Effects of Houttuynia Cordata thunb on the liver damage of TCDD-treated rats (TCDD를 투여한 rat의 간손상에 대한 어성초의 효과)

  • Houttuynia Cordata thunb has been used as folk medicine for analgesics, beriberi, edema, hepatitis and icterus etc. We investigated, the effects of Houttuynia Cordata thunb administration on protective in liver of 2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD) treated rats. Seven days after the injection of TCDD(1${\mu}g$/kg), Houttuynia Cordata thunb (200mg/kg) was administered into rats intraperitoneally for four weeks. We examined the antioxidative enzymatic activity by measuring the level of GOT, GPT in serum and MDA, GSH, GSSG, GPx, SOD and Catalase in liver tissue of rats. GOT activity of Houttuynia Cordata thunb and TCDD administered group(HTT) showed 49.00% of inhibitive effect compared to TCDD-treated abnormal group(TTA). GPT level of HTT group was decreased to the level of Non TCDD-treated group(NTT). MDA content in the TTA group was 1.27 times increased compared to NTT group. HTT group was inhibited by 69.53% compared to TTA group. GSH contents in HTT group was 1.91 times increased compared to TTA group. GSSG contents in HTT group was 46.72% decreased compared to TTA group. SOD and Catalase in TTA group were lower than in NTT group, but SOD and Catalase in HTT group were increased by 82% and 55.45% respectively compared to TTA group.

Quality characteristics of noodles added with Houttuynia cordata Thunb. powder (어성초 분말을 첨가한 국수의 품질특성)

  • The purpose of this study was to evaluate appropriate concentrations of Houttuynia cordata Thunb. powder, which has various functions, to dried wheat flour noodles. The viscosity of the composite flour and cooking quality of the noodle samples were measured, and sensory evaluations were conducted in this study. When measured by the amylograph, the composite flour had lower viscosity in the gelatinization points, maximum viscosity, and viscosity at a temperature of $95^{\circ}C$ than those of control. However, the gap between the maximum viscosity and viscosity after 15 minutes at a temperature of $95^{\circ}C$ was not great among all of the samples. While cooked noodles containing H. cordata Thunb. powder had lower weight and lower volume than those of control, the weight of two cooked noodle samples with 0.5% and 1.5% of H. cordata Thunb. powder was insignificantly different. Cooked noodles had higher concentrations of H. cordata Thunb. powder; therefore, the L value of cooked noodles decreased, while a and b color values of the noodles increased. Color, flavor, taste, and overall acceptability of the samples with H. cordata Thunb. powder were lower than those of wheat flour noodles via sensory evaluation. In the overall acceptability, however, samples with 0.5% and 1.5% of H. cordata Thunb. powder were insignificantly different from control. As a result, it was finally suggested that optimum level of the H. cordata Thunb. powder in the product was below 0.5%.

Anti-allergic Activity of the Extracts from Houttuynia cordata Thunb Fermented by Lactic Acid Bacteria (어성초(Houttuynia cordata Thunb) 유산균 발효물의 항알러지 활성)

  • This study was performed to evaluate the possibility of application of lactic acid bacteria fermentation to increase the anti-allergic activity of the extracts from Houttuynia cordata Thunb. H. cordata Thunb was fermented on 25, 30, 35 and $40^{\circ}C$ for 5 days by two species of lactic acid bacteria, Leuconostoc mesenteroides 4395 and Lactobacillus sakei 383. The anti-allergic activity of water extracts of H. cordata Thunb was then analyzed both before and after fermentation. Anti-allergic activity was determined in vitro assays by using 5-lipoxygenase (5-LO), cyclooxygenase-2 (COX-2) and ${\beta}$-hexoseaminidase release of RBL-2H3 cells (degranulation marker). The extracts fermented at $35^{\circ}C$ by both bacteria had the highest inhibitory activities against 5-LO, and also higher than the control, and the one fermented at $30^{\circ}C$ by both bacteria had the highest inhibitory activity against COX-2. The degranulation of RBL-2H3 cells induced by IgE-antigen complex was estimated as ${\beta}$-hexoseaminidase release rate as reference of 100%, the release rates were inhibited in $25{\mu}g/ml$ of the extracts fermented at 30, 35 and $40^{\circ}C$ only by L. mesenteroides 4395. These results indicate that anti-allergic activity of H. cordata Thunb is increased by lactic acid bacteria fermentation.

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata THUNB extracts on A549 (lung cancer), MDA-MB231 (breast cancer), SNU-C4 (colon cancer) and B16 (mouse melanoma) cell lines. We have determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The 150 $\mu$g/$m\ell$ concentration of methanol extract (63.81 %) of Houttuynia cordata THUNB was shown significantly antitoxic activity on A549 cell lines. (omitted)

Effect of drinking Houttuynia cordata Thunb extract supplement on growth performance and colony count outcomes in ICR mice

  • The purpose of this study was to investigate the safety of Houttuynia cordata Thunb extract (HCE) in Institute of Cancer Research (ICR) mice, to determine the effects of the extract on the growth performance and colony count, and to establish the optimal concentration of HCE. In total, 60 five-week-old male ICR mice with an average initial body weight (BW) of 27.24 ± 0.44 g were used in a four-week experiment. Mice were randomly allotted to four treatment groups (five replications per group, three mice per cage): 1) a control (CON) group fed with normal distilled water; 2) treatment group 1 (T1) fed with normal distilled water containing 0.05% HCE; 3) treatment group 3 (T3) fed with normal distilled water containing 0.1% HCE; and 4) treatment group 3 (T3) fed with normal distilled water containing 0.2% HCE. BW, feed intake (FI), and water intake were measured on the first, fourteenth, and eighteenth days. T2 showed a significant improvement (p < 0.05) in the feed conversion ratio (FCR) over the experimental period. However, water intake levels did not show significant differences among the groups. In the large intestine and feces, E. coli and Lactobacillus levels were significantly improved (p < 0.05) in the HCE treated group compared to the CON group. Supplying HCE via the drinking water improved the growth performance and colony count in ICR mice. Based on results of this study, utilizing HCE in livestock species is expected to be safe and feasible.

Increased Flavonoid Compounds from Fermented Houttuynia cordata using Isolated Six of Bacillus from Traditionally Fermented Houttuynia cordata

  • Flavonoids, which form a major component in Houttuynia cordata Thunb., display a wide range of pharmacological activities. The expression of plant flavonoids is partly regulated by fermentation. Therefore, we studied the effects of fermentation on H. cordata in order to identify the strains present during the fermentation process, and to determine whether fermented H. cordata could be used as a probiotic. Our results showed that all 6 of the bacterial strains isolated from fermented H. cordata (FHC) belonged to the genus Bacillus. As expected, fermenting H cordata also increased the flavonoid content as increases were observed in the levels of rutin, quercitrin, and quercetin. To test the effects of fermentation, we treated LPS-stimulated RAW264.7 cells with non-fermented H. cordata extracts (HCE) or FHC extracts (FHCE). Compared to the HCE-treated cells, the FHCE-treated cells showed increased viability. No cytotoxic effects were detected in the FHCE-treated groups in the 2 cell lines used in the study, namely, RAW264.7 and RBL-2H3. FHCE-treated HepG2 cells showed decreased growth, compared to HCE-treated HepG2 cells. These results indicate that the fermented H. cordata predominantly contained Bacillus strains. Furthermore, FHCE are able to prevent LPS-induced inflammatory effects and inhibit the growth of HepG2 cells.

Polyphenolic Profile of Fermented Houttuynia cordata Thunb. and Overall Contribution to Antioxidant and Lipolytic Activities

  • Polyphenol profiles, physicochemical properties, antioxidant activities, and inhibitory effect of adipocyte differentiation of Houttuynia cordata fermented with Lactobacillus brevis B84 were evaluated. Six polyphenols were characterized for this plant by using high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), and the results were compared with total phenolic content by a spectrophotometric method. The total amount of the identified polyphenols was lower than that determined by the spectrophotometric method. However, the fermentation process influenced polyphenol composition such as content of vanillic acid and caffeic acid. The phytochemical profiles were evaluated by high-performance liquid chromatography with UV and electrospray ionization mass spectrometry detection ($HPLC-DAD-ESI-MS^n$). Total sugar and reducing sugar contents decreased after fermentation. Antioxidant activities such as DPPH, ABTS, and superoxide anion radical scavenging and reducing power were evaluated to compare the beneficial effect after fermentation. Fermented H. cordata increased the lipolytic effect in 3T3-L1 adipocytes. Overall, the results indicate that the fermentation of H. cordata with L. brevis B84 produces changes of phenolic compounds, antioxidant activity, and lipolytic effect.

Effects of Houttuynia cordata Thunb on Lipidperoxide and Cholesterol in 2,3,7,8-TCDD-damaged Rats (2,3,7,8-tetrachlorodibenzo-p-dioxin(TCDD)로 처치된 흰쥐에서 어성초가 과산화지질 및 콜레스테롤에 미치는 영향)

  • TCDD, one of the notorious toxic environmental pollutants, damages various organs including liver and is regarded as an endocrine disrupter. To investigate the effects of Houttuynia cordata Thunb (HCT) on the biochemical parameters of function, liver and serum of TCDD-treated rats were used. Seven days after the injection of TCDD (1 $\mu\textrm{g}$/kg), HCT (200 mg/kg) was administered to rats on every other day for four weeks. The lipidperoxide activity was examined by measuring the level of total cholesterol, HDL-cholesterol, LDL-cholesterol, total lipid and triglyceride (TG) in serum, and malondialdehyde (MDA) in liver tissue of rats. Result showed that lipidperoxidation was inhibited In the significant level when 2,3,7,8-TCDD-damaged rats were treated with HCT.
