Single Nucleotide Polymorphism Exploring the 5'-Regions of Estrogen Receptor-${\alpha}$ Gene and Association With Reproduction Performance and Milk Yield in Hanwoo and Holstein Dairy Cattle (Estrogen Receptor-${\alpha}$ 유전자 5' 영역의 Single Nucleotide Polymorphism의 탐색과 한우와 Holstein에서 번식능력 및 산유능력과의 관계)

  • This study was conducted for SNPs in the 5'-regions of estrogen receptor-${\alpha}$ (ESR-${\alpha}$), and association with calving interval (CI), service per conception (SPC) and 305 days milk yield in Hanwoo and Holstein dairy cattle. The genetic improvement was incurred low reproduction performance. The objective of this study was to investigate connections between single nucleotide polymorphisms (SNP) of Estrogen receptor-${\alpha}$ (ESR-${\alpha}$) with reproduction performance (calving interval, service per conception, and 305 d milk yield) in Hanwoo and Holstein dairy cattle. Hanwoo and Holstein blood samples were collected from 183 and 124 dam of breeding farms and DNA was extracted. Primer design was based on NCBI GenBank (Accession No. AY340579). The PCR-RFLP method with Bgl I was used to genotype the cattle. The result showed two variants of the ESR-${\alpha}$ gene. The Bgl I cut the 492 bp amplification product into 322 bp and 170 bp fragments for allele G, while allele A remained uncut, resulting in two restriction fragments for homozygote G/G and three fragments for heterozygote A/G. We found two of different genotypes in these breeds, A/G and G/G. In Hanwoo, the A/G genotype frequency was 0.13, and G/G was 0.87. The CI of A/G was $382.18{\pm}10.03$ days, and G/G was $381.69{\pm}5.22$ days. The SPC of A/G was $1.62{\pm}0.16$, and G/G was $1.32{\pm}0.04$. While CI showed no significance difference, SPC exhibited significant difference (p<0.05). In Holstein cattle, the frequency of genotype A/G was 0.02 and G/G was 0.98. The 305 days milk yield of A/G was $7,253.00{\pm}936.00kg$ and of G/G was $8,747.51{\pm}204.88kg$, showing no significant difference.

  • The proteins of the bovine erythrocyte membrane were analyzed by polyacrylamide gel electrophoresis in sodium dodecyl sulfate, and their relations to the slow sedimentation rate of bovine erythrocytes were investigated by treating the erythrocytes with trypsin. The erythrocyte sedimentation rates of bovine erythrocytes from Holstein and Korean native cattle were very slow compared with the human one (1/7 as slow as the human one) as reported previously. However, when human and Holstein erythrocytes were treated with trypsin (0.2 and 0.5 mg/ml) for 1 hour at ${37^{\circ}C}$, their sedimentation rates were markedly accelerated while the sedimentation rate of Korean native cattle's erythrocytes were not affected. Although the general protein profiles of the bovine erythrocyte membranes were almost similar to that of human, bovine erythrocyte membranes showed one additional protein band, called band Q in this study, which migrated electrophoretically to the mid-position between band 2 and band 3 in human erythrocyte membranes. Treatment of Holstein and human erythrocytes with trypsin caused a decrease or disapperance of the band Q from the erythrocyte membrane. Although the band Q in Korean native cattle's erythroyte membrane was decreased by trypsin treatment of the erythrocytes, the magnitude of the decrement was not so pronounced as in the case of human and Holstein erythrocytes. The glycoprotein profiles of the bovine erythrocyte membranes revealed by periodic acid-Schiff stain showed a marked difference from that of human. The PAS-1 (glycophorin) and PAS-2 (sialoglycoprotein) present in human erythrocyte membrane were almost absent from the bovine erythrocyte membranes. Instead, the bovine erythrocyte membranes showed a strong PAS-positive band near the origin of the electrophorograms, which is named as PAS-B in this study. The PAS-B band was disappered completely by the trypsin treatment of Holstein erythrocytes whereas the PAS-B band in Korean native cattle's erythrocyte membrane still remained after the trypsin treatment. The trypsin treatment of Korean native cattle's erythrocytes, however, led to the appearance of small molecular weight peptides, indicating that the high molecular weight glycoproteins were degraded by trypsin as in human and Holstein ones. These results suggest that the slow sedimentation rate of bovine erythrocytes is due in part to the presence of band Q protein fraction and PAS-B glycoprotein in the bovine erythrocytes.

Genetic Evaluation of Somatic Cell Counts of Holstein Cattle in Zimbabwe

  • The objectives of the study were to examine non-genetic factors that influence somatic cell counts in dairy cattle and to estimate the genetic parameters of somatic cell counts. A total of 34, 097-test day somatic cell count records were obtained from the Zimbabwe Dairy Services Association (ZDSA). The data were from 5, 615 Holstein daughters of 390 sires and 2, 541 dams tested between May 1994 and December 1998. First lactation cows contributed 22, 147 records to the data set, while 11, 950 records were from second and later parity cows. The model for analysis included fixed effects of month of calving, year of calving, stage of lactation, calving interval and test date. Milk yield and age on test day were fitted in the model as covariates. The additive genetic effects pertaining to cows, sires and dams and the residual error were the random effects. The Average Information Restricted Maximum Likelihood algorithm was used for analysis. The heritability of somatic cell scores was low at $0.027{\pm}0.013$ for parity one cows and $0.087{\pm}0.031$ for parity two and above. Repeatability estimates were $0.22{\pm}0.01$ and $0.30{\pm}0.01$ for the two lactation groups, respectively. Genetic and phenotypic correlations between the somatic cell scores and test day milk production were small and negative. It seems that there is no genetic link between somatic cell counts and milk yield in Holstein cattle in Zimbabwe. The results also seem to indicate that somatic cell count is a trait that is mainly governed by environmental factors.

Effects of Monensin Administation on Mammary Function in Late Lactating Crossbred Holstein Cattle

  • An experiment was carried out to study the effect of monensin administration on mammary functions in crossbred Holstein cattle. Fourteen non-pregnant late lactating crossbred Holstein cattle, approximately 270 days postpartum, were selected for the experiment. They were divided into two groups of 7 animals each. Seven animals in the treated group were given sodium monensin orally in a slow-release capsule. Animals in both control and treated groups were fed the similar diet to maintain milk production and body score at 2.5. Rice straw was fed as a source of dietary fiber throughout the experimental period. After monensin administration, a significant increase in the molar percent of ruminal propionate (p<0.05) and a significant decrease in the molar percent of ruminal acetate (p<0.05) were apparent in comparison to the pretreated period. The ratio of acetate to propionate concentration decreased significantly after monensin administration (p<0.05), while it was maintained at the similar level throughout the period of experiment in the control group. Monensin did not affect the molar percent of ruminal butyrate and valerate. The concentration of milk allantoin between the control group and monensin treated group was not different. An excretion rate of allantoin in milk decreased in animals treated with monensin (p<0.05). Mammary blood flow did not show significant difference between control and monensin treated groups. The plasma glucose concentration, arteriovenous concentration difference and mammary gland uptake of glucose remained constant in both groups. Milk yield of the later stage of lactation in the control group declined during lactation advance while a tendency to increase in the milk yield was apparent after 21 days monensin administration. Milk compositions for concentration of lactose, fat and protein in both control group and monensin treated group did not change throughout the experimental periods. From these results, it can be concluded that the action of monensin could affect the ruminal fermentation pattern. Monensin could not increase milk yield in the late lactating period.

Random Regression Models Using Legendre Polynomials to Estimate Genetic Parameters for Test-day Milk Protein Yields in Iranian Holstein Dairy Cattle

  • The objective of this study was to estimate the genetic parameters of milk protein yields in Iranian Holstein dairy cattle. A total of 1,112,082 test-day milk protein yield records of 167,269 first lactation Holstein cows, calved from 1990 to 2010, were analyzed. Estimates of the variance components, heritability, and genetic correlations for milk protein yields were obtained using a random regression test-day model. Milking times, herd, age of recording, year, and month of recording were included as fixed effects in the model. Additive genetic and permanent environmental random effects for the lactation curve were taken into account by applying orthogonal Legendre polynomials of the fourth order in the model. The lowest and highest additive genetic variances were estimated at the beginning and end of lactation, respectively. Permanent environmental variance was higher at both extremes. Residual variance was lowest at the middle of the lactation and contrarily, heritability increased during this period. Maximum heritability was found during the 12th lactation stage ($0.213{\pm}0.007$). Genetic, permanent, and phenotypic correlations among test-days decreased as the interval between consecutive test-days increased. A relatively large data set was used in this study; therefore, the estimated (co)variance components for random regression coefficients could be used for national genetic evaluation of dairy cattle in Iran.

A Survey of the Internal Parasites of Holstein Cattle in Western Gyeongnam Area (경남지방의 젖소에 대한 내부기생충 조사)

  • This survey was conducted to investigate the prevalence of the internal parasites of Holstein cattle rearing in western Gyeongnam including Chinyang, Sachun, Namhae, Hadong, Sanchung, Hamyang, Geochang and Hapchun Area(Gun). A total of eight hundred and twenty five heads of Holstein cattle was randomly selected for the faecal examinations and the internal parasitisms were examined by floatation method using saturated salt solution and sedimentation method using 0.5% trio-soap solution. The relults obtained are summarized as follows: 1. The overall rate of internal parasite infections was 67.2% with infection of 554 cases and the non-infected rate was 32.8% with 271 cases from the total number of 825 samples. 2. A total fourteen species of parasites was identified from 825 faecal samples of Holstein cattle and the infection rate of each species of parasites was 3.0% in Haemonchus contortus, 2.7% in Cooperia spp., 1.8% in Bunostomum phlebotomum, 1.6% in Oesophagostomum radiatum, 1.2% in Ostertagia ostertagi, 1.0% in Trichuris bovis, 0.8% in Strongyloides papillsus, 0.4% in Nematodirus spp., 0.1% in Trichostrongylus spp., 57.3% in Paramohistomum spp., 31.2% in Fasciola hepatica, 0.6% in Eurytrema pancreaticum, 0.7% in Moniezia expansa and 5.1% in Eimeria spp. 3. The combined infection of internal parasites was 38.1% in single species, 27.0% in double and 2.1% in triple infections. 4. The EPG levels of each species of parasites were very low in all infected cases except trematodes showing no clinical symptoms.

  • The proportion of leukocyte subpopulation in the host is of a great importance in understanding their functions and disease progress. Many methods have been developed to seperate leukocytes and to measure their activities. Characterization of immune cell subpopulations in Korean native cattle was performed using a set of monoclonal antibodies specific which are specific to bovine leukocyte differentiation antigen. Peripheral blood leukocytes from fifty Korean native and ten Holstein cattle were collected and analyzed for the investigation of leukocyte subpopulation by using monoclonal antibodies and flow cytometry. The result indicated that Korean native cattle have significantly higher proportion of leukocyte subpopulations expressing MHC class II molecules and BoCD4 than Holstein cattle.

A Case of Lymphoma in a Holstein Cattle (젖소에서 발생된 림프종의 1증례)

  • A Holstein cattle with third pregnancy was found to have clinical emergency findings of anxiety, dyspnea and recumbency at just before parturition. So Cesarean section and observation of internal organs were taken. Macroscopic examination showed enlargement of th lymph nodes and spleen, and ascites. A large tumor mass with solid and white to gray-white and focal hemorrhage, 55$\times$120cm in dimension and weighing 25.5kg was found in pelivc cavity. For conventional light microscopy, tissue samples were fixed in 10% neutral formalin solution, and then paraffin embedding, sectining and H-E staining were performed. In microscopic findings, tumor tissue, lymph node and spleeen were composed of mainly lymphoid tumor cells and the number of white blood cells in peripheral blood were increased 10-fold more than in normal. We concluded this tumor was bovine lymphoma.

Antimicrobial Activities of Lactoferrin and its Hydrolysate Obtained from the Colostrum of Hanwoo and Holstein Cattle (한우와 젖소 초유로부터 분리한 Lactoferrin과 가수분해물의 항균활성)

  • The purpose of this study is to demonstrate antimicrobial activities of the lactoferrin and its peptic hydrolysates obtained from the colostrums of Hanwoo(Korean native cattle) and Holstein cattle. In the measurement of antimicrobial activity to E. coli O111 and other microorganisms, bovine lactoferrin showed a higher antimicrobial activity than that of Hanwoo cow's lactoferrin . The minimal inhibitory concentration (MIC) of lactoferrin against E. coli O111 was exhibited 1.5mg/ml(Holstein) and 2.75mg/ml (Hanwoo). The same result was also observed between bovine lactoferrin hydrolysate (0.12mg/ml) and Hanwoo cow's lactoferrin hydrolysate (0.25mg/ml). In addition of lysozyme, antimicrobial activity of lactoferrin was increased.

Studies on the Relationship between Number of Repeat and Duration of Estrous Behavior in Hanwoo and Holstein Cattle (한우와 젖소의 발정 행동 반복수와 지속 시간에 관한 연구)

  • The objective of this study was to investigate the relationship between behavioral signs and duration of estrous in Hanwoo and Holstein cattle for the prediction of optical insemination. Fifty two of 58 and 71 of 89 in Hanwoo and Holstein cows or heifers showed estrous exhibition within 72 h after 2 days following $PGF_{2}\alpha$ administration, respectively. The number of mounting and standing heat were 54.2 and 57.6, 59.4 and 53.5, 42.0 and 30.8 and 16.2 and 10.7 times in Hanwoo cows, Hanwoo heifers, Holstein cows and Holstein heifers, respectively. Duration of mounting were no significantly difference for Hanwoo cows ($21.2\pm11.3$), Hanwoo heifers ($19.9\pm11.4$), Holstein cows ($8.7\pm4.4$) and Holstein heifers ($16.9\pm8.0$). Duration of standing heat were significantly (p<0.05) shorter for Holstein cows ($5.4\pm3.4$) than for Hanwoo cows ($17.1\pm9.6$), Hanwoo heifers ($16.5\pm6.3$) and Holstein heifers ($15.0\pm7.2$). Time until mounting after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($56.3\pm11.45$) than for Hanwoo cows ($42.71\pm10.44$), Hanwoo heifers ($36.6\pm8.21$) and Holstein heifers ($40.70\pm6.15$). Time until standing heat after injection of $PGF_{2}\alpha$ were significantly (p<0.05) longer for Holstein cows ($61.6\pm8.92$) than for Hanwoo cows ($46.2\pm11.49$), Hanwoo heifers ($42.7\pm6.06$) and Holstein heifers ($44.1\pm6.72$). In the results of this study, duration of standing heat was the shortest in Holstein cows ($5.4\pm3.4$). The estimation of estrous with Holstein cows has more difficulty because to significantly shorter duration of standing heat than for Hanwoo cows, Hanwoo heifers and Holstein heifers. The standing heat can be a good predictor for time of ovulation but it is concluded that mounting behavior could be the best predictor for time of ovulation.
