Exosomes Secreted by Toxoplasma gondii-Infected L6 Cells: Their Effects on Host Cell Proliferation and Cell Cycle Changes

  • Toxoplasma gondii infection induces alteration of the host cell cycle and cell proliferation. These changes are not only seen in directly invaded host cells but also in neighboring cells. We tried to identify whether this alteration can be mediated by exosomes secreted by T. gondii-infected host cells. L6 cells, a rat myoblast cell line, and RH strain of T. gondii were selected for this study. L6 cells were infected with or without T. gondii to isolate exosomes. The cellular growth patterns were identified by cell counting with trypan blue under confocal microscopy, and cell cycle changes were investigated by flow cytometry. L6 cells infected with T. gondii showed decreased proliferation compared to uninfected L6 cells and revealed a tendency to stay at S or G2/M cell phase. The treatment of exosomes isolated from T. gondii-infected cells showed attenuation of cell proliferation and slight enhancement of S phase in L6 cells. The cell cycle alteration was not as obvious as reduction of the cell proliferation by the exosome treatment. These changes were transient and disappeared at 48 hr after the exosome treatment. Microarray analysis and web-based tools indicated that various exosomal miRNAs were crucial for the regulation of target genes related to cell proliferation. Collectively, our study demonstrated that the exosomes originating from T. gondii could change the host cell proliferation and alter the host cell cycle.

  • Alteration of genetic make-up of the isolates and mono-sporidial strains of Tilletia indica causing Karnal bunt (KB) disease in wheat was analyzed using DNA markers and SDS-PAGE. The generation of new variation with different growth characteristics is not a generalized feature and is not only dependant on the original genetic make up of the base isolate/monosporidial strains but also on interaction with host. Host determinant(s) plays a significant role in the generation of variability and the effect is much pronounced in monosporidial strains with narrow genetic base as compared to broad genetic base. The most plausible explanation of genetic variation in presence of host determinant(s) are the recombination of genetic material from two different mycelial/sporidia through sexual mating as well as through parasexual means. The morphological and development dependent variability further suggests that the variation in T. indica strains predominantly derived through the genetic rearrangements.

  • Insertions of transposable elements in or near a structural gene give rise to null phenotypes, reduced levels of gene expression, or alteration on the tissue-specific pattern of gene expression. Null phenotypes often result from insertions in exons. Reduced levels of gene expression results from insertions in various regions such as promoter region, 5' non-translated region, exon and intron. The maize allele of Adh1-3F1124 is an example of alteration in the tissue-specific patetern of gene expression. Adh1-3F1124 contains a Mu element inserted 31 bp 5' to the transcriptional start site of the wild-type Adh1 activity in seeds and anaerobically-treated seedlings but normal levels in the pollen. Upon the insertion of a transposable element a certain number of host DNA sequences at the insertion site is duplcated. When transposable elements excise, all element sequences are deleted. However, the duplicated host sequences may be left intact or deleted to various extents. This results in null phenotypes, restoration of original levels of gene expression, or altered levels of gene expression. On the basis of effects of transposable-element insertions or excisions on gene expression, the usefulness of transposable ellements for studies on gene expression is discussed.

Ultrastructure of the Rust Fungus Puccinia miscanthi in the Teliospore Stage Interacting with the Biofuel Plant Miscanthus sinensis

  • Interaction of the the rust fungus Puccinia miscanthi with the biofuel plant Miscanthus sinensis during the teliospore phase was investigated by light and electron microscopy. P. miscanthi telia were oval-shaped and present on both the adaxial and abaxial leaf surfaces. Teliospores were brown, one-septate (two-celled), and had pedicels attached to one end. Transmission electron microscopy revealed numerous electron-translucent lipid globules in the cytoplasm of teliospores. Extensive cell wall dissolution around hyphae was not observed in the host tissues beneath the telia. Hyphae were found between mesophyll cells in the leaf tissues as well as in host cells. Intracellular hyphae, possibly haustoria, possessed electron-dense fungal cell walls encased by an electron-transparent fibrillar extrahaustorial sheath that had an electron-dense extrahaustorial membrane. The infected host cells appeared to maintain their membrane-bound structures such as nuclei and chloroplasts. These results suggest that the rust fungus maintains its biotrophic phase with most mesophyll cells of M. sinensis. Such a nutritional mode would permit the rust fungus to obtain food reserves for transient growth in the course of host alteration.

진해 납석광상의 산상과 광물학적 특성 (Geological Occurrence and Mineralogy of Pyrophyllite Deposits in the Jinhae Area)

  • 진해지역에 위치하는 납석광상에 대하여, 광산 지질조사를 통해 산출상태를 파악하고, X-선회절분석, X-선형광분석, 전자현미분석, ICP분석 등을 통하여 변질광물의 광물학적 특성을 조사하였다. 그 결과, 이 납석광상에는 일라이트, 딕카이트, 엽납석, 다이어스포어, 녹니석, 황철석, 침철석 등의 다양한 광물들이 산출되었다. 이들 구성광물의 조합을 검토한 결과, A: 석영, B: 석영+ 일라이트, C: 석영 + 일라이트 + 딕카이트, D: 엽납석 + 일라이트 + 딕카이트 + 다이어스포어의 4개 광물조합으로 주로 구분되었다. 이들 광물조합의 산출상태로 보아 규화대, 일라이트변질대, 카올린변질대, 엽납석변질대로 구분할 수 있는 것으로 나타났다. 이 규화대의 대부분은 약 90% 이상의 높은 $SiO_2$ 함량을 보이고 작은 입자의 석영들로만 거의 구성되어 있는 특징을 보였다. 엽납석은 2M형의 폴리타이프에 해당되는 것으로 나타났다. 이 광상의 모암은 유문암질암, 안산암질 응회암 및 화산 각력암 등의 화산암이며, 모암의 차이에 따라 변질대 분포양상이 다소 다르게 나타났다. 이런 화산암이 수소이온 및 실리카의 활동도가 높은 산성열수용액에 의해 변질작용을 받아 납석광상이 형성되었으며, 그중에서 엽납석변질대가 가장 고온의 환경에서 생성된 것으로 사료된다.

전남 및 경상 열수변질 점토광상의 생성환경 비교 (Comparative Anatomy of the Hydrothermal Alteration of Chonnam and Kyongsang Hydrothermal Clay Alteration Areas in Korea)

  • 전남 및 경상점토열수변질지역은 백악기 유천층군의 화산암지대내 분포한다. 전남 변질지역의 모암은 산성화산암류이며 경상 변질지역의 모암은 산성 및 중성화산암류이나 중성화산암류가 우세하다. 두 변질지역의 열수변질작용을 비교 하였을 때 중요한 차이는 열수용액의 기원으로 생각된다. 경상 열수변질지대는 마그마수가 열수의 주 기원으로서 고온성 변질광물인 엽납석이나 홍주석이 우세하게 산출되며 마그마수에서 특징적인 붕소 함유 광물인 듀모티어라이트와 전기석의 산출이 특징적이다. 이에 반해서 전남 열수변질지역은 천수와 열수의 혼합용액이 열수의 주 기원으로 천수가 중요한 역할을 하였으며, 저온성 광물인 카오린, 명반석 등이 우세하게 산출된다. 또 다른 중요한 차이는 pH 와 같은 열수용액의 화학성 차이이다. 전남 열수변질지역의 명반석-카오런-석영 변질광물군은 저온의 강산성 열수용액으로서 "산-황산염형" 으로 특징되며 이에 반해 경상 열수변질지역의 견운모-석영 변질광물군은 "석영-견운모형" 에 해당되며 고온의 중성 내지 약산성 열수용액에 의한 변질특성을 나타낸다. 또한 두 지역은 열수변질대 모암인 화산암류의 지질구조 환경에서 차이를 찾아 볼 수 있다. 전남 열수변질지역은 산성암질 돔과 성인적으로 관련되나 경상 열수 변질지역은 칼데라와 관련된 특성을 보인다.

Verification of aecial host ranges of four Gymnosporangium species based on artificial inoculation.

  • Aecial host ranges of four Gymnosporangium species causing cedar-apple rust diseases, G. asiaticum, G. cornutum, 5. japonicum and G. yamadae, were investigated through artificial inoculation. Thirteen species of nine genera among Rosaceous plants, which have been reported as social hosts in Korea, were inoculated with fresh teliospores spores in early days of May of 2000 and of 2001, respectively. In the results, we re-confirmed that there was highly specific relationship between the rust species and aecial hosts and report new aecial hosts of four Gymnosporangium species. Teliospores of G. cornutum collected from Juniperus rigida successively produced spermogonia and aecia only on Sorbus alntifolia, the first report on host alteration of G. cornutum in Korea. Positive responses by teliospores of G. japonicum from J. chinenis of Suwon and from J. chinenis var. horizontalis of Jeju island were obtained only on P. villosa. Crataegus pinnatifida was confirmed as a new aecial host of G. viatium. Until this time, G. ymadae was believed to have Malus as the aecial host. However, teliospores of G. yamadae collected from J. chinensis var. kaizuka successively formed spermogonia and aecia on the leaves of Chaenomeles lagenaria, C. sinensis, Pyrus pyrtifolia var, culta, P. ussuriensis, Malus pumila and M. sileboldii. The date for maturation of spermogonia and aecia, and symptom development varied according to the rust fungi and aecial host plants, respectively.

충남 광천지역 석면의 산출 유형 및 광물학적 특징 (Occurrence types and mineralogical characteristics of asbestos for the Kwangcheon area, Chungnam)

  • This study is to characterize the occurrence types and mineralogical characteristics of asbestos for the Kwangcheon areas, Chungnam. The mine areas had been exploited as asbestos mines for several decades since 1930. Host rocks of the asbestos are serpentinites and altered rocks of the ultramafic rocks. Representative samples of the host rocks and minerals were sampled and were examined with microscopes. To confirm for the existences and compositions for the asbestos, the rock samples were analysed with EPMA, XRD and EDS. Chrysotile, tremolite and actinolite were found as asbestos and non-asbestos forms in these areas. Chrysotiles, as non-asbestos forms, occur in the host rocks with mesh and hourglass textures. They, as asbestos forms, are mainly found as the veins. The tremolite and actinolite, as asbestos types, occur as alteration products of the olivine and pyroxene within the host rocks. They, as asbestos types, are also found following the cracks and fractures. Overall results suggest that three types of the asbestos are found in the Kwangcheon and Kaewol mine areas. Based on the occurrence types for the asbestos, additional studies are required for the asbestos in the top soil and air.

  • In this study, Tri(1-phenylpyrazolato)iridium $(Ir(ppz)_3)$ was prepared for the pure blue phosphorescent dopant and various host materials were used for the appropriate energy alignment. Although the luminance was pure blue with the CIE coordinates of x = 0.158, y = 0.139, device efficiencies didn't improve yet. Instead of finding the proper host materials, the alteration of structure of OLEDs affected the improvement of electrical and optical characteristics of the devices. It was worthy that insertion the exciton formation zone with the host material between the emitting zone and the exciton blocking layer. The device with a structure of ITO/NPB/Ir(ppz)3 doped in CBP/CBP for the exciton formation zone/BCP/Liq/Al was fabricated and the characteristics were observed compared with the devices without the exciton formation zone. When CBP was used for the exciton formation zone, the device efficiency reached to over 0.25 cd/A. While the device used CBP only for the host showed the luminous efficiency of under 0.11 cd/A

가평 사문암 광산의 석면 산출의 산상과 모암의 특성 (Occurrences of Asbestos within Gapyeong Serpentinite Mines and Characteristics of Host Rocks)

  • Objectives: This study is for characteristics of asbestos occurrence (NOA, naturally occurring asbestos) from the Gapyeong area and its host rocks, serpentinites. Methods: Representative samples are collected from the serpentinite bodies, following degrees of hydrothermal alteration and metamorphism, after about 2 year field trips. Mineralogical, morphological and optical characteristics of the asbestos and host rocks are confirmed by PLM, XRD EPMA and SEM results. Results: The serpentinites are dunites and harzburgites, and host asbestos, including chrysotile, tremolite and actinolite. The asbestos chrysotiles are found as veins ranging from several millimeters to several centimeters in thickness, while asbestos-tremolite and -actinolite occur along cracks and fractures ranging up to ten centimeters in thickness. The chryostiles occur mainly as cross and slip fibers, while the amphibole asbestos is found as vein, slip and oblique fibers. More tremolitic grains are colorless and commonly show elongated or fiber shapes, whereas the magnesio hornblende grains mainly show light green and occur as subhedral to euhedral diamond grains. Conclusions: Overall characteristics of serpentinites from the Gapyeong area are similar to worldwide orogenic-related Alpine type ultramafic rocks serpentinized and serpentinites in South Chungcheong-do Province, Korea, and occurrences of asbestos are similar to those of the ultramafic bodies in South Chungcheong-do Province.
