Latitude and seasons influence the prevalence of Theileria orientalis and affect the hematology of non-grazed dairy cows in Korea

  • This pilot study aimed to investigate the effects of regional and seasonal variations on the prevalence of Theileria orientalis and the hematological profile of non-grazed dairy cows in Korea. A total of 365 clinically healthy lactating Holstein Friesian cows from 26 dairy farms in 7 provinces that were categorized into northern, central, and southern regions were sampled during the warm period from July to August and the cold period from October to December. The detection of T. orientalis major piroplasm surface protein gene and the hematology non-grazed dairy cows were analyzed using peripheral blood samples. The T. orientalis prevalence was 20.0% (73/365). The prevalence in the southern region was 35.9%, which was significantly higher than that in the central (21.6%) and northern (12.9%) regions (P<0.05). The prevalence during warm period was higher (43.0%) than that during the cold season (13.5%). The infected cows showed significantly lower erythrocyte counts in the southern region (5.8±0.6 M/㎕) and during the warm period (5.8±0.7 M/㎕) compared with those in the central and northern regions and during the cold season, which affected the extended RBC parameters, including hematocrit and hemoglobin concentrations. Our findings revealed the prevalence of T. orientalis in Korea, highlighting its high occurrence during warm periods and in certain geographical regions. Climatic factors could contribute to the health and productivity of cattle, as evidenced by the prevalence of T. orientalis and its negative impact on animals.

EFFECT OF BREEDING LENGTH ON GENETIC IMPROVEMENT IN JAPANESE HOLSTEIN POPULATION

  • The effect of breeding length of sire on genetic progress was examined in the Holstein dairy cattle population in Japan. Genetic progress was extimated by gene flow method. Breeding length of sires directly influences the replacement rates of sires and the selection intensity of sires because there are a fixed number of progeny tested young bulls per year. As breeding length of sires increased, rate of gene flow decreased and average proportions of genes deriving from selected animals had lower asymptotic values. When breeding length was short, average proportions of genes required a longer period to converge to asymptotic values. Changes of Rcow-sire's(sire to breed recorded cows) and Ncow-sire's(sire to breed non recorded cows) breeding length influenced not only transmission of their genes but also that of genes derived from all other selected animals. Irrespective of whether the discount rate was assumed to be 0 or 6%, longer term (${\geq}$ 20 years) expected total genetic improvement was maximized by a sire breeding length of five years. For shorter term assessment(10 years), genetic improvement was maximized by a sire breeding length of three years. There was a linear increase in the contribution of the sire to bulls pathway to the total genetic improvement, with increase in the term of assessment.

Effect of Imported Young Bulls with Higher Genetic Merit on Genetic Progress of Japanese Holstein Population

  • The effect of imported young bulls on the genetic progress was examined in the Holstein dairy cattle population in Japan. The effect of the difference of mean genetic merit between imported and domestic young bulls ("genetic difference") was recognized on the genetic progress of the domestic animals in the early stage of selection. On the other hand, the genetic progress of domestic animals were remarkably influenced by the genetic trend of imported young bulls ("genetic trend") in the later stage. Import of young bulls originated from high genetic level of young bulls originated from high genetic level population improved the genetic progress of domestic population. But, the increase of the immigration ratio of imported young bulls ("immigration ratio") did not influence linearly on the progress of the genetic merit of domestic animals. Even if "immigration ratio" was 100%, the genetic merit of domestic animals could not overcome the one of imported young bulls. In the later stage of selection, the genetic merit of domestic animals ran parallel to those of imported young bulls.

  • 본 연구는 국내 Holstein 젖소의 유생산 및 선형심사 형질의 유전적 특성을 파악하기 위해 실시하였다. 자료는 2009년 1월부터 2013년 4월까지 분만한 1산차인 Holstein 젖소 10,218두의 능력검정 자료와 유방 및 지제의 선형심사 자료를 이용하였고, 능력검정 자료 및 선형심사 자료는 각각 농협중앙회 젖소개량사업소 및 사)한국종축개량협회에서 수집한 자료이며, 사)한국종축개량협회의 33,436두에 대한 혈통정보를 분석에 활용하였다. 각 형질의 유전모수 추정은 Animal Model에 근거하여 개발 된 WOMBAT package를 이용하였으며, 추정된 유전분산 및 잔차분산을 이용하여 유전력을 계산하였다. 유량(MY), 유지방량(FY), 유단백량(PY), 유지방율(FP), 유단백율(PP) 및 체세포지수(SCS)의 유전력은 각각 0.128, 0.144, 0.100, 0.273, 0.333 및 0.090이었으며, 유방깊이(UD), 유방질(UT), 정중제인대(MS), 앞유방 붙음성(FUA), 앞유두 위치(FTP), 뒷유방 높이(RAH), 뒷유방 너비(RAW), 뒷유두 위치(RTP), 유두길이(FTL), 발굽기울기(FA), 뒤꿈치 깊이(HD), 뼈질(BQ), 옆에서 본 뒷다리(RLSV), 뒤에서 본 뒷다리(RLRV) 및 보행성(LC)에 대해 각각 0.179, 0.066, 0.104, 0.109, 0.127, 0.099, 0.059, 0.069, 0.154, 0.014, 0.010, 0.052, 0.065, 0.175 및 0.031로 추정되었다. MY와 UD, UT, FTP, RAW, FTL, FA 및 RLSV의 유전상관은 각각 0.334, 0.271, 0.445, 0.544, 0.076, -0.281 및 -0.228 이었고, PP와 MS, FTP, RTP, FTL, FA, BQ, RLSV, RLRV 및 LC의 유전상관은 각각 -0.147, -0.182, -0.262, -0.136, 0.355, 0.311, 0.135, 0.233 및 0.143으로 나타났다. 특히, MY는 RAW와 가장 높은 정의상관(0.544)을 나타낸 반면, SCS는 LC와 가장 높은 부의상관(-0.603)을 나타냈다. FP는 대부분의 유방 선형심사형질과 부의상관 관계를 나타낸 반면, FP는 지제 선형심사형질과 정의상관(0.056~0.355) 관계를 나타냈다.

Monitoring Feeding Adequacy in Dairy Cows Using Milk Urea and Milk Protein Contents under Farm Condition

  • The experiment was conducted on 264 crossbred Karan-Fries (Holstein Friesian${\times}$Tharparkar) cows, over one year to explore the possibility of using milk urea (MU) concentration and milk protein content to monitor feeding adequacy under farm condition and to investigate the effects of different animal factors and season on MU concentration. Individual noon (1200 to 1300 h) milk samples were collected once in every month and analysed for urea and protein contents. Representative feed samples were also collected on the same day of milk collection and were analysed for CP content. A significant positive association (p<0.01) between MU concentration and milk yield was observed. MU concentrations (mg/dl) were found to be significantly (p<0.01) higher and lower in first lactation (44.8${\pm}$0.7) and in early lactation stage (40.7${\pm}$0.5), respectively. Average MU values were found to be significantly (p<0.01) higher in winter (50.7${\pm}$0.3) and lower in summer (32.9${\pm}$0.6). During the investigation, of the total MU observations, 50.3% were within the range of 30 to 50 mg/dl, 21.4% were <30 mg/dl and only 7.5% were >60 mg/dl. MU concentration was found to be associated significantly (p<0.05) with CP content of forages rather than concentrate. A close positive association (p<0.01) between MU level and daily milk protein (DMP) yield was observed during the investigation. The regression equation, DMP yield (g) = -24.6+33.5 daily milk yield (kg) +0.9 MU (mg/dl) was developed to establish the reference level of DMP yield. The result indicates that the effect of parity and stage of lactation may be ignored while interpreting MU values. However, reference MU values may be standardised separately for high milk yielders as level of milk yield contributes significantly to the variation of MU. The study revealed that the MU values together with DMP yield and milk protein content could be used as a potential non-invasive pointer to monitor feeding adequacy in dairy cows under farm conditions.

국내 홀스타인젖소의 선형심사에 대한 보정계수 개발 (The Development of Adjustment Coefficients for Linear Classifications in the Korean Holstein Dairy Cattle)

  • 젖소의 선형심사형질은 젖소의 생물학적 변이를 심사자의 직관에 의하여 변이의 정도에 따라 점수를 부여한다. 심사형질에 대한 정의에 의하여 연령과 비유기에 상관없이 변이의 정도를 수치화 하기 때문에 형질에 따라서 필연적으로 연령과 비유기의 영향을 받을 수밖에 없다. 본 연구에서는 연령과 비유기가 선형심사형질에 미치는 영향을 조사하고 그를 보정하기 위한 보정계수를 추정하였다. 키 강건성, 체심, 엉덩이 너비, 뒷다리 옆모양과 앞유방 부착이 연령에 따라 증가 또는 감소하는 경향을 보이면서 크게 영향 받는 것으로 조사되었으며, 유용성, 뒷유방 높이, 뒷유방 너비와 정중제인대는 비유기에 따라 영향을 받았다. 유방깊이는 연령과 비유기 두 가지 요인 모두로부터 영향을 받았다. 추정된 보정 계수에 의한 각각의 보정계수를 적용한 결과 연령과 비유기에 영향 받는 모든 형질에서 분만년월, 심사자 등의 변이에는 작은 영향을 미치며, 보정하고자 하는 요인의 변이를 크게 줄여 보정효과가 양호한 것으로 나타났다. 유방깊이는 연령과 비유기에 대한 보정계수를 동시에 적용한 결과 두 요인 모두에서 변이를 크게 줄였다.

Genetic parameters of calving ease using sire-maternal grandsire model in Korean Holsteins

  • Objective: Calving ease (CE) is a complex reproductive trait of economic importance in dairy cattle. This study was aimed to investigate the genetic merits of CE for Holsteins in Korea. Methods: A total of 297,614 field records of CE, from 2000 to 2015, from first parity Holstein heifers were recorded initially. After necessary data pruning such as age at first calving (18 to 42 mo), gestation length, and presence of sire information, final datasets for CE consisted of 147,526 and 132,080 records for service sire calving ease (SCE) and daughter calving ease (DCE) evaluations, respectively. The CE categories were ordered and scores ranged from CE1 to CE5 (CE1, easy; CE2, slight assistance; CE3, moderate assistance; CE4, difficult calving; CE5, extreme difficulty calving). A linear transformation of CE score was obtained on each category using Snell procedure, and a scaling factor was applied to attain the spread between 0 (CE5) and 100% (CE1). A sire-maternal grandsire model analysis was performed using ASREML 3.0 software package. Results: The estimated direct heritability ($h^2$) from SCE and DCE evaluations were $0.11{\pm}0.01$ and $0.08{\pm}0.01$, respectively. Maternal $h^2$ estimates were $0.05{\pm}0.02$ and $0.04{\pm}0.01$ from SCE and DCE approaches, respectively. Estimates of genetic correlations between direct and maternal genetic components were $-0.68{\pm}0.09$ (SCE) and $-0.71{\pm}0.09$ (DCE). The average direct genetic effect increased over time, whereas average maternal effect was low and consistent. The estimated direct predicted transmitting ability (PTA) was desirable and increasing over time, but the maternal PTA was undesirable and decreasing. Conclusion: The evidence on sufficient genetic variances in this study could reflect a possible selection improvement over time regarding ease of calving. It is expected that the estimated genetic parameters could be a valuable resource to formulate sire selection and breeding plans which would be directed towards the reduction of calving difficulty in Korean Holsteins.

Odds Ratio and Probability of Conception of Holstein Friesian Dairy Cows in the Kingdom of Saudi Arabia

  • Logistic Regression Analysis was used to compute the odds ratio (OR) and probability of conception of Holstein dairy cows of AL-MARAIE company. Data consisted of 103,778 reproductive records collected from three farms in the central region in the Kingdom of Saudi Arabia. Records were classified according to lactation number, season of calving and milk level. At first lactation, OR of first service was 0.63 of other services and probability of pregnancy from first services was 0.39. Odds ratio increased to 1.72 at fourth lactation or probability of conception reached 0.63. The probability of conception increased from 0.39 for cows inseminated at first services to 0.75 at fifth service insemination. Odds ratio of cows calving in winter were higher than those calving in summer. Odds ratio of conception of low producing cows is about twice as likely to occur from first service as from other services. However, OR of conception of high producing cows was higher than one (probability=0.56) from first service and increased to 1.63 (probability=0.65) from third service. Odds ratio was in favor of the right uterus horn where probability of conception from first service was slightly greater than 50% in first and second lactations and less than 50% in favor of left horn in later lactations.

Blood and milk metabolites of Holstein dairy cattle for the development of objective indicators of a subacute ruminal acidosis

  • Objective: The purpose of this study was to perform a comparative analysis of metabolite levels in serum and milk obtained from cows fed on different concentrate to forage feed ratios. Methods: Eight lactating Holstein cows were divided into two groups: a high forage ratio diet (HF; 80% Italian ryegrass and 20% concentrate of daily intake of dry matter) group and a high concentrate diet (HC; 20% Italian ryegrass and 80% concentrate) group. Blood was collected from the jugular vein, and milk was sampled using a milking machine. Metabolite levels in serum and milk were estimated using proton nuclear magnetic resonance and subjected to qualitative and quantitative analyses performed using Chenomx 8.4. For statistical analysis, Student's t-test and multivariate analysis were performed using Metaboanalyst 4.0. Results: In the principal component analysis, a clear distinction between the two groups regarding milk metabolites while serum metabolites were shown in similar. In serum, 95 metabolites were identified, and 13 metabolites (include leucine, lactulose, glucose, betaine, etc.) showed significant differences between the two groups. In milk, 122 metabolites were identified, and 20 metabolites (include urea, carnitine, acetate, butyrate, arabinitol, etc.) showed significant differences. Conclusion: Our results show that different concentrate to forage feed ratios impact the metabolite levels in the serum and milk of lactating Holstein cows. A higher number of metabolites in milk, including those associated with milk fat synthesis and the presence of Escherichia coli in the rumen, differed between the two groups compared to that in the serum. The results of this study provide a useful insight into the metabolites associated with different concentrate to forge feed ratios in cows and may aid in the search for potential biomarkers for subacute ruminal acidosis.

Genetic parameters of milk and lactation curve traits of dairy cattle from research farms in Thailand

  • Objective: This study was aimed to estimate the genetic parameters, including genetic and phenotypic correlations, of milk yield, lactation curve traits and milk composition of Thai dairy cattle from three government research farms. Methods: The data of 25,789 test-day milk yield and milk composition records of 1,468 cattle from lactation 1 to 3 of Holstein Friesian (HF) and crossbred HF dairy cattle calved between 1990 and 2015 from three government research farms in Thailand were analysed. 305-day milk yield was estimated by the Wood model and a test interval method. The Wood model was used for estimating cumulative 305-day milk yield, peak milk yield, days to peak milk yield and persistency. Genetic parameters were estimated using linear mixed models with herd, breed group, year and season of calving as fixed effects, and animals linked to a pedigree as random effects, together with a residual error. Univariate models were used to estimate variance components, heritability, estimated breeding values (EBVs) and repeatability of each trait, while pairwise bivariate models were used to estimate covariance components and correlations between traits in the same lactation and in the same trait across lactations. Results: The heritability of 305-day milk yield, peak milk yield and protein percentage have moderate to high estimates ranging from 0.19 to 0.45 while days to peak milk yield, persistency and fat percentage have low heritability ranging from 0.08 to 0.14 in lactation 1 cows. Further, heritability of most traits considered was higher in lactation 1 compared with lactations 2 and 3. For cows in lactation 1, high genetic correlations were found between 305-day milk yield and peak milk yield (0.86±0.07) and days to peak milk yield and persistency (0.99±0.02) while estimates of genetic correlations between the remaining traits were imprecise due to the high standard errors. The genetic correlations within the traits across lactation were high. There was no consistent trend of EBVs for most traits in the first lactation over the study period. Conclusion: Both the Wood model and test interval method can be used for milk yield estimates in these herds. However, the Wood model has advantages over the test interval method as it can be fitted using fewer test-day records and the estimated model parameters can be used to derive estimates of other lactation curve parameters. Milk yield, peak milk yield and protein percentage can be improved by a selection and mating program while days to peak milk yield, persistency and fat percentage can be improved by including into a selection index.
