Monitoring of Possible Horizontal Gene Transfer from Transgenic Potatoes to Soil Microorganisms in the Potato Fields and the Emergence of Variants in Phytophthora infestans

  • To examine the possibility of horizontal gene transfer between transgenic potatoes and microorganisms in potato fields, the gene flow from transgenic potatoes containing the nucleoside diphosphate kinase 2 (NDPK2) gene to microorganisms in soils was investigated. The soil samples collected from the potato fields from March to October 2007 were examined by PCR, Southern hybridization, and AFLP fingerprinting. The NDPK2 gene from soil genomic DNAs was not detected by both PCR and Southern hybridization, indicating that gene transfer did not occur in the potato fields. In addition, no discrepancy was found in pathogenicity and noticeable changes for the appearance of variants of Phytophthora infestans in each generation when serial inoculations and the analysis of genomic DNAs by AFLP were conducted. Thus, these data suggest that transgenic potatoes do not give significant impacts on the communities of soil microorganisms and the emergence of variants, although continued research efforts may be necessary to make a decisive conclusion.

Pullout Resistance by Horizontal Spacing of Geosynthetic Strip (띠형 섬유보강재의 설치간격에 따른 인발저항 특성에 관한 연구)

  • In this study, the pullout tests are conducted to evaluate the pullout resistance of the geosynthetic strip with or without bearing resistance zone. The test results are indicated that the pullout resistance of the geosynthetic strip without bearing resistance zone is not affected by horizontal spacing. However, the horizontal spacing of reinforcement with bearing resistance zone affects the bearing resistance. In other words, it is indicated that the bearing resistance at spacing of 210mm is larger than that at spacing of 260mm. This means that the pullout strength at spacing of 210mm is larger than that at spacing of 260mm. Therefore.

  • The grating is the main purpose of drainage in civil engineering, architecture. This study examined the change and stress distribution by simulating three models of Vertical type grating structures that reduced the resistibility in respect of velocity rather than horizontal type grating and to design a grating with the optimal grating gap. The vertical type and horizontal type grating were compared in terms of the maximum stress and strain to identify the better product.

  • Dynamic analysis method is Presented for analyzing rectangular liquid storage structures excited by horizontal and vertical ground motions. The irrotational motion of invicid and incompressible ideal fluid in rigid rectangular liquid storage structures subjected to horizontal and vertical ground motions and the motion of fluid induced by structural deformation are expressed by analytic solutions. Analysis methods are obtained by applying analytic solutions of the fluid motion to finite element equation of the structural motion. The fluid-structure interaction effect is reflected into the coupled equation as added fluid mass matrix. The free surface sloshing motion, hydrodynamic pressure acting on the wall and structural behavior due to horizontal and vertical ground motions are obtained by the presented method.

  • We present for the first time the characteristics of upper atmospheric horizontal winds over the Korean Peninsula. Winds and their variability are derived using four-year measurements by the Korea Astronomy and Space Science Institute (KASI) meteor radar. A general characteristic of zonal and meridional winds is that they exhibit distinct diurnal and seasonal variations. Their changes indicate sometimes similar or sometimes different periodicities. Both winds are characterized by either semi-diurnal tides (12 hour period) and/or diurnal tides (24 hour period) from 80-100 km. In terms of annual change, the annual variation is the strongest component in both winds, but semi-annual and ter-annual variations are only detected in zonal winds.

  • Due to the oil price is increasing continuously, active researches on sources of renewable energy has been invigorated. Above all, ocean energy has high-usability because of ocean current has high density and large quantity compared to the wind energy. In this paper, efficiency enhancement of the wave power generation was described through horizontal plate installation at the break water wave power generation system that converts the ocean energy into electricity. The power-conversion efficiency can be improved by horizontal plate installation at existing system, but there has been insufficient studies domestically. The purpose of this paper is to analyze about the effects of the horizontal plate installation on the breakwater wave power generation system by wave basin experiment and to propose a position of horizontal plate installation.

Effects of Network Density on Gridded Horizontal Distribution of Meteorological Variables in the Seoul Metropolitan Area (관측망 밀도가 기상 자료의 격자형 수평 분포에 미치는 영향)

  • High-quality and high-resolution meteorological information is essential to reduce damages due to disastrous weather phenomena such as flash flood, strong wind, and heat/cold waves. There are many meteorological observation stations operated by Korea Meteorological Administration (KMA) in Seoul Metropolitan Area (SMA). Nonetheless, they are still not enough to represent small-scale weather phenomena like convective storm cells due to its poor resolution, especially over urban areas with high-rise buildings and complex land use. In this study, feasibilities to use additional pre-existing networks (e.g., operated by local government and private company) are tested by investigating the effects of network density on the gridded horizontal distribution of two meteorological variables (temperature and precipitation). Two heat wave event days and two precipitation events are chosen, respectively. And the automatic weather station (AWS) networks operated by KMA, local-government, and SKTechX in Incheon area are used. It is found that as network density increases, correlation coefficients between the interpolated values with a horizontal resolution of 350 m and observed data also become large. The range of correlation coefficients with respect to the network density shows large in nighttime rather than in daytime for temperature. While, the range does not depend on the time of day, but on the precipitation type and horizontal distribution of convection cells. This study suggests that temperature and precipitation sensors should be added at points with large horizontal inhomogeneity of land use or topography to represent the horizontal features with a resolution higher than 350 m.

Differential Horizontal Stress Ratio for Danyang Limestone with Vertical Transversely Isotropy (횡적등방성 특성을 갖는 단양 석회암의 수평응력차비 고찰)

  • To develope shale play which is one of unconventional energy resources, horizontal drilling and hydraulic fracturing are necessary and those are applied to the place where the differential horizontal stress ratio (DHSR) is low. The differential horizontal stress ratio is generally calculated by the minimum and maximum horizontal stress, but it is also calculated from dynamic elastic constants and anisotropic parameters. In this study we analyzed anisotropic properties through the core samples from Danyang limestone and calculated DHSR. The three types of core samples shaped in three directions (vertical, parallel and 45 degree to bedding) were used for laboratory test. We measured P-, S-wave velocities, and density and then calculated dynamic elastic constants, compliance and DHSR. According to the results of the core sample analysis the calculated DHSR is 0.185. Thomsen parameters of the Danyang limestone used in this study are characterized by the P- and S-wave velocities varying along the bedding symmetry axis. It is observed that the DHSR value is more affected by the change in compliance value than the Poisson's ratio. It is necessary to measure SH-wave velocity for more correct petrophysical properties.

High-temperature ultrasonic thickness monitoring for pipe thinning in a flow-accelerated corrosion proof test facility

  • In order to monitor the pipe thinning caused by flow-accelerated corrosion (FAC) that occurs in coolant piping systems, a shear horizontal ultrasonic pitch-catch waveguide technique was developed for accurate pipe wall thickness monitoring. A clamping device for dry coupling contact between the end of the waveguide and pipe surface was designed and fabricated. A computer program for multi-channel on-line monitoring of the pipe thickness at high temperature was also developed. Both a four-channel buffer rod pulse-echo type and a shear horizontal ultrasonic waveguide type for high-temperature thickness monitoring system were successfully installed to the test section of the FAC proof test facility. The overall measurement error can be estimated as ${\pm}10{\mu}m$ during a cycle from room temperature to $200^{\circ}C$.

  • This paper presents the method to provide a fast service for user in image manipulation such as zooming and panning of huge size high resolution satellite image (e.g. Giga bytes per scene). The proposed technique is based on the hierarchical structure that has 3D-Tiling in horizontal and vertical direction to provide the image service more effectively than 2D-Tiling technique in the past does. The essence of the proposed technique is to create tiles that have optimum level of horizontal as well as vertical direction on the basis of current displaying area which changes as user manipulates huge image. So this technique provides seamless service, and will be very powerful and useful for manipulation of images of huge size without data conversion.
