds, standard deviations $(SDs)={\pm}0.71$; however, clotting was observed in the control group 13.8 s, $SDs={\pm}0.52$. Thus, BV can be considered as containing anti-coagulation factors. Crude BV is composed 4 protein bands with molecular weights of 3, 15, 20 and 41 kilodalton (kDa), respectively. The $LD_{50}$ of the crude BV was found to be $177.8{\mu}g/mouse$. Conclusion: BV contains anti-coagulation factors. The fraction extracted from the Iranian bees contains proteins that are similar to anti-coagulation proteins, such as phospholipase $A_2(PLA_2)$ and melittin, and that can increase the blood clotting times in vitro.
We have developed a simple and accurate strip test that measures the blood glucose level semiquantitatively by visual observation, or qualitatively by using UltraScan spectrocolorimeter. The strip has solid phase reagents, including glucose oxidase, peroxidase, chromogen, affixed to a plastic support. The strip test is capable of measuring blood glucose level in the range of 0∼800 mg/dl and generating the results within 2 to 3 minutes. Human blood specimens obtained from normal individuals and the diabetic patients were evaluated by the new blood glucose strip and by the kit supplied by other commercial products. The test results exhibit the correlation coefficient of 0.964. The new test strip is proven simple and accurate, and it offers an alternative to the commercially available glucose tests.
