말에서 감염의 조기진단을 위한 haptoglobin 지표 연구 (Haptoglobin as an early indicator of infection in horses)

  • The objective of present study was to assess the diagnostic value of the concentration of haptoglobin, one of a series of acute phase proteins that is found in the blood of both humans and animals. A total of 79 random horse sera(10 dam and 69 foals) with clinically normal were analyzed by ELISA. The mean serum concentrations of haptoglobin of dam and foals were 1.67${\pm}$0.52mg/ml(range 1.08∼2.12 mg/ml), 1.65${\pm}$0.50 mg/ml(range 0.50∼2.66 mg/ml), respectively. These results may provide basic information for developing a useful marker system to monitor infections early in horses.

Microsatellite DNA형 분석을 이용한 더러브렛 말의 친자감정 (Parentage Testing for Thoroughbred Horse by Microsatellite DNA Typing)

  • The objective of present study was to ascertain parentage of Thoroughbred(TB) horses in Korea. A total of 2,029 TB horse samples including 993 foal samples for parentage testing were genotyped for nine international minimum standard markers(AHT4, 5, ASB2, HMS3, 6, 7, HTG4, 10, and VHL20). This method consisted of multiplexing PCR procedure, and showed reasonable amplification of all PCR products. Genotyping was performed with an ABI 310 genetic analyzer. The number of alleles per locus varied from 5 to 11 with a mean value of 7.33 in TB. Expected heterozygosity was ranged from 0.544 to 0.837(mean 0.709) and the total exclusion probability of 9 microsatellites loci was 0.9978. Of the 9 markers, ASB2, HMS7 and HTG10 loci have relatively high PIC value(>0.7). All of the 993 foals were qualified by compatibility according to Mendelian fashion in the present DNA typing for parentage testing. These results suggest that the present DNA typing has high potential for parentage verification of TB horses.

사람 및 동물에 대한 소 엔테로바이러스 항체 분포 조사 (Existence of antibodies against bovine enterovirus in humans and various animals in Korea)

  • Bovine enteroviruses (BEVs) were separated into two groups, BEV-1 and BEV-2. BEVs, found in cattle worldwide, usually cause asymptomatic infections and are excreted in the feces of infected animals. Antibodies against BEV have been found in different species including human, cattle, sheep, goats, dogs, horses and monkeys in the world. This study aimed to investigate prevalence of the neutralizing antibodies for BEVs in human and animals in Korea. Antibodies against BEV-1 in humans, cattle, pigs, goats, horses and dogs were shown to be 46.8%, 48.3%, 70.6%, 11.5%, 11.5% and 6.3% respectively. Also, antibodies against BEV-2 were shown to be 98.7%, 68.1%, 89.2%, 59.4%, 9.4% and 96.9% respectively. We found that the neutralizing antibodies against these viruses are common in Korea. The prevalences of antibodies against BEV-1 were lower than those against BEV-2 in humans and in all animals except horses. These results showed that the BEV is considered endemic in cattle in many regions in Korea.

  • This study was carried out to investigate a usefulness of the microsatellite DNA markers for parentage verification of Thoroughbred (TB) horses. 9 TB horses samples were genotyped for nine international minimum standard markers (AHT4, 5, ASB2, HMS3, 6, 7, HTG4, 10, and VHL20), and the additional panel of four markers, ASB17, CA425, LEX33, and TKY321. This methods consisted of multiplexing PCR procedures, and it showed reasonable amplification of all PCR products. Genotyping was performed with an ABI 310 genetic analyzer. Foal I was excluded according to principles of Mendelian genetics in AHT4 (H/K), ASB2 (Q/Q), HMS3 (I/P), HTG4 (M/O), HTG1O (K/R), VHL20 (M/P), ASB17 (F/N), LEX33 (M/O), and TKY321 (G/I) markets. Foal II was excluded with markers AHT5 (K/M), ASB2 (M/N), HMS7 (N/N), HTG1O (K/K), VHL20 (I/I), ASB17 (F/F) and TKY321 (G/I). Foal III was excluded with markers AHT4 (O/O), AHT5 (K/K), ASB2 (M/R), HMS6 (M/P), HMS7 (O/O), HTG10 (R/S), VHL20 (L/M), and ASB17 (N/O). These results suggest that the present DNA typing is so useful for parentage verification of TB horses.

Diaphragmatic hernia in a Jeju horse (crossbred) broodmare

  • Diaphragmatic hernias, whether congenital or acquired (traumatic), are rarely observed in the horse. Acquired diaphragmatic hernias typically occur secondary to trauma or an increase in intraabdominal pressure due to falling, heavy exercise, or parturition. Diaphragmatic herniorrhaphy is difficult to perform in adult horses and the horses with symptomatic diaphragmatic hernias usually die. A 10- year old, 340 kg, Jeju horse (crossbred) broodmare with sudden onset of gait disorder and a moderate emaciation was examined. Findings on physical examination included conjunctivitis, dehydration, shallow breathing, dyspnea, weaken heart beat, lack of auscultatable sounds from the gastrointestinal tract, and anorexia. Rectal temperature was $38.4^{\circ}C$ and respiratory rates were moderately increased. There were slight signs of acute colic. The broodmare died one day after non-specific treatment of fluids, nutriment, antibiotics and non-steroidal anti-inflammatory drug. The cause of death was strangulation of the small intestine through a diaphragmatic hernia. The rent was about 2 cm in diameter and located in the central right part of diaphragm. Around 60 cm of small intestine was protruded into thoracic cavity through the rent. The cause of the hernia could not be ascertained. The broodmare had been pastured with many other horses, and the groom had not noticed any aggressive behavior among them. It was, however, speculated that trauma by stallion's attack may have been the cause of the diaphragmatic hernia, because the new horse may be the object of behaviors ranging from mild threats to seriously aggressive kicking, squealing, rearing, and biting.

호이산화탄소성 Streptococcus equi subspecies equi에 의한 선역의 발생 (Outbreaks of Strangles due to Capnophilic Streptococcus equi subsp equi in South Korea)

  • 이 연구는 더러브렛 말에서 호이산화탄소성 Streptococcus equi subsp equi 에 기인한 선역의 발생에 관한 보고이다. 3개의 다른 농장에서 선역의 전형적인 증상인 림프절 화농이 있거나 없는 29두의 말에서 17 균주의 S equi subsp equi를 분리하였다. 이들 중 임상증상이 있는 1두에서 분리한2균주는 호기배양에서 분리되었으나, 임상증상이 있는 6두에서 분리한 7균주와 임상증상이 없는 22두에서 분리한 8균주는 호기조건에서 배양되지 않았다. 이 15 분리균주는 일반 호기배양기에서는 증식하지 않았으며, 5% $CO_2$배양기, 산소분압을 낮추는 candle jar 및 GasPak을 이용한 혐기배양에서 증식하는 호이산화탄소성, 산소민감성 또는 이산화탄소 요구성 S. equi subsp. equi로 판명되었다. 이 연구는 호이산화탄소성 S. equi subsp. equi 에 의한 선역발생에 관한 최초의 보고이다.

Exercise induced upregulation of glutamate-cysteine ligase catalytic subunit and glutamate-cysteine ligase modifier subunit gene expression in Thoroughbred horses

  • Objective: This study was performed to reveal the molecular structure and expression patterns of horse glutamate-cysteine ligase catalytic subunit (GCLC) and glutamate-cysteine ligase modifier subunit (GCLM) genes whose products form glutamate cysteine ligase, which were identified as differentially expressed genes in the previous study. Methods: We performed bioinformatics analyses, and gene expression assay with quantitative polymerase chain reaction (qPCR) for horse GCLC and GCLM genes in muscle and blood leukocytes of Thoroughbred horses Results: Expression of GCLC showed the same pattern in both blood and muscle tissues after exercise. Expression of GCLC increased in the muscle and blood of Thoroughbreds, suggesting a tissue-specific regulatory mechanism for the expression of GCLC. In addition, expression of the GCLM gene increased after exercise in both the blood and muscle of Thoroughbreds. Conclusion: We established the expression patterns of GCLC and GCLM in the skeletal muscle and blood of Thoroughbred horses in response to exercise. Further study is now warranted to uncover the functional importance of these genes in exercise and recovery in racehorses.

Nutritional Composition in Bone Extracts from Jeju Crossbred Horses at Different Slaughter Ages

  • The effects of slaughter age (28, 32, or 38 mon) on the proximate composition, collagen content, fatty acid composition, amino acid content, and mineral contents of horse leg bone (femur and tibia) extracts (HLBE) obtained from Jeju crossbred horses (n=14) in Korea. HLBE was extracted for 24 h with boiling water. At those ages, the respective proximate compositions of the HLBE were found as follows: 5.20-6.42% crude protein; fat 0.61-1.65% crude; and 0.10-0.22% crude ash. At 32 and 38 mon, the HLBE showed higher levels of both crude protein and collagen than at 28 mon. The major fatty acids of the horse leg bone extract powder (HLBP) were palmitic acid and oleic acid. Palmitoleic acid and the essential fatty acids were higher in the HLBP at 38 mon compared to that at 28 mon. Nearly all the amino acids were found at higher levels in the HLBP at 38 mon than at 28 mon, except histidine. The P, K, Zn, Se and Fe contents in the HLBP increased significantly with age. These results suggest that some nutrients in the HLBE increase with age, and that extracts from horses older than 32 mon would be more nutritious for human consumers.

더러브렛 말의 혈액형에 관한 연구 (Studies on blood types in Thoroughbred horses)

  • The present study was carried out to investigate the blood markers of Thoroughbred horses (TB) The blood red cell types and blood protein types (biochemical polymorphisms) were tested from 1,125 Thoroughbred horses by serological and electrophoretic procedures, and their phenotypes, gene frequencies, heterozygosity, polymorphic information content values and exclusion probability were estimated. The blood group and biochemical polymorphism phenotypes observed with high frequency were Aaf(91.7%), Ca(94.7%), K-(94.5%), Ua(75.9%), P-(50.6%), Qabc(82.6%), ALB-BB(67.7%), GC-FF(92.7%), AIB-KK(99.6%), ES-II(77.9%), TF-DF1(23.6%), PI-LL(23.2%), HB-B2B2(73.6%), PGD-FS(45.4%) and genotypes Dcgm/dk(16.9%), Dbcm/cgm(13.6%), Dbcm/dk(11.9%), Dcegmn/cegmn(10.0%), Dcgm/cgm(8.7%) in TB. Alleles observed with high frequency were Aaf(0.796), Ca(0.769), Ddk(0.266), Dcgm(0.261), Dbcm(0.211), K-(0.972), P-(0.710), Qabc(0.565), Q-(0.368), Ua(0.509), $HB^{B2}$(0.858), $PGD^F$(0.634), $ALB^B$(0.825), $GC^F$(0.927), $AIB^K$(0.998), $ES^I$(0.881), $TF^{F1}$(0.346), $TF^D$(0.319), $TF^{F2}$(0.184), $PI^L$(0.479), $PI^N$(0.214), $PI^U$(0.116) in TB. The heterozygosity, polymorphic information content (PIC) and exclusion probability (PE) were calculated. The mean heterozygosity and PIC value were 0.3899 and 0.3375, respectively. The highest heterozygosity and PIC were estimated 0.7834 and 0.7492 in blood group D locus, respectively. The cumulated PE obtained by blood groups and biochemical polymorphisms was 0.9813.

Analysis of cross-population differentiation between Thoroughbred and Jeju horses

  • Objective: This study was intended to identify genes positively selected in Thoroughbred horses (THBs) that potentially contribute to their running performances. Methods: The genomes of THB and Jeju horses (JH, Korean native horse) were compared to identify genes positively selected in THB. We performed cross-population extended haplotype homozygosity (XP-EHH) and cross-population composite likelihood ratio test (XP-CLR) statistical methods for our analysis using whole genome resequencing data of 14 THB and 6 JH. Results: We identified 98 (XP-EHH) and 200 (XP-CLR) genes that are under positive selection in THB. Gene enrichment analysis identified 72 gene ontology biological process (GO BP) terms. The genes and GO BP terms explained some of THB's characteristics such as immunity, energy metabolism and eye size and function related to running performances. GO BP terms that play key roles in several cell signaling mechanisms, which affected ocular size and visual functions were identified. GO BP term Eye photoreceptor cell differentiation is among the terms annotated presumed to affect eye size. Conclusion: Our analysis revealed some positively selected candidate genes in THB related to their racing performances. The genes detected are related to the immunity, ocular size and function, and energy metabolism.
