The Effects of Hominis Placenta Herbal-Acupuncture Solution on the Alzheimer's Disease Model Induced by ${\beta}A$ (자하차(紫河車) 약침(藥鍼)이 ${\beta}A$로 유도(誘導)된 Alzheimer's Disease 병태(病態) 모델에 미치는 영향(影響))

  • Objective: Hominis Placenta is used in many cure, mainly treats a weak, chronic disease, especially senile. This research investigates the effect of the Hominis Placenta Herbal-Acupuncture Solution on Alzheimer's disease. Method: The effects of the Hominis Placenta Herbal-Acupuncture Solution on (1) $IL-1{\beta}$ protein, $TNF-{\alpha}$ protein, MDA, and CD68/CD11b (2) the behavior (3) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with 13A were investigated. Results: 1. For the Hominis Placenta Herbal-Acupuncture Solution group a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by ${\beta}$ A in the Morris water maze experiment, which measured stop-through latency, and distance movement-through latency. 2. The Hominis Placenta Herbal-Acupuncture Solution group suppressed the over-expression of $IL-1{\beta}$ protein, $TNF-{\alpha}$ protein, MDA, and CD68/CD11b, in the mice with Alzheimer's disease induced by ${\beta}A$. 3. The Hominis Placenta Herbal Acupuncture Solution group reduced the infarction area of hippocampus, and controlled the injury of brain tissue in the mice with Alzheimer's disease induced by ${\beta}A$. 4. The Hominis Placenta Herbal-Acupuncture Solution group reduced the Tau protein, GFAP protein, and presenilin1/2 protein, beta-secretase protein, (immunohistochemistry) of hippocampus in the mice with Alzheimer's disease induced by ${\beta}A$. Conclusion: These results suggest that the Hominis Placenta Herbal-Acupuncture Solution group may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the Hominis Placenta Herbal-Acupuncture Solution for Alzheimer's disease is suggested for future research.

  • This experiment was designed to investigate the effect of Mominis Placenta herbal acupuncture solution(HP-HAS) on immune cells and cytokines in murine asthma model. In vivo C57BL/6 mice were sensitized and challenged with OVA for 12 weeks. The experimental group was treated with Hominis Placenta herbal acupuncture solution(HP-HAS) at P'yesu(BL13) for the later 8 weeks(3 times a week) and analyzed by ELISA, flow cytometer. The results were obtained as follows Eosinophils in BALF(bronchoalveolar lavage fluid) of HP-HAS group decreased significantly compared with that of control group. IL-4, IL-5, IL-13, IgE in BALF of HP-HAS group decreased significantly compared with that of control group. Number of $CD3e^-/CCR3^+$, $CD69^+/CD3e^+$, $CD11b^+/Gr-1^+$ cells in the HP-HAS group decreased compared with that of control group.

A Pilot Study on Single-dose Toxicity Testing of Hominis placenta Pharmacopuncture in Sprague-Dawley Rats

  • Objectives: This study was performed to analyze the toxicity and to find the lethal dose of the test substance Hominis placenta pharmacopuncture when used as a single-dose in 6 week old, male and female Sprague-Dawley (SD) rats. Methods: All experiments were conducted at Biotoxtech (Chungwon, Korea), an institution authorized to perform non clinical studies, under the regulations of Good Laboratory Practice (GLP). SD rats were chosen for the pilot study. Doses of Hominis placenta pharmacopuncture extracts, 0.125, 0.25 and 0.5 mL, were administered to the experimental group, and 0.5 mL doses of normal saline solution were administered to the control group. This study was conducted under the approval of the Institutional Animal Ethics Committee. Results: No deaths or abnormalities occurred in any of the groups. Also, no significant changes in body weights were observed among the groups, and no significant differences in hematology/biochemistry, necropsy, and histopathology results were noted. Hematologically, some changes in the male rats in two experimental groups were observed, but those changes had no clinical or toxicological meaning because they were not dose dependent. Histopathological tests on the injected parts showed cell infiltration in the male rats in one of the experimental groups; however, that result was due to spontaneous generation and had no toxicological meaning. Therefore, this study showed that Hominis placenta pharmacopuncture had no effect on the injected parts in terms of clinical signs, body weight, hematology, clinical chemistry, and necropsy. Conclusion: As a result of single-dose tests of the test substance Hominis placenta pharmacopuncture in 4 groups of rats, the lethal dose for both males and females exceeded 0.5 mL/animal. Therefore, the above findings suggest that treatment with Hominis placenta pharmacopuncture is relatively safe. Further studies on this subject are needed.

Microarray Analysis of Gene Expression in Raw 264.7 Cells Treated with Hominis Placenta Herbal-Acupuncture Solution (자하거(紫河車) 약침액(藥鍼液)이 Lipopolysaccharide로 처리된 RAW 264.7 대식세포주(大食細胞柱)의 유전자(遺傳子) 발현(發顯)에 미치는 영향(影響))

  • Hominis Placenta has a broad array of clinical applications in Korean medicine, including treatment of inflammatory conditions such as rheumatoid arthritis. The purpose of this study is to explore the global gene expression profiles in human RAW 264.7 cell lines treated with Hominis Placenta herbal-acupuncture solution (HPHAS) using microarray analysis. The RAW 264.7 cells were treated with lipopolysaccharide (LPS), HPHAS, or both. Of the 8,170 genes profiled in this study, with a cut-off level of two-fold change in the expression, 72 genes (CTD1, regulating synaptic membrane exocytosis 2, etc.) were upregulated and 135 genes(splicing factor, arginine/serine-rich 1, actinin, alpha 1, etc.) downregulated following LPS treatment. One gene (acrosin) was upregulated and 12 genes (phospholipase A2, group IB, neurofilament, heavy polypeptide 200kDa, etc.) were downregulated following HPHAS treatment. Eleven genes (RAB27A, member RAS oncogene family, eosinophil peroxidase, etc.) were upregulated and 16 genes (V-maf musculoaponeurotic fibrosarcoma oncogene homolog G (avian), RW1 protein, etc.) were downregulated following co-stimulation of HPHAS and LPS. It is thought that microarrays will play an ever-growing role in the advance of our understanding of the pharmacological actions of HPHAS in the treatment of arthritis. Further studies, however, are required to concretely prove the effectiveness of HPHAS.

Scavenging Effect of Hominis Placenta Herbal Acupuncture Solution on Nitric Oxide (자하거 약침액의 Nitric Oxide에 대한 소거 효과)

  • 목적 : 본 연구는 최근 임상에서 많이 사용하는 자하거 약침액의 nitric oxide(NO)에 대한 소거 효과를 분석하기 위하여 실행되었다. 방법 : 양성대조군으로 비타민C와 실험군으로 자하거 약침액에 NO를 분비하는 S-nitroso-N-acetylpenicillamine(SNAP)을 투여한 후 NO의 농도를 540nm 파장의 자외선 흡수량을 측정하여 평가하였다. 결과 : 실험에 사용된 자하거 약침액의 NO 소거 효과는 강력한 항산화제인 비타민C보다 우수하지는 못하였으나 0.005 및 0.001mg/ml 농도에서 12시간 동안 SNAP를 투여한 후 생성된 NO를 유의성 있게 소거하는 효과가 나타났다. 결론 : 이상의 결과를 통하여 자하거 약침액이 NO를 소거하는 효과가 있는 것이 확인이 되었으나 추후 농도별 실험과 다른 시료와의 비교실험이 더 요구된다.

  • Rheumatoid arthritis (RA) is a chronic inflammatory autoimmune disease. characterized by leukocyte infiltration, a chronic inflammation of the joint, a pannus formation and the extensive destruction of the 3Iticular caJ1ilage and bone. Several proinflammatory cytokines such as tumor necrosis factor-${\alpa}$(TNF-${\alpa}$), interleukin-1${\beta}$ (IL-1${\beta}$) and interleukin 6 (IL-6) have been implicated in the pathological mechanisms of synovial tissue proliferation, joint destruction and programmed cell death in rheumatoid joint. In the Korean traditional medicine, Hominis placenta (HP) as an herbal solution of herb-acupuncture has been widely used to treat the inflammatory diseases including RA. In order to study the medicinal effect of HP herb-acupuncture on rheumatoid joint, an adjuvant-induced arthritis (AlA) was generated by the injection of 1.5 mg uf Mycobactelium tuberculusis. emulsified in squalene, 10 the base of the tail of Sprague-Dawley(SD) rats. After onset stage of polyarthritis, HP was daily injected to the Zusanti (ST36) acupuncture points in both of rat lags and the expression pattems of cytokines such as TNF-{\alpa}$, IL-1${\beta}$, and 1L-6 at the knee joint were analyzed using immunostaining and RT-PCR. The HP herb-acupuncture was found to be effective to alleviate the arthritic symptums in adjuvant-induced arthritic rats as regards the joint appearance and the expression profiles of inflammatory cytokines. In conclusion, therapeutic effects of HP herb-acupuncture on the rat with AlA might be related to anti inflammatory activities of the hurb-acupuncture.
