  • The observation of calcium salts in dentinogenesis was performed with Von Kossa-toluidine blue method and metachromasia were observed on the portion of border that is beginning the calcification in dentin and predentin, which is positive Von Kossa reaction.

폐렴우와 Klebsiella pneumoniae 실험적 감염 랫드의 기관 및 기관지 점막층에서 Globule leukocyte의 출현양상 (Appearance of globule leukocytes in tracheal and broncheal mucosa in Korean native cattle with pneumonia and rats experimentally infected with Klebsiella pneumoniae)

  • The study was aimed at identifying the globule leukocytes (GL) of tracheal mucosa layer of Korean native cattle showing symptom of pneumonia which have died as enterotoxemia and normal Korean native cattle in Kyungpook local area. In another set of experiment, Isolated Klebsiella pneumoniae from suddenly died cattle specimens was subjected to rat for a determining globule leukocyte appearance by using histochemical and immunohistochemical method. In histochemical study, globule leukocytes generally was existed in all the case of postmortem of Korean native cattle and 3 heads of slaughtered cattle which showing symptoms of pneumonia and it showed significant increase in tracheal mucosa of rats experimentally infected with Klebsiella pneumoniae. These increased number of globule leukocytes was moderately remained on early infection stage and gradually decreased in timedependent manner after infection. The granule patterns were also determined as an acidmucopolysaccharide. In immunohistochemical study, serotonin intensity in the treacheal mucosaepithelial cells of rat experimentally infected observed a strong immunoreactivity during early infection and gradually decreased in dependent of infection stage while no IgE immunoreactivity observed. These data show that globule leukocytes were increased in a pneumonia, therefore it was considered as a valuable cell that was associated with early stage of inflammatory response.

척추동물 뇌조직의 Energy 대사에 관한 계통학적 비교연구 (A Comparative Study on the Energy Metabolism of Brains of Several Vertebtrates with Respect to Their Phyogeny)

  • The present paper deals with the comparative study on phylogenic difference in the patterns of energy metabolism of brain slices of several vertebrate species by measuring oxygen consumptionwith glucose-6-phosphate, glucose-1-phosphate, glyceraldehyde-3-phosphate or glutamate as respiratory substrate employing Warburg's manometric method, by determination of the utilization rate of glucose using glucose-1-C14 by analyzing patterns of free amino acid distribution , and by histochemical determination using glucose-1-C14 by analyzing patterns of free amino acid distribution acid distribution , and by histochemical determination of glycogen contents. 1. Glucose enhances the oxygen consumption of brain slices of animals belinging to reptile, aves and mammalia while it shows a tendency to decrease that of animals belonging to pisces and amphibia. 2. Glucose-6--phosphate increase oxygen consumption more than glucose in every species examined, while glucose-1-phosphate and glyceraldehyde-3-phosphate increase that of Rana nigromaculata only . In general m, it appears that phosphosugars are more effective as a respiratory substrate to those species which have less endogenous respiration than to those having larger endogenous respiration. 3. Similar patterns of free amino acid distribution and the relative amount are found among the species and in every species examined glutamic acid is detected in the larges amount . ${\gamma}$-Amino butyric acid, glycine, alanine and aspartic acid are found in every species. 4. Ophicephalus showed less oxygen consumption than endogenous respiration when glutamate was added to the medium. When sodium fluoride was added, the oxygen consumption was some what increased . Such phenomenon wasnot found in the frog. 5. The result of histochemcial analysis of the brain showed that glycogen was abundantly present in the fish , amphibia , and especially in the reptile and that no distinctive grains of glycogen were found in the bird and mammal . From these facts, it may be supposed that anaerobic glycolysis as energy source dominates in fish and amphibia and aerobic respiration through the oxidation of glucose dominates in bird and mamal , the reptile occupying transitional position between these two categories. The way of obtaining energy for brain activity by the oxidation of glucose supplied from the circulating blood is seemed to be first acquired by reptile and the function is completed both in aves and mammal.

Imidazole 염색에 의한 분비지질의 조직화학적 연구 (Histochemical Study of Secretory Lipids by Imidazole Staining)

  • Lipophilic glandular trichomes form secretory vesicles that accumulate in a distended noncellular secretory cavity . Imidazole-buffered osmium tetroxide solution was used to visualize unsaturated lipids in glands of Cannabis sativa. This method of staining revealed two kinds of secretory vesicles in the cavity of glands. Some smaller and rounded vesicles in the secretory cavity and secretory cells were positively stained with the imidazole, and they appeared electron-dense. The other type of vesicles which have bigger sizes more or less were not reacted, however, they appeared transparent. A high contrast of the cuticles which cover the gland was also strongly reacted with that processing. Those result suggest that the dark vesicles in the cavity may contribute to enlarge the subcuticular wall and cuticle when contents of these vesicles are dispersed into wall.

Methionine투여의 백서구강점막에 관한 조직학적연구 (HISTOCHEMICAL STUDY ON THE ORAL MUCOSA OF METHIONINE ADMINISTERED RATS)

  • The buccal mucosa was observed histochemically in male rats which were injected with methionine. The staining methods were proceed by means of periodicacid-Schiff reaction, toluidine blue stain, alloxan-Schiff reaction, Mallory's aniline blue stain, Lillie's modification of Bielschowsky method and hematoxylin-eosin stain. The results of the experiment were summarized as follows: 1) PAS reactions of basement membrane and lamina propria increased after 3 and 5 days of methionine administration. 2) Metachromasia of stratum spinosum increased after 3 days of methionine administration. 3) Alloxan-Schiff reactions of stratum granulosum and stratum spinosum increased after 7 days of methionine administration. 4) In the lamina propria, aniline blue staining of collagenous fibers increased after 7, 10 and 14 days of methionine administration.

STABLE TRANSFORMATION OF CULTURED CHICKEN CELLS

  • A plasmid vector, $RSVLTR/{\beta}G2$, containing lacZ gene under the control of the RSVLTR promoter were transfected into chicken embryo fibroblasts by three different transfection methods. Calcium phosphate, lipsome and DEAE-dextran techniques were applied for transfection of chicken cells. A histochemical assay with X-gal was used as a simple method for screening transfected cells. Plasmid $RSVLTR/{\beta}G2$ was expressed proficiently in the chicken embryo fibroblast. Calcium phosphate-DNA precipitate transfection resulted in the highest efficiency for transient expression of $RSVLTR/{\beta}G2$. Transfected cells formed colonies on the 9th day of incubation indicating stable transformation of the inserted plasmid.

  • 서울주걱흡충의 냉동절편에서 효소조직화학법을 이용하여 조직융해구(trlbocytic organ)와 맹장에 acid phosphatase, non-specific esterase 활성도가 있음을 알 수 있었다. 이 두 기관이 표피 이외에 중요한 흡수기관일 것이다. 이 흡충의 우리말 이름을, 이미 회원들에 의해 제안된 "쥐주걱흉충"은 쥐만 적절한 숙주로 생각할 수 있으므로 발견 장소가 학명에 포함된 것을 감안하여 "서울주걱흡충"이라고 고쳐 쓰고 싶다. 준말로 쓸 때는 "서울흡충"이 나을 것이다.

In vitro Cartilage Regeneration using Primary Chondrocytes Cultured within Porous Poly(lactide-co-glycolide) Scaffolds

  • Cartilage injuries are frequent nowadays. The previous surgical treatment of cartilage defect was limited. Another approach in the treatment of cartilage injuries is the use of reconstitute cartilage consisting of chondrocytes cultured in suitable biodegradable scaffolds. Current studies have demonstrated the compatibility of chondrocytes with different biomaterials and the chondrogenesis in various types of porous scaffolds. The cell ingrowth into the porous scaffolds is modulated by initial cell loading efficiency. Therefore, well-interconnected pore structure and even pore distribution of the scaffolds are essential for efficient cell seeding. According to our previous work, well-interconnected macroporous scaffolds can be prepared by gas-foaming/salt-leaching method using ammonium bicarbonate salt as porogen additives. In this work, primary chondrocytes were cultured in PLGA 65/35 scaffolds fabricated by using our method. Cells seeded in the scaffolds showed well distribution by agitated seeding method. Histochemical staining of proteoglycans present in the scaffolds was used to visualize the chondrocyte ingrowth in the scaffolds. At 3 weeks, the population of chondrocytes was increased for the most part of the scaffolds, and extra cellular matrix (ECM) secretion was increased as culture periods progressed.

전침자극이 Spontaneously Hypertensive Rat의 대뇌겉질, 뇌줄기, 소뇌 부위의 Nitric Oxide Synthase 신경세포에 미치는 영향 (Difference in NOS between 2 Hz and 100 Hz EA in cerebral cortex, brain stem and cerebellum of spontaneously hypertensive rats)

  • Background and Objetive : The aim of this study was to investigate the effect of various electroacupuncture stimulation on NADPH-diaphorase in cerebral cortex, brain stem, cerebellum of spontaneously hypertensive rats. Materials and Methods : We evaluated the changes of NADPH-d-positive neurons using a histochemical method. The staining intensity of NADPH-d-positive neurons was assessed in a quantitative fashion using a microdensitometrical method based on optical density by means of an image analyzer. Results and Conculsion : The average optical density of NADPH-d-positive neurons of 100 Hz (bipolar square wave 0.2 ms duration and 100 Hz frequency) electroacupuncture treatment group significantly increased in most cortical areas comparison between the manual acupuncture and 2 Hz (bipolar square wave 0.2 ms duration and 2 Hz frequency) electroacupuncture groups. In the brain stem, the optical density of NADPH-d-positive neuron at only superficial gray layer of the superior colliculus area was same as cerebral cortex. We conclude that the morphological evidence for NADPH-d-positive neurons may be have regional change in cerebral cortex brain stem and cerebellum according to various electroacupuncture stimulations.

  • Objective: This study examines the effects of acupoint combination on NADPH-diaphorase and neuronal nitric oxide synthase(nNOS) in the brainstem of spontaneously hypertensive rats. Methods: The changes of NADPH-d-positive neurons using a histochemical method and the changes of nNOS-positive neurons using an immunohistochemical method were evaluated. The optical densities of NADPH-d-positive neurons and nNOS-positive neurons of the Choksamni(ST36) Umnungchon(SP9) groups were significantly increased in all brainstem areas as compared to the Choksamni and Choksamni Kokchi(LI11) groups and decreased, with the exeption of the nNOS-positive neurons in the superficial gray of superior colliculus, as compared to the normal group. Results: Our results demonstrated that electroacupuncture changes the activity in the NO system in the brainstem of SHR and the acupoint combination is one of the important parameters for this effect.
