Guided Selection of Human Antibody Light Chains against TAG-72 Using a Phage Display Chain Shuffling Approach

  • To enhance therapeutic potential of murine monoclonal antibody, humanization by CDR grafting is usually used to reduce immunogenic mouse residues. Most humanized antibodies still have mouse residues critical for antigen binding, but the mouse residues may evoke immune responses in humans. Previously, we constructed a new humanized version (AKA) of mouse CC49 antibody specific for tumor-associated glycoprotein, TAG-72. In this study, to select a completely human antibody light chain against TAG-72, guided selection strategy using phage display was used. The heavy chain variable region (VH) of AKA was used to guide the selection of a human TAG-72-specific light chain variable region (VL) from a human VL repertoire constructed from human PBL. Most of the selected VLs were identified to be originated from the members of the human germline VK1 family, whereas the VL of AKA is more homologous to the VK4 family. Competition binding assay of the selected Fabs with mouse CC49 suggested that the epitopes of the Fabs overlap with that of CC49. In addition, they showed better antigen-binding affinity compared to parental AKA. The selected human VLs may be used to guide the selection of human VHs to get completely human anti-TAG72 antibody.

  • 본 논문에서는 시퀀스 레이블링 문제(sequence labeling problem)인 개체명 인식에 사용할 새로운 태깅 포맷인 Delimiter tag (D-tag)를 소개한다. 시퀀스 레이블링 문제에서 사용하는 BIO-tag 포맷은 개체명 레이블을 B (beginning)와 I (inside) 의미의 레이블로 확장하여 타겟 클래스의 수가 2배 증가한다. 또한 BIO-tag 포맷을 사용할 경우, 모델이 B와 I 를 잘못 분류하는 문제가 발생하며, 레이블 수가 많은 세부 분류 개체명의 경우에는 label confusion을 야기한다. 본 논문에서 제안한 D-tag 포맷은 타겟 클래스의 수를 증가시키지 않기 때문에 앞서 언급한 문제를 해결할 수 있다. 실험 결과, D-tag를 사용하여 학습한 모델이 BIO-tag를 사용한 경우보다 더 좋은 성능을 보여, 유망함을 확인하였다.

  • This study aims to improve the access and retrieval of images and to find a way to effectively generate tags as a tool for providing explanation of images. To do this, this study investigated the features of human tagging and machine tagging, and compare and analyze them. Machine tags had the highest general attributes, some specific attributes and visual elements, and few abstract attributes. The general attribute of the human tag was the highest, but the specific attribute was high for the object and scene where the human tag constructor can recognize the name. In addition, sentiments and emotions, as well as subjects of abstract concepts, events, places, time, and relationships are represented by various tags. The tag set generated through this study can be used as basic data for constructing training data set to improve the machine learning algorithm.

  • In this paper, we proposed a Hangul type chipless RFID tag, which has better legibility than the conventional chipless RFID tag not only to a computer but also to a human. We made consonant model, vowel model and whole character model by WIPL tool and checked the applicability of Hangul type chipless RFID tag. We obtain the RCS pattern of each character by simulation. Finally, We classify the character from input data in noisy environment using a variance of the data.

  • 의미 TAG는 한국어 기초어휘에 대한 개념지식을 구축하는 데 기본이 될 뿐만 아니라, 문장 분석시의 구조적 모호성과 단어 의미 모호성을 해소하는 중요한 단서를 제공할 수 있다. 이러한 의미 TAG가 실용적으로 여러 응용 시스템에서 사용되기 위해서는 광범위하고 타당한 자료를 바탕으로 하여 객관적인 방법으로 설정 되어야 한다. 국어사전의 뜻풀이말에서의 상위개념을 표제어의 상위어로 선정하는 bottom-up 방식으로 구축하였던 한국어 명사의미체계는 근본적으로 사전편찬자의 비일관적인 뜻풀이말의 기술에 따른 여러 문제점이 있었다. 본 연구에서는 이러한 문제점들을 해결하기 위해서 사전 뜻풀이말에서 상위개념을 수식하는 어절과 용언의 의미호응관계에서 상위개념의 의미속성을 추출하고, 이들 의미속성에 의한 명사 의미체계를 구축하여 이를 바탕으로 명사의미 TAG를 설정할 수 있도록 하였다.

  • Recently the tagSNP selection problem has been researched for reducing the cost of association studies between human's diversities and SNPs. General approach for this problem is that all of SNPs are separated into appropriate blocks and then tagSNPs are chosen in each block. Marsel in this paper is the system that involved the concept of linkage disequilibrium for overcoming the problem that the existing block partitioning approaches have short of biological meanings. In most approaches, the contiguous regions, which recombinations have LD coefficient |D'| and then tagSNP selection step is performed. And MarSel guarantees the minimum tagSNP selection using entropy-based optimal selection algorithm when tagSNPs are chosen in each block, and enables chromosome-level association studies using efficient memory management technique when input is very large-scale dataset that is impossible to be processed in the existing systems.

A Tag-based Music Recommendation Using UniTag Ontology (UniTag 온톨로지를 이용한 태그 기반 음악 추천 기법)

  • In this paper, we propose a music recommendation method considering users' tags by collaborative tagging in a social music site. Since collaborative tagging allows a user to add keywords chosen by himself to web resources, it provides users' preference about the web resources concretely. In particular, emotional tags which represent human's emotion contain users' musical preference more directly than factual tags which represent facts such as musical genre and artists. Therefore, to classify the tags into the emotional tags and the factual tags and to assign weighted values to the emotional tags, a tag ontology called UniTag is developed. After preprocessing the tags, the weighted tags are used to create user profiles, and the music recommendation algorithm is executed based on the profiles. To evaluate the proposed method, a conventional playcount-based recommendation, an unweighted tag-based recommendation, and an weighted tag-based recommendation are executed. Our experimental results show that the weighted tag-based recommendation outperforms other two approaches in terms of precision.

A Study on Rule Schemas of User Interface in HCI Devices (HCI 장치의 사용자 인터페이스 규칙스키마에 관한 연구)

  • As humans perform many tasks using computers, HCI(Human-Computer Interaction) has received much attention these days. One objective of HCI research is to propose how to design more consistent systems. In order to evaluate the consistency of HCI devices from the user's perspective, several models such as TAG(Task Action Grammar), GOMS(Goals-Operators-Methods-Selection Rule), and GTN(General Transition Network) have been developed. TAG specifies actions to perform tasks in terms of rule schema. It has been verified that the less the number of rule schema is, the better users perform the tasks due to assumably higher consistency. This paper hypothesizes that the consistency of systems depends not only on the number of rule schema but also on the distances between rule schema. That is, the closer the rule schema are, the easier it is to acquire the whole set of rule schema. An experiment supported this hypothesis. Therefore, distances between rule schema should be considered as well as the number of rule schema when designing systems.

Enhanced Tumor-targeted Gene Delivery by Immunolipoplexes Conjugated with the Humanized Anti-TAG-72 Fab' Fragments

  • Cationic immunoliposomes were prepared by conjugation of Fab' fragments of the recombinant humanized monoclonal antibody (HuCC49) against tumor-associated glycoprotein (TAG)-72 to sterically unilamella liposomes. The cationic immunoliposomes are composed of cationic lipid (O,O'-dimyristyl-N-lysyl aspartate, DMKD), cholesterol, and 1,2-dipalmitoyl-sn-glycero-3-phosphoethanolamine-N-[maleimide(polyethyleneglycol)$_{2000}$] (DPPE-PEG-maleimide) with a molar ratio of 0.5:0.47:0.03. Plasmid DNA was effectively condensed by addition of transferrin (Tf) during the formation of anti-TAG-72 PEG-immunolipoplexes (PILPs). These anti-TAG-72 PILPs were able to adhere to the surface of TAG-72-overexepressing LS174T human colon cancer cells more effectively than conventional liposomes, thereby facilitating gene delivery in vitro. Furthermore, intravenous administration of the anti-TAG-72 PILPs into the tumor-carrying mice exhibited efficient localization of the reporter gene in the tumor tissues.

Expression of Human Serine Palmitoyltransferase Genes for Antibody Development (Antibody 제작을 위한 human serine palmitoyltransferase 유전자의 발현)

  • For antibody development of human serine palmitoyltransferase (SPT, EC 2.3.1.50), SPTLC1 and SPTLC2 genes were subcloned in pRset vector and expressed in E. coli BL21 (DE3)pLys cells. Eucaryotic SPT is a membrane-bound heterodimer enzyme, while all other members are soluble homodimer enzymes. cDNA library were obtained from total RNA from human embryo kidney cell line, HEK293, using RT-PCR and PCR with specific primers was carried out for preparing SPTLC1 and SPTLC2 genes. pRset vector which can express hexahistidine-tag fusion protein was used and the DNA sequences of pRsetB/SPTLC1 and pRsetA/SPTLC2 were confirmed. Recombinant BL21 cells with SPTLC subunits were selected with LB plate containing ampicillin and chroramphenicol. SPTLC1 and SPTLC2 proteins were induced with 1 mM IPTG and seperated on 10% SDS-PAGE gel. Expressed proteins were confirmed by western blotting with His-tag antibody.
