Evaluation of the Neurological Safety of Epidural Milnacipran in Rats

  • Background: Milnacipran is a balanced serotonin norepinephrine reuptake inhibitor with minimal side effects and broad safety margin. It acts primarily on the descending inhibitory pain pathway in brain and spinal cord. In many animal studies, intrathecal administration of milnacipran is effective in neuropathic pain management. However, there is no study for the neurological safety of milnacipran when it is administered neuraxially. This study examined the neurotoxicity of epidural milnacipran by observing behavioral and sensory-motor changes with histopathological examinations of spinal cords in rats. Methods: Sixty rats were divided into 3 groups, with each group receiving epidural administration of either 0.3 ml (3 mg) of milnacipran (group M, n = 20), 0.3 ml of 40% alcohol (group A, n = 20), or 0.3 ml of normal saline (group S, n = 20). Results: There were no abnormal changes in the behavioral, sensory-motor, or histopathological findings in all rats of groups M and S over a 3-week observation period, whereas all rats in group A had abnormal changes. Conclusions: Based on these findings, the direct epidural administration of milnacipran in rats did not present any evidence of neurotoxicity in behavioral, sensory-motor and histopathological evaluations.

Acute Toxicity Study of the Glyoxal by Intratracheal Instillation in Male Sprague-Dawley Rats (수컷 랫드(Sprague-Dawley)에서 글리옥살(glyoxal)의 단회 기도내 투여에 따른 급성 독성시험)

  • Objectives: The present study was performed to obtain acute toxicity information on glyoxal in male rats after intratracheal instillation. Methods: In order to calculate the LD50 of glyoxal using Probit analysis with SAS, the test article was one intratracheal instillation to male Sprague-Dawley rats at dose levels of 0, 225, 451 or 902 mg/kg. During the test period, mortality, clinical signs, and body and organ weights were examined. At the end of the 14-day observation period, all animals were sacrificed and complete gross postmortem and histopathological examinations were performed. Results: Four animals of the 902 mg/kg group died within one week after the administration of glyoxal. All treatment group in a dose dependent manner, decreased body weight was found during the study period. The absolute and relative lung weight, and histopathological changes (bronchiolar-alveolar hyperplasia, chronic inflammation) of lung exhibited an increased in glyoxal treated groups in a dose dependent manner. However, there were no changes on the organ weights and histopathological changes of any other organ except lung. Conclusions: The results obtained in the present study suggest that the LD50 in male Sprague-Dawley rats after a single intratracheal instillation of glyoxal was considered to be 866.9 mg/kg and the lung was found to be the target organ for glyoxal.

Giardiasis in Black-tailed Prairie Dogs (Cynomys ludovicianus) (검은 꼬리 프레리 독(Cynomys ludovicianus)의 편모충증)

  • Five black-tailed prairie dogs showed weakness. emaciation and anemia during the lairage period in May 2005. Among them, one adult male and one female prairie dogs were found dead. The stamp smear for revealed a tremendous number of Giardia trophozoites. Histopathological examinations of the intestine showed slightly inflammatory changes and cystic enlargement of the crypts in the duodenal and jejunal mucous membranes. From these findings, this disease was diagnosed as giardiasis in black-tailed prairie dog.

  • Desmoplastic variant of ameloblastoma is a new and unusual variant of ameloblastoma with extensive stromal desmoplastic proliferation. The authors experienced a case of desmoplastic variant of ameloblastoma with moderate-defined radiolucency on the right maxillary anterior area in 62-year-old female. As a result of careful analysis of clinical, radiological, histopathological examinations, we diagnosed it as desmoplastic variant of ameloblastoma, and the following results were obtained : 1. Main clinical symptoms were nontender bony swelling with normal intact overlying mucosa on the right maxillary anterior area. 2. Radiographically, moderate-defined, multilocular radiolucency on the right maxillary anterior area were shown, and severe cortical bony thinning and expansion to labial and palatal sides were also observed. And this lesion was shown to be extended to the right nasal cavity. 3. Histopathologically, follicle-like epithelial islands with densely abundant collagenous stroma were morphologically compressed.

Unusual Intracranial Parasagittal Meningioma Extending into the Internal Jugular Vein through the Sinuses

  • Meningiomas frequently invade cerebral venus sinuses, especially parasagittal meningioma to superior sagittal sinus. However, most invasions do not reach internal jugular vein. We present a case of parasagittal meningioma extending into the internal jugular vein through the sinuses. Radiological investigation revealed that the tumor was invading the sagittal, transverse, sigmoid sinus and junction of the internal jugular vein to subclavian vein, which was filled with tumor. The histopathological examinations revealed that both the cerebral tumor and mass in the internal jugular vein contributed to the transitional meningioma. This is a rare case of a meningioma extending into the internal jugular vein through the sinuses. According to this case, the frontal parasagittal meningioma could invade directly the internal jugular vein. The significance of this association to cerebral venus sinuses and internal jugular vein are discussed.

A Typical Case of Lipomatosis Occurred in a Korean Native Cow (한우에 발생한 전형적인 지방종증의 1례)

  • A Patient (Korean native cow, 4 years old, 300 kg, female), with chief complaint of anorexia for long period was admitted to the Chungnam national University Veterinary Medical Teaching Hospital. At necropsy findings, hard masses of necrotic fat were located in the omentum, rectum, kidney, colon, heart and thoracic cavity, respectively. The yellowish changes also were observed in liver, spleen and lung, respectively. In histopathological findings, fatty accumulation was observed by microscopic observation in each organ of fatty masses. Hamatological analysis revealed the low levels of Hb, PCV and neutrophil. Blood chemical analysis indicated the high concentration of Cholestelol and Triglyceride. That patient was diagnosed as typical case of lipomatosis by clinical, blood, blood chemical and pathological examinations.

  • A 13-year-old, male Maltese was refered to Veterinary Teaching Hospital of Konkuk University with inappropriate urination and hematuria. CBC, serum chemistry profile, urinalysis and urine culture were performed. Malignant cells were found in the urine sediment. Thoracic and abdominal radiography, retrograde positive-contrast cystography, retrograde double-contrast cystography, excretory urography, ultrasonography were performed. On the retrograde positive-contrast cystography examination, irregular filling defects were present on the left side of the cranial part of the bladder. Partial cystectomy performed and then medical therapy was conducted. On histopathological examinations, the lesion was confirmed as a transitional cell carcinoma of the urinary bladder.

  • Effects on the healing process of the wound was compared between acupuncture and medicament anesthesia. Ten adult dogs were used and far medicament anesthesia thiopental sodium (Ravonal (R)) was injected. Incisions were made under anesthesia and thereafter clinical signs were observed and histopathological examinations were performed. Experimental results were summanized as following; No differences were found in the clinical signs between the two groups. Macroscopically and microscopically, the healing process was a little faster in the wounds under acupuncture anesthesia than under medicament anesthesia.

A Case of Canine Hydrocephalus with Nonsuppurative Encephalitis (비화농성 뇌염을 동반한 개 수두증 1례)

  • A Shih Tzu Puppy had clinical onset of anotexia crying and progression of neurological sings when enlargement of the cranial vault at 1 month old and died after showing clinical signs during 2 months period. Radiological and pathological examinations were performed. Radiological findings were homogeneous appearance of the calvaria with cortical thinning, loss of the normal convolutional skull markings and persistent fontanelles. Grossly enlargement of the cranial vault thinning of the bone and defective closure of the fontanelles were also observed. The entire subcortical area of the cerebral hemispheres with severe, dilatation of ventricles and cerebrospinal fluid(CSF) wits absent. There was parenchyma atrophy affecting chiefly in the white mater and the cerebral cortices, axon degeneration and necrosis and gitter cell infiltration in the whiter matter and the subependymal area. Mononuclear perivacular cuffing in the cerebrum and the pons was shown. Based on the radiological, gross and histopathological findings, this case was believed to have congenital hydrocephalus with nonsuppurative encephahitis. Possible etiology on the case is also discussed.

  • Pulmonary malakoplakia is a rare lung lesion more frequently found in immunocompromised patients than in immunocompetent individuals. In this study, we report the challenging case of a young immunocompetent patient with an irregular pulmonary nodule with peripheral cysts who, after undergoing surgery, was diagnosed with malakoplakia. Due to the rarity of the disease and the similarity of this condition to malignant neoplasms, cytopathological or histopathological examinations are necessary for the correct diagnosis. A description of pulmonary malakoplakia with peripheral cysts has not been previously published in the literature.
